도시림 복원 및 관리 기술의 개발에 관한 연구 - 원식생 복원과 생물다양성 증진을 중심으로 - (A Study on the Development of Techniques for Urban Forest Restoration and Management - Focus on the Restoration of Origin Vegetation and Improvement of Biodiversity -)

  • This study aims at restoring urban forest destructed and eliminated by industrialization, urbanization, and city development and presenting a direction to manage remaining urban forest ecologically. To this end, an experiment zone where Populus tomentiglandulosa T. Lee were selective cutted and a control zone where Populus tomentiglandulosa T. Lee were kept intact were created in Chongdam Park located in Kangnam-ku, Seoul. Then, the structural changes of herbaceous plant species, the growth of targeted woody plants, and the increase of the number of insect and bird species were examined. The conclusions reached in this study are as follows. First, for the sake of ecological restoration and management of urban forest, it is good to selective cutting. Although timing, frequency, and methods may vary depending on the features and types of urban forest, the study revealed that selective cutting contributes to the restoration speed of origin vegetation and the enhancement of biodiversity including plants and insects. Second, as for the correlations of selective cutting and the appearance of plant species, the growth of origin vegetation, and insect distribution, the study showed that the impact of meteorological environment such as brightness is much greater than that of soil environment. Third, in order to manage urban forest, tramping pressure needs to be controlled efficiently. The efficient control of tramping pressure would contribute in the appearance of herbaceous plants. It would also be beneficial in promoting biodiversity of birds by removing the impact of people using routes. Fourth, in order to enhance the overall biodiversity of urban forest, diverse environment needs to be provided. In particular, it is necessary to supply water that is insufficient in urban forest. Providing habitats such as forest wetland performs an important function to amphibians and birds that require water as well as the appearance of aquatic plants and insects. Therefore, ways to introduce water efficiently should be initiated.

  • 상업적으로 유통되는 젤라틴 캡슐 제품은 소비자의 건강(예: 광우병) 및 종교적 신념에 대한 우려를 야기시킬 수 있다. 젤라틴은 대부분 소, 돼지 등에서 유래한 원료물질을 가공한 것으로서, 가공 후 그 원료 물질을 분석하는 것은 대단히 어렵다. 따라서 정부 규제기관의 표시사항 준수여부 모니터링 연구가 주기적으로 필요하다. 본 연구에서는 인터넷에 유통되는 건강기능식품(n = 181)을 대상으로 젤라틴 캡슐의 원료 물질을 종 특이 PCR 방법으로 분석했다. 55개 제품의 경우 표시사항에 젤라틴 캡슐원료 물질에 대한 정보를 명시하였으나(예: bovine-, fish-and plant-derived gelatin), 126개 제품의 경우 사용 원료에 대한 정보 없이 "gelatin"으로 표시하였다. 이 126개 제품의 젤라틴 캡슐 분석 결과 51개 제품은 소 유래의 젤라틴을, 31개 제품은 돼지 유래의 젤라틴을, 그리고 44개 제품은 소와 돼지의 원료를 혼합하여 제작한 블렌딩 젤라틴을 사용한 것으로 밝혀졌다. 따라서 소비자의 알 권리, 종교적 신념 및 건강을 보호하기 위해 젤라틴 캡슐에 사용된 원료 물질을 표시사항에 제공하는 것은 매우 중요하다.

Clonal plant as experimental organisms - DNA mutation rate evaluation in the radiation contaminated area of Fukushima Daiichi NPP accident

  • The Fukushima Daiichi Nuclear Power Plant accident in March 2011 caused severe radioactive contamination in the surrounding environment. Since the accident, much attention has been paid to the biological and genetic consequences of organism inhabiting the contaminated area. The effect of radiation exposure on genetic mutation rates is little known, especially for low doses and in situ conditions. Evaluating DNA mutation by low levels of radiation dose is difficult due to the rare mutation event and lack of sequence information before the accident. In this study, correlations with air dose levels and somatic DNA mutation rates were evaluated using Next Generation Sequencer for the clonal plant, Phyllostachys edulis. This bamboo is known to spread an identical clone throughout Japan, and it has the advantage that we can compare genetic mutation rate among identical clone growing different air dose levels. We collected 94 samples of P. edulis from 14 sites with air dose rates from $0.04{\sim}7.80{\mu}Gy/h$. Their clonal identity was confirmed by analysis using 24 microsatellite markers, and then, sequences among samples were compared by MIG sequence. The sequence data were obtained from 2,718 loci. About ~200,000 bp sequence (80 bp X 2,718 loci) were obtained for each sample, and this corresponds to about 0.01% of the genome sequence of P. edulis. In these sequences, 442 loci showed polymorphism patterns including recent origin mutation, old mutation, and sequence errors. The number of mutations per sample ranged from 0 to 13, and did not correlate with air dose levels. This result indicated that DNA mutations have not accumulated in P. edulis living in the air doses levels less than $10{\mu}Gy/h$. Our study also suggests that mutation rates can be assessed by selecting an appropriate experimental approach and analyzing with next generation sequencer.

Himalayan Medicinal Resources: Present and Future. A Case Study: Andidiabetic Activity of Shilajit

  • A major portion of drugs used in Ayurvedic system of medicine which has been practiced since the early human civilization in Indian subcontinent were of plant origin. It should be noted that 70% of the population in this region depends on Ayurveda for their medical treatment and 60% of the drug resources are obtained from the Himalayan region. Therefore, Nepal becomes a potential source of plant drug resource since it occupies a major portion of the Himalaya. In the present paper, in general a current status of medicinal plant resources of Himalayan region especially Nepal will be discussed. In addition to this, a typical example of antidiabetic activity of Shilajit will be taken for the discussion. Shilajit is one of the crucial elements in several formulations including those of Rasayana, a therapy in Ayurveda, which has been practiced in the prevention of ageing and mental disorder. Although, Shilajit is widely used for the treatment of diabetes, no satisfactory scientific reports are available up to now. The crude Shilajit in the market is a dark brown or black rock-like substance collected from the Himalayan region with a strong smell of cow's stale urine. In our studies, Shilajit (collected in the central Himalayan region) prevented the diabetes in nonobese diabetic (NOD) mice model. Shilajit also prevented the diabetes in the rats against the action of multiple low-dose (10 mg/kg, i.v., 5 times) of streptozotocin. On the other hand, Shilajit did not show antioxidative activity. The preventive action of Shilajit on diabetes is mainly focused on the Thl and Th2 cell activities, since Th2 cells activity was found to be significantly upregulated. Shilajit, however, showed a mild action in controlling the blood sugar level in young, old, and mild diabetic rats, but not in the severe diabetic rats. It also stimulated the nitric oxide production in macrophages. Based on these evidences, the antidiabetic activities of Shilajit appear to be immunomodulative probably by protecting or strengthening insulin-producing b-cells in the pancreas. Further systematic research on constituents of Shilajit and its quality evaluation is necessary to enable the use of natural medicines in the treatment of diabetes.

Occurrence and Evolutionary Analysis of Coat Protein Gene Sequences of Iranian Isolates of Sugarcane mosaic virus

  • Sugarcane mosaic virus (SCMV) is one of the most damaging viruses infecting sugarcane, maize and some other graminaceous species around the world. To investigate the genetic diversity of SCMV in Iran, the coat protein (CP) gene sequences of 23 SCMV isolates from different hosts were determined. The nucleotide sequence identity among Iranian isolates was more than 96%. They shared nucleotide identities of 75.5-99.9% with those of other SCMV isolates available in GenBank, the highest with the Egyptian isolate EGY7-1 (97.5-99.9%). The results of phylogenetic analysis suggested five divergent evolutionary lineages that did not completely reflect the geographical origin or host plant of the isolates. Population genetic analysis revealed greater between-group than within-group evolutionary divergence values, further supporting the results of the phylogenetic analysis. Our results indicated that natural selection might have contributed to the evolution of isolates belonging to the five identified SCMV groups, with infrequent genetic exchanges occurring between them. Phylogenetic analyses and the estimation of genetic distance indicated that Iranian isolates have low genetic diversity. No recombination was found in the CP cistron of Iranian isolates and the CP gene was under negative selection. These findings provide a comprehensive analysis of the population structure and driving forces for the evolution of SCMV with implications for global exchange of sugarcane germplasm. Gene flow, selection and somehow homologous recombination were found to be the important evolutionary factors shaping the genetic structure of SCMV populations.

Prevalence of Tobacco mosaic virus in Iran and Evolutionary Analyses of the Coat Protein Gene

  • The incidence and distribution of Tobacco mosaic virus (TMV) and related tobamoviruses was determined using an enzyme-linked immunosorbent assay on 1,926 symptomatic horticultural crops and 107 asymptomatic weed samples collected from 78 highly infected fields in the major horticultural crop-producing areas in 17 provinces throughout Iran. The results were confirmed by host range studies and reverse transcription-polymerase chain reaction. The overall incidence of infection by these viruses in symptomatic plants was 11.3%. The coat protein (CP) gene sequences of a number of isolates were determined and disclosed to be a high identity (up to 100%) among the Iranian isolates. Phylogenetic analysis of all known TMV CP genes showed three clades on the basis of nucleotide sequences with all Iranian isolates distinctly clustered in clade II. Analysis using the complete CP amino acid sequence showed one clade with two subgroups, IA and IB, with Iranian isolates in both subgroups. The nucleotide diversity within each subgroup was very low, but higher between the two clades. No correlation was found between genetic distance and geographical origin or host species of isolation. Statistical analyses suggested a negative selection and demonstrated the occurrence of gene flow from the isolates in other clades to the Iranian population.

대추나무 미친병에 관한 연구 (I) -병식물의 내외형태학적 특징 및 그 명명에 대해서-

  • Since the peculiar virus disease of chinese date tree (Zizyphus jujuba Mill. var. inermis Rehd.) has been noted in South Korea around 1950, 70% to 80% of the economically important trees have been either completely destroyed or infected with the virus, severe damage has been noted, particularly, across the area ranged from middle east to the middle part of Korea, including Seoul area. Yoon-Koock-Byung in 1958 first reported the disease and descirbed it might be caused by a kinds of yellows. But he did not conform in his paper that the disease is pecisely caused by yellows virus. The authors, hereby intend to identify the true cause of the desease of the chinese data tree by studying the external symptoms of the disease and the internal morphological characteristics of the diseaset plant which shows various abnormalities in contrast to the healthy checks. In view of fact that leaves of the infected plants become yellowish in color similar to the peach yellows, aster yellows, it is likely to be identifiable as the common yellows. Furthermore, the abnormal characteristics observed by the authors are as follow: The floral organs such as petals, sepals, stamens, and pistil turn into vegetative leaves, the leaves on heavily infected plant appear as small sized one and also showing as a common witch's broom like symptom. There are also an occuring of numerous advantitious shoots developed from both of stems and roots. The amount of photosynthetic starch grains increases in parenchymatous cells, necrosis takes place in mesophyll, Particularly, Palisade Parenchyma in the leaves of infected plants are distinguished in contrast to the healthy checks. From the symptoms and the present experimetns described above, the authors are believed that the disease of chinese data tree is not caused by the yellows. It appears the disease is rather similar to the symptoms of sandal spike virus which was noted in India early in this centry. But the host plant of standal disease, Santalum albun L. and the insect vector, Jassus indicus Wal., have never been reported in Korean flora and the founa. The termperature and the otehr environmental factors is quite different Korea and India. Thus the authors believe that the peculiar disease must be an endemic new virus origin in Korea and must be called as "shoot cluster disease of chinese date tree."

DNA 바코드 분석을 통한 소계(小薊) 및 대계(大薊) 기원식물 감별과 종간 유연관계 분석 (Molecular Authentication and Phylogenetic Analysis of Plant Species for Breeae and Cirsii Herba based on DNA barcodes)

  • Objectives : The origin of Breeae Herba (So-gye) and Cirsii Herba (Dae-gye) is differently prescribed in Korean and Chinese modern pharmacopoeia. Since the similar morphological characteristics and chaotic plant names, moreover, the aerial part of Carduus crispus have been used as the Cirsii Herba. To develop a reliable method for correct identification of these herbal medicines and to evaluate the genetic relationship of these closely related plant species, we analyzed sequences of DNA barcode regions. Methods : Thirty-one samples of 6 medicinal plants (B. segeta, B. setosa, C. japonicum var. maackii, C. setidens, C. chanroenicum, and C. crispus) were collected from different habitate and nucleotide sequences of DNA barcode regions (rDNA-ITS, matK, and rbcL) were analyzed after amplification using appropriate primers reported in previous studies. The nucleotides of species-specific authentic marker and phylogenetic relations were estimated based on the entire sequences of DNA barcodes by the analysis of ClastalW and UPGMA, respectively. Results : In comparative analysis of DNA barcode sequences, we obtained specific nucleotides to discriminate the medicinal plant of Breeae/Cirsii Herba in species level and evaluated the phylogenetic relationship of these species. Futhermore, we identified distinct marker nucleotides enough to authenticate respective species. These sequence differences at corresponding positions were avaliable genetic markers to determine the botanical origins of Breeae Herbal as well as Cirsii Herba. Conclusions : These marker nucleotides would be useful to identify the official herbal medicines by providing of definitive information that can identify each plant species and distinguish from unauthentic adulterants and substitutes.

한반도 동해안 석호의 관속식물상 (Floristic study of lagoon areas on the eastern coast in Korean peninsula)

  • 동해안 석호는 해수와 담수가 섞인 독특한 형태의 기수호이며, 수변식생이 발달되어 생물다양성이 풍부한 지역이다. 본 연구는 국내에 분포하는 17개소의 석호에 대한 식물상과 식물 종조성 등의 조사를 통하여 석호 식생에 대한 안정적인 보전계획 수립을 위한 기초자료를 제시하고자 한다. 17개소의 석호에 대한 식물상 조사결과, 관속식물은 118과 378속 655종 13아종 46변종 6교잡종, 총 720분류군이 확인되었다. 이중에서 한반도 고유종이 11분류군, 멸종우려 식물이 18분류군, 식물구계학적 특정식물인 IV-V등급 식물이 23분류 군이었다. 남방계식물은 4분류군, 북방계식물은 8분류군으로 조사되었고, 특히 남한 내 희귀식물인 갯활량나물(Thermopsis lupinoides), 기름당귀(Ligusticum hultenii), 벼룩아재비(Mitrasacme alsinoides), 통발(Utricularia australis), 검정납작골풀(Juncus fauriei), 새방울사초(Carex vesicaria), 천도미꾸리광이(Puccinellia kurilensis) 등이 동해안 석호에서 생육하고 있었다. 외래식물은 96분류군이 발견되었다. 한반도 석호는 식물다양성이 풍부하고, 희귀식물 및 식물지리학적 중요한 식물이 관찰된다. 석호의 효과적인 관리와 생물다양성 보전을 위해 체계적인 사항들이 논의되었다.
