Egg Shell and Oyster Shell Powder as Alternatives for Synthetic Phosphate: Effects on the Quality of Cooked Ground Pork Products

  • This study aimed to determine the optimal ratio of natural calcium powders (oyster shell and egg shell calcium) as synthetic phosphate replacers in pork products. Ground pork samples were subjected to six treatments, as follows: control (-) (no phosphate added), control (+) (0.3% phosphate blend added), treatment 1 (0.5% oyster shell calcium powder added), treatment 2 (0.3% oyster shell calcium powder and 0.2% egg shell calcium powder added), treatment 3 (0.2% oyster shell calcium powder and 0.3% egg shell calcium powder added), and treatment 4 (0.5% egg shell calcium powder added). The addition of natural calcium powders resulted in an increase in the pH values of meat products, regardless of whether they were used individually or mixed. The highest cooking loss was observed (p<0.05) in the negative control samples, whereas the cooking loss in samples with natural calcium powder added was similar (p>0.05) to that in the positive control samples. CIE $L^*$ values decreased as the amount of added egg shell calcium powder increased. CIE $a^*$ values were higher (p<0.05) in samples containing natural calcium powder (treatments 1, 2, 3, and 4) than in the positive control. The combination of oyster shell calcium powder and egg shell powder (treatment 2 or 3) was effective for the improvement of textural properties of the pork products. The findings show that the combined use of 0.2% oyster shell calcium and 0.3% egg shell calcium should enable the replacement of synthetic phosphate in the production of cooked pork products with desirable qualities.

Mutagenic Characterization of a Conserved Functional Amino Acid in Fuculose-1-Phosphate Aldolase from Methanococcus jannaschii, a Hyperthermophic Archaea

  • To elucidate the putative role of the amido group in the metal binding of the fuculose-1-phosphate aldolase from Methanococcus jannaschii, we have examined a potential targen using site-directed mutagenesis. The replacement of asparagine 25 with leucine or threonine was shown to have a negative effect, not only on catlytic efficiency, but also on substrage recognition as well. The Hill coefficient values yeilded a value of =1. All metals used with the wild-type aldolases exhibited higher activity than that of the mutants. The spectra of the mutants were quite different from the wild-type aldolase. A highly conserved amino acid of asparagine 25 in a related family of aldolase odes not appear to provide sufficient evidence for evolution.

Effect of environmental and nutritional conditions on $H_2$ production from glucose by the chemoheterotropic facultative bacterium, Citrobacter sp. Y19

  • Citrobacter sp. Y19 was studied for $H_2$ production from glucose in batch culture. Important conditions studied include phosphate concentration, temperature, glucose concentration, and gas-phase replacement. Optimal $H_2$ production was observed at 140 - 180 mM of phosphate and $36^{\circ}C$. When glucose concentration increased from 0.1 to 5% (w/v), $H_2$ production increased up to 2% and remained constant thereafter. Intermittent purging of the reaction bottle with Ar gas stimulated the $H_2$ production by alleviating the inhibition by $H_2$. The maximum productivity was observed to be 113.2 ml $H_2$/h-1.

  • RhodopseudolllOllas palustris P4 was studied for $H_2$ production from glucose in batch culture. Important conditions studied include phosphate concentration, initial pH, temperature, glucose concentration, and gas-phase replacement. Optimal $H_2$ production was observed at 60 - 300 mM of phosphate and 7.8 - 8.6 of initial pH. The effect of culture temperature was negligible When glucose concentration increased from 0.1 to 5% (w/v), $H_2$ production increased up to 2% and remained constant thereafter. Intermittent purging of the reaction botlle with Ar gas stimulated the Hl production by alleviating the inhibition by $H_2$. The maximum productivity was 111.1 ml $H_2$/h-1.

  • Building structures are generally large in size and have a long life, and the construction of such structures requires the investment of a huge amount of money and social infrastructure. Furthermore, building structures are closely related to people's life. Recently, however, the rapid development of society has been worsening air pollution, which is in turn accelerating the degradation of building structures. Thus, the safety of building structure is emerging as a critical issue. To cope with this problem, the government enacted "The Special Act on Safety Control for Infrastructure" but we need engineers' higher concern over the maintenance and reinforcement of existing structures. Recently researches are being made actively on repair mortar using ultra rapid hardening cement for recovering the performance of structures. The present study conducted an experiment on the basic physical properties of ultra rapid hardening mortar for repairing and reinforcing building structures using magnesia cement and mono-ammonium phosphate. In the experiment, we changed the water-cement ratio and carried out replacement at different ratio of MAP/MgO(%). We used retarder to have working life, and made comparative analysis through evaluating working life and fluidity and measuring strength by age.

  • The hydroxyapatite with different amounts of strontium was synthesized with wet chemical technique. The structural, morphological and chemical characterizations for the prepared hydroxyapatite with various strontium amounts were carried out with XRD, SEM, EDS, and FT-IR. Strontium is quantitatively incorporated into hydroxyapatite where its substitution for calcium provokes a well distribution in the EDS mapping and a linear shift of the infrared absorption bands of the hydroxyl and phosphate groups, coherent with the greater ionic radius of strontium.

In Vitro N-Glycan Mannosyl-Phosphorylation of a Therapeutic Enzyme by Using Recombinant Mnn14 Produced from Pichia pastoris

  • Enzyme replacement therapy for lysosomal storage diseases usually requires recombinant enzymes containing mannose-6-phosphate (M6P) glycans for cellular uptake and lysosomal targeting. For the first time, a strategy is established here for the in vitro mannosyl-phosphorylation of high-mannose type N-glycans that utilizes a recombinant Mnn14 protein derived from Saccharomyces cerevisiae. Among a series of N-terminal- or C-terminal-deleted recombinant Mnn14 proteins expressed in Pichia pastoris, rMnn1477-935 with deletion of N-terminal 76 amino acids spanning the transmembrane domain (46 amino acids) and part of the stem region (30 amino acids), showed the highest level of mannosyl-phosphorylation activity. The optimum reaction conditions for rMnn1477-935 were determined through enzyme assays with a high-mannose type N-glycan (Man8GlcNAc2) as a substrate. In addition, rMnn1477-935 was shown to mannosyl-phosphorylate high-mannose type N-glycans (Man7-9GlcNAc2) on recombinant human lysosomal alpha-glucosidase (rhGAA) with remarkably high efficiency. Moreover, the majority of the resulting mannosyl-phosphorylated glycans were bis-form which can be converted to bis-phosphorylated M6P glycans having a superior lysosomal targeting capability. An in vitro N-glycan mannosyl-phosphorylation reaction using rMnn1477-935 will provide a flexible and straightforward method to increase the M6P glycan content for the generation of "Biobetter" therapeutic enzymes.

Clinical Experience of the Brushite Calcium Phosphate Cement for the Repair and Augmentation of Surgically Induced Cranial Defects Following the Pterional Craniotomy

  • Objective : To prevent temporal depression after the pterional craniotomy, this study was designed to examine the safety and aesthetic efficacy of the brushite calcium phosphate cement (CPC) in the repair and augmentation of bone defects following the pterional craniotomy. Methods : The brushite CPC was used for the repair of surgically induced cranial defects, with or without augmentation, in 17 cases of pterional approach between March, 2005 and December, 2006. The average follow-up month was 20 with range of 12-36 months. In the first 5 cases, bone defects were repaired with only brushite CPC following the contour of the original bone. In the next 12 cases, bone defects were augmented with the brushite CPC rather than original bone contour. For a stability monitoring of the implanted brushite CPC, post-implantation evaluations including serial X-ray, repeated physical examination for aesthetic efficacy, and three-dimensional computed tomography (3D-CT) were taken 1 year after the implantation. Results : The brushite CPC paste provided precise and easy contouring in restoration of the bony defect site. No adverse effects such as infection or inflammation were noticed during the follow-up periods from all patients. 3D-CT was taken 1 year subsequent to implantation showed good preservation of the brushite CPC restoration material. In the cases of the augmentation group, aesthetic outcomes were superior compared to the simple repair group. Conclusion : The results of this clinical study indicate that the brushite CPC is a biocompatible alloplastic material, which is useful for prevention of temporal depression after pterional craniotomy. Additional study is required to determine the long-term stability and effectiveness of the brushite calcium phosphate cement for the replacement of bone.

항바이러스제에 의한 저인산성 골연화증으로 진단된 난치성 요통 (Recalcitrant Low Back Pain Diagnosed as Hypophosphatemic Osteomalacia Induced by Antiviral Medication)

  • We report a rare case of anti-viral agent induced hypophosphatemic osteomalacia presented with localized and radicular pain. A 51-year-old man, who had been taking adefovir for chronic hepatitis, had experienced low back pain radiating to his right thigh for 2 years. With impression of lumbar disc herniation, he underwent magnetic resonance imaging and found multi-level disc herniation with facet joint synovial cysts. He received transforaminal epidural steroid injections, however, symptoms did not improve. To find other possible causes, additional tests were performed. Blood tests revealed hypophosphatemia and increased serum alkaline phosphatase, and osteoporosis was noted in dual-energy X-ray absorptiometry with multiple hot uptakes in bone scan. After replacement of adefovir to entecavir and supplement of phosphate and vitamin D, phosphate level and the clinical symptoms were improved. This is the first to report the presentation of osteomalacia due to anti-viral agent as radicular low back pain with facet synovial cysts.
