Isolation, Identification and Characterization of Phytophthora katsurae, Causing Chestnut Ink Disease in Korea

  • Since July 2005, survey of chestnut ink disease was carried out in chestnut stands located at southern parts of Korea. Dead chestnut trees showing inky ooze on necrotic trunks were found in two different locations. In order to isolate and identify the causal fungus, infected tissues and soil samples around dead or dying trees were collected and placed on Phytophthora-selective medium. Rhododendron and chestnut tree leaves were used as a bait to isolate the fungus from soil samples by attracting zoospores in soil suspensions. On V-8 culture medium, the isolates produced homothallic oogonia with protuberances ($34.0-46.2{\times}21.9-26.7{\mu}m$) abundantly, but did not produced sporangia. Mass production of sporangia was possible by immersing agar plugs with actively growing mycelium in the creek water at $18^{\circ}C$ for 3 days. Sporangia were papillate, and ovoid to obpyriform ($17.0-38.9{\times}14.6-29.2{\mu}m$) in shape. Comparison of the ITS sequences revealed that the isolates had 100% identity to the P. katsurae isolates from Japan and New Zealand and 99.6% identity to other P. katsurae isolates. All of the examined isolates from Korea were completely identical to each other in ITS sequence. Numerous sporangia were formed in filtered as well as unfiltered creek water, but no sporangia formed in sterilized distilled water. Light induced sporangia formation, but has no influence on oospore formation. Amendments of ${\beta}$-sitosterol in culture media have no significant effect on mycelial growth but significantly stimulate oospore and sporangia formation.

Occurrence of Stem Canker on Rape Caused by Leptosphaeria biglobosa in Korea

  • Stem canker symptoms were observed in a rape field in Muan, Korea during a disease survey in May 2006. A total of 15 isolates of Phoma sp. were obtained from the infected stems of the plant. All isolates were identified as Leptosphaeria biglobosa based on their morphological and cultural characteristics. The Korean isolates of L. biglobosa were assigned to 'brassicae' among six subclades of L. biglobosa complex based on the entire ITS sequences of rDNA. Pathogenicity of the fungal isolates was confirmed on leaves and stems of rape by artificial inoculation. This is the first report that Leptosphaeria biglobosa causes stem canker of rape in Korea.

Effect of Polyvinyl Tunnel after Mulching on the Occurrence of Phytophthora Blight and Anthracnose of Red Pepper (비닐터널 유인재배가 고추 역병과 탄저병의 발생에 미치는 효과)

  • This survey was conducted to investigate effect of covering with polyvinyl ($120{\times}90cm$) on incidence of Phytophthora blight and anthracnose and growth. Early growth of pepper plant was superior when was covered with polyvinyl after planting. Incidence of anthracnose and Phytophthora blight was lower 26.9% and 60.3% in covering system than those in conventional culture system, respectively. A yield of dried red pepper was more 48.5% in cultured with covering system than that in conventional culture system.

  • Ginseng is popularly known to be the king of ancient medicines and is used widely in most of the traditional medicinal compositions due to its various pharmaceutical properties. Numerous studies are being focused on this plant's curative effects to discover their potential health benefits in most human diseases, including cancer- the most life-threatening disease worldwide. Modern pharmacological research has focused mainly on ginsenosides, the major bioactive compounds of ginseng, because of their multiple therapeutic applications. Various issues on ginseng plant development, physiological processes, and agricultural issues have also been studied widely through state-of-the-art, high-throughput sequencing technologies. Since the beginning of the 21st century, the number of publications on ginseng has rapidly increased, with a recent count of more than 6,000 articles and reviews focusing notably on ginseng. Owing to the implementation of various technologies and continuous efforts, the ginseng plant genomes have been decoded effectively in recent years. Therefore, this review focuses mainly on the cellular biomolecular sequences in ginseng plants from the perspective of the central molecular dogma, with an emphasis on genomes, transcriptomes, and proteomes, together with a few other related studies.

Green Tobacco Sickness Among Tobacco Harvesters in a Korean Village

  • Background: Green tobacco sickness (GTS), an occupational disease in tobacco harvesters, is a form of acute nicotine intoxication by nicotine absorption through the skin from the wet green tobacco plant. We carried out a questionnaire survey and measured cotinine concentration, the metabolic product of nicotine, to determine the prevalence, incidence, and risk factors of GTS in Korean tobacco harvesters. Methods: We measured cotinine concentrations, and administered a questionnaire survey to tobacco harvesters in Cheongsong-gun, Gyeongsangbuk-do, Korea. We repeatedly measured urine cotinine concentration five times with a questionnaire survey. Results: Cotinine concentration at dawn was significantly higher than that at other times; it was significantly lower during the nonharvesting period than during the harvesting period. However, little change in cotinine concentration was detected in the daytime during the harvesting period. Study participants included 20 men and 20 women. The prevalence of GTS was 37.5% and was significantly higher in women than in men (55.0% vs. 20.0%, p < 0.01). GTS incidence according to number of workdays was 3.4 occurrences/100 person days. Conclusion: In this study, nicotine exposure and metabolism were experimentally determined from the time of cotinine exposure, and biological monitoring was performed in each season. In the future, this information may be valuable for medical decision-making in GTS prevention.

Food behaviors accounting for the recent trends in dietary fatty acid profiles among Korean adults

  • BACKGROUND/OBJECTIVES: This study examined the changes in food behaviors of dietary fatty acids over 2007-2018 among Korean adults. SUBJECTS/METHODS: This study used data from the 4th (2007-2009), 5th (2010-2012), 6th (2013-2015), and 7th (2016-2018) Korea National Health and Nutrition Examination Surveys. A total of 46,307 adults aged 19-64 yrs were selected and dietary data were obtained from a single 24-h recall. In the 4th and 7th data, the major food sources for each fatty acid based on the contributing percentage of the food item were compared. The consumption trends in the major food sources were presented as grams per day over 2007-2018 and compared across the survey periods using the multiple regression model. RESULTS: From 2007 to 2018, for total fat, saturated fatty acid (SFA), and monounsaturated fatty acid, the contribution of animal food sources, including beef, chicken, and eggs increased but plant food sources (e.g., tofu, soybean, and plant oil) decreased. As polyunsaturated fatty acid sources, mayonnaise, eggs, and bread showed higher contributions, whereas soybean and tofu showed lower contributions in the 7th data compared to the 4th data. For n-3 fatty acids, the contribution of fish decreased between the 4th and 7th data. Over 12 yrs, the significant increases in the absolute amount of consumption from animal sources were observed. In contrast, decreases in the consumption from plant sources and fish were seen across the survey periods. CONCLUSIONS: In Korean adults, increases in the intake of dietary fatty acids along with changes in the food behaviors during 2007-2018 have evoked great concern for SFA intake, which is a cardiovascular disease risk factor. Healthy food sources of dietary fatty acids should be emphasized in this population.

Application and utilization of marker assisted selection for biotic stress resistance in hybrid rice (Oryza sativa L.)

  • Development of disease resistant plant is one of the important objectives in rice breeding programs because biotic stresses can adversely affect rice growth and yield losses. This study was conducted to identify lines with multiple-resistance genes to biotic stress among 173 hybrid rice breeding lines and germplasms using DNA-based markers. Our results showed that one hybrid rice line [IR98161-2-1-1-k1-3 (IR86409-3-1-1-1-1-1/IRBB66)] possessed 5 bacterial blight resistance genes (Xa4, xa5, Xa7, Xa13 and Xa21) while two hybrid rice lines [IR98161-2-1-1-k1-2 (IR86409-3-1-1-1-1-1/IRBB66) and 7292s (IR75589-31-27-8-33S(S1)/IR102758B)] possessed 3 bacterial blight resistance genes (Xa4, Xa7 and Xa21, and Xa3, Xa4 and xa5). Molecular survey on rice blast disease revealed that most of these lines had two different resistant genes. Only 11 lines possessed Pib, Pi-5, and Pi-ta. In addition, we further surveyed the distribution of insect resistant genes, such as Bph1, Bph18(t), and Wbph. Three hybrid breeding lines [IR98161-2-1-1-k1-3 (IR86409-3-1-1-1-1-1/IRBB66), IR98161-2-1-1-k1-2 (IR86409-3-1-1-1-1-1/IRBB66), and 7292s (IR75589-31-27-8-33S(S1) /IR102758B)] contained all three resistance genes. Finally, we obtained four hybrid rice breeding lines and germplasms [IR98161-2-1-1-k1-2 (IR86409-3-1-1-1-1-1/IRBB66), Damm-Noeub Khmau, 7290s, and 7292s (IR75589-31-27-8-33S(S1)/IR102758B)] possessing six-gene combination. They are expected to provide higher level of multiple resistance to biotic stress. This study is important for genotyping hybrid rice with resistance to diverse diseases and pests. Results obtained in this study suggest that identification of pyramided resistance genes is very important for screening hybrid rice breeding lines and germplasms accurately for disease and pest resistance. We will expand their cultivation safely through bioassays against diseases, pests, and disaster in its main export countries.

Wilt of Perilla Caused by Fusarium spp.

  • A survey of Fusarium wilt of perilla was conducted in 12 locations in Korea from 1999 to 2001. The disease occurred in 74 out of 187 fields in the 12 locations surveyed, and incidence of the disease reached up to 30% at its maximum in some perilla fields in Seosan and Dangjin. Incidence of the disease in the other locations ranged from 0.2 to 20%. A total of 327 isolates of Fusarium spp. were obtained from stems and roots of the diseased perilla plants. The isolates were identified based on their morphological characteristics. Out of the 327 isolates of Fusarium, 277 isolates from 12 locations were identified as F. oxysporum, 11 isolates from three locations as F. solani,17 isolates from two locations as F. equiseti, 4 isolates from one location as F. avenaceum and 6 isolates from one location as F. subglutinans. The other 12 isolates of Fusarium from four locations were unidentified. Twelve isolates of F. oxysporum and two isolates each of the other Fusarium spp. were tested for their pathogenicity to five cultivars of perilla. Seven isolates of F. oxysporum were strongly pathogenic to some perilla cultivars, but the other five isolates were weakly or not pathogenic. One isolate of F. solani was strongly pathogenic to all the perilla cultivars tested, but another isolate was not pathogenic. All the isolates of F. equiseti, F. avenaceum, and F. Subglutinans tested were not pathogenic to any of the perilla cultivars tested. Symptoms on the perilla plants induced by artificial inoculation with strongly pathogenic isolates of F. oxysporum and F. solani appeared as wilt, stem blight, and root yet, which were similar to those observed in the fields. The isolates which induced symptoms by artificial inoculation were re-isolated from the lesions of the perilla plants inoculated. All the isolates of F. oxysporum tested were not pathogenic to eight other crops inoculated. Results of this study reveal that F. oxysporum is the main pathogen of perilla wilt and that it is host specific to perilla. forma specialis of F. oxysporum causing wilt of perilla is proposed as perillae.

Survey of Viral Diseases Occurrence on Major Crops in 2007 (2007년 우리나라 주요 작물 바이러스병 발생 상황)

  • The severe damage induced by the important viruses of Rice stripe virus (RSV), Cucumber green mottle mosaic virus (CGMMV), Melon necrotic spot virus (MNSV), Tomato spotted wilt virus (TSWV) and Tomato bushy stunt virus (TBSV) was described on major crops in Korea. In 2007, the plot incidence rate of RSV was 100% on the precocious rice cultivars at the Western coastal provinces of Gyeonggido, Chungcheongnamdo, Jellabugdo and Jellanamdo, and Jejudo. RSV occurred in 2,441 ha with incidence rate of 70% over at 5 areas of Seocheon, Seosan, Boryung, Hongsung and Buyou in Chungcheongnamdo. At 4 areas of Buan, Gimje, Gunsan and Gochang in Jellabukdo, RSV occurred in 2.016 ha. CGMMV occurred on watermelon in 4.6 ha at Cheongyang area, and its outbreak was also 890 ha on oriental melon for 120 farmers with the incidence area of 23% against total cultivation areas of Seongju. MNSV was recorded firstly on watermelon in 2006 at Andong and it spread to 3 areas of Hapcheon, Gochang and Yanggu. TSWV occurred firstly at Danggin in Chungcheongnamdo in 2005. TSWV in 2006 spread to 6 areas; Taian, Hongsung and Seosan in Chungcheongnando, Namwon in Jellabukdo, and Sunchon and Kwangju in Jellanamdo. In 2007, TSWV covered 17 areas of western and southern parts; the 5 area including Taian in Chungcheongnamdo, Kwangju in Jellanamdo, Bucheon in Gyunggido, and so forth. TBSV was described firstly on table tomato at Sacheon in Kyungsangnamdo in 2004. TBSV occurred on cherry tomato at Chungju in 2006 and on table tomato at Busan area.

Incidence of Wildfire Disease on Soybean of Korea during 2014-2015 (2014-2015년 국내 콩 들불병 발생 상황)

  • Wildfire caused by Pseudomonas amygdali pv. tabaci has been seen a steady increase in the incidence and prevalence on soybean in Korea. But up to know there is no official report on the prevalence of the disease in Korea. The nationwide survey of the occurrence of soybean wildfire showed that 19 out of 30 regions in 2014 and 9 out of 28 regions in 2015 had been infected with P. amygdali pv. tabaci. The severity of wildfire varied according to the cultivar, cultivation regions and climates. However, cultivar seoeitae was susceptible in both 2014 and 2015. The disease severity of 2014 was twice that of 2015. Since the temperature of 2014 and 2015 were very similar but the precipitation of 2014 was more than twice that of 2015, the precipitation of 2014 had a great impact on the wildfire disease severity.
