국내 분리 porcine circovirus 2의 이유자돈에 대한 병원성 시험연구 (Pathogenesis and pathogenicity for the porcine circovirus 2 Korean isolates in weaned pigs)

  • This study was carried out to investigate the pathogenesis and pathogenicity of the porcine circovirus type 2 (PCV2) Korean isolate from weaned pigs. Twenty four weaned pigs, PCV2, porcine reproductive and respiratory syndrome virus (PRRSV) and porcine parvovirus (PPV) antibodies free, were allocated to 4 groups (n = 6). Six pigs were inoculated intranasally with PCV2 alone, 6 with PCV2 and PRRSV, 6 with the combined PCV2/PRRSV/PPV inoculum, and 6 were remained as a uninoculated negative control. Pigs were killed 3 and 6 weeks after inoculation and tissue samples examined for gross and microscopic lesions and for the presence of PCV2 antigens and nucleic acids. Experimentally inoculated pigs were evaluated for 3 considerations: 1. development of postweaning multisystemic wasting syndrome (PMWS), 2. distribution of viral antigens by immunohistochemistry and polymerase chain reaction (PCR), and 3. cytokine mRNA levels in lymph nodes. Pigs inoculated with PCV2/PRRSV/PPV showed typical clinical signs, gross findings, and histopathologic characteristics of PMWS. In the PCV2/PRRSV/PPV inoculated group, the PCV2 antigen was widely distributed in various parenchymal organs such as brain, spinal cord, tonsil, lymph nodes, lung, heart, liver, kidney, spleen, and peyer's patch. Lymph node mRNA expression of IL-$1{\alpha}$, IL-2R and IL-8 was determined by real-time PCR. The pigs of PCV2/PRRSV and PCV2/PRRSV/PPV inoculation group, the mRNA expression was characterized by a decrease of IL-$1{\alpha}$, IL-2R and IL-8. The decrease of cytokine mRNA represent the state of T cell immuno-suppression in pig, and nicely support the evidence for the impairment of immune system in pigs with PMWS. In conclusion, PCV2 infection and some additional infectious causes such as PRRSV and/or PPV are warranted for the presence of PMWS in weaned pigs in Korea.

  • The analysis of membrane receptors for hormones and neurotransmitters has progressed considerably by pharmacological and biochemical means and more recently through the use of specific antibodies. To generate and characterize a moloclonal antibody against $\beta$-adrenergic receptor, a synthetic $\beta$2-adrenergic receptor peptide (Phe-Gly-Asn-Phe-Trp-Cys-Phe-Trp-Thr-Ser-lle-Asp-Val-Leu) which may comprise part of $\beta$-adrenergic receptor ligand binding pocket was coupled to Keyhole Limpet Hemocyanin (KLH) and used as an immunogen. Male BALB/C mice were immunized with this antigen and the immunized spleen was fused with myeloma SP2/0-Ag14 cells to produce monoclonal antibodies. Two clones were obtained but one of monoclonal antibodies, mAb5G09, was used throughout in this study because the other clone, mAb5All showed weak immunoreactivity against KLH as well. The mouse monoclonal antibody mAb5G09 produced in this study showed immunoreactivity to peptide-KLH conjugates and also to human A43l cells and guinea pig lung $\beta$2-adrenergic receptor as revealed by ELISA and western blot. In the course of determination of the effects of mAb5G09 on $\beta$-receptor ligand binding, it was observed that mAb5G09 specifically bound $\beta$-adrenergic radioligand [$^3$H]dihydroalprenolol (DHA) with a dissociation constant (Kd) of 60 nM. The [$^3$H]DHA binding activity of mAb5G09 had characteristics of immunoglobulins and the binding activity was not observed in the control anti-KLH monoclonal antibody. The monoclonal antibody, mAb5G09 produced in this study may provide useful models for the study of the structure of receptor binding sites.

피루브산염과 아스파라진산염을 첨가한 위스콘신대학 용액의 심근보호 효과 (Effect of Pyruvate and Aspartate Enriched University of Wisconsin Solution on Myocardial Protection)

  • 배경: 심장이식에 있어서 허혈-재관류 손상은 이식심장의 기능회복이나 장기생존을 좌우하는 중요한 요소이다. 본 연구에서는 세포질내의 NADH/NAD$^{+}$ 비율의 조절에 관여하는 피루브산염과 아스파라진산염을 현재 일반적으로 흔히 쓰이고 있는 장기 보존액인 위스콘신대학 용액에 첨가하여 심근을 보호하고 재관류 후의 심장 보존능의 효과를 증명하고자 하였다. 대상 및 방법: 생후 3일 이내의 신생돈의 심장을 4$^{\circ}C$ 위스콘신대학 용액(대조군, n=8)과 피루브산염과 아스파라진산염을 첨가한 위스콘신대학 용액(실험군, n=8)으로 심정지를 유도하여 적출하고 4$^{\circ}C$ 동일한 용액에서 24시간 동안 허혈상태로 보존한 후, 성돈을 교차순환 재관류혈 공급원으로 사용하여 좌심단순작업성 관류모델(left-sided working heart model)로 재관류 시킨 다음, 관류심장에서 일정한 시간 간격으로 박출작업계수(stroke work index)를 측정하였고, 관류가 끝난 후, 고 에너지 인산함유량(high-energy Phosphate stores), 심근의 수분 함유량(myocardial water content)을 측정하여 두 군을 비교하였다. 결과: 재관류가 시작되고 60분과 120분이 경과된 후에 좌심실 확장기말 압력(LVEDP)이 3, 6, 9, 및 12mmHg일 때 각각 박출작업계수를 측정하였는데, 60분이 경과된 후에 측정한 박출작업계수는 실험군에서 통계적으로 유의하게 높았고 [n=8, p<0.05, 대조군(16.3 $\pm$8.3$\times$1,000 erg/g) vs. 실험군(33.1 $\pm$ 15.1$\times$1,000 erg/g)], 120분이 경과된 후에도 실험군에서 통계적으로 유의하게 높게 나타났다 [n=8, p<0.05, 대조군(15.8$\pm$8.0$\times$1,000 erg/g) vs. 실험군(35.1$\pm$16.3$\times$l,000 erg/g)〕고 에너지 인산 중, AMP, ADP, 및 ATP의 함유량은 실험군에서 높게 측정되었다 [n=8, p<0.05, AMP -대조군(30.8$\pm$8.7 micromo1/g myocardium) vs. 실험군(53.1$\pm$11.1),ADP -대조군(52.6$\pm$7.3) vs. 실험군(91.3$\pm$20.9), ATP -대조군(67.5$\pm$23.8) vs. 실험군(156.5$\pm$45.8)]. 그러나, creatine phosphate 함유량 [n=8, p>0.05, 대조군(546.6$\pm$197.0 micromol/g myocardium) vs. 실험군(595.5$\pm$179.6) 과 심근 수분 함유량 [n=8, p>0.05, 대조군(87.2$\pm$5.5%) vs. 실험군(82.4$\pm$10.1)] 은 두 군간에 유의한 차이가 없었다. 결론: 이상의 결과는 피루브산염 와 아스파라진산염이 첨가된 위스콘신대학 용액이 위스콘신대학 용액만을 사용한 경우보다, 박출작업계수와 심근에너지 보존 측면에서 평가하기로는 심근보호 기능이 더 우수하다는 것을 나타내고 있다. 그리고, 대사과정에서 NADH/NAD$^{+}$의 대사와 관련이 없는 creatine phosphate의 보존에는 차이가 없다는 결과는 피루브산염과 아스파라진산염이 세포질내의 NADH/NAD$^{+}$의 조절에 관여하여 심근을 보호하리라는 가설을 가능하게 한다.

돼지에서 Tiletamine-Zolazepam 단독과 Azaperone, Tiletamine-Zolazepam 합제의 마취 효과에 대한 비교 (Comparison of Anesthetic Effects Induced by Tiletamine-Zolazepam and Azaperone Plus Tiletamine-Zolazepam in Growing Pigs)

  • 본 연구에서는 돼지에서 tiletamine-zolazepam (TZ) 단독 사용과 azaperone, tiletamine-zolazepam (ATZ) 합제 사용시의 마취효과를 규명하고, 양군에서의 생리학적 parameters를 비교하였다. 두개의 군으로 구분하였으며 건강한 6개의 랜드레이스와 요크셔 교잡종 돼지를 실험에 사용하였다. 교차실험을 하였으며, 각 군 사이의 휴약기간은 2주로 하였다. 1군 (TZ 군): 돼지 6두 ($31.4{\pm}4.83$) kg에 TZ 4.4 mg/kg을 투여하였다. 2군 (ATZ 군): 동일한 돼지 6두 ($43.6{\pm}4.31$ kg)에 azaperone 2 mg/kg을 투여하고, 20분 후에 TZ 4.4 mg/kg을 투여하였다. 모든 마취약물들은 등세모근에 근육주사를 하였다. 약물 투여 24시간 전부터 절식을 시켰다. 도입 및 회복 시간을 측정하였다. 심박수, 호흡수 체온, $pO_2,\;pCO_2$ 및 pH를 투여 전, 투여 후 5분, 25분, 45분, 65분 및 85분에 측정하였다. 도입시간에서는 ATZ 군이 TZ군에 비하여 더 빠른 도입을 나타내었다 (p<0.01). 회복에 있어서, 흉와자세 시간, 기립시간 및 보행시간은 ATZ군이 TZ군에 비하여 더 긴 시간을 나타내었다 (p<0.01). 심박수, 호흡수, $pO_2,\;pCO_2$, 및 pH는 두 군간에서 유의차가 인정되지 않았다. 그럼에도 불구하고, 체온은 두 군간 유의성 있는 변화가 인정되었다 (p<0.05). 이상의 결과를 종합하여 볼 때에, ATZ군이 TZ군에 비하여 빠른 도입과 긴 마취시간을 나타내었다. 따라서, ATZ 병용은 돼지의 화학적 보정에 유용하게 사용될 수 있을 것으로 판단되었다.

돼지 대동맥, 폐동맥의 근위부 기하학적 구조 측정을 통한 판막 구조 수치의 계량화와 판막 도안에 관한 연구 (Measurement of Porcine Aortic and Pulmonary Valve Geometry and Design for Implantable Tissue Valve)

  • 배경: 사람의 수명이 연장되면서 판막 질환이 점차 증가하는 가운데, 과거에는 심장 판막 이식에 기계 판막을 많이 사용하였으나, 혈액 응고 장애로 인한 출혈, 임신 시 태아의 기형 가능성, 판막의 급성 기계적 작용 부전, 평생 약을 복용해야 하는 불편함 등의 여러 합병증이 동반되는 단점이 있어, 조직 판막 사용이 증가하고 있고, 그 내구성을 높이기 위한 연구와 관심이 점차 증가하고 있다. 현재 사용하는 조직 판막은 외국계 회사의 수입된 제품으로서 그 구조나 도안에 이론적인 제안 등이 부족한 실정이다. 이에 본 연구는 돼지의 대동맥 및 폐동맥 판막의 구조에 대한 기하학적 분석을 시행하고, 각 수치를 계량화, 분석하여 그 3차원적 구조를 이해하여 대동맥 판막과 폐동맥 판막을 만들 수 있는 이론적 배경을 제시하고자 하였다. 대상 및 방법: 도축한 25마리의 돼지에서 채취한 대동맥 판막과 폐동맥 판막을 신선한 상태에서, 그리고 글루타알데하이드(GA) 용액으로 고정한 상태에서 판막의 각 구조물 수치를 2차원적으로 측정하여 그 배율을 계량화하고 분석하였다. 신선한 상태와 글루타알데하이드(GA) 용액으로 고정한 상태에서 판막엽 간 길이, 접합면 간 길이, 판막엽의 높이, 접합면 높이를 각각 측정하였다. 또한 이식 가능한 판막을 제작하기 위하여 스텐트의 도안으로 모양, 유순도에 따른 스텐드의 두께, 높이, 간격 및 판막 크기에 따른 심낭 판막엽의 크기, 모양, 두께 등을 도안하였다. 결과: 돼지 대동맥과 폐동맥 판막의 수치 및 각 구조물들의 배율을 구하였다. 대동맥 판막 및 폐동맥 판막은 각각 우측 관상동맥 첨판 및 우향 관상동맥 첨판의 크기가 상대적으로 제일 컸다. 무접합 관상동맥 첨판이 가장 작았으며(우향 관상동맥 첨판: 무접합 관상동맥 첨판: 좌향 관상동맥 첨판=1.00 : 0.88 : 1.00), 판막의 높이는 첨판의 크기에 비례하였다. 스텐트를 사용한 돼지 대동맥 판막은 외측 직경 크기를 19 mm부터 33 mm까지 정하였고, 이전에 정한 비율 및 스텐트 두께, 그리고 실제 판막 제작에 필요한 봉합 두께와 판막 주변 벽의 두께 등을 고려하여 스텐트의 높이 ($R{\times}1.4$, R: radius, 직경), 폭, 간격 수치를 정하였으며, 심낭을 이용한 판엽은 판막의 직경, 각 첨판 길이 ($2{\times}R$), 높이 ($R{\times}1.4$)와 첨판의 최소 접합면을 고려하여 스텐트 크기와 함께 모양을 도안하였다. 결론: 25개의 돼지 대동맥 판막과 폐동맥 판막의 구조물에 대한 수치를 측정하여 판막들의 기하학적 비율을 계산하고, 여려 크기의 이식형 돼지 대동맥 판막을 만들어 볼 수 있었으며, 또한 소의 심낭을 이용하여 다양한 표기의 판막을 만들어 볼 수 있는 이론적 근거를 만들 수 있었다. 향후 이러한 도안의 판막 제작 후 충분한 생체 내외의 실험 및 추가 연구가 필요할 것으로 생각된다.

저지방 고품질 돈육 생산을 위한 돼지 복강 및 피하지방 항체 개발 (Development of Polyclonal Antibodies to Abdominal and Subcutaneous Adipocyte for Producing Fat-reduced High Quality Pork)

  • 본 연구는 저지방 고품질 돈육의 생산을 위해 돼지 복강 및 피하지방 감소를 위한 다클론 항체의 개발 및 실제 세포에 미치는 세포독성효과를 확인하고자 실시되었다. 항원으로써 돼지의 복강 및 피하지방세포 PMP를 분리하여 면양에 3회에 걸쳐 수동면역 주사하고, 면역 주사 전 및 후에 비면역혈청과 항체를 생산하였다. 돼지 AAb 및 SAb 항체의 역가와 생체 주요 장기 조직인 심장, 신장, 간장, 폐, 근육 및 비장세포의 PMP에 대한 타장기 교차반응성을 조사하였다. 돼지 부위별 지방 조직에서 지방세포를 분리하고 각각 primary 배양시킨 후 개발된 AAb 및 SAb를 주입한 뒤 media 내 LDH 방출 수준을 조사하였다. 희석배율 1:1,000배를 기준으로 비면역혈청은 항원-항체 결합반응이 거의 측정되지 않았고, AAb 및 SAb는 희석배율 1:16,000배까지 각각 항원-항체 반응이 감지되었으며, 이는 본 연구에서 생산한 부위별 지방 감소 다클론 항체가 지방세포 PMP에 대해 강한 역가를 가진 항체임을 시사한다. 또한 AAb 및 SAb는 타 장기들과는 특이한 반응을 나타내지 않았다. 본 연구에서 개발한 두 항체들은 모두 항원으로 이용된 부위의 지방세포 PMP와 가장 높은 반응을 나타내었으며, 비면역혈청에 비해 유의적으로 높은 세포독성 효과가 있음을 확인할 수 있었다. 이상의 결과를 종합할 때 본 연구에서 개발된 AAb 및 SAb는 높은 역가, 타 장기 안전성 및 in vitro 지방 감소 효과가 있었으며 향후 영양생리 안전성 및 생체 지방 감소 효과 등의 추가 연구가 지속될 경우 저지방 고품질 돈육의 생산이 가능하리라고 판단된다.

돼지-염소 이종이식모델에서 냉동 및 무세포화 혈관이식편의 조직학적 비교분석 (Histological Comparison of Vascular Grafts in a Pig to Goat Xenotransplantation Model)

  • 배경: 현재까지 개발된 인조혈관들은 소구경 혈관에서는 개통성을 유지하기 어려워 사용에 제한이 따른다. 이에 저자는 이종혈관이식편이 소구경 혈관을 대신할 수 있는지 시험하고자 돼지의 혈관을 채취하여 냉동보관과 무세포화의 두 가지 방법으로 전처치한 뒤 이를 염소에게 이식하고 일정기간 동안 그 변화를 비교 분석하였다. 대상 및 방법: 돼지의 양측 경동맥을 적출한 뒤 하나는 바로 조직 보관액에 담아 냉동 보관하였고 하나는 NaCl-SDS 용액을 이용하여 무세포화(acellularization)한 뒤 냉동 보관하였다. 동일한 염소의 양측 경동맥에 냉동보관만 했던 이식편과 무세포화한 이식편을 각각 삽입하였다. 3마리의 염소에게 이를 시행하였고 술 후 각각 1, 3, 6개월째에 이식편을 적출하였다. 관찰기간 동안 주기적으로 초음파검사를 시행하여 개통성 여부를 확인하였으며, 이식 편의 적출 후에는 육안소견 및 광학현미경 소견을 비교 분석하였다. 결과: 3마리 실험동물 모두 혈전색전증과 같은 별다른 문제없이 예정된 적출시기까지 생존하였다. 초음파검사상 모든 혈관이식편에서 관찰기간 동안 우수한 개통성을 보였다. 육안소견상 이식편의 내강에서는 혈전의 생성없이 매끈한 표면을 관찰할 수 있었다. 현미경 검사상 6개월째의 혈관이식편에서 세포가 재구성되고 있는 것을 확인할 수 있었다. 이식 후 초기였을 때 무세포화이식편보다 단순 냉동이식편에서 염증반응이 활발한 것으로 보였으나 유의한 거부반응의 증거는 관찰되지 않았다. 결론: 돼지-염소간의 이종이식모델에서 단순 냉동혈관이식편과 무세포화혈관이식편 사이에 이식 후 6개월까지는 임상적인 차이를 야기하지 않았다. 이는 동일한 모델의 혈관이식에 있어 무세포화의 과정을 생략할 수 있다는 가능성 때문에 고무적인 결과라 할 수 있으나, 관찰기간이 비교적 짧고, 실험동물의 수가 많지 않아 향후 추가적인 연구가 뒷받침되어야 하리라 생각된다.

미니돼지에서 99mTc-Tin colloid와 99mTc-DISIDA를 사용한 간신티그라피의 비교 연구 (Comparison of 99mTc-Tin colloid colloid and 99mTc-DISIDA Hepatoscintigraphy in Miniature Pigs)

  • 동물 모델에서의 간기능의 비침습적 평가는 하나의 과제로 남아있다. 간신티그라피는 간의 크기 및 모양의 변화에 대한 정보를 제공할 뿐 아니라 간의 전체적인 기능을 파악할 수 있게 해 주며 간이식의 주요 합병증인 간염, 거부반응 및 담도계 합병증의 진단에도 편리하게 사용될 수 있다. 이에 본 연구에서는 미니돼지의 간 기능을 평가하기 위해 사람에서 주로 사용되는 방사성의약품인 $^{99m}Tc-Tin$ colloid와 $^{99m}Tc-DISIDA$를 이용하여 간 및 간담도 신티그라피를 시행하였다. 그 결과, 사람의 간신티그라피와 달리 미니돼지에서는 $^{99m}Tc-Tin$ colloid가 폐, 간, 위벽 및 신장에 섭취되고 폐와 간의 영상이 겹쳐져 간의 관심영역을 설정할 수 없었다. 그러나 $^{99m}Tc-DISIDA$를 사용한 간담도 신티그라피에서는 심장, 폐, 간, 담낭 및 십이지장으로의 연속적 방사능섭취를 관찰할 수 있었다. 따라서 미니돼지에서의 $^{99m}Tc-Tin$ colloid의 사용은 비특이적 결합력으로 인해 간 기능 평가를 위한 영상에는 적합하지 않으나 $^{99m}Tc-DISIDA$는 간담도 기능을 파악하는데 있어 좋은 수단이 될 수 있을 것으로 생각된다.

  • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70$\pm$1.32mmHg/min)compared to CF dialyzers(4.32$\pm$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The CF 15-11 HFDs which failed APD test did not show changes in the UFR compared to normal dialyzers indicating that APD is a more sensitive test than UFR test to evaluate the integrity of the fibers. 30527 T00401030527 ^x For quantitative measurement of reflected light from a clinical diagnostic strip, a prototype old reflectance photometer was designed. The strip loader and cassette were made to obtain more accurate reflectance parameters. The strip was illuminated at 45˚c through optical fiber and the intensity of reflected light was determined at rectanguLat angle using a photodiode. The kubelka-munk coefficient and reflection optical density were determined ar four different wavelengths(500, 550, 570 and 610nm) for blood glucose strip. For higher concentration than 300mg/41 about glucose, a saturation state of abforbance was observed at 500, 550 and 570nm. The mice were divided into two groups. One group was injected with Photofrin II, intravenously (2.5mg/kg body weight). The other group received no Photofrin II. Mice from both groups were irradialed for approximately 15 minutes at 100, 300, or 500 mW/cm2 with the argon (488nm/514.5 nm), dye(628nm) and gold vapor (pulsed 628 nm) laser light. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. High Resolution HC$_3$N Observations toward the Central Region of Sagittarius B2

  • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70${\pm}$1.32mmHg/min)compared to CF dialyzers(4.32${\pm}$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The CF 15-11 HFDs which failed APD test did not show changes in the UFR compared to normal dialyzers indicating that APD is a more sensitive test than UFR test to evaluate the integrity of the fibers. 30527 T00401030527 ^x For quantitative measurement of reflected light from a clinical diagnostic strip, a prototype old reflectance photometer was designed. Stimulus patterns of our stimulator were designed to simulate electromyographic activity monitored during locomotion of normal subjects. Muscle types of the lower extremity were classified according to their mechanical property of contraction, which are slow muscle (msoleus m.) and fast muscle (medial gastrocneminus m., rectus femoris m., vastus lateralis m.). Optimal parameters of electrical stimulation for slow muscles were 20 Hz, 0.2 ms square pulse. For fast muscle, 40 Hz, 0.3 ms square pulse was optimal to produce repeated contraction.
