In Vitro Assessment on Biological Activities of Methanol Extracts from Several Compositae Edible Plants

  • Phytotoxicity, antioxidant activity, and cytotoxicity of the aqueous or methanol extracts from the young sprouts of the six Compositae medicinal plants were determined. Aqueous leachates at 40g dry tissue $L^{-1}$ (g $L^{-1}$) Cirsium japonicum and Aster yomena showed the highest inhibitory effect on alfalfa (Medicago sativa L.). Total phenolic content showed the highest amount in methanol extracts from Ixeris dentata, and followed by A. yomena, and Cephalonoplos segetum. Methanol extracts of C. segetum and I. dentata at 25${\mu}g$ m$L^{-1}$ exhibited the highest DPPH radical scavenging activity by 87.2, and 52.8%, respectively. By means of HPLC analysis, MeOH extracts of C. segetum had the highest amount of antioxidant chlorogenic acid. Based on MTT assay, the methanol extracts from Y. sonchifolia ($IC_{50}$ = 65.7${\mu}g$ $mL^{-1}$) showed the highest cytotoxicity against Calu-6. These results suggest that plant extracts had a dose-dependent biological potentials including phytotoxicity, antioxidant activity, and anticancer activity, and that their activities exhibited differently depending on plant species.

Free Radical Scavenging and Antioxidant Activities of Water Extracts from Amannia multiflora, Amannia coccinea, Salix gracilistyla Inhabiting Along the Nakdong River (Republic of Korea)

  • Plant-derived antioxidants are used as a healthy diet and are known to inhibit various human diseases. In this study, we investigated free radical scavenging and antioxidant activity of extracts from three plants (Ammannia multiflora, Ammannia coccinea and Salix gracilistyla) with the most DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging activity from 196 plant extracts inhabiting along Nakdong River in Republic of Korea. The three extracts also have strong total antioxidant activity. Moreover, the extracts inhibited hydrogen peroxide (H2O2)-induced reactive oxygen species production and depolarized mitochondrial membrane potential in RAW264.7 macrophages. In zebrafish larvae, 2',7'-dichlorodihydrofluorescein diacetate (DCFDA) fluorescent intensity, induced by H2O2, was markedly reduced by the extracts of A. multiflora, A. coccinea and S. gracilistyla. Meanwhile, the extracts were upregulated Nrf2 and HO-1 expression, and an HO-1 inhibitor reversed the extract-induced oxidative responses both in vivo and in vitro. The data suggest that the extracts of A. multiflora, A. coccinea, and S. gracilistyla exert potential free radical scavenging and antioxidant capacities both in vivo and in vitro by activating the Nrf2/HO-1 signaling pathway.

한국 약식동원 식물자원의 항산화 활성 비교 (The Screening of Antioxidant Activity in some Korean Medicinal Plants)

  • This study was conducted to find the powerful free radical scavenging effect in methanol extracts from 35 kinds of Korean medicinal plants obtained from the Plant Extract Bank as natural plant products. Antioxidant activity of the plant extracts was determined by measuring the scavenging effect of 1,1-diphenyl-2-picryl hydrazyl radical (DPPH). In the screening of plant extracts tested, at 200 ppm level, almost all plantex tracts showed significantly high antionidant activity. In the reinvestigation of 15 kinds of Korean medicinal plants at 50 ppm level selected as good sources with good free radical scavenging effect at 200 ppm level, the DPPH scavenging effects were similar to those of L-ascorbic acid in whole plant extract of Fragaria yezoensis (90.4%), in the plant stems and roots extract of Gingko biloba, and the leaves extract of Rhododendron mucronulatum var. ciliatum (90%). The fruits extract of Cornus officinalis showed the strongest antioxidant activity (95%). These results suggest that the methanol extracts from Korean medicinal plants can be a potential source of antioxidants that can be used to control oxidative stress in the body, But further study is needed to measure their safety for consumption

Antibacterial and virucidal activity of 28 extracts from plants endemic to Korea against Bacillus cereus, Staphylococcus aureus, and murine norovirus

  • Antibacterial activity against foodborne bacteria (Bacillus cereus, Staphylococcus aureus, Salmonella Enteritidis) and inhibitory activity against murine norovirus, a human norovirus surrogate, of 28 extracts from plants endemic to Korea were investigated in this study. All plant extracts showed antibacterial activity only against gram-positive bacteria, B. cereus and S. aureus. Extracts from Callistemon speciosus and Nymphaea tetragona showed inhibition zones of 16.54 and 24.35 mm against B. cereus and S. aureus, respectively, presenting the highest antibacterial activities recorded in this study. Among all samples, Ardisia japonica extract at concentrations of 100 and 200 ㎍/mL showed the highest virucidal activities of 96.6 and 100.0%, respectively. Ardisia japonica, Duchesnea indica, Polygonum aviculare, and Geum japonicum extracts showed high antibacterial and virucidal activity simultaneously without Raw 264.7 cell cytotoxicity. These plant extracts may serve as potential antimicrobials to control foodborne infections.

An Assessment of Allelopathic Potential of Korean Black Soybean Plant Parts

  • A series of aqueous extracts and residues from leaves, stems, roots, pods and seeds of Korean black soybean (Glycine max (L.) Merr.) were assayed against alfalfa (Medicago sativa) and barnyard grass (Echinochloa crus-galli) to determine their allelopathic activities through petri-dish and greenhouse experiments, and the results showed highest inhibition in the extracts or residues from the seeds, and followed by pods. The extracts of 40g dry tissue $\textrm{L}^{-1}$ applied on filter paper in petri-dish bioassay significantly inhibited root growth of alfalfa, and especially extracts from seeds and pods reduced root length of alfalfa more than those from leaves, stems, or roots. Plant height, root length, shoot and root dry weights of barnyard grass were reduced significantly by residue incorporation of seeds and pods as the incorporated amount increased. These results suggest that black soybean plants had herbicidal potential, and their activities were exhibited differently depending on plant parts.

PRELIMINARY STUDIES ON NATURAL PLANT EXTRACTS AS SUNSCREEN AGENTS

  • The aim of this study is to evaluate several plant extracts with a view to developing UV sunscreen agents. In this study, 150 plant extracts were screened to elucidate their UV spectra using spectrophotometric method. Several plant extracts such as Phellodendron amurense, Morus alba, Rhododendron mucronulatum, Brassica alba have strong absorbency at UVA region (350nm), the suntanning wavelength. And Sophora flavescens, Caesalpinia sapper, Morus alba, Phellodendron amurense, showed absorption plateau value at UVB region (308nm), the erythema action wavelength. These extracts have a good absorbency property as synthetic filter and could be served as substitutes for synthetic UV sunscreen agents.

Inhibition of Biofilm Formation in Yersinia enterocolitica by Edible Plant Extracts Including Polygoni Multiflori Radix

  • Yersinia enterocolitica, which causes yersiniosis, is a bacterium that produces biofilms effectively. The inhibition of biofilm formation provides a method for preventing infections with Y. enterocolitica. In this study, the inhibitory activity of Y. enterocolitica biofilm formation was investigated in a library of 140 edible plant methanol extracts including forest products. It was identified that the biofilm formation could be inhibited by 12 extracts of plants, Agastachis Herba, Agrimoniae Herba, Diospyros kaki leaves, Elsholtziae Herba, Ginkgonis Semen, Lycopi Herba, Melonis Pedicellus, Menthae Herba, Mori Radicis Cortex, Polygoni Multiflori Radix, Prunellae Spica, and Schizonepetae Spica. After changing the solvent to ethanol and water, the greatest inhibition of biofilm formation was produced by a 50% ethanol extract of Polygoni Multiflori Radix. A method to effectively prevent yersiniosis can be developed using the edible plant extracts identified in this study.

In Vivo Antifungal Activities of 57 Plant Extracts Against Six Plant Pathogenic Fungi

  • Methanol extracts of fresh materials of 57 plants were screened for in vivo antifungal activity against Magna-porthe grisea, Corticium sasaki, Botrytis cinerea, Phyto-phthora infestans, Puccinia recondita, and Blumeria graminis f. sp. hordei. Among them, seven plant extracts showed disease-control efficacy of more than 90% against at least one of six plant diseases. None of the plant extracts was highly active against tomato gray mold. The methanol extracts of Chloranthus japonicus (roots) (CjR) and Paulownia coreana (stems) (PcS) displayed the highest antifungal activity; the CjR extract controlled the development of rice blast, rice sheath blight, and wheat leaf rust more than 90%, and tomato gray mold and tomato late blight more than 80%. The PcS extract displayed control values of more than 90 % against rice blast, wheat leaf rust, and barley powdery mildew and more than 80% against tomato gray mold. The extract of PcS also had a curative activity against rice sheath blight and that of CjR had a little curative activity against rice blast. On the other hand, the extract of Rumex acetocella roots reduced specifically the development of barley powdery mildew. Further studies on the characterization of antifungal substances in antifungal plant extracts are underway and their disease-control efficacy should be examined under greenhouse and field conditions.

두충나무, 황벽나무 등을 포함하는 수목추출물의 항여드름 및 항아토피 효과 (Anti-acne and Anti-atopic Dermatitis Effect of Plant Extracts Including Eucommia ulmoides Oliv and Phellodendron amurense)

  • 본 연구에서는 두충나무, 황벽나무 등을 포함하는 수목추출물의 소재특성을 시험하여 화장품 및 피부질환 치료소재로서의 응용 가능성을 조사하였다. 수목추출물의 항산화력은 DPPH 자유라디칼 소거법으로 측정되었으며 저농도에서는 항산화력이 낮았으나, $1000{\mu}g/ml$의 고농도에서는 우수한 항산화제인 비타민 C와 유사하였다. 항균력은 원판확산법으로 측정되었으며, 수목추출물은 피부상재균인 Staphylococcus epidermidis에 대해서는 낮은 항균력을 보이지만, 여드름 유발균인 Propionibacterium acne에 대해서는 높은 항균력을 보여주는 선택성 항균력이 있어, 여드름소재로서 매우 우수한 특성을 나타내었다. 무모생쥐를 이용한 항아토피 시험결과, 수목추출물은 DNCB에 의해 유도된 피부염증을 14일 째 거의 정상상태로 회복시켰으며, 피부내 항체인 IgE의 농도도 대조군에 비해 16% 낮추어주어 항아토피 효과가 우수한 것으로 나타났다. 따라서 수목추출물은 항여드름효과 및 항아토피 효과가 뛰어나서 화장품 혹은 피부질환 치료소재로서 응용 가능성이 매우 높음을 알 수 있었다.

식물 추출물의 화장품 오염 미생물에 대한 생육억제 활성 (Screening for Inhibition Activity of Plant Extracts on Microorganism Contaminating in Cosmetics)

  • A total of 708 methanol extracts from 599 species in 126 families(e.g. Aceraceae) of plant resources were screened for inhibition of cosmetics contaminating microbe, Staphylococcus aureus, Pseudomonas aeruginosa and Candida albicans using 96-well microplate bioassay. Four plant extracts including Celastrus orbiculatus inhibited the growth of Staphylococcus aureus over 90%, 38 extracts including Acer palmatum var. nakaii inhibited the growth of Pseudomonas aeruginosa over 60% and 10 extracts including Ilex crenata var. microphylla inhibited growth of Candida albicans over 60%. Plant extracts showing growth inhibition activity against S. aureus, P. aeruginosa and C. albicans could be used as a reference guide for the future development of natural preservatives for cosmetics.
