Stromata of the white root rot fungus, Rosellinia necatrix, were produced on diseased roots although they were reported to develop rarely in nature. Forty-two (42) out of 47 samples produced synnemata while 23 developed stromata. Forty-seven (47) isolates obtained from diseased root samples were divided into 24 mycelium compatibility groups (MCGs). Sixteen (16) out of 24 MCGs produced stromata. Single ascospore isolates from 10 stroma samples produced dsRNA-containing isolates from diseased tissue beneath stromata. The frequency of synnema production on axenic culture varied among isolates with different origin. The dsRNA was not transmitted vertically to the ascospore offspring despite the infection of various dsRNA in the parental isolates. The dsRNA was absent in 35 ascospore isolates in two stroma samples that originated from the isolates, in which dsRNA was not eliminated by hyphal tip isolation. Consequently, sexual reproduction in the white root rot fungus was suggested to produce propagules as a new infection source and to have the function to eliminate infectious factors such as mycoviruses.
Gemcitabine is an anti-cancer drug with clinically uses in the treatment of various neoplasms, including breast, ovarian, non-small cell lung, pancreaticand cervical cancers, T-cell malignancies, germ cell tumours, and hepatocellular carcinomas. However, it has also been reported to have many adverse effects. Naturally occurring anti-mutagenic effects, especially those of plant origin, have recently become a subject of intensive research. The present study was therefore designed to investigate the anti-mutagenic effects of Salvia merjamie (Family: Lamiaceae) plant extracts against the mutagenic effects of gemcitabine. The anti-mutagenic properties of Salvia merjamie were tested in Inbred SWR/J male and female mice bone marrow cells. The mice were treated in four groups; a control group treated with 30 mg/kg body weight gemcitabine and three treatment groups, each with 30 mg/kg body weight gemcitabine together with, respectively, 50, 100 and 150 mg/kg body weight Salvia merjamie extract. Chromosomal aberration and mitotic index assays were performed with the results demonstrating that Salvia merjamie extract protects bone marrow cells in mice against gemcitabine induced mutagenicity. This information can be used for the development of a potential therapeutic anti-mutagenic agents.
We determined the complete genome sequence of a Vietnamese isolate of Southern rice black-streaked dwarf virus (SRBSDV). Whole genome comparisons and phylogenetic analysis showed that the genome of the Vietnamese isolate shared high nucleotide sequence identities of over 97.5% with those of the reported Chinese isolates, confirming a common origin of them. Moreover, the greatest divergence between different SRBSDV isolates was found in the segments S1, S3, S4 and S6, which differs from the sequence alignment results between SRBSDV and Rice black streaked dwarf virus (RBSDV), implying that SRBSDV evolved in a unique way independent of RBSDV. This is the first report of a complete nucleotide sequence of SRBSDV from Vietnam and our data provides new clues for further understanding of molecular variation and epidemiology of SRBSDV in Southeast Asia.
Beet curly top virus (BCTV) mutant has been constructed in vitro that contain G-to-T transversions at nucleotide 2727 within overlapping open reading frames (ORFs) L1 and L4. The mutations introduce termination codon in ORF L4 without affecting the amino acid encoded by ORF L1. When agroinoculated into Arabidopsis thaliana the mutant caused mild stunting and stem curling, but not the callus induction and hyperlasia on infected tissues of Sei-O ecotype. However, this mutant was not infectious on Col-O. Levels of single stranded DNA forms were similar in mutant and wild type BCTV infections. The DNA quantitation data showed that the DNA of BCTV-L4 mutant virus was accumulated in shoot tips, infection origin and roots with similar levels to those of wild type virus infected. Three tissues of asymptomatic ecotype Col-O also had as much as virus DNA from wild type virus infections. In both ecotypes infected with BCTV-Logan and BCTV-L4 mutant, root tissues contained more virus DNA than any other tissues by the Southern hybridization data. The results suggest that ORF L4 encodes a functional protein that is a major determinant of pathogenesis that might affect the hyperplastic response of the host to BCTV infection.
Variability in virulence of Calonectria ilicicola isolates form different hosts and geographic origin provides important information for breeding cultivars resistant to red crown rot. A wide range in virulence for 25 isolates of C. ilicicola from soybean and peanut was observed on six soybean cultivars. Soybean isolates were pathogenic on soybean although some were less virulent. Virulence of isolates was not affected greatly by cultivar and relatively consistent regardless of virulence level. Soybean isolates were more virulent on soybean than were peanut isolates. When virulence of two soybean and tow peanut isolates was compared between laboratory and greenhouse tests, it was stable across a range of cultivars. Mycelial growth of isolates from either soybean or peanut was reduced significantly on potato dextrose chlorate showed significant reduction of fungal growth than isolates from peanut did although their growth on potato dextrose agar was not significantly different. Evidence for physiologic specialization was not recognized in this system. However, the findings that soybean isolates of C. ilicicola were more virulent on soybean and reduction of fungal growth on potato dextrose chlorate than were peanut isolates suggest that host specialization may exist in this fugus.
Phytophthora infestans populations collected from various geographical locations of Korea in 1991 and 1993 were analyzed for mating types and responses to metalaxyl. Both A1 and A2 mating type isolates were detected in 1991. The majority of the isolates were A2 mating type, but no A1 mating type was detected in 1993. About 40% of the isolates collected in 1991 were resistant to metalaxyl, and the distribution of metalaxyl-resistant isolates of P. infestans was strongly associated with their geographic origins in Korea. Metalaxyl-resistant isolates with EC50 values > 50$\mu\textrm{g}$/ml were collected from the northern provinces of Kangwon, Kyungbuk, and Chonbuk, but not from the southern provinces of Kyungnam, Chonnam, and Jeju in 1991. The drastic increase in the degree of quantitative resistance to metalaxyl was detected among the isolates from the southern provinces during 1991~1993. More than 50% of the isolates collected from the southern provinces of Kyungnam and Chonnam in 1993 had EC50 values >50$\mu\textrm{g}$/ml. The province of Kangwon had isolates with the greatest resistance to metalaxyl. this alpine areas might be the origin of metalaxyl-resistant isolates of P. infestans in Korea. The A2 genotype with metalaxyl resistance appears to be displacing the A1 genotype which is presently the predominant genotype in Korea.
Objectives : The purpose of this study was to characterize the effect of the fraction of Taraxacum officinale and T.coreanum on the learning and memory impairments induced by scopolamine. Methods : The cognition-enhancing effect of Taraxacum officinale and T.coreanum was investigated using a passive avoidance test, the Morris water maze test and Y-maze test in mice. Drug-induced amnesia was induced by treating animals with scopolamine (1 mg/kg, i.p.). Results : The results showed that the Aug harvested T.offiicinale extract-treated group (200 mg/kg, p.o.) and the tacrine-treated group (10 mg/kg, p.o.) significantly ameliorated scopolamine-induced amnesia based on the Passive avoidance Y-maze test and Watermaze test. And these results are same manner in DPPH radical scavenger effect and Acetylcholineseterase inhibition effect. Conclusions : These results suggest that Taraxacum officinale extract maybe a useful cognitive impairment treatment, and its beneficial effects are depending on the collecting time and origin plants. As a result, Taraxacum officinale harvested in August improve memory most.
The calluses formed on the surface of a quarter-girdled Robinia pseudoacacia stems have been shown to originate from immature xylem cells and preexisting cambial cells. The cellus is not only formed by periclinal and anticlinal divisions of radial cells, but also axial cells. In tangential view, the callus at initial stage showed heterogeneous structure composed of long and short cells and then homogeneous one with short cells. Some cells of homogeneous structure in middle region of callus at early stage is later elongated and others mainly divided in trasverse plane. In the result the homogeneous structure becomes into a heterogeneous one. Subsequently, the long cells in heterogeneous structures elongated further and became fusifrom initials, and the short cells divided transversely became ray initials. The appearence of homogeneous and heterogeneous structure in the callus on debarked stem without organ elongation is almost similar to that of the structure in the procambium of young stem which is elongating extensively. Eventually, the ontogeny of vascular cambium in wound callus resembles that of a young stem grown normally, although the debarked stem does not grow in length but in girth and the young stem elongates activity. These findings mean that the active intrusive growth of short procambial cells occurs during the differentiation of fusiform cambial cells.
Glycinin is the major storage protein in soybean. It has been known that a molecule of glycinin is composed of 6 subunits, each of which consists of two different kinds of polypeptides, acidic (A) and basic (B) one (NW 39K and 19K, respectively). To study the molecular origin and the relationship of glycinin subunit polypeptides, antibodies against A-and B-polypeptide were obtained by immunizing rabbits with either of the antigens purified by gel filtration and preparative electrophoresis. Each antibody was not only specific for its own antigen polypeptide in soybeans but also recoginzed the precursor which was synthesized in vivo and in vitro. The polyadenylated mRNAs were isolated from immature seeds and leaves and were translated in vitro using wheat germ extract. One of the seed-specific translation products. MW 60K, was identified to be the precursor of glycinin subunit by immunoprecipitation with antibodies against glycinin A- and B-polypeptide. Mature A- and B-polypeptides were not detected in the translte in vitro. These results suggest that the precursor polypeptide is synthesized from the mRNA and is cleaved to yield A- and B-polypeptides which from a glycinin subunit in the cell. Glycinin genes were expressed with the maturation of soybean seeds in a tissue-specific and developmental stage-specific manner.
Radix Angelicae Gigantis is sweet and pungent in flavor, warm in property. Its effects are tonifying the blood, promoting blood circulation, relieving pain and moistening the bowels. Its indications are blood deficiency syndrome characterized by sallow complexion, dizziness, irregular menstruation, amenorrhea, pains due to blood stasis, and rheumatic arthralgia. Using genes of A. gigas, A. acutiloba, and A. sinensis, the origin of which is identified, as criteria, we analysed many kinds of Angelica with RAPD and RFLP on ITS region, in order to compare and discriminate genes extracted from crude drugs ‘Dang-gui’, that are produced in Korea on the one hand and imported on the other hand. We reached the following conclusion. 1. We could extract DNA from both original plant and dried plant. 2. Especially Uniprimer #1, Uniprimer #2, Uniprimer #4 and Uniprimer #9 were useful. 3. Among the restriction enzymes Sma I, Msp I, Hae III, and Hinf I, used in this experiment, four restriction enzymes except Hinf I could be used properly in discriminating all samples used as A. gigas. We think that this result can be used as a method of discriminating crude drug of Angelica L. related drugs, and used in controlling quality and circulation.
