Serological and virological investigation of pestiviruses in Korean black goats

  • Blood samples were collected from 672 goats in 60 farms from five provinces of Korea between November 2009 and August 2011. The prevalence of antibodies to pestiviruses was investigated. The examination for antibodies was performed using an enzyme-linked immunosorbent assay (ELISA) detecting antibodies to the bovine viral diarrhea virus (BVDV) and border disease virus (BDV). All blood samples were screened using reverse transcription-polymerase chain reaction (RT-PCR) with primer pairs specific to common pestivirus genome regions. The observed individual seroprevalence was 1.49% and herd seroprevalence was 11.67%. Also, the specific genomes to pestiviruses were detected in 3 out of the 915 clinical samples (0.45%). Based on the nucleotide sequence data, detected pestiviruses were belonged to two BVDV type-1 and one BVDV type-2. The pestivirus infection has been occurred among Korean black goats. However, our results indicate that the prevalence of pestiviruses in black goats was not significantly higher on farms with cattle.

김포 매립지와 그 근린 지역의 식물상 분석 (Floral Analysis in the Kimpo Landfills and Its Peripheral Region)

  • Floral analysis about vegetation of Kimpo landfills and its periphery region was carried out. The study area was defined to a square $(81km^2)$ of which center was located at 250m in front of Andongpo, Komdan-myon, Kimpo-gun in the northwest part of the Kyunggi Province. This study was accomplished by analyzing five qualitative traits such as ecological strategy, reproductive strategy, distribution type, native/foreign division and life-form throughout actual investigation of the flora. The flora was composed of 536 taxa which comprise 105 families, 343 genera, 458 species, 1 subaspecies, 70 varieties and 7 forms (including 92 species of horticultural plant). Lythrum salicaria community, Spiraea salicifolia community, ottelia alismoides community and woods of Alnus japonica were recorded ? 새 retrictive distribution according to habitat characteristics. Owing to severe anthropogenic interferences such as construction of the Kimpo landfills in the study areas, a high proportion of ruderal plants and neophytes in the floral composition was recognized. The proportion of individual distribution type and therophyte was very high throughout whole study areas, and plant communities in the reclaimed areas were characterized by high proportion of phalanx plant species. Analysis of the correlations between meshes with ecological traits of plant species showed that Kimpo landfills and its periphery region was divided into two vegetation types, coastal type and inland type, as a result of possible saline effects.

Towards Improved Performance on Plant Disease Recognition with Symptoms Specific Annotation

  • Object detection models have become the current tool of choice for plant disease detection in precision agriculture. Most existing research improves the performance by ameliorating networks and optimizing the loss function. However, the data-centric part of a whole project also needs more investigation. In this paper, we proposed a systematic strategy with three different annotation methods for plant disease detection: local, semi-global, and global label. Experimental results on our paprika disease dataset show that a single class annotation with semi-global boxes may improve accuracy. In addition, we also studied the noise factor during the labeling process. An ablation study shows that annotation noise within 10% is acceptable for keeping good performance. Overall, this data-centric numerical analysis helps us to understand the significance of annotation methods, which provides practitioners a way to obtain higher performance and reduce annotation costs on plant disease detection tasks. Our work encourages researchers to pay more attention to label quality and the essential issues of labeling methods.

Induction of in vitro root tubers in Holostemma annulare (Roxb.) K. Schum. for the production of bioactive metabolites

  • Holostemma annulare (Family Asclepiadaceae) is an invaluable vulnerable medicinal plant; the root tubers are used in Ayurveda medicine and by folk healers to treat various ailments. In this study, Schenk and Hildebrandt medium fortified with the cytokinins 6-benzyl adenine, kinetin, and auxins, including indole 3-butyric acid, indole 3-acetic acid, α-naphthaleneacetic acid, and 2,4-dichlorophenoxyacetic acid, were checked for their efficiency on root tuber induction from different explants. Adventitious root tubers were more successfully induced from in vitro leaf segments and shoots when cultured in Schenk and Hildebrandt medium supplemented with 0.5 mg/l of α-naphthaleneacetic acid. In addition, preliminary phytochemical analysis of in vitro root tubers and identification of different secondary metabolites were conducted. Thin layer chromatography and high performance thin layer chromatography analysis of the crude methanolic extracts of the in vitro root tuber identified the presence of lupeol, a bioactive triterpene. Adventitious root tuber induction offers a novel method for the in vitro production of bioactive metabolites that can be scaled up by bioreactors, thus ensuring the conservation and sustainable utilization of H. annulare. The study warrants further scale-up production and pharmacological investigation that can be extended for pharmaceutical needs.

Inhibition Effects of Persicaria amphibia (L.) Delarbre on Oxidative DNA Damage via ATM/Chk2/p53 pathway

  • Persicaria amphibia as an England native plant, is a rhizomatous perennial, one of the rather amphibious plants. Its aquatic form contains water-soluble sugars, starch, and protein. P. amphibia have up to 18% tannins in stems and rhizomes. Previous studies have confirmed the anti-inflammatory activity of live bacteria roots, but no studies on bioactivity are known. DNA damage responses (DDRs) pathways are considered a crucial factor affecting the alleviation of cellular damage. The ataxia-telangiectasia mutated and Rad3 related (ATM) and checkpoint kinase 2 (Chk2) pathways are the main pathways of DNA damage response. Also, p53 is a key integrator of cellular response to oxidative DNA damage, contributing repair, or leading transcription including apoptosis. In the present study, we conducted an investigation into the inhibitory effects of P. amphibia on oxidative DNA damage for confirming potential to complementary medicine and therapies. In conclusion, P. amphibia can provide protective effects against double-stranded DNA break (DSB) caused by oxidative DNA damage.

Feasible Management of Southern Corn Leaf Blight via Induction of Systemic Resistance by Bacillus cereus C1L in Combination with Reduced Use of Dithiocarbamate Fungicides

  • Dithiocarbamate fungicides such as maneb and mancozeb are widely used nonsystemic protectant fungicides to control various plant fungal diseases. Dithiocarbamate fungicides should be frequently applied to achieve optimal efficacy of disease control and avoid either decline in effectiveness or wash-off from leaf surface. Dithiocarbamates are of low resistance risk but have the potential to cause human neurological diseases. The objective of this study was to develop a strategy to effectively control plant disease with reduced use of dithiocarbamtes. Southern corn leaf blight was the model pathosystem for the investigation. When corn plants were drench-treated with Bacillus cereus C1L, a rhizobacterium able to induce systemic resistance in corn plants against southern leaf blight, frequency of spraying dithiocarbamate fungicides could be decreased. The treatment of B. cereus C1L was able to protect maize from southern leaf blight while residues of dithiocarbamates on leaf surface were too low to provide sufficient protection. On the other hand, frequent sprays of mancozeb slightly but significantly reduced growth of corn plants under natural conditions. In contrast, application of B. cereus C1L can significantly promote growth of corn plants whether sprayed with mancozeb or not. Our results provide the information that plant disease can be well controlled by rhizobacteria-mediated induced systemic resistance in combination with reduced but appropriate application of dithiocarbamate fungicides just before a heavy infection period. An appropriate use of rhizobacteria can enhance plant growth and help plants overcome negative effects caused by dithiocarbamates.

홍수시 4대강에서 유입되는 부유폐기물 성상 조사 (Investigation of Floating Debris Characteristics Drained from 4 Big River on a Flooding)

  • 본 조사는 장마나 홍수로 인하여 강에서 해양으로 유입되는 부유폐기물의 성상을 파악하여, 해양폐기물 저감대책을 마련하기 위한 자료로 활용하기 위해 한국의 4대강인 한강, 낙동강, 금강, 영산강을 대상으로 2002년 7원과 8원 두 달간 수행하였다. 부유폐기물의 유입은 강우의 정토에 따라 차이가 있으나, 일일 강수량 150mm 이하의 일시적인 호우경보 시에도 부유폐기물의 유임은 미미하였으며, 지속적이며 다량의 강우량이 발생하거나 국지성 호우에 의하여 홍수가 발생될 때 대부분의 부유폐기물이 유입됨을 알 수 있었다 따라서 강에서 유입되는 부유폐기물은 지속적으로 해양에 유입되는 것이 아니라, 일시에 특히 홍수시 다량으로 며칠에 걸쳐서 유입된다. 또한 발생된 부유폐기물 모두가 해양으로 유입되지 않고 폐기물의 상당량이 강변에 집적되는 것을 확인하였다. 각 강의 특성에 따라 부유물의 구성비는 달랐지만 부유물들의 전반이상은 식물 쓰레기이다. 그러므로 해양에 유입되는 부유폐기물 수거 방안마련을 위해서는 식물쓰레기에 의한 영향을 고려해야 한다.

소면적 재배작물인 당귀밭에 발생하는 잡초현황과 잡초에 의한 피해 (Occurrence of Weed Flora and Their Yield Loss in Angelica gigas Upland Fields of Minor Crop in Korea)

  • 충북 제천, 경북 봉화를 중심으로 당귀밭에 발생하는 잡초를 조사하여 당귀밭 문제잡초의 현황을 파악하고 잡초방제의 기초자료로 활용하기 위하여 2015년 5월과 6월에 실시하였다. 당귀밭에 발생하는 잡초는 37과 105초종으로 국화과 20종, 화본과 9종, 두과, 마디풀과, 사초과 각각 6종, 꿀풀과, 메꽃과 각각 5종 등의 순이었다. 이들 잡초를 생활형으로 구분하면, 일년생 53종, 월년생 24종, 다년생 28종이었다. 우점잡초로는 바랭이, 쇠비름, 흰명아주, 참방동사니, 망초, 털별꽃아재비 등이었다. 외래잡초는 27종으로 25.7%를 점유하고 있으며, 흰명아주, 망초, 털별꽃아재비, 개비름, 서양민들레, 개망초 순으로 우점도가 높았다. 잡초에 의해 당귀 수량은 49.6% 감소되었다.

국내 가축에서 브루셀라병에 대한 혈청학적 모니터링 (Serological monitoring on brucellosis in livestock of Korea)

  • In Korea, brucellosis has been reported periodically in cattle and rarely in dogs; however, it has not previously been screened in domestic animals such as elk, pigs and goats. To investigate the serological prevalence, serum samples were taken from the aforementioned animals annually during 2007-2013 and screened by the rose-bengal test (RBT) or modified RBT, after which positive sera were evaluated by the standard tube agglutination test (STAT). Finally, RBT and STAT-positive sera were confirmed by competitive-ELISA. Brucella abortus biovar 1 was isolated from three elk that were shown to be positive serologically in 2008. There was no evidence of brucellosis in pigs. Based on serological monitoring and investigation of etiological agents, there is no evidence of outbreak of brucellosis in elk, pigs or goats of Korea since 2008. However, the possibility for brucellosis from cattle to affect these other livestock exists; therefore, extensive and continuous serological monitoring is required to maintain their brucellosis-free status.

당진화력발전소 녹지공간의 식재현황 및 생태적 특성 분석 (Planting Structure and Ecological Characteristics of Green Spaces at the Dangjin Steam Power Plant)

  • This study was conducted to analyze the planting structure and ecological characteristics of greon spaces on the grounds of e Dangjin steam power plant. To achieve these goals, we surveyed existing vegetation, plant community structure, Plant volume and growth rate. Based on e vegetation analysis, existing vegetation was classified into six types: herbaceous species $(70.54\%)$, evergreen coniferous trees $(21.17\%)$, deciduous broad-leaved trees $(5.10\%)$, deciduous coniferous trees $(1.47\%)$, shrubs $(0.12\%)$, and other types $(1.59\%)$. The coal storage, office, and playground areas were community is the natural forest area where a Pinus thunbergii / Pinus densiflora community is distributed in terms of vegetation structure, species diversity, plant volume, and growth rate. The artificial green spaces(near the coal storage, office, playground areas) had a single-layer structure. Species diversity indices of the artificial green areas were $0.1655\~0.4807$ compared to 0.8628 in the natural forest, which presented a good growth environment. Also, the plant volume in the artificial green space was lower than that of the natural green space. Therefore, it would be desirable to develope a multi-layer structure similar to that of the vegetation in the natural green space in order to improve the amount of plant volume. The plant-damage ratio of Pinus thunbergii was $52.48\%$ in the coal storage area, and $8.48\~ 11.52\%$, in the other survey areas. Also, the vitality of Pinus thunbergii was $15.45k{\Omega}$ in the coal storage areas, which indicates bad growing conditions. This suggests that soil characteristics and dust have a bad impact on growth. The investigation into deciduous tres' growth status showed that appropriate plants would be Albizzia julibrissin, Acer palmatum var. sanguineum, Acer palmatum, Malus spp., Prunus sargentii.
