Recently, according as people who have sensitive skin increase, we've been giving more importance to the safety of cosmetics. Especially, preservative is known to be one of the main stimuli which cause side-effects of cosmetics. However, there have been few reports describing cell cytotoxicity, skin penetration, oil-aqueous phase partition, anti-microbial activity of preservatives and their correlation with skin irritation. The study is aimed to develop low irritable preservative system with phenoxyethanol, one of the most commonly used preservatives in cosmetics, considering various factors mentioned above. According to our results of cell cytotoxicity against human normal fibroblasts by means of MTT assay, phenoxyethanol showed the lowest cytotoxicity when compared to other preservatives tested (cytotoxicity: pro-pylparaben > butylparaben > ethylparaben > methylparaben > triclosan > phenoxyethanol), but human patch test for assessing shin primary irritation revealed that phenoxyethanol has higher skin irritation than methylparaben and triclosan. We performed in vitro skin penetration test using horizontal Franz diffusion cells with skin membrane prepared from hairless mouse (5 ${\~}$ 8 weeks, male) to evaluate the rate of skin penetration of preservatives. From the results, we found that the higher irritable property of phenoxyethanol in human skin correlates with its predominant permeability (skin penetration: phenoxyethanol > methylparaben > ethylparaben > propylparaben > butylfaraben > triclosan). Therefore, we made an effort to reduce skin permeability of phenoxyethanol and found that not only the rate of skin penetration of phenoxyethanol but also its skin irritation is dramatically reduced in formulas containing oils with low polarity. In the experiments to investigate the effect of oil polarity on the oil-aqueous phase partition of phenoxyethanol, more than $70\%$ of phenoxyethanol was partitioned in aqueous phase in formulas containing oils with low polarity, while about $70 {\~} 90\%$ of phenoxyethanol was partitioned in oil phase in formulas containing oils with high polarity. Also, in aqueous phase phenoxyethanol showed greater anti-microbial activity. Conclusively, it appears that we can develop less toxic preservative system with reduced use dosage of phenox-yethanol and its skin penetration by changing oil composition in formulas.
We investigated whether ${\beta}$-galactosidase (${\beta}$-gal)-containing Escherichia coli cells could transfer a galactose to 2-phenoxyethanol, resulting in 2-phenoxyethanol galactoside (PE-Gal). PE-Gal was confirmed by liquid chromatography-mass spectrometry. In addition, we also confirmed that a galactose molecule was covalently bonded with PE during thin-layer chromatography analysis of the ${\beta}$-gal hydrolysate of PE-Gal. The yield for PE-Gal synthesis was about 37.5% (weight basis), which was about 7-8 times greater than that of a previous report. In addition, the concentration of ${\beta}$-gal (0.96 U/ml) used in this PE-Gal synthesis was about 20 times less than that in a previous report.
The efficiency of clove oil, MS-222, and 2-phenoxyethanol was evaluated as anesthetics in juvenile Scomber japonicus. Stage A5 of anesthesia was assumed to be sufficient for conducting routine aquaculture procedures in less than 3 min, with recovery (stage R5) in less than 5 min. The lowest effective doses of the three anesthetics were 50 mg $L^{-1}$ clove oil (anesthetic time of 71.3 s and recovery time of 167.0 s), 100 mg $L^{-1}$ MS-222 (anesthetic time of 70.7 s and recovery time of 115.7 s), and 400 mg $L^{-1}$ 2-phenoxyethanol (anesthetic time of 86.7 s and recovery time of 95.0 s). Anesthetic times decreased with increasing doses for all three anesthetic agents, and fish anesthetized with clove oil exhibited the longest recovery times. After 30 min, the highest plasma cortisol and lactate levels were detected with the use of clove oil, whereas the lowest values were observed with 2-phenoxyethanol. In addition, high glucose levels were maintained during recovery with clove oil, but the treatments did not significantly differ. The most effective of the three anesthetic agents was 2-phenoxyethanol, although all were considered acceptable for use in cultures of juvenile Scomber japonicus.

We investigated the growth-inhibitory activities of cosmetics and their preservatives against pathogens and resident skin bacteria. Of the tested cosmetics, preservatives such as parabens, 1,2-hexanediol, phenoxyethanol-contained toner, emulsion, cream and baby cream exhibited potent antibacterial effects against Staphylococcus aureus, Escherichia coli and Pseudomonas aeruginosa. Parabens, 1,2-hexanediol and phenoxyethanol inhibited the growth of pathogens, as well as skin-resident bacteria such as Staphilococcus epidermidis, Shigella flexneri, Enterobacter aerogenes and so on. The application of a basic cream containing phenoxyethanol to human skin was shown to disturb the skin microbiota: at the phylum level, Proteobacteria increased and at species level, 4P004125_s increased and Propionibacterium humerusii decreased. Based on these findings, parabens, 1,2-hexanediol and phenoxyethanol have antimicrobial activity and cosmetics containing phenoxyethanol may disturb skin microbiota.

This study was performed to develop a new natural antimicrobial materials by analyzing the effect of extracts obtained from Ten Lauraceae Species on the inhibitory activity against Propionibacterium acnes. The plant materials were collected from Wando and Jeju islands, and the antimicrobial activity of the crude extracts was examined by the agar diffusion method with different part (i.e., leaf and branch), solvents (i.e., distilled water, 80% ethanol, and 100% methanol) and at different ultrasonic extracting times (i.e., 15, 30, and 45 minutes). The control agents used were synthetic antimicrobials, methylparaben and phenoxyethanol, at concentrations of 0.4, 1, 2, and 4 mg/disc. Altogether, extracts of 10 species used in the study showed inhibitory activity, which confirmed their antimicrobial action against acnes. Among these, leaves of Laurus nobilis L. which was extracted in 80% ethanol for 45 min showed the largest clear zone (19.8 mm). Leaves of L. nobilis L., showing highest antimicrobial activities among 10 species, were successively reextracted with n-hexane, chloroform, ethylacetate and n-butanol. As a results, in all fractions except butanol, clear zone above 10 mm were formed. The ethyl acetate fraction showed the highest inhibitory activity (13.3 mm) and the inhibitory activity was significantly higher than that of crude extract (10.2 mm) and phenoxyethanol as a control (12.5 mm).

Sensory irritation is one of the important side effects of cosmetics and it is required to develop new products that are more tolerable to the consumers. There are lots of cosmetic ingredients known to induce sensory irritation such as lactic acid, glycolic acid, ethanol, preservatives, fragrances and menthol etc. It is also known that sensory irritation increases by change of pH as well as additional occlusive conditions. The aim of this study is to know various factors affecting sensory irritation due to preservatives (methylparaben, propylparaben, phenoxyethanol and chlorophenecin). We also wanted to investigate the effect of preservatives to sensory irritation according to change of formulations. Our results showed that sensory irritation increased with the conditions of increasing absorption such as packs. We have also found that sensory irritation increased synergistically when to apply two different preservatives together. For example, phenoxyethanol and chlorphenesin themselves have weak sensory irritation potentials, but we observed phenoxyethanol combined with chlorpenesin synergistically increase of sensory irritation potentials. In conclusion, formulation and combination of different preservatives should be considered to reduce the unwanted sensory irritation of preservatives.
In this study, partition coefficients(Kw) of phenoxyethanol in oil-water phase according to type of oils was investigated. Partition coefficients(Kw) were experimented by classified oils that hydrocarbones, fatty alcohols, fatty acids, esters, triglycerides and silicones. It was found that partition coefficients(Kw) of hydrocarbons and silicones were low, but partition coefficients(Kw) of fatty acids, fatty alcohols, esters and triglycerides were high. It was known that the emulsions which were made of oils having low partition coefficients value had a good antimicrobial effects. Thus, the cosmetics stability of microorganisms and skin safety of phenoxyethanol could be improved by using the oils which have low partition coefficients value.

We investigated the purifications of PE-gal and CPN-gal, synthesized by transgalactosylation reaction using recombinant ${\beta}$-gal. The reaction mixture containing PE and PE-gal was first mixed with EA, and thereafter PE and PE-gal were distributed in two-phase (EA/water) system. In this system, PE and PE-gal was selectively moved into EA and water phase, respectively. Then, the water phase was collected, and silica gel chromatography was carried out using the collected water phase. Finally, we compared two purified PE-gal samples using HPLC and TLC analysis, in which the one was purified only by silica gel chromatography and the other was purified by EA extraction followed by silica gel chromatography. In the latter case, the residual PE was almost completely removed, whereas, in the former case, the residual PE was remained remarkably. Additionally, the purification yield of PE-gal was about 21% on the basis of mole. In the same purification protocol, CPN-gal was able to be purified using EA extraction followed by silica gel chromatography, in which the residual CPN was almost removed when CPN-gal was purified by EA extraction followed by silica gel chromatography.
Three synthetic chemicals, benzoyl chloride, 2-propyn-l-ol, and 2-phenoxy ethanol were selected for genotoxicity testing, based on production quantity and available genotoxic data. In our previous report, benzoyl chloride induced chromosomal aberrations in Chinese hamster lung (CHL) fibroblast in vitro with and without metabolic activation, while 2-propyn-l-ol and 2-phenoxy ethanol induced only with metabolic activation. To compare the genotoxicity of chromosome aberration assay, the single cell gel electrophoresis (comet) assay subjected using CHL cells. As a result, statistically significant differences of tail moment values of benzoyl chloride, 2-propyn-1-ol, and 2-phenoxy ethanol were observed compared with control values on almost all concentrations with S9 or without S9 metabolic activation system. This results suggest that genotoxic results of the comet assay and the chromosome aberration assay show correlationship of genotoxicity in the CHL fibroblast. In summary, the positive result of chromosome aberration of benzoyl chloride, 2-propyn-l-ol, and 2-phenoxy ethanol was also induced DNA damages in comet assay with same cell line. Consequently, comet assay will be useful and more accurate tool to detect and to confirm the genotoxicity especially DNA damages in CHL fibroblast.

The productionof the highly relible softwae systems and theirs performance evaluation hae become important interests in the software industry. The software evaluation has been mainly carried out in ternns of both reliability and performance of software system. Software reliability is the probability that no software error occurs for a fixed time interval during software testing phase. These theoretical software reliability models are sometimes unsuitable for the practical testing phase in which a software error at a certain testing stage occurs by causes of the imperfect debugging, abnornal software correction, and so on. Such a certatin software testing stage needs to be considered as an outlying stage. And we can assume that the software reliability does not improve by means of muisance factor in this outlying testing stage. In this paper, we discuss Bavesian software reliability growth modeling and estimation procedure in the presence of an imidentitied outlying software testing stage by the modification of Jehnski Moranda. Also we derive the Bayes estimaters of the software reliability panmeters by the assumption of prior information under the squared error los function. In addition, we evaluate the proposed software reliability growth model with an unidentified outlying stage in an exchangeable model according to the values of nuisance paramether using the accuracy, bias, trend, noise metries as the quantilative evaluation criteria through the compater simulation.
