Cromakalim Blocks Membrane Phosphoinositide Activated Signals in the Guinea Pig Lung Mast Cells Stimulated with Antigen-Antibody Reactions

  • Cromakalim (BRL 34915), known as an airway smooth muscle relaxant, inhibited the releases of mediators in the antigen-induced mast cell activation. It has been suggested that cromakalim, in part, inhibited mediator releases by inhibiting the initial increase of 1,2-diacylglycerol (DAG) produced by the activation of the other phospholipase system which is different from phosphatidylcholine-phospholipase D pathway. The aim of this study is to further examine the inhibitory mechanism of cromakalim on the mediator release in the mast cell activation. Guinea pig lung mast cells were purified by using enzyme digestion and percoll density gradient. In purified mast cells prelabeled with $[^3H]PIP_2$, phospholipase C (PLC) activity was assessed by the production of $[^3H]$insitol phosphates. Protein kinase C (PKC) activity was assessed by measuring the protein phosphorylated from mast cells prelabeled with $[{\gamma}-32P]ATP$, and Phospholipase $A_2\;(PLA_2)$ activity by measuring the lyso-phosphatidylcholine produced from mast cell prelabeled with 1-palmitoyl-2-arachidonyl $phosphatidyl-[^{14}C]choline$. Histamine was assayed by fluorometric analyzer, and leukotrienes by radioimmunoassay. The PLC activity was increased by activation of the passively sensitized mast cells. This increased PLC activity was decreased by cromakalim pretreatment. The PKC activity increased by the activation of the passively sensitized mast cells was decreased by calphostin C, staurosporine and cromakalim, respectively. The $PLA_2$ activity was increased in the activated mast cells. The pretreatment of cromakalim did not significantly decrease $PLA_2$ activity. These data show that cromakalim inhibits histamine release by continuously inhibiting signal transduction processes which is mediated via PLC pathway during mast cell activation, but that cromakalim does not affect $PLA_2$ activity related to leukotriene release.

Isolation and structure elucidation of a catechin glycoside with phospholipase $A_2$ inhibiting activity from Ulmi cortex

  • 식물에서부터 새로운 소염작용제를 개발하기 위하여 여러식물을 대상으로 염증반응의 초기단계에서 중요한 역할을 하는 것으로 알려진 Phospholipase $A_2$에 저해활성을 갖는 물질을 검색하였고 그 중 강한 억제활성을 보인 유근피에서 유효성분을 분리하였다. 유근피의 에칠 아세테이트 분획에 대하여 실리카겔 크로마토그래피, Sephadex LH-20 크로마토그래피, 분취 박막 크로마토그래피를 수행하여 phenol성 -OH기를 갖는 활성성분인 PSH-II-84-1를 분리하였다. $^1$H-NMR 신호의 양상과 짝지움 상수 값에서 분리된 물질은 (+)-catechin 의 당 유도체로 확인되었다. $^{13}$C-NMR 자료를 분석하여 치환된 당은 D-apiofuranose로 확인되었다. 방향족환의 $^{l3}$C-NMR 신호들은 extended Huekel theory를 응용하여 얻은 net charge 계산 값과 상관시켜 할당하였다. 이상의 구조연구 결과 이 물질은 (+)-catechin-7-0-$\beta$-D-apiofuranoside로 밝혀졌다. (+)-catechin-7-0-$\beta$-D-apiofuranoside의 효소억제활성은 Type II Phospholipase $A_2$에 대하여 $IC_{50}$/이 600$\mu$M이었다.다.

한국산 인삼의 Polyphenol 분획물의 항산화, Phospholipase $A_2$ 및 암세포증식 억제효과 (Antioxidantive, Phospholipase $A_2$ Inhibiting, and Anticancer Effect of Polyphenol Rich Fractions from Panax ginseng C. A. Meyer)

  • 한국산 인삼을 60% acetone으로 추출하여 Sepadex LH-20 gel column chromatography, MCI-CHP 20 gel column chromatography, ${\mu}-Bondapack\;C_{18}$ gel column chromatography을 이용하여 polyphenol 분획물을 3종류를 분리하여 항산화 효과, phospholipase $A_2$ 저해효과 및 암세포 증식억제 효과를 검토하였다. 항산화 효과에서 분획물 I은 150 ppm에서 $Cu^{2+},\;KO_2$ 그리고 $H_2O_2$에 대하여 각각 48.16%, 79.71%, 43.55%를 나타내었고 DPPH 수소공여능은 분획물 II가 200 ppm에서 35.17%의 라디칼소거능을 보였다. 분획물 III는 $Fe^{2+}$, OH가 존재 시 150 ppm에서 48.49%, 25%의 항산화효과를 나타내었다. Phospholipase $A_2$ 저해활성은 분획물 III이 $60\;{\mu}/ml$에서 48.9%의 활성을 나타냈다. HT 29 cell에 대한 인삼의 세포 독성은 분획물 II에서 0.25 mg/ml에서 73.29%의 가장 높은 억제능을 나타내었다.

Inhibitory effects of thromboxane A2 generation by ginsenoside Ro due to attenuation of cytosolic phospholipase A2 phosphorylation and arachidonic acid release

  • Background: Thromboxane A2 ($TXA_2$) induces platelet aggregation and promotes thrombus formation. Although ginsenoside Ro (G-Ro) from Panax ginseng is known to exhibit a $Ca^{2+}-antagonistic$ antiplatelet effect, whether it inhibits $Ca^{2+}-dependent$ cytosolic phospholipase $A_2$ ($cPLA_{2{\alpha}}$) activity to prevent the release of arachidonic acid (AA), a $TXA_2$ precursor, is unknown. In this study, we attempted to identify the mechanism underlying G-Ro-mediated $TXA_2$ inhibition. Methods: We investigated whether G-Ro attenuates $TXA_2$ production and its associated molecules, such as cyclooxygenase-1 (COX-1), $TXA_2$ synthase (TXAS), $cPLA_{2{\alpha}}$, mitogen-activated protein kinases, and AA. To assay COX-1 and TXAS, we used microsomal fraction of platelets. Results: G-Ro reduced $TXA_2$ production by inhibiting AA release. It acted by decreasing the phosphorylation of $cPLA_{2{\alpha}}$, p38-mitogen-activated protein kinase, and c-Jun N-terminal kinase1, rather than by inhibiting COX-1 and TXAS in thrombin-activated human platelets. Conclusion: G-Ro inhibits AA release to attenuate $TXA_2$ production, which may counteract $TXA_2-associated$ thrombosis.

Saucerneol F, a New Lignan Isolated from Saururus chinensis, Attenuates Degranulation via Phospholipase Cγ1 Inhibition and Eicosanoid Generation by Suppressing MAP Kinases in Mast Cells

  • During our on-going studies to identify bioactive compounds in medicinal herbs, we found that saucerneol F (SF), a naturally occurring sesquilignan isolated from Saururus chinensis (S. chinensis), showed in vitro anti-inflammatory activity. In this study, we examined the effects of SF on the generation of 5-lipoxygenase (5-LO) dependent leukotriene $C_4$ ($LTC_4$), cyclooxygenase-2 (COX-2) dependent prostaglandin $D_2$ ($PGD_2$), and on phospholipase $C{\gamma}1$ ($PLC{\gamma}1$)-mediated degranulation in SCF-induced mouse bone marrow-derived mast cells (BMMCs). SF inhibited eicosanoid ($PGD_2$ and $LTC_4$) generation and degranulation dose-dependently. To identify the molecular mechanisms underlying the inhibition of eicosanoid generation and degranulation by SF, we examined the effects of SF on the phosphorylation of $PLC{\gamma}1$, intracellular $Ca^{2+}$ influx, the translocation of cytosolic phospholipase $A_2$ ($cPLA_2$) and 5-LO, and on the phosphorylation of MAP kinases (MAPKs). SF was found to reduce intracellular $Ca^{2+}$ influx by inhibiting $PLC{\gamma}1$ phosphorylation and suppressing the nuclear translocations of $cPLA_2$ and 5-LO via the phosphorylations of MAPKs, including extracellular signal-regulated protein kinase-1/2 (ERK1/2), c-Jun N-terminal kinase (JNK), and p38. Taken together, these results suggest that SF may be useful for regulating mast cell-mediated inflammatory responses by inhibiting degranulation and eicosanoid generation.

위령선약침이 Collagen, Adjuvant, LPS 및 PLA2 유발 류머티스성 관절염에 미치는 영향 (The Effect of $Clematidis$ Radix Herbal-acupuncture Solution, on Collagen, Adjuvant, Lipopolysaccharide and Phospholipase $A_2$ Induced Rheumatoid Arthritis in Mice)

  • Objectives : The purpose of this study is to investigate the effect of $Clematidis$ radix herbal-acupuncture solution, on collagen, adjuvant, lipopolysaccharide and phospholipase A2 induced rheumatoid arthritis in mice. Methods : Arthritis index was measured for mouse that was injected subcutaneously in solution mixed chicken type II collagen with Freund's complete adjuvant. We injected Freund's complete adjuvant into right posterior part of the sole of a ICR mouse foot, which was measured by plethysmometer. The solution mixed $CRHS$ with Tris-HCI, $CaCl_2$, substrate, enzyme was done a chemical action for thirty minutes, and then inhibitory activity of PLA2 enzyme was expressed with inhibition percentage by utilizing isolated arachidonic acid. COX-2 was induced by adding LPS to RAW 264.7 cell, and COX-2 activity was measured by western blot analysis and $PGE_2$ Biotrak kit. Results : $CRHS$ also inhibited Freund's complete adjuvant induced chronic rheumatoid arthritis in mice. $CRHS$ showed significant inhibition of type I and type II $PLA_2$ activities in a dose dependent manner. Furthermore, $PGE_2$ production was decreased with $CRHS$ and lipopolysaccharide-induced COX-2 protein expression was significantly inhibited by $CRHS$. Conclusions : These results suggest that $CRHS$ has an therapeutic effect on drug induced-rheumatoic arthritis by inhibiting $PLA_2$ and COX-2 activities.

넙치 (Paralichthys olivaceus) 뇌로부터 phospholipase D 활성 억제 단백질의 정제 및 특성 규명 (Purification and Characterization of Proteins Inhibiting Phospholipase D Activity from Flounder (Paralichthys olivaceus) Brain)

  • 본 연구에서는 넙치, Pafalichthys olivaceus 뇌 조직에서 인지질가수분해효소 D (phospholipase D, PLD) 활성의 특성 규명 및 이 활성을 억제하는 단백질을 분리 정제하여 그 특성을 규명하였다. 넙치 뇌 조직에서 PLD 활성이 관찰되었으며, 이 활성은 포스파티딜 이노시톨 비스인산염 (phosphatidyl-inositol 4,5-bisphos-phate, $PIP_2$)에 대해서 의존성을 나타내었으나, ADP-rebosylation factor (ARF)에 의해서는 영향을 받지 않았다. PLD 억제물은 넙치 뇌 조직의 세포질 분획물을 사용하여 여러 종류의 칼럼을 통하여 분리 정제하였고, 그 억제물의 분자크기 및 기작의 특성을 규명하였다. 마지막 chromatography을 통하여 여섯 개의 억제를 나타내는 분획물을 얻었으며, 이 중 두 개의 분획물인 IIA IIB는 $PIP_2$-phosphatase activities를 나타내었다. 이 중 IIA 분획물은 면역화학적 분석을 통하여 inositolpolyphosphate 5-phosphatase family로 알려져 있는 신경말단 단백질인 synaptojanin으로 동정되었다. 그리고 IIB fraction은 Superose 12 gel filtration chromatography을 통하여 158-kDa의 크기로 확인되었으며, 이것은 면역화학적 분석을 통하여 synaptojanin과는 별개의 단백질로 판명되었다. 또한, IIB 분획물은 $PIP_2$ phosphatase activity 확인 실험에서 대사산물로서 phosphatidylinositol phosphate (PIP)만을 생성하였다. 이 결과는 IIB 분획물이 $PIP_2$의 4혹은 5 위치의 인산 (phosphate) 중 어느 하나만을 선택적으로 가수분해시킨다는 것을 암시한다. 이상의 연구 결과들을 종합하여 보면, 넙치 뇌 조직에는 다양한 형태의 $PIP_2$-phosphatases가 존재하며, 이들은 $PIP_2$-의존적인 PLD 활성의 억제조절과정에서 중요한 역할을 할 것으로 사료된다.

Improved Therapeutic Profiles of PLA2-Free Bee Venom Prepared by Ultrafiltration Method

  • Bee venom (BV) has long been used in traditional Eastern and Western medicine for chronic inflammation, pain and skin therapy. Human exposure to BV, however, often causes unwanted adverse effects and is even fatal in some cases. Phospholipase $A_2$ ($PLA_2$) of BV is now suspected to play a key role in these adverse effects. We investigated the potential use of $PLA_2$-free bee venom (PBV) as a replacement for BV in cosmetic products. PBV prepared by molecular weight cut-off ultrafiltration exhibits a superior profile in comparison with regular BV, by inhibiting elastase activity and suppressing the induction of nitric oxide (NO) and metalloproteinase-9 (MMP-9), while retaining the effects of cell proliferation and protection against ultraviolet B (UVB)-induced damage in human dermal fibroblast cells. PBV thus appears to be more promising than BV as a cosmetic ingredient with a reduced potential for adverse reactions in the recipient.
