Effect of the methanol extract of Dansameum on the ammonia-induced peptic ulcer in rats (단삼음(丹參飮)이 흰쥐의 소화성 궤양에 미치는 영향)

  • Objective : his study was carried out to investigate the effect of Dansameum (DS) on the gastrohemorrhagic lesion induced by ammonia in rats. Methods : Rats were pretreated with DS extract 25 mg/kg for 10 days and then were given amonia through gastric tube. The animals were killed 1 hr after amonia treatment. Results : DS extract siginificantly reduced the gastrohemorrhagic lesion score, the gastric lipid peroxide level, the gastric urease activity, the gastric myeloperoxidase activity, the gastric acid phosphatase activity, xanthine oxidase activity and type conversion, and increased the gastric glutathione level considerably. In photomicrographs of stomach tissue in rat, we could see the gastrohemorrhagic lesion induced clearly. conclusions : These results suggest that DS extract may be effective in peptic ulcer.

Bone Changes in Femoral Bone of Mice Using Calcein Labeling (Mice에서 Calcein 표지를 이용한 골 변화 관찰)

  • In vivo labeling of bone with fluorochromes is a widely used method for assessment of bone formation and remodeling processes. In particular, calcein is used as a marker for identification of bone growth, which is indicated by a green color. Calcein green is a calcium chelator that adheres to regions of mineralizing bone thereby allowing localization of new bone. Bone formation and remodeling in vivo can be assessed by calcium-binding calcein labeling. In this study, changes in the femoral bone of a normal mouse model at both 4 and 8 weeks were evaluated using calcein labeling. Intense deposition of calcium in the bone was observed after application for 8 weeks. A mouse model is suitable for application in in vivo experiments using genetically modified mice, such as knock-out mice, however data regarding femoral cross sectional bone in young mice are limited. The current study confirmed calcein as a useful marker for identification of bone growth, which was indicated by a green color on photomicrographs. This methodological process may provide basic information for interpreting bone formation and regeneration to pharmacologic or genetic manipulation in mice.

The Effect of Etching Time on the Biaxial Flexural Strength of IPS Empress® 2 Ceramic (불산 처리 시간이 IPS Empress® 2 세라믹의 2축 굴곡강도에 미치는 영향에 대한 연구)

  • Fluoric acid etching is an essential procedure in cementation of reinforced ceramics to tooth surface. But there have been few studies about the changes of surface structure and flexural strength of IPS $Empress^{(R)}$ 2 ceramic according to the etching time. The objectives of this study were to examine the surface structure changes and the difference in biaxial flexural strength of IPS $Empress^{(R)}$ 2 ceramic according to various etching times. Sixty one disk-shaped specimens of IPS $Empress^{(R)}$ 2 ceramic($14mm{\times}1.2mm$) were fabricated for the biaxial flexural strength test and SEM analysis according to the manufacturer's recommendations. Sixty specimens were divided into 6 groups(n=10) according to the time of HF acid etching(0, 20, 180 and 300s)and silane/resin cement application. Each disk was loaded using a piston-on-3 ball biaxial configuration in a universal testing machine. The failure loads(N) were recorded, and the biaxial flexural strength for each disk was calculated. A one-way analysis of variance and independent t-test on transformed fracture strength data were used to determine significant differences between groups. The groups of no cementation showed a trend toward progressive weakening with increasing the etching time. However, this was not statistically significant at p=0.05 level. The groups of resin cementation exhibited no apparent trend in their mean strength values. SEM photomicrographs showed very different results of etching. Within the conditions of this study, alteration of surface topography by acid etching does not have a deleterious effect on the biaxial flexural strength of IPS $Empress^{(R)}$ 2 ceramic.

Effect of Microwave Reheating on Quality of Aseptic-Packaged Cooked Rice (무균포장법의 Microwave Reheating 후의 품질 특성에 관한 연구)

  • Qualify changes of microwave-reheated cooked rice aseptic-packaged with different lid materials and oxygen absorbers were determined during storage at $20^{\circ}C,\;30^{\circ}C$ and $40^{\circ}C$. Springiness cohesiveness and adhesiveness increased as storage temperature increased, while gumminess and hardness decreased. Lid materials oxygen absorbers, and storage temperatures showed no significant effect on the textural properties of cooked rice after microwave reheating. Degree of gelatinization was increased as storage temperature increased and was restored after microwave reheating. Scanning electron micrograph showed that the surface of cooked rice became rough and hollowed after microwave reheating for 1.5 min, indicating starch was regelatinized, while there was no difference with respect to lid materials. Photomicrographs of the cross-section of aseptic-Packaged cooked rice showed that a compact and solid structure increased and retrogradation occured in starch as storage temperature decreased, while a compact and solid structure was decreased and starch was regelatinized after microwave reheating.

Inhibitory Effects of Kimchi Extracts on the Growth and DNA Synthesis of Human Cancer Cells

  • Effect of solvent extracts and juice supernatants from kimchis on the growth of various human cancer cells was studied, comparing with the actions on the normal cells. Inhibitory effect of kimchi extracts on[3H] thymidine incorporation n cancer cells was also investigated. The methanol extract, hexane extract and methanol soluble fraction (MSF) of 3-week fermented kimchi did not have growth inhibitory effect on Ac2F rat normal liver cells at the concentrations of 0.5~2%. However, marked decrease in the growth of AGS human gastric cancer cells was shown by the treatment of those extacts. The juice from the kimchi samples also suppressed the growth of K-562 human leukemia cells and MG-63 human osteosarcoma cells. Especially, the juice of 3-week fermented kimchi exhibited the strong growth inhibitory effect in MG-63 human osteosarcoma cells. At the photomicrographs, growth inhibition and morphological change of the cells treated with kimchi juice were observed. And the solvent extracts of 3-week fermented kimchi suppressed the growth of cancer morethan the extracts or juices from fresh and 6-week fermented kimchi. When AGS human gastric cancer cels were treated with the extracts of 3-week fermented kimchi, [3H] thymidine incorporation in the cells also decreased. These results showed that kimchi extracts and juices had growth inhibitory effects on human osteosarcoma, leukemia and gastric cancer cells, but had no toxicity to the normal cells. We suggest that kimchi might have anticancer effect in part due to inhibition of the growth and DNA synthesis of cancer cells.

Effect of Fasciculus Vascularis Luffae Herbal-Acupuncture at St36 on OVA-induced Asthma in Mice (사과락(絲瓜絡) 약침(藥鍼)의 천식억제(喘息抑制) 및 면역조절효과(免疫調節效果)에 대(對)한 실험적(實驗的) 연구(硏究))

  • Objectives : The purpose of this study is to confirm the suppression effect of Asthma and Immune response improvement of Fasciculus Vascularis Luffae Herbal-acupuncture into Chok-samni(St36) on ovalbumin-induced asthma in mice. Methods : C57BL/6 mice were sensitized and challenged with OVA(ovalbumin) for 12 weeks Two experimental groups were treated with different concentrations(1%, 0.1%) of FVL-HA at Chok-samni(St36) for the later 8 weeks(3times/week). Results : 1. Lung weigh of the mice group treated with FVL-HA decreased significantly compared with that of control group. 2. Total cells of lung, total Leukocytes and Eosinophils in BALF of the mice group treated with FVL-HA decreased significantly compared with those of control group. 3. Eosinophils in BALF of the mice group treated with FVL-HA in Photomicrographs decreased significantly compared with those of control group. 4. The concentration of IL-l3, IgE, IL-4 In serum and IL-4 in BALF of the mice group treated with FVL-HA decreased significantly compared with that of control group. 5. The number of Gr-1+/CB11b+, CD3-/CCR3+, CD4+, CD8+, CD3e+/CD69+, IgE+/B220+ cells in the lungs of the mice group treated with FVL-HA decreased significantly compared with those of control group. 6. The cytokine's manifestation of mRNA of the mice group treated with FVL-HA with RT-PCR decreased significantly compared with that of control group. Conclusion : These result suggests that Fasciculus Vascularis Luffae Herbal-acupuncture Choksamni(St36) in C57BL/6mice may be an effective part to OVA-induced asthma in C57BL/6 mice.

Shear bond strength of a layered zirconia and porcelain according to treatment of zirconia liner (치과용 지르코니아 이장재 처리에 따른 지르코니아와 도재의 전단결합강도 비교)

  • Purpose: Physical and chemical properties of gold is most suitable to be restored of teeth to its original state. Recently zirconia was used instead of gold because of esthetical and intimacy of human body. Because of high strength and high abrasion resistance of zirconia, all zirconia artificial tooth lead to wear the original tooth of opposite site. To preserve this original tooth, zirconia artificial tooth covered with dental ceramic glass was used. When joining the zirconia core and dental ceramic glass, difference of their thermal expansion coefficient and wetting ability is generated the residual stress at interface lead to crack. In order to solve this problem, intermediate layer what is called zir-liner was imported to decrease the residual stress and increase the bonding strength. Methods: In this study, to identify the optimum conditions for manufacturing process, various methods to rough the surface of zirconia core were adopted, and vary the thickness of interlayer, and analyzed bond strength. Results: Bond strength of sanding specimens group showed higher than that of non-sanding specimens group, and once applied intermediate layer with sanding specimens showed highest bond strength with 28 MPa. SEM photomicrographs of zirconia cores fired at $1500^{\circ}C$ showed parallel straight lines in sanding and pockmarked surface in blasting surfaces as abrasion traces. Observation of the destruction section after shear test by SEM were carried out. Liner applied non-sanding group and non-liner applied sanding group all showed interfacial crack. Sandblasting group with non-liner showed remained dental ceramic glass on the surface of zirconia. Sandblasting group with once applied liner showed partially remained liner and dental ceramic glass on the surface of zirconia. XRD analysis revealed that sandblasting group showed higher monoclinic peaks than other specimens group and this result was due to the high collision energy for stress induced phase transformation. Conclusions: A study on the improvement of bonding strength between zirconia and dental ceramic glass steadily carried out for the future to practical use.

Effect of Continuous Oil Phase on Preparation of Silver Halide Nanoparticles using AOT-Based W/O Microemulsions (AOT W/O 마이크로에멀젼을 이용한 할로겐화은 나노입자 제조에서 연속상 오일의 영향)

  • Effects of continuous oil phase on silver halide nanoparticles were investigated where nanoparticles were prepared using two different types of water-in-oil(W/O) microemulsions containing silver and halide, respectively. Phase behavior experiments for ternary systems containing AOT surfactant, hydrocarbon oil and aqueous solution of an inorganic salt showed that the region of one phase W/O microemulsion was found to be broadened with an increase in the alkyl chain length of a hydrocarbon mainly due to an increase in hydrophilic nature of a surfactant. With the information of phase behavior experiments, silver halide nanoparticles were prepared using different AOT-based microemulsion systems and photomicrographs obtained by transmission electron microscopy indicated that about 10 nm size particles of relatively spherical shape were obtained. It has been found that an increase in alkyl chain length of a hydrocarbon results in a decrease in particle size because of higher intermicellar exchange rate among microemulsion drops. The average particle size was also found to increase with the inorganic salt composition of initial aqueous solution.

The use of auxiliary devices during irrigation to increase the cleaning ability of a chelating agent

  • Objectives: This study investigated the cleaning ability of ultrasonically activated irrigation (UAI) and a novel activation system with reciprocating motion (EC, EasyClean, Easy Equipamentos $Odontol\acute{o}gicos$) when used with a relatively new chelating agent (QMix, Dentsply). In addition, the effect of QMix solution when used for a shorter (1 minute) and a longer application time (3 minutes) was investigated. Materials and Methods: Fifty permanent human teeth were prepared with K3 rotary system and 6% sodium hypochlorite. Samples were randomly assigned to five groups (n = 10) according to the final irrigation protocol: G1, negative control (distilled water); G2, positive control (QMix 1 minute); G3, QMix 1 minute/UAI; G4, QMix 1 minute/EC; G5, QMix 3 minutes. Subsequently the teeth were prepared and three photomicrographs were obtained in each root third of root walls, by scanning electron microscopy. Two blinded and pre-calibrated examiners evaluated the images using a four-category scoring system. Data were statistically analyzed using Kruskal-Wallis and Dunn tests (p < 0.05). Results: There were differences among groups (p < 0.05). UAI showed better cleaning ability than EC (p < 0.05). There were improvements when QMix was used with auxiliary devices in comparison with conventional irrigation (p < 0.05). Conventional irrigation for 3 minutes presented significantly better results than its use for 1 minute (p < 0.05). Conclusions: QMix should be used for 1 minute when it is used with UAI, since this final irrigation protocol showed the best performance and also allowed clinical optimization of this procedure.

Preparation and Electrical Properties of Manganese-incorporated Neodymium Oxide System

  • Manganese-incorporated neodymium oxide systems with a variety of Mn mol% were prepared to investigate the effect of doping on the electrical properties of neodymium oxide. XRD, XPS, SEM, DSC, and TG techniques were used to analyze the specimens. The systems containing 2, 5, 8, and 10 mol% Mn were found to be solid solutions by X-ray diffraction analysis and the lattice parameters were obtained for the single-phase hexagonal structure by the Nelson-Riley method. The lattice parameters, a and c, decreased with increasing Mn mol%. Scanning electron photomicrographs of the specimens showed that the grain size decreased with increasing Mn mol%. The curves of log conductivity plotted as a function of 1/T in the temperature range from 500 to 1000$^{\circ}C$ at $PO_2$'s of $10^{-5}$ to $10^{-1}$ atm for the specimens were divided into high-and low-temperature regions with inflection points near 820-890$^{\circ}C$. The activation energies obtained from the slopes were 0.53-0.87 eV for low-temperature region and 1.40-1.91 eV for high-temperature region. The electrical conductivities increased with increasing Mn mol% and $PO_2$, indicating that all the specimens were p-type semiconductors. At $PO_2$'s below $10^{-3}$ atm, the electrical conductivity was affected by the chemisorption of oxygen molecule in the temperature range of 660 to 850$^{\circ}C$. It is suggested that electron holes generated by oxygen incorporation into the oxide are charge carriers for the electrical conduction in the high-temperature region and the system includes ionic conduction owing to the diffusion of oxygen atoms in the low-temperature region.
