Growth Response of Strawberry Seedlings to Application of Solid Chlorella (고형 클로렐라 시비에 대한 딸기유묘의 생육 반응)

  • A liquid-type chlorella (LC) produced by self-cultivation is used for strawberry cultivation by farmers in Korea. This study aimed to investigate seedling growth in two strawberry cultivars 'Soraya' and 'Haruhi' after applying solid-type chlorella (SC) in the greenhouse for 30 d. The treatments were as follows: Control, LC (0.2% of 1.0 × 107 cells/mL), ¼ SC (12.5 g/m2), ½ SC (25 g/m2), and SC (50 g/m2). Compared to the control, in the ½ SC treatment, dry weight of the seedlings increased at 30 days after treatment (DAT). Phosphorus content in the leaves of 'Soraya' seedlings of the SC treatment increased compared to that of the control. At 10 and 30 DAT, photosynthetic pigments including chlorophylls and carotenoids in the seedlings of both cultivars increased with the ½ SC treatment. These results indicate that the application of SC improved the growth of strawberry seedlings and could replace LC.

Impact of salt stress on the α-tocopherol, carotenoid derivatives and flocculation efficiency of Euglena sp., Indonesian Strain

  • Tocopherol, carotenoids, and chlorophyll are the primary components of the antioxidative response in microalgae. Conditions of stress, such salt stress, can trigger the processes responsible for the accumulation of tocopherol and carotene. It has been found that the most difficult part of culturing microalgae is keeping it affordable. This study investigated the effects of different salt types and concentrations on the amount of α-tocopherol, carotenoid derivatives, and flocculation efficiency of Euglena sp. Cultures of Euglena sp. was developed under salt stress conditions of NaCl 200 mM and KCl 200 mM. UV-VIS spectrophotometry was used to confirm the presence of α-tocopherol and carotenoid derivatives under thirteen days of salt stress testing. Increasing salinity has a significant effect on Euglena sp., causing spherical cell morphologies with aspect ratio 1.385 ± 0.031 for NaCl 200 mM and 1.414 ± 0.040 for KCl 200 mM. Increasing salinity also slowing down development with specific growth rate value of 0.171 ± 0.006 per day and 0.122 ± 0.029 per day for NaCl and KCl 200 mM, respectively. Nevertheless, the amount of α-tocopherol in Euglena sp. increases with a high salt concentration; algal cells flocculated more successfully when increasing the salt concentrations (NaCl 200 mM and KCl 200 mM) was added. Due to the inhibition of photosynthetic activity in salt-stressed cells, the control group exhibited higher levels of carotenoid derivatives (ranging from 0.5-1 ㎍/mL) and pheophytin a and b (0.0062 ± 0.001 ㎍/mL and 0.0064 ± 0.001 ㎍/mL) than the group treated with salt stress. In conclusion, salt stress was an effective way to raises the concentration of α-tocopherol and significantly reduce the expense of harvesting Euglena sp.

Growth, Photosynthesis and Chlorophyll Fluorescence of Chinese Cabbage in Response to High Temperature (고온 스트레스에 대한 배추의 생장과 광합성 및 엽록소형광 반응)

  • In order to gain insight into the physiological responses of plants to high temperature stress, the effects of temperature on Chinese cabbage (Brassica campestris subsp. napus var. pekinensis cv. Detong) were investigated through analyses of photosynthesis and chlorophyll fluorescence under 3 different temperatures in the temperature gradient tunnel. Growth (leaf length and number of leaves) during the rosette stage was greater at ambient $+4^{\circ}C$ and ambient $+7^{\circ}C$ temperatures than at ambient temperature. Photosynthetic $CO_2$ fixation rates of Chinese cabbage grown under the different temperatures did not differ significantly. However, dark respiration rate was significantly higher in the cabbage that developed under ambient temperature relative to elevated temperature. Furthermore, elevated growth temperature increased transpiration rate and stomatal conductance resulting in an overall decrease of water use efficiency. The chlorophyll a fluorescence transient was also considerably affected by high temperature stress; the fluorescence yield $F_J$, $F_I$, and $F_P$ decreased considerably at ambient $+4^{\circ}C$ and ambient $+7^{\circ}C$ temperatures, with induction of $F_K$ and decrease of $F_V/F_O$. The values of RC/CS, ABS/CS, TRo/CS, and ETo/CS decreased considerably, while DIo/CS increased with increased growth temperature. The symptoms of soft-rot disease were observed in the inner part of the cabbage heads after 7, 9, and/or 10 weeks of cultivation at ambient $+4^{\circ}C$ and ambient $+7^{\circ}C$ temperatures, but not in the cabbage heads growing at ambient temperature. These results show that Chinese cabbage could be negatively affected by high temperature under a future climate change scenario. Therefore, to maintain the high productivity and quality of Chinese cabbage, it may be necessary to develop new high temperature tolerant cultivars or to markedly improve cropping systems. In addition, it would be possible to use the non-invasive fluorescence parameters $F_O$, $F_V/F_M$, and $F_V/F_O$, as well as $F_K$, $M_O$, $S_M$, RC/CS, ETo/CS, $PI_{abs}$, and $SFI_{abs}$ (which were selected in this study), to quantitatively determine the physiological status of plants in response to high temperature stresses.

Influence of the Mesophyll on the Change of electrical Potential Difference of Guard Cells Induced by Red-light and CO2 in Commelina communis L. and Tradescantia virginiana L. (닭의장풀과 자주달개비에서 적색광과 이산화탄소에 의해 유도된 공변세포의 전위차 변화에 미치는 엽육세포의 영향)

  • The effects of light and $CO_2$ on the electrophysiological characteristics of guard cells in the intact leaf and in the detached epidermis have been investigated. Guard cells in intact leaves showed the membrane hyperpolarization in response to light. The biggest induced change of the membrane potential difference (PD) in the guard cells of the intact leaf was 13 m V by light and 42 mV by $CO_2$ in Commelina communis. Similar results were obtained with Tradescantia virginiana. However, there were no changes of membrane PD in detached epidermis. In order to determine the influence of the mesophyll on the changes of membrane PD, infiltration of the mesophyll cells with photosynthetic inhibitors was performed. In CCCP infiltrated leaf discs the guard cell membrane was depolarized slightly by red-light and hyperpolarized by $CO_2$, but in leaf discs infiltrated with DCCD and DCMU the guard cell membrane was hyperpolrized by both red-light and $CO_2$ as the control leaf discs. In azide infiltrated leaf discs the guard cell membrane showed no response to light and there was a much reduced membrane hyperpolarization by $CO_2$ compared to other responses. It was likely that azide caused leaf damage and the activity of cell metabolism was decreased greatly, resulting in small membrane PD changes by $CO_2$ and no changes by redlight. Therefore, it can be suggested that red light was sensed by the mesophyll and the light induced guard cell membrane hyperpolarization was related to energy produced by cyclic-photophosphorylation, but ${CO_2}-induced$ guard cell membrane hyperpolarization was not related to photosynthesis. Alkalisation of the vacuole was observed when the intact leaf was exposed to $CO_2$, indicating that membrane hyperpolarization was mainly the result of proton efflux.efflux.

Effete of Ozone Uptake Rate on Photosynthesis and Antioxidant Activity in the Leaves of Betula Species (자작나무류 잎의 오존흡입량이 광합성 및 항산화효소 활성에 미치는 영향)

  • This study was conducted to compare the physiological and biochemical responses of four Betula species in response to ozone, and to find out the relationship between ozone uptake rate and photosynthesis or antioxidant activity. One-year-old seedlings of four Betula sp, B. costata, B. davurica, B. platyphylla var, japonica, and B. ermani, exposed to 100 pub ozone concentration for 8h day$^{-1}$ for 5 weeks in fumigation chamber. Ozone uptake rate, photosynthesis, SOD and GR activity were measured in the leaves of four species once a week. Cumulative ozone uptake rate was largest in the loaves of B. costata(53.8 mmol m$^{-2}$ ), smallest in the leaves of B. davurica(35.5 mmol m$^{-2}$ ). Photosynthesis of four Betula sp. exposed to ozone reduced relative to control, but the photosynthetic responses with changing stomatal conductance were different among species. Ozone exposure increased SOD activities of four species at the early exposing period, but after a critical point SOD activity decreased gradually. GR activity of B. costata was similar to the change of SOD activity, but the others showed the different patterns from B. costata. In conclusion, decreasing both SOD and GR activity at the critical point, B. costata may be sensitive species in response to ozone. In contrast, the others may be resistant species, which gradually increase GR activity following ozone exposure. GR activity was not always in accord with the change of SOD activity against ozone uptake, and the different responses between species were supposed to be affected by the cumulative ozone uptake.

Effects of the Light Source of LEDs on the Physiological and Flowering Response of Endangered Plant Silene capitata Kom. (LED광질에 따른 분홍장구채(Silene capitata Kom.)의 생리 및 개화 반응)

  • We examed physiological and flowering response of S. capitata, the endangered plant in Korea, under LED light conditions in plant factory to cultivate artificially for conservation. We cultivated S. capitata and measured its physiological responses and the number of flowers under red, blue, white, red+far-red mixed, red+blue mixed, and red+blue+white mixed light. The results showed that its photosynthetic rate and chlorophyll content were recorded relatively high in red+blue+white and red+blue mixed light respectively. Transpiration rate and stomatal conductance appeared relatively high in the white single light while water use efficiency was no difference. Photochemical efficiency of photochemical photosystem II by minimum and maximum chlorophyll fluorescence was the highest in the red+blue+white mixed light condition than other ones. The number of flowers of S. capitata was at its peak under the red light or red+far-red mixed light. Therefore, we conclude that the most efficient way to grow for flowering of S. capitata is to provide red light or red+far-red mixed light in the plant factory.

Physiological Response and Cadmium Accumulation of MuS1 Transgenic Tobacco Exposed to High Concentration of Cd in Soil: Implication to Phytoremediation of Metal Contaminated Soil (토양 중 고농도 카드뮴에 노출된 MuS1 형질전환 담배 (Nicotiana tabacum cv. Xanthi)의 생리적 반응 및 카드뮴 축적: 식물학적 오염토양정화를 위한 형질전환 식물 탐색)

  • The objective of this study was to understand the physiological response and cadmium accumulation of MuS1 transgenic tobacco exposed to high concentration of Cd in soil. For this, a pot experiment was carried out in a greenhouse for a month, with two lines of MuS1 transgenic tobaccos (S4 and S6) and non-transgenic tobacco cultivated in the soils spiked at three different Cd concentrations (0, 60 and 180 mg $kg^{-1}$). Both transgenic and non-transgenic tobacco showed visible toxic symptoms such as chlorosis and leaf roll as treated concentration increased. The net photosynthetic rates of MuS1 plants (S4 and S6) exposed at 180 mg $kg^{-1}$ Cd were 6.3 and $7.7{\mu}mol\;m^{-2}s^{-1}$, being higher than those of the non-transgenic plant ($4.8{\mu}mol\;m^{-2}s^{-1}$). Values of stomatal conductance of MuS1 transgenic plants (0.05 and 0.008 mmol $H_2O\;m^{-2}s^{-1}$) were also higher than those of non-transgenic plant (0.03 mmol $H_2O\;m^{-2}s^{-1}$). In addition, fresh and dry weights of MuS1 transgenic plants were heavier than those of non-transgenic plant. Likewise, MuS1 transgenic plants appeared to be better physiological performance than non-transgenic tobacco when exposed at high concentration of Cd in soil. With regard to metal accumulation, MuS1 transgenic tobaccos accumulated more Cd in their roots than non-transgenic tobacco implying that MuS1 transgenic tobacco is suggested to be used for phytostabilization of heavy metals.

Growth and Physiological Response in Container of Daphniphyllum macropodum by Shading Level (피음 수준에 따른 굴거리나무의 용기 내 생장 및 생리적 반응)

  • This study was carried out in order to closely examine the growth and the physiological response by shading levels of a container seedling of Daphniphyllum macropodum which is known as the species of having shade tolerance in a warm temperature evergreen broad-leaved tree. The shading treatment was regulated with the shading level of full sunlight, and 35%, 55%, 75% of full sunlight. As a result of surveying the height and the root collar diameter growth by the shading level, the height was the highest with 13.2 cm under 35% of shading. The root collar diameter was the highest with 4.99 mm under full sunlight. The organ-based (leaves, shoot and root) dry mass production and the whole dry mass production were higher with 1.38 g, 0.47 g, 0.87 g, 2.72 g, respectively, in order of 35% shading. As for chlorophyll a, b, and the total chlorophyll content, the highest chlorophyll content was shown under 75% of shading with the relatively highest shading level. The ratio of chlorophyll a to chlorophyll b (a/b) was the highest under 35% of shading. The photosynthetic rate, stomatal conductance and water use efficiency were the highest with $12.3{\mu}molCO_2{\cdot}m^{-2}{\cdot}s^{-1}$, $0.27molH_2O{\cdot}m^{-2}{\cdot}s^{-1}$, $3.53{\mu}molCO_2{\cdot}mmol^{-1}H_2O$, respectively, under 35% of shading. As a result of surveying the whole experiment, D. macropodum grows nicely by maintaining 35% shading.

Photosynthesis and Growth of Southern-type Garlic (Allium sativum L.) in Response to Elevated Temperatures in a Temperature Gradient Tunnel (온도구배터널 내 상승온도에 의한 난지형 마늘(Allium sativum L.)의 광합성 및 생육 특성의 변화)

  • This study assessed clove germination, shoot growth, photosynthesis and bulb development of southern-type garlic (Allium sativum L.) in a temperature gradient tunnel (TGT), to examine the impacts of increases in temperature on the growth of garlic and find a way to minimize them. The temperatures in the middle and outlet of the TGT were 3.2℃ and 5.8℃ higher, respectively, than the ambient temperature at the tunnel inlet. The germination of garlic cloves was late at temperatures of ambient+3℃ (in the middle of the TGT) and ambient+6℃ (at the outlet) than at ambient temperature (at the inlet). However, bolting and the timing of maximum leaf number per plant were faster at ambient+3℃ or +6℃ than at ambient temperature. Shoot growth was generally greater at ambient temperature. Bulb growth did not significantly differ according to cultivation temperatures, but fresh and dry weights were slightly higher at ambient temperature and ambient+3℃ in the late growth stage. The photosynthesis rate (A), stomatal conductance (gs), and transpiration rate (E) were higher at ambient+3℃ than at ambient temperature. Furthermore, at ambient+3℃, the net photosynthetic rate (Amax) was high, while the dark respiration rate (Rd) was low. At ambient temperature and ambient+3℃, bulb development was healthier, resulting in better productivity and more commercial bulbs, while at ambient+6℃, the bulbs were small and secondary cloves developed, resulting in low commercial value. Therefore, at elevated temperatures caused by global warming, it is necessary to meet the low-temperature requirements before clove sowing, or to delay the sowing time, to improve germination rate and increase yield. The harvest should also be advanced to escape high-temperature stress in the bulb development stage.

Seasonal Variation in Photosynthetic Characteristics and Chlorophyll Content of the Loranthus tanakae, Viscum album var. coloratum and its Hosts in Korea (계절에 따른 국내 자생 꼬리겨우살이, 겨우살이 및 기주목의 광합성 특성과 엽록소 함량)

  • We investigated seasonal variation in photosynthetic characteristics and chlorophyll content of the Loranthus tanakae, Viscum album var. coloratum and its hosts in Korea. The maximum photosynthesis and transpiration rate of L. tanakae were $9.36{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ at $941{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ PAR (photosynthetically active radiation) in June, $5.06{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ at $1,596{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ PAR in July, respectively. The maximum photosynthesis and transpiration rate of V. album var. coloratum were $6.51{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ at $418{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ PAR in Autumn, $3.91{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ at $1,735{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ PAR in Autumn, respectively. V. album var. coloratum was able to conduct photosynthesis in November whereas its host and L. tanakae were not able to conduct photosynthesis. Especially transpiration rate of L. tanakae were always higher than its host and V. album var. coloratum. The chlorophyll a+b contents of L. tanakae was $8.23mg{\cdot}g^{-1}$ in July, V. album var. coloratum was $10.27mg{\cdot}g^{-1}$ in June, and chlorophyll a/b ratio of L. tanakae was 1.7~3.7, V. album var. coloratum was 1.1~4.5, depend on season.
