Photocatalytic-Photooxidation of Halogen Derivatives of Phenols in Aqueous Solution (방향족 탄화수소 할로겐 유도체의 광촉매-광산화)

  • Industrial waste which highly loaded by halogenide phenols was photooxidized by laboratory-scale photooxidation of these organic impurities in the presence of aerotropic and titaniumdioxide as photocatalyst. The disapperance of organic compounds was determined as a function of the irradiation time. Some contaminants such as 2-chlorophenol, 2-bromphenol, 3-bromphenol, 4-bromphenol, 2,4-dibromophenol and 2,6-dibromophenol were photodegraded separately to obtain information on the reaction rates, reactivities, and reaction mechanisms of the photooxidation, and on the stoichiometric correlation between organic reactant and inorganic products concentration in the course of the photocatalytic photoreaction.

  • Singlet oxygen-mediated photooxidation and subsequent triplet oxygen insertion of 11-hydro-13-(methyloxy)carbonylartemisinate (4) yielded a diketo compound 5 as the major product instead of the expected 11-hydro-13-(methyloxy)carbonylartemisinin (6). The formation of the unexpected product was in part consistent with the mechanism proposed by Acton and Roth and is the first isolation of the diketo compound from the photooxidation of artemisinate.

  • This study was investigated and analyzed the physiological reaction mechanisms and the factors of the chlorophyll bleaching phenomenon on leaf burning-disease of the Ginseng (Panax ginseng C.A. Meyer). Chlorophyll bleaching phenomenon was mainly caused by the photooxidation of singlet oxygen and the autooxidation of hydrogen peroxide($H_2O_2$) accumulation resulted from inactivation of catalase and peroxidase. Chlorophyll bleaching phenomenon was remarkably accelerated by addition of saponin.

  • γ -Glutamyl transpeptidase (EC 2.3.2.2) plays a key role in glutathione metabolism by catalyzing the transfer of the γ -glutamyl residue and hydrolysis of glutathione. The functional residues at the active site of the rat kidney γ -glutamyl transpeptidase were investigated by kinetic studies at various pH, the treatment of diethylpyrocarbonate (DEPC), and photooxidation in presence of methylene blue. An ionizable group affecting the enzymatic activity with an apparent pKa value of 7.1, which is in the range of pKa values for a histidine residue in protein, was obtained by examining the pH-dependence of kinetic parameters. The pH effect on the photoinduced inactivation rate of the enzyme corresponds to that expected for the photooxidation of the free histidine. The involvement of a histidine in the catalytic site of the enzyme was further supported by DEPC modification accompanied by an increase in absorbance at 240 nm, indicating the formation of Ncarbethoxyhistidine. The histidine located at the position of 382 in the precursor of the enzyme is primarily suspected based on the amino acid sequence alignment of the transpeptidases from various organisms.

Effects of Light and Photosynthetic Electron Transport System on the Generation of Singlet Oxygen ($^1$O$_2$) in Ginseng Thylakoid Membrane (인삼 틸라코이드에서 Singlet Oxygen($^1$O$_2$) 생성에 미치는 전자전달계의 영향)

  • In order to Investigate the mechanism of the leaf-burning disease of ginseng (Panax ginseng C.A. Meyer), studies on the generation of singlet oxygen (1O2) and the photooxidation of the pigments were carried out in comparison with the ones of soybean (G1ycine max L). The studies were mainly focalized on the effects of light intensity, light intensity, inhibitor and electron donor/acceptor of the Photosynthetic electron transport system. When we measured the amounts of 1O2 generated in the thylakoids of ginseng and soybean by the irradiation of light (300 w/m2) as a function its time. It was identified that a higher amount of 1O2 was formed in the ginseng thylakoid than the case of soybean. A generation ratio of lO2 between ginseng and soybean sltbstantially identical in the range of light intensities 50∼150w/m2 However much higher amount of 1O2 was generated in ginseng by irradiation of strong intensity of light (200 500w/m2). Wave length dependency on the generation of 1O2 and the pigment photooxidation was observed on ginseng thylakoids; red light (600-700 nm) gave a maximum effect in the contrast with blur green light (400-60 nm). When the ginseng thylalioid was treated with the electron donor (Mn2+) and acceptors (DCPIP, FeCy) of the photosynthetic electron transport system. a drastic inhibition of 1O2 generation was observed. However, treatment with its inhibitors (DCMU, KCW) activated 1O2 generation. An interesting fact that an electron donor or acceptor of the photosystem II(P680) Inhibited 1O2 generation, suggests an intimate relationship between 1O2 generation and photosystem II.

  • Self-assembled monolayers (SAMs) formed by the adsorption of alkanethiols, $HS(CH_2)_nX$, where X is an organic functional group, onto gold surfaces have attracted widespread interest as templates for the fabrication of molecular and biomolecular microstructures. Previously photopatterning has been thought of as being restricted to the micron scale, because of the wellknown diffraction limit. So, we have explored a novel approach to nanofabrication by utilizing a femtosecond laser coupled to a near-field scanning optical microscope (NSOM).
