Physiologically-based pharmacokinetic predictions of intestinal BCRP-mediated drug interactions of rosuvastatin in Koreans

  • It was recently reported that the $C_{max}$ and AUC of rosuvastatin increases when it is coadministered with telmisartan and cyclosporine. Rosuvastatin is known to be a substrate of OATP1B1, OATP1B3, NTCP, and BCRP transporters. The aim of this study was to explore the mechanism of the interactions between rosuvastatin and two perpetrators, telmisartan and cyclosporine. Published (cyclosporine) or newly developed (telmisartan) PBPK models were used to this end. The rosuvastatin model in Simcyp (version 15)'s drug library was modified to reflect racial differences in rosuvastatin exposure. In the telmisartan-rosuvastatin case, simulated rosuvastatin $C_{maxI}/C_{max}$ and $AUC_I/AUC$ (with/without telmisartan) ratios were 1.92 and 1.14, respectively, and the $T_{max}$ changed from 3.35 h to 1.40 h with coadministration of telmisartan, which were consistent with the aforementioned report ($C_{maxI}/C_{max}$: 2.01, $AUC_I/AUC$:1.18, $T_{max}:5h{\rightarrow}0.75h$). In the next case of cyclosporine-rosuvastatin, the simulated rosuvastatin $C_{maxI}/C_{max}$ and $AUC_I/AUC$ (with/without cyclosporine) ratios were 3.29 and 1.30, respectively. The decrease in the $CL_{int,BCRP,intestine}$ of rosuvastatin by telmisartan and cyclosporine in the PBPK model was pivotal to reproducing this finding in Simcyp. Our PBPK model demonstrated that the major causes of increase in rosuvastatin exposure are mediated by intestinal BCRP (rosuvastatin-telmisartan interaction) or by both of BCRP and OATP1B1/3 (rosuvastatin-cyclosporine interaction).

국내산 황색 방울토마토의 항산화활성 및 암세포 생육억제 효과 (Antioxidant and Anticarcinogenic Effects of Domestic Yellow Cherry Tomato)

  • 본 연구에서는 황색 방울토마토의 생리활성 식품소재로서의 가능성을 알아보기 위하여 추출 건조물의 폴리페놀 및 플라보노이드 함량, 항산화 효과 및 암세포에 대한 생육억제 효과를 검증하였다. 폴리페놀 및 플라보노이드 함량은 각각 10.96±1.57 및 4.12±0.41 mg/g이었다. 항산화 활성은 DPPH 및 ABTS 라디칼 소거능을 측정하여 확인하였고 free radical을 50% 감소시키는 농도인 RC50은 각각 490.83±17.35 ㎍/mL과 355.90±0.79 ㎍/mL이었다. 추출 건조물은 정상 간세포(Chang)에 대해서는 세포 독성을 보이지 않았고 A549 폐암세포에 대해서는 생육 억제 활성을 나타내지 않았으나 자궁경부암세포(HeLa)와 간암세포(HepG2)에 대해서는 100 ㎍/mL 농도로 처리하였을 때 각각 15.2% 및 18.4%의 생육 억제 효과를 보였다. 본 연구의 결과로부터 황색 방울토마토의 항산화 활성과 자궁경부암세포와 간암세포 등 일부 암세포에 대한 억제 활성이 검증되어 생리활성 식품소재로서 가능성을 확인하였다.

Plasma, tissue and urinary levels of aloin in rats after the administration of pure aloin

  • Aloin is a physiologically active anthraquinone present in aloe. There are two isomers of aloin, aloin A and aloin B, occurring as a mixture of diastereomers. The objective of this study was to determine the bioavailability and tissue distribution of aloin. Rats were gavaged with 11.8g/kg aloin, and the levels of aloin and its conjugates were measured in plasma, tissues, and urine. Plasma aloin level showed a peak at 1hr after the administration and the concentration was $59.07{\pm}10.5\;ng/ml$. The 24 h cumulated urinary aloin was 0.03% of the initial dose. These results suggest that aloin is absorbed and reaches a peak plasma level within 1-1.5 h after the administration and a significant portion is possibly metabolized or is excreted in feces. These results can apply to the determination of the adequate intake level of aloe and aloe products to achieve the desired biological effect, and to interprete in vitro study results.

  • Functionally and physiologically active peptides are produced from several food proteins during gastrointestinal digestion and fermentation of food materials with lactic acid bacteria. Once bioactive peptides (BPs) are liberated, they exhibit a wide variety of physiological functions in the human body such as gastrointestinal, cardiovascular, immune, endocrine, and nervous systems. These functionalities of the peptides in human health and physiology include antihypertensive, antimicrobial, antioxidative, antithrombotic, opioid, anti-appetizing, immunomodulatory and mineral-binding activities.

Analysis of reactive species in water activated by plasma and application to seed germination

  • The use of plasma has increased in bio-application field in recent years. Particularly, water treated by arc discharge or atmospheric pressure plasma has been actively utilized in bio-industry. In this study, we have developed a plasma activated water generating system. For this system, two kinds of plasma sources; dielectric barrier discharge (DBD) plasma and arc discharge plasma have been used. The discharge energy was calculated using the breakdown voltage and current, and the emission spectrum was measured to investigate the generated reactive species. We also analyzed the amount of reactive oxygen and nitrogen species in water using the chemical methods and nitric oxide sensor. Finally, the influence of plasma generated reactive species on the germination and growth of spinach (Spinacia oleracea) was investigated. Spinach is a green leafy vegetable that contains a large amount of various physiologically active organic compounds. However, it is characterized with a low seed germination rate.

  • Orthostatic stress in human cardiovascular system following spaceflight remains a critical problem in the current lifercience space program. The study presented in this paper is part of an ongoing effort to use mathematical models to investigate the effects of gravitational stresses on the cardiovascular system of normals and microgravity adapted individuals. We employ a twelve compartment lumped parameter representation of the hemodynamic system coupled to set-point models of the arterial baroreflex and the cardiopulmonary reflex to investigate the transient response of heart rate to orthostatic stress. We simulate current hypotheses concerning the mechanisms underlying postspaceflight orthostatic intolerance over a range of physiologically reasonable values and compare the simulations to astronaut stand-test data pre-and postflight.

Effects of Medicinal Plant Extracts on Antioxidant System in Ethanol-intoxicated Rats

  • Four medicinal plants selected from preliminary screening study were evaluated in the aspects of their antioxidant activities in alcohol-intoxicated rats. Rats fed 1% ${\alpha}-tocopherol-supplemented$ diet as positive control and ones done ${\alpha}-tocopherol-deficient$ diet as negative control were compared with ones done the plant extract-supplemented diet (n=8). After the administration of the experimental diets for 4 weeks, typical increments in activities of manganese-superoxide dismutase (Mn-SOD) and glutathione peroxidase (GSH-px) indicated in alcohol-intoxicated rats, were not observed in ones fed Lagerstroemia and Ulmus extract-supplemented diet. The content of thiobarbituric acid reactive substance (TBARS), the product of lipid peroxidation, did not increased in rats fed plant extracts-supplemented diet except for Terminalia. From the results, it is concluded that Lagerstroemia and Ulmus have physiologically efficient antioxidant activities.

정상 좌심실을 가진 대동맥 판막 폐쇄증에서의 일차성 완전교정술 (Single Stage Anatomic Repair of Aortic Atresia with Normal Left Ventricle)

  • Four to seven percent of infants born with aortic atresia have a normal-sized left ventricle in association with a ventricular septal defect and a normal mitral valve. In contrast to the more common group of infants with aortic atresia whose left ventricle is hypoplastic, this important subgroup has potential for complete operative correction involving both the right and left ventricle. Our approach was to perform complete repair at one stage. The patient who underwent single-stage repair was discharged from the hospital in good condition. Achieving a physiologically normal circulation simplifies the postoperative management of this condition.

  • Biological monitoring, analyses of internal dose for exposure to toxicants, has been thought as one of the belt approaches for risk assessment. As the amount detected in human samples is generally very low, typically in the parts-per-bilion (ppb) or parts-per-trillion (ppt) range, analytic technologies such at HPLC, GC/MS, LC/MS, and LC/MS/MS have been continuously developed. In addition, route specific and sensitive exposure biomarkers have been developed for proper biological monitoring. PBPK modeling, particularly reverse dosimetry, has been emphasized as an useful method via interpretation of biological monitoring results for regulation of toxicants. Thus, this review is focused on the use of PBPK dosimetry models for toxicology research and risk assessment in Korea.

경혈점과 유발점에 의한 근막통증후군의 임상적용에 대한 연구 (Study of Clinic application of Myofascial Pain Syndrome with Acupucture and Trigger Point)

  • Myofascial pain syndrome is one of the major cause of chronic pain and trigger point injection, stretching, spray and electrical therapy are often used in clinical situation for treatment of myofascial pain syndrome. Myofascial pain syndrome is characterzied by the existence of a hypersensitive region, called the trigger point in a muscle or in the connective tissue, together with palpable noble, stiffness, limitation of motion and referred pain when trigger point is stimulated. Physiologically, they represent a self-sustaining vicious cycle of pain-spasm-pain. The purpose of this study is to illustrate mechanisms of pain by stimulation of acupuncture and trigger point, to introduce clinic application of orient and western stimulative point (acupuncture, trigger point)for treatment of MPS(myofascial pain syndrome), to make physiotherapist use both stimulative points for treatment of MPS.
