Purification and Partial Characterization of Thermostable Carboxyl Esterase from Bacillus stearothermophilus L1

  • A bacterial strain L1 producing a thermostable esterase was isolated from soil taken near a hot spring and identified as Bacillus stearothermophilus by its microbiological properties. The isolated thermostable esterase was purified by ammonium sulfate fractionation, ion .exchange and hydrophobic interaction chromatographies. The molecular weight of the purified enzyme was estimated to be 50,000 by SDS-PAGE. Its optimum temperature and pH for hydrolytic activity against PNP caprylate were $85^{\circ}C$ and 9.0, respectively. The purified enzyme was stable up to $70^{\circ}C$ and at a broad pH range of 4.0-11.5 in the presence of bovine serum albumin. The enzyme was inhibited by phenylmethylsulfonyl fluoride and diethyl p-nitrophenyl phosphate, indicating the enzyme is a serine esterase. The enzyme obeyed Michaelis-Menten kinetics in the hydrolysis of PNPEs and had maximum activity for PNP caproate ($C_6$) among PNPEs ($C_2-C_12$) tested.

  • Leuconostoc citreum is one of the representative strains of Leuconostoc spp. that show fast growth rates in fermented vegetables. Sequential experimental designs including the Plackett-Burman design, fractional factorial design, steepest ascent analysis, central composite design and response surface methodology were introduced to optimize and improve the medium for L. citreum. Fifteen medium ingredients were examined and glucose ($20 g/\ell$), yeast extract ($12.5g/\ell$), sodium acetate trihydrate ($6.12g/\ell$), potassium phosphate ($42.55g/\ell$), and dibasic ammonium citrate ($4.12g/\ell$), were chosen as the best components to give a critical and positive effect for cell-growth. The biomass was increased to ($2.79g/\ell$), (169%), compared to the $1.65g/\ell$ in MRS medium.

  • Riboflavin is a water-soluble vitamin, which serves as a precursor to flavin mononucleotide and flavin adenine dinucleotide. This study aimed to develop a simple and rapid liquid chromatography-tandem mass spectrometry (LC-MS/MS) analysis for the quantification of riboflavin in the Beagle dog plasma. This method utilized simple protein precipitation with acetonitrile and 13C4, 15N2-riboflavin was used as an internal standard (IS). For chromatographic separation, a hydrophilic interaction liquid chromatography (HILIC) column was used with gradient elution. The mobile phase consisted of 0.1% (v/v) aqueous formic acid with 10 mM ammonium formate and acetonitrile with 0.1% (v/v) formic acid. Since riboflavin is an endogenous compound, 4% bovine serum albumin in phosphate buffered saline was used as a surrogate matrix to prepare the calibration curve. The quantification limit for riboflavin in the Beagle dog plasma was 5 ng/mL. The method was fully validated for its specificity, sensitivity, accuracy and precision, recovery, and stability according to the US FDA guidance. The developed LC-MS/MS method may be useful for the in vivo pharmacokinetic studies of riboflavin.

  • By the successive enrichment culture, methanol-utilizing bacteria of 213 strains were isolated from soil samples collected from various places. Among them one strain showing excellent growth was selected. The organism isolated was obligate methylotroph and identified as Methylomonas methanolica on the basis of its mophological and physiological characteristics of the cell. The medium have been to be collected for the maximum biomass productivity. The microorganism was capable of growing satisfactorily on a medium containing only methanol 0.8% (v/v), ammonium sulfate 0.6%, magnesium sulfate 0.1%, phosphate salts, but did not require growth factor.

  • The optimal culture conditions on bacteriocin producing of Lactobacillus sp. FF-3 isolated from Korean Dongchimi, were studied for enhancing its production with regard to environmental and nutritional factors. The optimal cultivation time, initial pH and temperature were 21 hours, pH 7.0 and 30∼37$^{\circ}C$ respectively. Optimal compositions of culture medium for bacteriocin production were glucose 3% as carbon source, tryptone 4% as nitrogen source, and manganese sulfate 0.005% as inorganic salt with other basal components. The maximum antimicrobial activity was 484 BU/mL under the optimal culture condition.

The Survey of Urinary Calculi Following Long Feeding Period in Korean Native Steers (거세한우 장기비육에 따른 비뇨기결석 발생조사)

  • We surveyed the incidence of urinary calculi for feeding period, urinary lesion, composition of calculi and changes of serum chemistry with Korean native castrated steers, which are growing until 26 through 31 months. Without showing any clinical signs such as urethral obstruction, hydronephrosis and strangury for all group, $37.8\%$ (34 out of 90) of Korean native steers carried urinary calculi in carcass. The incidence of urine calculi far different feeding stages from 26 to 31 months were observed as $13.3-60.0\%$er, no significant differences were detected. The distribution of calculi were kidney $(24.4\%)$, kidney and bladder $(10\%)$ and kidney, bladder and urine $(3.3\%)$. Congestion of urinary bladder was shown in $26.6\%$ of Korean native steers. Korean native steers with urinary calculi $(41.2\%)$ had more possibilities (P<0.05) to bring outbreaks of congestion of urinary bladder than those of without calculi $(17.8\%)$. The main composition of calculi were struvite (magnesium ammonium phosphate crystals), calcium phosphate, calcium carbonate, calcium oxalate, non-oxalate calcium, ammonia and phosphate. The distribution of urine pH was $71.7\%$(above pH 8.0), $12.2\%$ (between pH 7.0 and pH 8.0) and $16.6\%$ (under pH 7.0). Like shown in herbivores, most of the urine pH was distributed as alkaline urine. Serum creatinine was shown significant difference (P<0.05) according to incidence of calculi but other serum chemistry didn't show any difference in serum chemistry. These results suggest that feeding until 26 to 31 months in Korean native steers castrated at 6 months of the age elicits urinary calculi and congestion of urinary bladder, but not clinical abnormality. And adjustment of the dietary Can ratio to a level of 2:1 or greater during feeding period may reduce the possibility of incidence of urinary calculi in Korean native steers.

Analysis for benzimidazoles in eggs (식용란중 벤지미다졸계 구충제 분석)

  • This stud was attempted to detect six benzimidazoles[thiabendazole(TBZ), oxfendazole(OFZ), mebendazole(MBZ), flubendazole(FLBZ), albendazole(ABZ), and febendazole(FBZ)] in eggs using high performance liquid chromatography(HPLC) with photodiode array detector(DAD) simultaneously. The eluates were determined by HPLC on a waters X-Terra$^{TM}\;C_{18}$ reverse-phase column($4.6{\times}250nm,\;5{\mu}m$) with DAD at 295nm. The mobile phase was 0.04M ammonium phosphate(pH 7.5)/ACN(62.28 v/v) run isocratically. The calibration curves were linear(r>0.999) for six benzimidazoles. Average recovery rate from eggs were 94.54% for TBZ, 97.71% for OFZ, 88.82% for MBZ, 93.71% for FLBZ, 78.61% for ABZ, and 75.09% for FBZ, respectively. The limits of detection and quantification were 2.27ppb, 3.88 for TBZ, 9.34ppb, 15.61ppb for OFZ, 28.53ppb, 45.15ppb for MBZ, 27.39ppb, 40.95ppb for FBZ, 8.61ppb, 13.95ppb for ABZ and 12.79ppb, 22.34ppb for FBZ, respectively.

Effect of $Cr^{6+}$ Stress on Photosynthetic Pigments and Certain Physiological Processes in the Cyanobacterium Anacystis nidulans and Its Chromium Resistant Strain

  • A MNNG (N-methyl-N'-nitro-N-nitrosoguanidine) induced chromium resistant strain ($Cr^{r}18$) of unicellular cyanobacterium Anacystis nidulans has been isolated and characterized. The resistant strain could grow (although restricted to $50\%$ of control) in chromium concentration (180${\mu}M$) lethal to the wild-type. Sublethal ($160{\mu}M$) concentration of $Cr^{6+}$ significantly reduced (13-$37.5$) all the photosynthetic pigments of A. nidulans with maximum reduction in phycoerythrin followed by ChI $\alpha$. Pigments of A. nidulans were drastically decreased in lethal concentration of Cr^{6+} with maximum reduction in phycoerythrin ($75\%$) and allophycocyanin ($67.5\%$). Resistant strain $Cr^{r}18$ resisted toxic effects of sublethal and lethal concentrations of $Cr^{6+}$ on photosynthetic pigments as revealed by less decrease in pigments as compared to A. nidulans. Effect of $Cr^{6+}$ stress was also studied on nitrogen assimilation and phosphate uptake. Sublethal concentration of $Cr^{6+}$ drastically reduced ($71.5\%$) nitrate uptake by A. nidulans while a decrease of $29\%$ was observed in strain $Cr^{r}18$. Short (2 day) exposure of A. nidulans and its resistant strain $Cr^{r}18\;to\;Cr^{6+}$ did not affect nitrate reductase and glutamine synthetase (transferase), whereas longer (10 day) exposure to $Cr^{6+}$ lowered activities of both enzymes in A. nidulans but not significantly in the strain $Cr^{r}18$. Ammonium uptake by both strains was not affected by $Cr^{6+}$. Thus, $Cr^{6+}$ affected photosynthetic pigments, nitrogen assimilation, and phosphate uptake of A. nidulans, while strain $Cr^{r}18$ was able to resist toxic effects of the metal. Advantages of using strain $Cr^{r}18$ for bioremediation purposes have been evaluated by studying $Cr^{6+}$ removal from the solution. Resistant strain $Cr^{r}18$ was able to remove $33\%$ more $Cr^{6+}$ than A. nidulans and thus it can prove to be a good candidate for bioremediation of $Cr^{6+}$ from polluted waters.

  • This study was carried out to investigate the influence of chemical type and its retention in the fire-retardant treated plywoods on the static bending strength, a property peculiar to plywood. Being soaked in 20% aqueous solution of $(NH_4)_2SO_4$, $NH_4H_2PO_4$, $(NH_4)_2HPO_4$. Borax-Boric acid and Minalith for 3 to 12 hours at three-hour intervals and redried at $120^{\circ}C$ in hot press, the treated plywoods were put to static bending test. The values of chemical treated plywoods in Stress at proportional limit, Modulus of elasticity, Modulus of rupture and Work per unit volume to proportional limit were widely higher than those of water treated plywoods(control) and Borax-Boric acid treatment showed the highest value in the four mechanical data. And the bending strength of fire-retardant treated plywoods increased with the extension of soaking time or the increase of chemical retention in themselves. Borix-Boric acid was the desirable fire-retardant for thin plywood in view of mechanical strength and soaking defects in this study.

Changes in The Sensitive Chemical Parameters of the Seawater in EEZ, Yellow Sea during and after the Sand Mining Operation (서해 EEZ 해역에서 바다모래 채굴에 민감한 해양수질인자들)

  • Eight comprehensive oceanographic cruises on a squared $30{\times}30\;km$ area have been made to investigate the short and long-term impacts on the water qualities due to the sand mining operations at Exclusive Economic Zone (EEZ) in the central Yellow Sea from 2004 to 2007. The area was categorized to 'Sand Mining Zone', 'Potentially Affected Zone', and 'Reference Zone'. The investigation covered suspended solids, nutrients (nitrate, nitrite, ammonium, phosphate), and chlorophyll-a in seawater and several parameters such as water temperature, salinity, pH, and ORP. Additionally, several intensive water collections were made to trace the suspended solids and other parameters along the turbid water by sand mining activities. The comprehensive investigation showed that suspended solids, nitrate, chlorophyll-a and ORP be sensitively responding parameters of seawater by sand mining operations. The intensive collection of seawater near the sand mining operation revealed that each parameter show different distribution pattern: suspended solids showed an oval-shaped distribution of the north-south direction of 8 km wide and the east-west direction of 5 km wide at the surface and bottom layers. On the other hand, phosphate showed so narrow distribution not to traceable. Also ammonium showed a limited distribution, but its boundary was connected to the high nitrate and chlorophyll-a concentrations with high N/P ratios. From the last 4 years of the comprehensive and intensive investigations, we found that suspended solids, ammonium, nitrate, chlorophyll-a, and ORP revealed the sensitive parameters of water quality for tracing the sand mining operations in seawater. Especially suspended solids and ORP would be useful tracers for monitoring the water qualities of remote area like EEZ in Yellow Sea.
