Selecting Appropriate Seedling Age for Restoration Using Comparative Analysis of Physiological Characteristics by Age in Abies koreana Wilson

  • The aim of this study was to investigate the sensitivity to environmental stress, and changes in the photosynthesis capacity in Abies koreana seedlings by age and to suggest the most effective age for restoration. To identify these physiological characteristics of A. koreana, the chlorophyll fluorescence and photosynthetic capacity of 1-, 2-, 3-, 5- and 6-year-old A. koreana seedlings were observed from June 2020 to June 2021. The maximum quantum efficiency of Photosystem II (Fv/Fm), a chlorophyll fluorescence measurement parameter, was strongly positively correlated with the monthly average temperature (1-year-old seedling: r=0.8779, 2-year-old seedling: r=0.8605, 3-year-old seedling: r=0.8697, 5-year-old seedlings: r=0.8085, and 6-year-old seedlings: r=0.8316). The Fv/Fm values were the lowest in winter (November 2020-March 2021). In addition, the Fv/Fm values of 1-, 2-, and 3-year-old seedlings in winter were lower than that of 5- and 6-year-old seedlings, while the Fv/Fm values in summer were relatively higher than those in winter. Further, the Fv/Fm values of seedlings of all ages decreased in August 2020, when the monthly average temperature was the highest. In particular, 1-year-old to 3-year-old seedlings showed Fv/Fm values less than 0.8. Further, the photosynthetic capacity measured in August 2020 increased with increasing seedling age. The analysis of variance results for summer Fv/Fm values showed significant differences in age-specific averages (p<0.05), and Duncan's multiple range test showed significant differences between 5- and 6-year-old seedlings and 1-, 2-, and 3-year-old seedlings (p<0.05). These results suggested that the 5- and 6-year-old seedlings were less sensitive to environmental stress and showed better photosynthetic capacity than the 1-, 2-, and 3-year-old seedlings. Therefore, 5-year-old or older A. koreana seedlings can be used as restoration materials because they can show increased adaptability and stable growth during transplantation due to their relatively high environmental resistance and photosynthetic capacity.

  • Five different types of catalases from photosynthetic bacterium Rhodospirillum rubrum S1 grown aerobically in the dark were found in this study, and designated Catl (350 kDa), Cat2 (323 kDa), Cat3 (266 kDa), Cat4 (246 kDa), and Cat5 (238 kDa). Analysis of native PAGE revealed that Cat2, Cat3, and Cat4 were also produced in the cells anaerobically grown in the light. It is notable that only Cat2 was expressed much more strongly in response to the anaerobic condition. Enzyme activity staining demonstrated that Cat3 and Cat4 had bifunctional catalase-peroxidase activities, while Catl, Cat2, and Cat5 were typical monofunctional catalases. S1 cells grown aerobically in the presence of malate as the sole source of carbon exhibited an apparent catalase Km value of 10 mM and a Vmax of about 705 U/mg protein at late stationary growth phase. The catalase activity of Sl cells grown in the anaerobic environment exhibited a much lower Vmax of about 109 U/mg protein at late logarithmic growth phase. The catalytic activity was stable in the broad range of temperatures (30$\^{C}$-60$\^{C}$), and pH (6.0-10.0). R. rubrum S1 was much more resistant to H$_2$O$_2$in the stationary growth phase than in the exponential growth phase regardless of growth conditions. Cells of stationary growth phase treated with 15 mM H$_2$O$_2$for 1 h showed 3-fold higher catalase activities than the untreated cells. In addition, L-glutamate induced an 80-fold increase in total catalase activity of R. rubrum S1 compared with magic acid. Through fraction analyses of S1 cells, Cat2, Cat3, Cat4 and Cat5 were found in both cytoplasm and periplasm, while Catl was localized only in the cytoplasm.

Effects of Hexaconazole on Growth and Antioxidant Potential of Cucumber Seedlings under UV-B Radiation

  • The present study was conducted to determine the effect of hexaconazole (HEX), a triazole fungicide, on the growth, yield, photosynthetic response and antioxidant potential in cucumber (Cucumis sativus L.) plants subjected to UV-B stress. UV-B radiation and HEX were applied separately or in combination to cucumber seedlings. The growth parameters were significantly reduced under UV-B treatment, however, this growth inhibition was less in HEX treated plants. HEX caused noticeable changes in plant morphology such as reduced shoot length and leaf area, and increased leaf thickness. HEX was quite persistent in inhibiting shoot growth by causing a reduction in shoot fresh and dry weight. HEX noticeably recovered the UV-B induced inhibition of biomass production. Significant accumutation in anthocyanin and flavonoid pigments in the leaves occurred as a result of HEX or UV-B treatments. HEX permitted the survival of more green leaf tissue preventing chlorophyll content reduction and higher quantum yield for photosystemII under UV-B exposure. HEX treatment induced a transient rise in ABA levels in the leaves, and combined application of HEX and UV-B showed a significant enhancement of ABA content which activates $H_2O_2$ generation. UV-B exposure induced accumulation of $H_2O_2$ in the leaves, while HEX prevented UV-B induced increase in $H_2O_2$, indicating that HEX serves as an antioxidant agent able to scavenge $H_2O$ to protect cells from oxidative damage. An increase in the ascorbic acid was observed in the HEX treated cucumber leaves affecting many enzyme activities by removing $H_2O_2$ during photosynthetic processes. The activities of antioxidant enzymes including catalase(CAT), ascorbate peroxidase(APX), superoxide dismutase(SOD) and peroxidase(POD) in the leaves in the presence of HEX under UV-B stress were higher than those under UV-B stress alone. These findings suggest that HEX may participate in the enhanced tolerance to oxidative stress. From these results it can be concluded that HEX moderately ameliolate the effect of UV-B stress in cucumber by improving the components of antioxidant defense system.

The Effects of Salt Stress on Photosynthetic Electron Transport and Thylakoid Membrane Proteins in the Cyanobacterium Spirulina platensis

  • The response of Spirulina (Arthrospira) platensis to high salt stress was investigated by incubating the cells in light of moderate intensity in the presence of 0.8 M NaCl. NaCl caused a decrease in photosystem II (PSII) mediated oxygen evolution activity and increase in photosystem I (PSI) activity and the amount of P700. Similarly maximal efficiency of PSII (Fv/Fm) and variable fluorescence (Fv/Fo) were also declined in salt-stressed cells. Western blot analysis reveal that the inhibition in PSII activity is due to a 40% loss of a thylakoid membrane protein, known as D1, which is located in PSII reaction center. NaCl treatment of cells also resulted in the alterations of other thylakoid membrane proteins: most prominently, a dramatic diminishment of the 47-kDa chlorophyll protein (CP) and 94-kDa protein, and accumulation of a 17-kDa protein band were observed in SDS-PAGE. The changes in 47-kDa and 94-kDa proteins lead to the decreased energy transfer from light harvesting antenna to PSII, which was accompanied by alterations in the chlorophyll fluorescence emission spectra of whole cells and isolated thylakoids. Therefore we conclude that salt stress has various effects on photosynthetic electron transport activities due to the marked alterations in the composition of thylakoid membrane proteins.

Photosynthetic, Morphological and Growing Characteristics by Shading Materials in Panax ginseng C. A. Meyer (차광재 종류가 묘삼의 광합성, 형태 및 생육에 미치는 영향)

  • This study was conducted to response the photosynthetic rate and anatomical characters in one year ginseng plant against the light transmission ratio (10%, 20% and 30% in natural light) under shading materials by polyethylene (PE) net. The chlorophyll content and the photosynthesis of ginseng according to the light transmissivity showed decreasing trend as the light transmission ratio increased. At the higher light transmission ratio condition, leaves showed the chlorosis due to the physiological disorder, stems formed hypodermis, and phloem development inside the vascular tissue shrunk remarkably. Also, roots showed the epidermis destruction and less cambium development at the higher light intensity. The root weight also decreased as the light transmission ratio increased.

Differential Response in Photosynthesis and Respiration of Rice Cultivars as Affected by Oxyfluorfen and Bensulfuron-methyl (Oxyfluorfen 및 Bensulfuron-Methyl 내성수도품종의 광합성 및 호흡저해 반응)

  • The study was objected to compare the differential responses in photosynthesis and resipiration by selected cultivars as tolerant or susceptible to oxyfluorfen and bensulfuron-methyl. Clark-typed oxygen elect rude system (Rank Brothers Co., Bottisham. UK.) was used to detect the releasing oxygen from the leaf disks. By increasing oxyfluorfen concentrations. both cultivars were disposed to decrease in photosynthetic O$_2$ generation and increase in respiratory O$_2$ generation. Comparing of both rice cultivars. cv. Mushakdanti (selected at 10$\^$-3/M as the susceptible) failed down in photosynthetic O$_2$ generation as lower as 43% of the control but cv. Aichiasahi to 55%, respectively. However. no significant difference in respiratory O$_2$ generation were detected between both cultivars. As for bensulfuron-methyl. almost same tendency was conferable with general responses on O$_2$ generation between both selected cultivars, namely cv. IR 1846 failed down in photosynthetic O$_2$ generation as lower as 55% of the control but cv. Chinsurah Boro II to 77%. respectively.

Effects of Light Enviroment on Photosynthetic Rate and Chlorophyll Contents of Three Broad-leaved Species Growing in the Forest (임분의 광환경이 활엽수 3수종의 광합성율 및 엽록소 함양에 미치는 영향)

  • To seek for proper regeneration methods with under-planting, Photosynthetic rate and chlorophyll contents of three broad-leaved species seedlings, Kalopanax pictum, Fraxinus rhynchophylla and Cornus controversa, were measured and compared between growing sites, below the canopy of Larix kaemferi and open land in the forest. Nursery stocks was planted in open nurs- ery in the forest in late April, 1997, and under-planting below the canopy of Larix kaemferi was conducted in late April, 1998. This experiment examined the relationships between shade-tolerance and physiological response to different light environment by tree species. To adapt the shade environment, leaves of Kalopanax pictum might be increased the more chlorophyll con- tents and photosynthetic ability than other species. From these results, shade-tolerance of the tree might be ordered Kalopanax pictum, Fraxinus rhynchophylla, and Cornus controversa.

Photosynthetic Responses of four Oak Species to Changes in Light Environment (광환경 변화에 대한 네 참나무 수종의 광합성 반응)

  • This study was conducted to investigate the photosynthetic responses of four Oak species (Quercus mongolica, Q. serrata, Q. acutissima and Q. variabilis) by shading treatment. We investigated light response curve, photosynthesis (A)-intercellular $CO_2$ concentration (Ci) curve, leaf growth and chlorophyll content at the level of 35, 55 and 75% shading treatments and under the full sunlight. In our results, Q. variabilis and Q. acutissima showed increased leaf growth, chlorophyll content and net apparent quantum yield but reduced chlorophyll a/b and carboxylation efficiency under the low light intensity. Therefore, light absorption and light utilization efficiency were improved under the low light intensity. Q. mongolica showed the similar responses that Q. variabilis and Q. acutissima showed, but net apparent quantum yield was reduced. The effects of shading treatment on Q. serrata were lower than those of other three species.

The Growth Effects on Interior Landscape Plants by Optical Fiber Lighting System (광섬유 조명체계가 실내조경식물의 생육에 미치는 효과)

  • This study was carried out to obtain fundamental information on the growth response of interior landscape plants under fluorescent light, sunlight and optical fiber lighting indoors. Saintpaulia ‘Delaware’, ‘Kalanchoe blossfeldiana, Anthurium scherzerianum and Ardisia crenata were examined using light intensity of 5001ux and 1,0001ux of fluorescent light, sunlight and optical fiber lighting in an interior environment. Results of experiments are as follows; 1) Plant growth status showed the best results under optical fiber lighting compared with fluorescent light or sunlight. 2) Plant growth status was better under 1,0001ux light intensity than 5001ux light intensity and in cases of the same light intensity, the highest growth increase was under optical fiber lighting. while it was showed relatively different according to the different plant species between a fluorescent light and sunlight. 3) The deep pinkish red color of Saintpaulia ‘Delaware’flower was obtained first under an optical fiber lighting and a fluorescent light, a sun light in that order. 4) Regarding interred activity, photosynthetic rate and transpiration rate, intercellular CO, water absorption rate showed a similar tendency generally in spite of a little difference. Namely, transpiration rate and intercellular CO, $CO_2$ a absorption rate increased according to increase of photosynthetic rate. 5) Photosynthetic rate of test plants except Anthurium scherzerianum increased according to increase of light intensity and increased highest under optical fiber lighting in the same light intensity condition. Increases differed under fluorescent light and sun light. That of Saineaulia ‘Delaware’and Anthurium scherzerianum increased in the order of optical fiber, fluorescent light and sun light, but that of Kalanchoe blossfeldiana and Ardisia pusilla increased in the order of optical fiber lighting, sun light and fluorescent light. Summing up these results, In visual value or internal health status of all experimental plants we obtained the highest result under an optical fiber lighting. Finally, we need to introduce an optical fiber lighting in interior landscape space as main light source.

Eco-physiological Responses of Roadside Tree Species to Contamination of Soil with Lead (토양 납 오염에 대한 가로수 식물종의 생리생태적 반응)

  • Heavy metal pollution in soil, such as lead contamination, has become an area of interest in Korea because of urbanization and atmospheric deposition from neighboring countries. Therefore, in this research, eco-physiological responses such as chlorophyll contents, antioxidant enzyme activity, photosynthetic rate, biomass and phytoaccumulation abilities were investigated for 4 commonly used native roadside tree species to suggest suitable tree species to cope with lead contamination. The target species, Ginkgo biloba, Prunus yedoensis, Zelkova serrata and Chionanthus retusus showed lead toxicity by significant changes of chlorophyll contents and antioxidant enzyme activities on treatments over 200 mg Pb/kg. However, biomass and photosynthetic rates only showed significant responses of plants in the highest level (5,000 mg/kg) treatment. Especially, G. biloba did not show any significant changes of antioxidant enzyme activity, photosynthetic rate, and biomass even in the highest level treatment. In low level - environmentally realistic treatments, G. biloba and P. yedoensis showed the highest phytoaccumulation rate of lead from soil. Selecting and planting species like G. biloba which have good phytoaccumulation abilities and resistance to lead contamination by further research will be required to deal with emerging lead contamination.
