Flavone (2-phenyl chromone) is a well-known plant flavonoid, but its bioactivity has been little explored. Treatment of Caenorhabditis elegans or C. brissage with flavones induced embryonic and larval lethality that was pronounced in early larval stages. This anti-nematodal effect was also observed in the pinewood nematode, B. xylophilus. $LD_{50}$ values were approximately $100{\mu}M$ for both B. xylophilus and C. elegans. Our results indicate that flavone is an active nematicidal compound that should be further investigated with the aim of developing a potent drug against B. xylophilus.

There is an adsorption method using activated carbon as a typical method for removing elemental mercury. Physical characteristics of activated carbon such as specific surface area and volume of pore (micro and meso) have positive effect for mercury adsorption. Activated carbon is carbon-based material with a high specific surface area. This activated carbon can be manufactured through carbonization and activation process. In this process, physical characteristics of specific surface area and pore distribution are changed by controlling operating parameters like temperature, time and reagent of activation. In this study, we evaluated characteristics of activated carbons manufactured from pinewood and coal with the operating parameters. We evaluated mercury adsorption capacities of the activated carbons having excellent physical characteristics and compared those to the commercial activated carbon.
Korean pine (Pinus koraiensis Sieb. et Zucc.), a five-needle pine, has recently been suffering pine wilt disease caused by non-native pinewood nematode, Bursaphelenchus xylophilus. Three-year-old Korean pine seedlings were inoculated with 10,000 pathogenic nematodes in a greenhouse to investigate disease development, water content and the density of nematodes in stems. Needle dehydration, xylem drying and pith browning started 20 days after inoculation (DAI). There were significant differences between seedlings inoculated with nematodes and control seedlings in the relative water content of stems and leaves at 20 and 30 DAI. At 60 DAI, all remaining seedlings inoculated with nematodes had died, but control seedlings all remained alive. The average number of nematodes recovered from stems of Korean pine dramatically increased from 10 to 20 DAI, and then decreased at the end of the experiment at 60 days. This study suggests that the relative water content of stems and leaves in current-year branches could be used as a useful physiological indicator for early diagnosis of pine wilt disease.
Japanese pine sawyer, Monochamus alternatus, is a primary vector of pinewood nematode, Bursaphelenchus xylophilus in Korea. Emergence characteristics of the adults were studied in Jinju, Korea using dead pine logs in which the larvae of the sawyer overwintered from 1999 to 2002. Emergence data showed that the adults began to emerge out of the logs in sunny place from May the 15th in average of the four years. Cumulative emergence ratio (CER) reached at 50% at mid-June, and the emergence terminated by early or late July depending on years. In shady place, however,50% CER was reached 17 days later than in sunny place. The 50% CER of males reached 1 to 4 days earlier than that of females depending on years. Of the adults emerged from the logs collected from February to April in 2001, 97.6% was univoltine, and the rest was biennial which emerged from May to July of the next year. Female and male adults emerged throughout 24 hours; 32.3% of the total adults emerged from 8 to 12 a.m. It took 68.0 seconds for the adults to escape from pine logs.

The pinewood nematode Bursaphelenchus xylophilus (Nematoda: Parasitaphelenchidae) poses a serious threat to pine forests in Europe and East Asia, leading to a debilitating pine wilt disease. Infected pine trees in Korea are generally fumigated or crushed to small wood chips after felling. Although pine wilt disease often recurs in pest management sites, there are no adequate means to monitor the effectiveness of pest control measures in those sites. Recently, a male-produced aggregation pheromone, 2-(undecyloxy)ethanol, was shown to be useful for attracting several Monochamus species, which are vectors for the pinewood nematodes. In this study, we investigated the abilities of 2-(undecyloxy)ethanol at three different doses (175, 350, and 700 mg), as well as host plant volatiles (α-pinene and ethanol), to attract M. alternatus (Coleoptera: Cerambycidae) at a pine forest in Pohang, Korea where infected pine trees had been cut down and fumigated. Twenty-seven M. alternatus were captured in cross-vane panel traps made of polyethylene terephthalate bottles and acrylic sheets. The results indicate that a high dose of 2-(undecyloxy)ethanol (700 mg per trap) is the most effective for attracting M. alternatus. The aggregation pheromone could be used to monitor the effectiveness of pest control measures as well as M. alternatus populations.
Physical characteristics of Korean red pine (Pinus densiflora) were investigated with different cultivation locations in Taebaek Mountains as Goseong-gun, Hongcheon-gun and Bonghwa-gun as experimental sites. Moisture content based on air-dried or green (artificial) wood was no significant differences with different cultivation places. Specific gravities of both sapwood and heartwood of red pine from Bonghwa-gun were higher than those from other two sites (Hongcheon-gun or Goseong-gun). Specific gravity of heartwood of red pine from Goseong-gun was higher than it from Hongcheon-gun, but this trend was opposite in case of sapwood. Higher specific gravity of red pine heartwood from Goseong-gun maybe resulted in higher strength than those of other sites. In shrinkage ratio, there was no significant difference among different cultivation places in radial directions, but red pine from Bonghwa-gun was higher than those of other sites in tangential directions. Hygroscopicity was no significant differences with different cultivation places. These results will be helpful information for efficient use of pinewood and good quality of pinewood production for genetical breeding improved.
In this study fire control characteristics for inflammable materials of water curtain system are experimentally analyzed. Heat release rate for pinewood and gasoline was calculated using Room Corner Tester (RCT) and fire test apparatus for water curtain system is manufactured. Nozzles (180 degree of injection angle, 8.2 mm of orifice diameter) are installed at the nearby ceiling of place at 5 m distance from fire originate and temperature profile as well as transmission are obtained from the fire experiment of pinewood and gasoline in the water curtain system. Based on the results, parameters of engineering importance for fire control characteristics of water curtain system such as generation of high temperature smoke and thermal phenomena of fluid flow by injection nozzle are identified.

During a survey of the activities of fungi in steamed sweet potato, stored garlic, agricultural by-products for mushroom cultivation media, and pinewood chips from a pinewood nematode-infected tree, numerous fungal samples were isolated and identified. This study identified five species that have not been previously reported in Korea, namely Geomyces pannorum, Neopestalotiopsis javaensis, Penicillium allii, Penicillium chermesinum, and Ophiognomonia setacea. For all identified species, the cultural features of colonies formed on growth media, their morphological characteristics observed by a light microscope, and their molecular phylogenetic relationships based on nucleotide sequences of the internal transcribed spacer rDNA region or calmodulin gene were described.
RNA interference (RNAi) was performed on several essential genes in the pinewood nematode Bursaphelenchus xylophilus, which causes pine wilt disease. Double-stranded RNA (dsRNA) was delivered to larvae or adult worms by soaking, electroporation, or microinjection. Soaking and electroporation of L2-L3 stage worms in solutions containing dsRNA for essential genes induced over 25% lethality after 5 days, and gene-specific phenotypes were observed. This lethality agreed with significant reductions of the targeted transcripts, as assayed by reverse-transcription coupled with real time PCR. Microinjection was the most efficient route as measured by the hatching rate of F1 embryos, which was reduced by 46%. When adult worms were soaked in dsRNA, lethality was induced in the F1 larvae, revealing the persistence of knockdown phenotypes. The penetrance of the RNAi phenotypes for essential genes was relatively low but consistent, indicating that RNAi should be useful for studying the in vivo functions of B. xylophilus gene products.
