Analysis of algal spatial distribution characteristics using hyperspectral images and machine learning in upstream reach of Baekje weir (초분광영상과 머신러닝을 이용한 백제보 상류구간 조류 공간분포 특성분석)

  • 부영양화된 호수나 유속이 느린 하천에서 발생하는 녹조의 과도한 발생은 하천 생태계 훼손, 동식물의 건강, 담수의 오염 등 환경 사회 경제적으로 큰 피해를 준다. 현재 수질 측정망은 정해진 지점에서 Chlorophyll-a(Chl-a), Phycocyanin(PC)을 대표농도로 산정하고 조류경보에 활용하고 있으나, 일주일에 한번씩 샘플링을 통해 Chl-a 및 PC를 측정하여 시공간적인 신뢰성의 문제가 제기될 수 있다. 본 연구에서는 기존 점단위 조류 모니터링의 한계점을 개선하기 위해 초분광영상 자료를 머신러닝 기법에 적용하여 Chl-a 및 PC 산정 알고리즘을 개발하였다. 이를 위해 Chl-a와 PC의 최대 흡수, 반사 파장대, 주요 물 흡수 파장대 자료를 조합하여 9개의 파장비를 구축하였으며, 기존 연구에서 활용한 머신러닝 기법인 Partial Least Square, Random Forest, Gradient Boosting, Support Vector Machine, K-Nearest Neighbor, Artificial Neural Network를 검토하여 최적 모델을 선정하였다. 학습된 머신러닝의 성능을 R2, NSE, RMSE 목적함수를 이용해 평가하였으며, 그 결과 ANN이 각각 PC 0.801, 0.755, 11.774 mg/m3, Chl-a 0.733, 0.622, 8.736 mg/m3로 가장 우수한 성능을 보였다. 최적화 된 ANN 모델을 백제보 상류 2016-2017년 항공 초분광영상에 적용하여 시공간에 따른 조류 분포변화를 평가하고자 한다.

Influence of Rainfall on Cyanobacterial Bloom in Daechung Reservoir

  • The water quality and algal communities in the Daechung Reservoir, Korea, were monitored from summer to autumn in 1999 and 2001. Although the average weekly precipitations during June and July were very similar in 1999 and 2001, they were much different during August and September, the so-called blooming season. The rainfall in 1999 increased about 70% after late August, whereas it decreased to the one-fifth level in 2001. The higher concentrations of chlorophyll- a, phycocyanin, and cyanobacteria were observed in 2001, which resulted in the dense algal bloom. In addition, in 2001, the cyanobacterial percentage remained above 80% during the investigation period, and the cyanobacteria were exclusively composed of Microcystis spp. Conversely, there was no report on the algal bloom in 1999. However, the peak bloom seasons were the same for both years, from late August to early September, irrespective of the amount of precipitation. These results suggest that the magnitude and duration of rainfall before bloom season are important factors determining the extent of cyanobacterial bloom in this system.

  • Recently, it has been shown that quantum coherence appears in energy transfers of various photosynthetic lightharvesting complexes at from cryogenic to even room temperatures. Because the photosynthetic systems are inherently complex, these findings have subsequently interested many researchers in the field of both experiment and theory. From the theoretical part, simplified dynamics or semiclassical approaches have been widely used. In these approaches, the quantum-classical Liouville equation (QCLE) is the fundamental starting point. Toward the semiclassical scheme, approximations are needed to simplify the equations of motion of various degrees of freedom. Here, we have adopted the Poisson bracket mapping equation (PBME) as an approximate form of QCLE and applied it to find the time evolution of the excitation in a photosynthetic complex from marine algae. The benefit of using PBME is its similarity to conventional Hamiltonian dynamics. Through this, we confirmed the coherent population transfer behaviors in short time domain as previously reported with a more accurate but more time-consuming iterative linearized density matrix approach. However, we find that the site populations do not behave according to the Boltzmann law in the long time limit. We also test the effect of adding spurious high frequency vibrations to the spectral density of the bath, and find that their existence does not alter the dynamics to any significant extent as long as the associated reorganization energy is changed not too drastically. This suggests that adopting classical trajectory based ensembles in semiclassical simulations should not influence the coherence dynamics in any practical manner, even though the classical trajectories often yield spurious high frequency vibrational features in the spectral density.

Quality Characteristics of Pan Bread with Spirulina Powder (스피루리나를 첨가한 식빵의 품질 특성)

  • This study was conducted to investigate the quality characteristics of bread prepared with spirulina powder (0, 0.4, 0.8, and 1.2%). The volume and specific volume of bread decreased with increasing spirulina powder content. The pH of the bread with 1.2% spirulina powder was the highest. Content of phycocyanin increased with addition of spirulina powder. For color values, increasing spirulina powder content resulted in decreased lightness, redness, and yellowness in the crust. Amylogram showed that spirulina addition increased initial gelatinization temperature, decreased breakdown, and lowered the setback and consistency, which prevented retrogradation. Moreover, the hardness of spirulina bread decreased as spirulina addition increased. Further, springiness, cohesiveness, gumminess, chewiness. and resilience of bread with 0.8% spirulina powder were the lowest. Sensory results showed that the scores of overall preference and buying intention in the bread with 0.8% spirulina were the highest among the control and other groups.

Geosmin and Morphological Characteristics of Anabaena circinalis, Obtained from the Bukhan River (북한강에서 출현한 Anabaena circinalis의 형태학적 특성 및 지오스민(geosmin) 발생 양상)

  • This study was carried out in the Bukhan River in the summer of 2014 and 2015, to identify the relationship between geosmin and the morphological changes in Anabaena. Identification of Anabaena was conducted using morphological and molecular analyses. Anabaena in this study was similar to Anabaena circinalis, A. crass, and A. spiroides with regard to regular coils, vegetative cell, akinete shape, and size, hoever, it was distinguishabl from A. crass and A. spiroides because of its larger trichome coil size. Additionally, the sequences of phycocyanin (PC) gene from Anabaena showed a 99% genetic similarity with A. circinalis NIES-1647 strain. The coil diameter of trichome ranged from 106 to $899{\mu}m$, and the diameter and abundance showed an insignificant positive correlation (r=0.544, p<0.05). The result of relationship between the coil diameter and the cell number per 360-degree rotation was kept at $33.8{\pm}5.2$ cells per $100{\mu}m$ diameter despite variable diameter. The average geosmin concentrations in 2014 and 2015 were investigated to be 99 ng/L and 35 ng/L, respectively. A. circinalis cell density contributed considerably to the change in geosmin and was positively correlated with geosmin concentration (2014; r=0.599, p<0.01, 2015; r=0.559, p<0.01). Our results suggest that geosmin and coil diameter could be estimated with the help of cell density.

Quality characteristics and antioxidant activities of rice crispy cereal added with dried laver (마른 김 첨가 크리스피 쌀 과자의 이화학적 품질특성 및 항산화활성)

  • This study was performed to evaluate the quality characteristics and antioxidant activities of rice crispy cereal supplemented with dried laver powder. Reducing sugar content decreased in the dried laver-supplemented group. According to the Hunter color system, the L, a, and b values of rice crispy cereal decreased with an increase in the amount of dried laver. The LP5 (rice crispy cereal added with 5% dried laver) group showed the highest phycocyanin and chlorophyll contents, which tended to increase with increased amounts of added dried laver. Total phenolic and flavonoid contents increased with the amount of dried laver, as did 2,2-diphenyl-1-picrylhydrazyl (DPPH) and hydroxyl radical-scavenging activities. Intensity test results showed that the color, flavor, and taste of rice crispy cereal improved with the addition of dried laver. The LP5 group scored the highest on overall satisfaction. These results suggest that the addition of dried laver to rice crispy cereal improves sensory properties by increasing antioxidant activities.

Chronic Toxicity of the Triazole Fungicide Tebuconazole on a Heterocystous, Nitrogen-Fixing Rice Paddy Field Cyanobacterium, Westiellopsis prolifica Janet

  • This study explored the chronic effects of different doses of the triazole fungicide tebuconazole on the growth, and metabolic and enzymatic functions of the filamentous paddy field cyanobacterium, Westiellopsis prolifica Janet. The growth of the cyanobacterium was determined by an estimation of the change in pigment contents. Chlorophyll-a, carotenoids, and accessory pigments such as phycocyanin, allophycocyanin, and phycoerythrin were shown to decline over a 16-day period by a factor of 92%, 93%, 83%, 95%, and 100%, respectively, with increasing doses of the fungicide. Metabolic and enzymatic activities were also adversely affected. Over the 16 days, a gradual rise in total phenol content was recorded when Westiellopsis prolifica Janet was treated with 60 ppm of the fungicide, despite the reduction in carbohydrates, proteins, and amino acids by 96%, 92%, and 90%, respectively. Moreover, the enzymes nitrate reductase (NR), glutamine synthetase (GS), and succinate dehydrogenase (SDH) also registered reductions of 93%, 90%, and 98%, respectively. This study indicates that tebuconazole, although an important fungicide used extensively in rice fields, exhibits an inhibitory effect on the growth and metabolic activities of Westiellopsis prolifica Janet and hence possibly on other varieties as well.

Seasonal and Spatial Diversity of Picocyanobacteria Community in the Great Mazurian Lakes Derived from DGGE Analyses of 16S rDNA and cpcBA-IGS Markers

  • The seasonal and spatial diversity of picocyanobacteria (Pcy) in lakes of the Great Mazurian Lakes (GLM) system was examined by DGGE analysis of molecular markers derived from the 16S-23S internal transcribed spacer (ITS) of the ribosomal operon and the phycocyanin operon (cpcBA-IGS). The study of nine lakes, ranging from mesotrophy to hypereutrophy, demonstrated seasonal variance of Pcy. The richness and Shannon diversity index calculated on the basis of both markers were higher in spring and lower in early and late summer. No statistically significant relationships were found between the markers and trophic status of the studied lakes or Pcy abundance. There were, however, statistically significant relationships between the diversity indices and sampling time. The analysis pointed to a different distribution of the two markers. The ITS marker exhibited more unique sequences in time and space, whereas a greater role for common and ubiquitous sequences was indicated by the cpcBA-IGS data. Examination of the Pcy community structure demonstrated that communities were grouped in highly similar clusters according to sampling season/time rather than to the trophic status of the lake. Our results suggest that time is more important than trophic status in shaping the diversity and structure of Pcy communities. The seasonal changes in picocyanobacteria and differences in diversity and community structures are discussed in the context of well-established ecological hypotheses: the PEG model, intermediate disturbance hypothesis (IDH), and horizontal gene transfer (HGT).

Metaproteomic analysis of harmful algal bloom in the Daechung reservoir, Korea

  • The present study aimed to analyze the metaproteome of the microbial community comprising harmful algal bloom (HAB) in the Daechung reservoir, Korea. HAB samples located at GPS coordinates of 36°29'N latitude and 127°28'E longitude were harvested in October 2013. Microscopic observation of the HAB samples revealed red signals that were presumably caused by the autofluorescence of chlorophyll and phycocyanin in viable cyanobacteria. Metaproteomic analysis was performed by a gelbased shotgun proteomic method. Protein identification was conducted through a two-step analysis including a forward search strategy (FSS) (random search with the National Center for Biotechnology Information (NCBI), Cyanobase, and Phytozome), and a subsequent reverse search strategy (RSS) (additional Cyanobase search with a decoy database). The total number of proteins identified by the two-step analysis (FSS and RSS) was 1.8-fold higher than that by one-step analysis (FSS only). A total of 194 proteins were assigned to 12 cyanobacterial species (99 mol%) and one green algae species (1 mol%). Among the species identified, the toxic microcystin-producing Microcystis aeruginosa NIES-843 (62.3%) species was the most dominant. The largest functional category was proteins belonging to the energy category (39%), followed by metabolism (15%), and translation (12%). This study will be a good reference for monitoring ecological variations at the meta-protein level of aquatic microalgae for understanding HAB.

Quality Characteristics of Spirulina Tofu Prepared with Different Coagulants (응고제에 따른 스피루리나 첨가 두부의 품질 특성)

  • Quality characteristics and antioxidant activity of spirulina with added tofu were investigated in the presence of different coagulants ($CaSO_4$, $MgCl_2$, Glucono delta lactone (GDL), vinegar). The yield of the spirulina tofu was the highest when coagulated with GDL. The pH of spirulina tofu with $MgCl_2$ was the highest compared to those of other coagulants. The tumidity of spirulina tofu was the highest with $MgCl_2$. In the color analysis of spirulina tofu, the L (lightness) value was the highest with $CaSO_4$, whereas a (redness) value was the highest with $MgCl_2$. C-phycocyanin contents were the highest with the $CaSO_4$. Phycoerythrin of spirulina tofu for vinegar was 0.194 mg/mL, and GDL content was 0.214 mg/mL. Additionally, the antioxidant activity of spirulina tofu was the highest with $MgCl_2$ $IC_{50}$ values of the DPPH radical scavenging activity of spirulina tofu with $MgCl_2$ was 89.2 mg/g, GDL 93.5 mg/mL, $CaSO_4$ 116.7 mg/mL, and vinegar 118.0 mg/mL. The texture analyses (TPA) results showed that hardness was in the order of vinegar>$MgCl_2$>GDL>$CaSO_4$. For the overall acceptability of the preference test, the spirulina tofu that was made using $MgCl_2$ coagulant had the highest score. Based on these results, it is suggested that $MgCl_2$ is the proper coagulant for the preparation of spirulina with added tofu.
