Soybean plants inoculated Bradrhizobium japonicum MN 110 were grown in outdoor perlite pots with nitrogen free nutrient solution containing 1.0 mM-P(control) and 0.05 mM-P(stress) and harvested at 28, 35, 42 and 49 days after transplanting (DAT) to examine the effect of phosphorus deficiency on ureide concentration of and distribution to diffrent plant organ in nitrogen fixing soybean plant during the vegetative growth. Total dry mass of control plants increased 8.9 fold and that of phosphorus deficient plant increased 2.7 fold during the experimental period. Phosphorus deficiency reduced total phosphorus and nitrogen accumulation by 80%,40% respectively, at 28 DAT and 93%, 84%, respectively, at 49 DAT. Nitrogen concentration was reduced by phosphorus deficiency in all tissues with leaf and stem tissues affected to a greater degree than nodule and root tissues at every sampling date. Phosphorus deficiency significantly reduced soluble reduced-N and ureide-N concentration in leaf and stem but did not affect those in root. The proportion of soluble reduced-N in leaf was reduced from 60% to 50% but increased from 10% to 20% in the roots. The proportion of ureide-N in leaf of control plants was higher than that in phosphorus deficient plants, whereas, roots of phosphorus deficient plants contained a higher propotion of ureide-N than those of control plants. These indicated that phosphorus deficiency not only inhibit nitrogen fixation of nodules but also restrict the translocation of fixed nitrogen out of the root system into the xylem.(Received April 4, 1997; accepted May 2, 1997)
Most of the lakes in Korea are artificial, and their limnological characteristics are significantly different from those of natural lakes in other countries. In this study, the relationship between trophic state parameters was investigated, based on summer average data of the upper layer, in 81 lakes in Korea, 2013-2017. Compared with trends of foreign natural lakes, chlorophyll a (Chl.a) concentration was slightly lower at the same total phosphorus (TP) concentration, and transparency (Secchi depth, SD) was noticeably lower at the same Chl.a concentration. This is because of excessive allochthonous loading of non-algal material during the monsoon period, and the reduction in phosphorus availability to algal growth, by light limitation and short hydraulic residence time. Considering these characteristics, we suggested site-specific thresholds of trophic state classification for Chl.a, TP and SD, based on annual average data at the upper layer of lakes ($3-10{\mu}g\;L^{-1}$ of Chl.a measured by UNESCO method; $13-33{\mu}g\;L^{-1}$ of TP; 1.6-3.2 m of SD for mesotrophic state class, respectively). The threshold value of TP for each trophic state class, corresponded to the upper value of previously reported range, and that of SD was out of the range. We suggested applying only TP and Chl.a in assessment of trophic state of lakes in Korea, excluding SD.
This study was conducted to determine limiting nutrients and the physiological characteristics of phytoplankton in response of nutrients in Lake Paldang from March 2002 to October 2002. A field research was conducted along with laboratory batch culture experiment to find the limiting nutrients and the growth kinetics. According the results of Chl. a TP relationship, TN/TP ratio, and nutrient addition bioassay, phosphorus appeared to be a major limiting nutrient in Lake Paldang and thus the lake productivity was greatly influenced by it. P limitation for the phytoplankton of Lake Paldang varied with season, and the possibility of limitation by nitrogen and silica also occurred. The degree of P limitation was greatest during spring when the concentration of dissolved phosphorus is relatively much lower than summer and autumn. The maximum growth rate (${\mu}_{max}$) and half saturation concentration ($K_u$) of Lake Paldang phytoplankton ranged from 0.8${\sim}$1.1$day^1$ and from 0.1${\sim}$O.8${\mu}M$, respectively. $K_u$ was highest during May ($0.8{\mu}M$) and the lowest during September ($0.1{\mu}M$). Such result may be induced by the phytoplankton cell quota that showed the lowest concentration ($0.13{\mu}gP/{\mu}gChl.$ a) during May. The growth kinetics showed that phytoplankton growth in Lake Paldang was faster during summer and autumn than spring, suggesting that the Potential of algal bloom is high after the summer monsoon season.

Wastewater-adapted microalgae such as Chlorella vulgaris AG10032, Ankistrodesmus gracilis SAG278-2 and Scenedesmus quadricauda AG10308 are useful biological resources for recovering biofuel and other bio-based materials from wastewater because of their efficient removals of nitrogen and phosphorus from wastewater and their high fatty acid contents in biomass. Although the concentrations of phosphorus typically vary in wastewater environment, very little is known about the effect of phosphorus concentration, especially phosphorus starvation, on microalgal fatty acid synthesis. This is partially due to the lack of methodological establishment for algal fatty acid analysis. In this study, we compared the analysis performances of microalgal fatty acids by two different methods; one is a non-polar GC (gas chromatography) column based method, which is generally used for microbial fatty acids, and the other is a polar WAX-type GC column method, which is typically used for plant fatty acids. And then, we explored the effect of phosphorus concentration levels on fatty acid production in microalgae cultivated from wastewater. As results, the polar WAX-type column method has better ability to separate poly unsaturated fatty acids (PUFA) including $C_{18:3}$ (linolenic acid), and was found to be more applicable in analyzing fatty acids from wastewater-cultivated microalgae than the non-polar column method. The fatty acid characterization by the WAX-type column method revealed little effect of phosphorus starvation on the quantity and composition of fatty acids from wastewater-cultivated microalgae.
To study the environmental features causing a bloom of the novel dinoflagellate Heterocapsa circularisquama (Dinophyceae), hydrographic and chemical aspects were measured in the Uranouchi Bay, Kochi Prefecture, Japan, from January to December, 1997. The cell density of H. circularisquama increased rapidly in early October, and dropped sharply in mid-October. Growth rate of H, circularisquama during bloom period appeared 1.50 division day$^{-1}$ under high water temperature (25$^{\circ}C$) and salinity (32 psu) conditions. Althought the result from hydrographic aspect indicated good condition for their growth, dissolved inorganic phosphorus (DIP) concentration in surface layer before bloom formation was less than 0.70uM, which is lower than their half saturation constant(Ks). Dissolved inorganic nitrogen(DIN): DIP ratio was > 30, indicating potential P-limitation. However, before bloom formation period of H. circularisquama, DIP concentrations were high in bottom layer (> 4.0 uM). Some studies reported that H. circularisquama had the ability to migrate vertically and to utilize dissolved organic phosphorus (DOP). Thus, DIP in bottom layer might have been utilized by H. circularisquama for their growth.DOP might have weakly affected their growth because of low reactive DOP concentrations owing to low DOP concentration (ca. 0.39 uM). Thus, if nutrient condition of bottom layer in Uranuchi Bay is not improved, the outbreaks of H. circularisquama red tides may became an annual feature.
Background: Previous numerous studies on watershed scale demonstrated that the constructions of upper dams may influence the below dams due to modifications of flow regime and nutrient inputs. Little is known about how the dam constructions influence the downstream lakes or reservoirs in the regional scale. This study demonstrates how the construction of upper dam (i.e., Yongdam Dam) influences nutrient regime, trophic relations, and empirical models in Daechung Reservoir (DR). Yongdam Dam was constructed at the upstream region of DR in year 2000. Results: The analysis of hydrological variables showed that inflow and discharge in the DR were largely reduced after the year 2000. The construction of upper dam construction also resulted in increases of water temperature, pH and conductivity (as an indicator of ionic content) in the DR. Empirical models of TP-CHL and N:P ratio-CHL suggested that stronger responses of CHL to the phosphorus were evident after the upper dam construction, indicating that algal production at a unit phosphorus increased after the upper dam construction. Mann-Kendall tests on the relations of N:P ratios to TN showed weak or no relations ($t_{au}=-0.143$, z = -0.371, p = 0.7105) before the dam construction, while the relation of N:P ratios to TP showed strong in the periods of before- ($t_{au}=-0714$, z = -2.351, p = 0.0187) and after the construction ($t_{au}=-0.868$, z = -4.270, p = 0.0000). This outcome indicates that TP is key determinant on N:P ratios in the reservoir. Scatter Plots on Trophic State Index Deviations (TSIDs) of "TSI(SD) - TSI(CHL)" against "TSI(TP) - TSI(CHL)" showed that the dominance of clay turbidity or light limitation was evident before the upper dam construction [TSI(TP) - TSI(CHL) > 0 and TSI(SD) - TSI(CHL) > 0] and phosphorus limitation became stronger after the dam construction [(TSI(TP) - TSI(CHL) < 0 and TSI(SD) - TSI(CHL) > 0]. Conclusions: Overall, our analysis suggests that the upper dam construction modified the response of trophic components (phytoplankton) to the nutrients or nutrient ratios through the alteration of flow regime, resulting in modifications of ecological functions and trophic relations in the low trophic levels.

We estimated the limiting nutrient and DIP limiting history based on alkaline phosphatase (APase) activity during the spring of 2017 in the Southern Sea, Korea. In the frontal area, concentration of dissolved inorganic phosphorus (DIP), dissolved inorganic nitrogen (DIN): DIP ratio and Chlorophyll a (Chl-a) were < $0.2{\mu}M$, 23.2 and $2.2{\mu}g/L$, respectively, indicating high productivity despite DIP limiting. The relationship between APase and DIP indicates that the study area had limited DIP because of a strongly reverse correlation (r= -0.81; P<0.001). Relationship between APase and Chl-a (r=0.61, p<0.001) also indicated that APase may have been induced by phytoplankton (ca. 60 %) and bacteria (ca. 40 %). In DIP limiting history in this study area, frontal area and non-frontal areas might have induced long-term DIP limitation and the recent relief from DIP-limitation, respectively, based on distributions of dissolved APase and particulate APase. Thus, these results suggest that by measuring the enzyme that hydrolyzes organic matter such as APase in frontal area, it is possible to estimate temporal and spatial characteristics of limiting nutrient, thereby improving our understanding of biogeochemistry cycles.
Increased harmful algal blooms by cyanobacteria are threatening public health and limiting human activities related with freshwater ecosystems. Phosphorus (P) has long been suggested as a critical nutrient for cyanobacterial bloom through field research in Canada during the 1970s, proposing a P-based freshwater management guideline. However, recently, nitrogen (N) has also been highlighted as an impacting nutrient on cyanobacterial harmful algal blooms (CyanoHABs). Due to the intensive and frequent observation of Microcystis, this kind of paradigm shift from P limitation to season-dependent N or P limitation has an important implication for a dual nutrient management strategy in eutrophic freshwaters. Through recent international researches, general strategies to control CyanoHABs in lakes and reservoirs are as follows: a dual nutrient (N & P) reduction, wastewater collection and treatment, pre-treatment of influent water in buffer zones, dredging of sediment, reduction of residence time, algal collection, and precipitation and flocculation of cyanobacteria. In addition, sustainable and integrative freshwater algae management should be carried out, based on the ecological aspect, because cyanobacteria are not the target organism to be eradicated, but an essential microbial member in the freshwater ecosystem.
Carbon ($^{14}CO_2$) and nitrogen ($^{15}NH_4$ and $^{15}NO_3$) uptake were measured at two stations in the hypertrophic zone of the Seonakdong River, where Microcystis aeruginosa explosively bloomed in September 1998. Significant nitrogen limitation occurred in the period of Microcystis bloom, while phosphorus limitation was common in the river. The specific nitrogen ($NH_4$ + $NO_3$) uptake was 12-50 $\mu$mol mg chl-a$^{-1}$ hr$^{-1}$ at two stations, showing substantially higher than for any other freshwaters. The specific nirtogen uptake was higher at the GAR Station of the nitrogen-limited area and this high nirtogen uptake resulted in low $^{14}C:^{15}N$ atomic ratios of algal uptake. Carbon uptake was dependent upon irradiance, decreasing gradually toward the bottom in the euphotic zone, whereas the nitrogen uptake increased slightly toward the bottom. $NH_4$ preferable uptake against $NO_3$ was hardly discemilble due to the fact that it exceeded the $NH_4$ ambient concentraiton. The $^{14}C:^{15}N$ atomic ratios of algal uptake in the surface waters approached the Redfield C:N ratio.
Nutrient Enrichment Bioassays (NEBs) were conducted in the laboratory during June $22{\sim}28$, 2006 in order to determine primary limiting factor on the phytoplankton growth. For the NEBs, the water was sampled using a 10L polyethylene-lined container and dispensed into 2.5L container in the laboratory. The algal growths response in the control (C) and three treatments of phosphorus (P), 2-fold phosphorus (2P), and nitrate nitrogen $(NO_3-N)$ were monitored during 7 days. In the cubitainers which were spiked as P (T1) and 2P (T2) Chl-${\alpha}$ concentrations were decreased during the test period and the final concentrations was low than initial values. However, Chl-${\alpha}$ in the cubitainers which were spiked as $NO_3$(T3) and $P+NO_3$(T4) showed significant increases compared to the initial values, indicating that in the short-term experiments, nitrogen seemed to be a primary limiting nutrient during the periods of NEBs experiment. Long-term ambient nutrient data of TP and TN, and TN:TP mass ratios, however, showed a potential phosphorus limitation on phytoplankton growth and previous other researchers showed a variations of limiting nutrients by nitrogen or phosphorus depending on the seasons sampled and locations. In this study nitrogen as primary limiting nutrient in the NEBs seem to be an seasonal effect rather than the consistent nitrogen limitation.
