Electron beam (EB) irradiation was tested to determine the dose required to eradicate plant pathogens, such as Botrytis cinerea and Agrobacterium rhizogenes, from the infected seeds without affecting the germination rate of the irradiated vegetable seeds, including crown daisy, cucumber, hot pepper, green onion, leaf lettuce, and radish seeds. EB irradiation of 1.5 kGy and 2 kGy was sufficient to kill 100% of hairy root disease bacteria and gray mold conidia, respectively. EB irradiation showed no effect or minimal effect on the germination rate of the crown daisy, cucumber, green onion, and radish seeds. However, the germination rate of the hot pepper and leaf lettuce seeds was significantly reduced by using 2 kGy of EB irradiation. Difference in susceptibility to EB irradiation appears not to be related to the weight of each seed, but to the intrinsic characteristic of each plant. Conclusively, EB irradiation might be a useful way to decontaminate crown daisy, cucumber, green onion, and radish seeds.
Salinity has been a threat to agriculture in some parts of the world; and recently, the threat has grown. Plant growth-promoting rhizobacteria (PGPR) may benefit plant growth, either by improving plant nutrition or producing plant growth hormones. The effects of rhizobacterial strains to attenuate the salinity stress on the germination of Chinese cabbage seeds were tested using four different concentrations of NaCl (50, 100, 150, and 200 mM). Also, PGPR strains were tested to enhance the early germination of Chinese cabbage seeds under normal conditions. Azotobacter chroococcum performed best with enhancing the radicle length of 4.0, 1.2, and 1.0 times at treatments of 50, 100, and 150 mM of NaCl, respectively. Additionally, significant differences were found in plumule length, A. chroococcum and Lactobacillus sp. showed remarkable activation either in normal or under stress conditions. Co-inoculation by three rhizobacterial strains (LAPmix) indicated synergistic effect to enhance the early germination of the seeds. The results of this study are promising for application of rhizobacterial strains that possess plant growth promoting traits to enhance the plant tolerance against salinity.

One key step in the plant restoration process is the collection of seeds from the field. For the selection of source populations of target plant species for translocation purposes (reintroduction or reinforcements), several approaches are possible. A practical method involves the use of data from reciprocal transplant studies. If no direct data are available, knowledge of population genetics and the phylogeography of the target species can serve as an alternative. In this short review, we briefly propose guidelines for those collecting seeds for plant species restoration based on population genetics theory, focusing on two main questions: Where does the plant material come from and how are sources designated, and how are seeds efficiently collected from local populations? While genetic data on a larger scale (phylogeography and population genetics) are needed to form a reply to the first question, similar data on a smaller scale (fine-scale genetic structures within populations) are necessary to shed light on the second issue.
An antagonistic bacterium, Pseudomonas flurorescens MC07 inhibited the mycelial growth of Rhizoctonia solani, Pythium ultimum, Fusarium oxysporum, and Phytophthora capsici in on potato dextrose agan (PDA) and other media. The strain MC07 conlonizes various plant roots and possesses antifungal activity. To determine the role of antifungal activity of the bacterium in disease suppression, a mutant Okm3-4 which lost its activity was isolated after screening 2,500 colonies generated by Omegon-Km insertions. The mutant Okm3-4 showed diminished growth inhibition of R. solani, P. ultimum, F. oxysporum, and Ph. capsici in vitro and had reduced suppressive effects on sesame damping.-off compared to the parental strain. In soils, accumulation of the pathogens by continuous cropping, 90% of sesame plants were killed by natural infection of damping-off whereas, only 29% of plants grown from seeds treated with MC07 were killed. On the other hand, 85% of plants died when sesame seeds were treated with the Okm3-4 cells. This indicated that antifungal activity of MC07 in vitro is directly responsible for the suppression of damping-off disease. Emergence rates of sesame seeds in pots containing diseased soil were 33%. However, MC07 treatments on seeds significantly improved emergence rates, which has similar effects of Benomyl treatment. The mutant Okm3-4 exhibited 53% of emergence rate. This indicated that antifungal activity of MC07 also affects the emergence rate of sesame seeds.
The present study was carried out to investigate the effects of cadmium(Cd), mercury(Hg) and lead(Pb) on the respiration of the germinating seeds. Two kinds of seeds namely, mungbean (Paseolus radiatus L.) and buckwheat (Fagopyrum esculentum M ench) were used for the plant materials. The concentrations of Cd, Hg, and Pb applied for the treatment were 0.1, 1, 10, 100 and 1,000ppm. The germination test was carried out in the dark at 27$^{\circ}C$. Regardless of the kinds of heavy metals, the respiration of the germinating seeds was greatly increased for 24hrs absorbing the heavy metals, and subsequently the respiration was increased average 0.69mg/g.hr.CO2 for mungbean and 0.7mg/g.hr.CO2 for buckwheat. In the lower concentrations of cadmium and lead than 10ppm the germinating rates, seedling concentraitons. The germinating rates, the seedling growth and the respiration were not increased in ay concentrations of mercury, but were decreased with increase of the mercury concenration. The inhibition of seeds respiration by mercury was 3 times higher than by cadmium, and 5 times higher than that by lead. The toxicity of three heavy metals on the respiration was showed from the highest to the lowest in the order of Hg, Cd, and Pb.

Protocol was established for production of virus free healthy seeds using meristem ($0.3-0.5\;\cal{mm}$ in size) culture and field management under net house condition in tomato. The isolated meristem was found well established in MS liquid medium containing $0.1\;\cal{mg}\;1^{-1}\;of\;GA_3$. For shoot and root development either from primary meristem or from nodal segment of meristem derived plants, semisolid MS medium having $0.5\;\cal{mg}\;1^{-1}$ of IBA was found most effective. The elimination of the studied viruses (ToMV, CMV, ToLCV) in meristem-derived plants was confirmed by DAS-ELISA test. For field management of the virus eradicated meristem-derived plants, use of net house was found very effective measures to check viral vector visit and eventually infection. The meristem-derived plants were vigor and high yielder than the native seed derived plants and produced healthy seeds. Due to stop vector visit, no viral symptoms were observed in both $R_1\;and\;R_2$ plants cultivated in net house condition. Starting of viral infestation was observed in $R_2$ generation when they were planted in open house condition without control of vector visit. Therefore, for management of viral diseases, use of virus free meristem derived plantlets and their subsequent cultivation in soil under net house condition without using any vector killing insecticide can be recommended for producing healthy seeds in tomato. The developed protocol for environmentally healthy tomato seed production in Bangladesh may be used in the countries having similar tropical like environment conducive for viral vector visit.

The equilibrium moisture contents (EMC) in seeds of four Echinochloa (E. crus-galli var. crus-galli, E. crus-galli var, echinata, E. crus-galli var. praticola, E. esculenta) were measured at $20^{\circ}C$ with equilibration over a series of lithium chloride solutions with relative humidities ranging from 0.11 to 0.8 to determine sorption isotherms and safe storage relative humidity. Standard seed sorption isotherm models i.e. modified Henderson, modified Chung-Pfost, modified Halsey, modified Oswin and Guggenheim-Anderson-deBoer (GAB) equations were adopted to evaluate the goodness of fit to sorption isotherms. This study indicated that EMC of seeds was significantly different in four Echinochloa species at various relative humidity. The modified Oswin equations for E. crus-galli var. crou-galli, E. crus-galli var, echinata, E. esculenta and GAB equation for E. crus-galli var. praticola were adequate models for the EMC data. Seeds of four Echinochloa species have monolayer moisture contents when stored at RH < 0.1. These results show that seed moisture isotherm model should be selected according to genetic variation.

In this study, seven grain seeds(sorghum maize, buckwheat, soy bean, mung bean, red bean, and barngrass) and germinated seven grain seeds were examined the fibrinolytic activity through fibrin plate assay and SDS-PAGE. The results obtained were as follows : 1. In the fibrin plate assay, the extracts of maize, barngrass, sorghum and buckwheat showed fibrinolytic activity. Especially, Maize of them showed fibrinolytic activity that was almost similar to plasmin, fibrinolytic enzyme used as a positive control. 2. In the SDS-PAGE of seven grain seeds, fibrinolytic activity was remarkably shown in mung bean and red bean. 3. In the fibrin plate assay of germinated grain seeds, buckwheat(5 mm), buckwheat(10 mm) and soy bean(10 mm) showed a level of fibrinolytic activity that was about 0.3 fold than 1.0 unit of plasmin, Also maize(10 mm) of them showed a level of fibrinolytic activity that was about 0.5 fold than 1.0 unit of plasmin. As a result, maize of grain seeds was found that it has a strong fibrinolytic activity.

This study was carried out to investigate of the biologically active components in germinating Mung bean(Phaseolus radiata L.) and Buckwheat (Fagopyrum esculentum Moench) seeds. During the initial germination, germination ratio of 24 hours pre-soaking Mung bean and Buckwheat seeds were higher about 2∼3% than that of non-soaking. This experiment also was peformed to observe cytotoxic effect of the germinating seeds(germination length : 2, 5, 10mm) extracts against cancer cell lines including human lung carcinoma(Calu-6), human breast adenocarcinoma(MCT-7), human great intestine carcinoma(Caco-2) and human leukemia carcinoma(AML-2/WT). The growth of the cancer cells in medium containing Mung bean and Buckwheat extracts were significantly inhibited degree in proportion to the length of germination seeds, Especially, the results show that a significant shrinkage of Calu-6 cells was observed when the cells were exposed into extract of 10mm germination seeds in germinating Mung bean and Buckwheat seeds.
This study was conducted to control the seed borne pathogens (Alternaria spp.) of carrot and to examine the seed germination rate by using 13 environment friendly agricultural materials instead of conventional chemicals for organic cultivation. The growth inhibiting effects on pathogens showed the different responses according to each agricultural material and effective 7 materials against seed borne pathogens were selected. Among 7 materials, the carrot seeds sterilized with plant extracts, Tanger Stop and Land Saver were not germinated at all. The germination rate of seeds sterilized with other materials showed the similar levels with reference chemical (Benlate-T) and non-treated seeds. Infection rates of seeds sterilized with seaweed extract, Bellopper for controlling A. radicina and plant extract, Ssial-100 for A. dauci were similar or low infection rate compared with reference chemical. The germination rate and root length of seeds sterilized with Bellopper and Ssial-100 was similar or superior to those of chemical sterilized seeds. The optimal condition seemed to be a little bit different depending on the concentration of materials. As a result, the sterilization of carrot seeds by using the environment friendly materials could be effectively utilized as a technology to inhibit the infection of seed-borne pathogens.
