Selection of bactericides for control of potato Blackleg disease in Korea (감자흑각병 (Potato Blackleg Disease) 방제를 위한 살균제 선발)

  • Potato blackleg disease caused by Erwinia carotovora subsp. atroseptica has been a serious problem in Korea. It was previously reported that four mixtures [streptomycin (9.3 ppm) + copper oxide (171.6 ppm)/copper hydroxide (146.3 ppm), streptomycin sulfate (7.0 ppm)+copper oxide (171.6ppm)/copper hydroxide (146.3 ppm)] were effective for the control of E. carotovora subsp. atroseptica. in in vitro test. Using those four mixtures and two antibiotics [streptomycin (81.4 ppm) and streptomycin sulfate (61.3 ppm)], the effectiveness of control for E. carotovora subsp. atroseptica. was conducted in the field. Two antibiotics showed over 60% of control efficacy under different soil conditions, while mixtures of two antibiotics with copper compounds did not show any control effects on the infected seed potato. Two mixtures [streptomycin (27.9 ppm)+copper hydroxide (438.9 ppm), streptomycin sulfate (21.0 ppm) + copper oxide (514.8 ppm)] were effective in the control of potato blackleg disease on the infected potato plants under different climate conditions.

Comparison of Acute Symptoms Between a Alpine Agricultural Workers and General Workers in Gangwon-do (강원도 일부지역 고랭지 농업 농민과 일반 농업 농민의 농약중독 증상 비교)

  • Objectives: This study was performed to evaluate the pesticide exposure status and acute pesticide poisoning symtoms among agricultural workers at Gangwon-do province alpine area. Alpine area was defined as the area higher than 400m. Methods: We analyzed 257 interviwed questionnaire about pesticide exposure, acute pesticide poisoning symptoms and other variables. Results: The result was shown that agricultural workers at alpine area used more pesticide than general agricultural workers for annual usage days(35.9 days vs 14.4 days), daily usage hours(6.7 hrs vs 2.8hrs, p<0.05). But there was no difference between general and alpine agricultural worker's herbicide exposure. Moreover, the alpine agricultural worker's acute pesticide poisoning symtom score was higher than general agricultural worker's. These difference was also found at result of regression analysis, under control the age, sex, monthly income. Most frequently suffered symptom was headache and dermatological problems. The symptom prevalence of dermatological problem, headache, general weakness, eye irritation, nausea were higher among alpine agricultural workers than general agricultural workers. Conclusions: As a result, agricultural workers at alpine area were more exposed to pesticide and suffered from pesticide poisoning symptoms. To prevent the symptoms and disease from pesticide exposure among agricultural workers at alpine area, more research and political effort will be needed.

Development of an Official Analytical Method for Determination of Imazapyr in Agricultural Commodities using HPLC-UVD (HPLC-UVD를 이용한 농산물 중 Imazapyr의 공정분석법 확립)

  • A chromatographic method for the determination of imazapyr, a non-selective herbicide, in agricultural commodities was developed to use safety control of pesticide residue on crops, and was fully validated as an official method for residue analysis. Agricultural commodities, mandarin (fruit), hulled rice (cereal grains), pepper (vegetables), potato (potatoes) and soybean (beans) were extracted with methanol and partitioned with dichloromethane to remove the interference obtained from sample extracts, adjusting pH to 2.5 by 4N hydrochloric acid. Finally, they were analyzed by high performance liquid chromatography coupled to UV detector (HPLC-UVD). The developed method had the linearity in the range of test concentrations with coefficients of determination ($r^2$) more than 0.99. Recovery studies were carried out at three concentration levels (LOQ, 10LOQ, and 50LOQ) performing five replicates at each level. Recoveries were ranged between 72.1 to 108.0%, with relative standard deviations less than 10%. A consistent recovery was determined according to the CODEX guidelines (CAC/GL40, 2003). Finally, LC/MS with selected ion monitoring was also applied to confirm the suspected residues of imazapyr in agricultural samples. This developed method for determination of imazapyr residues in agricultural commodities. can be used as an official method.

Control Effects of different Concentrations and Mixtures of the commercial Bacillus thuringiensis Products against Moths (Bacillus thuringiensis 제품의 농도 및 혼합 조건에 따른 나방류 방제 효과)

  • Beet armyworm (Spodoptera exigua), diamondback moth (Plutella xylostella) and tobacco cutworm (S. litura) are insect pests causing damage in many economically important vegetables in Korea. Bacillus thuringiensis, commonly known as Bt, has been available as an alternative insecticide for many years. Five commercial Bt products and the mixtures evaluate the control efficacy against three kinds of moths. These commercial Bt products had high control efficacy against $2^{nd}$ instar of diamondback moth, but didn't show high mortality against beet armyworm and tobacco cutworm. Mixtures of Bt products didn't have synergistic effects to the tested moths. Also application of twice and four times the recommended concentration didn't improve the control effects against the tested three species of moths.

Effects of Minute Pirate Bug, Orius strigicollis (Hemiptera: Anthocoridae) on Control of Thrips on Hot Pepper in Greenhouse (하우스고추에서 Orius strigicollis Poppius를 이용한 총채벌레류 방제효과)

  • Biological control of thrips with Orius strigicollis Poppius was evaluated in a hot pepper greenhouse. To study the biological control strategy of thrips on hot pepper grown in greenhouse, three plots were established: natural enemy removed plot(CNT), pesticide treated plot(PAT) and O. strigicollis released plot with no pesticide application (NRT). The nymphs of O. strigicollis were found on the leaves of lower stratum in CNT and NRT at 13 days after the first release. The density of thrips was suddenly dropped and maintained at low level since 15 day after transplanting in NRT. The densities of thrips on hot pepper flowers was also maintained at much less level in NRT than in CNT. The thrips and O. strigicollis, which were aggregated in flowers, may have resulted in improved predator searching. Average damage indices of CNT, PAT and NRT were 0.31, 0.05 and 0.08 and the percentage of damaged fruit were 80.0, 17.1 and 24.8% respectively. The damage index and percentage of damaged fruit of NRT was slightly higher than PAT, but very lower than CNT. The introduction of the adults of O. strigicollis, which was the biological control agent for the control of thrips, was effective on hot pepper.

Immunoassay for Monitoring Pesticide Contamination in Agricultural Products

  • Much of the increase in agricultural productivity over the past half century has been due to the control of the pests with synthetic pesticides. The use of these pesticides has caused environmental problems and public health concern. The guidelines of maximum residue levels of pesticides in agricultural products has been well documented but more careful monitoring of their residues is required. Pyrethorid class pesticides are dominant in modern agricultural industry but public health concerns have been recently considered. The major route of pesticide exposure is the diet and with improved surveillance of pyrethorid residues in agricultural products their exposure should be controlled and minimized. In suitable products with reduced matrix effects such as agricultural products, aqueous samples, fruits and vegetables the use of immunoassays for pyrethorid residue monitoring could satisfy this requirement. Immunoassays have several advantages, namely they are highly sensitive, selective and cost-effective and enable large-scale sample handling and analysis in the laboratory.

Effect of Aqueous Solutions of Pesticides on Survival of Entomopathogenic Nematodes (농약이 곤충병원성 선충의 생존에 미치는 영향)

  • The toxic effects of four pesticides on the entomopathogenic nematodes. Heterorhabditis bacteriophora NC strain, Steinernema glaseri NC and Dongrae strain were tested by checking the mortality of infective juveniles(Ijs) in aqueous solution of pesticide. Mortality of Ijs was influenced by pesticide and concentration and soaking time. The herbicide alachlor and insecticide fenitrothion were toxic to Ijc of three entomopathogenic nematodes. But, the fungicides mepronil and tolclofos-methyl were nontoxic to Ijs. Tolclofos-methyl showed significantly very week toxicity at 28 days after soaking for S. glaseri NC strain. H. bacteriophora NC strain was more highly sensitive to high temperature condition then were the other nematodes.

Artificial diminution of the residual pesticides on horticultural crops using photosensitizers (감광제에 의한 원예작물중 잔류농약의 인위적 경감)

  • Photosensitizing activities of some photosensitizers (PS) for the artificial diminution of pesticide residues on horticultural crops were investigated. Five fungicides, iprodione, bitertanol, chlorothalonil, myclobutanil, and dichlofluanid were sprayed on apple and cucumber, followed by the application of each selected photosensitizer, and the samples were collected 0, 1, 3, 7, 15 days after the photosensitizer application and analyzed for the residual amounts. Of the 40 photosensitizers tested, six selected on the basis of the eliminating effect of pesticide residues were PS-1 (aromatic ketone), PS-2 (aromatic amine), PS-3 (quinone), PS-4 (inorganic compound), PS-5 (organic acid salt), and PS-6 (semiconductor photocatalyst). The residual amount of iprodione after 15 days of the application of PS-1 was 74% of that of the control. For bitertanol, the residual amount after 15 days of the application of PS-1 accounted for 78% of that of the control. The residual amounts of chlorothalonil after 1 day of the application of PS-1 and PS-2 accounted for 56 and 54% of those of the control, respectively. The residual amounts of iprodione on cucumber after 3 days of the application of the photosensitizers PS-1 and PS-2 were 44 and 67% of those of the untreated control, respectively. For myclobutanil, the residual amount after 15 days of the application of PS-6 accounted for 45% of that of the control. In case of dichlofluanid, the residual amount after 3 days of the application of PS-1 accounted for 44% of that of the control. Based on the results, PS-1 turned out to be the most promising photosensitizer for the accelerated photodegradation of the above fungicides on apple and cucumber.

Control Effect of Environmental-friendly Organic Materials against Major Pear Diseases (친환경 유기농자재를 이용한 배 주요병해 방제효과)

  • This study was conducted to investigate the control effects of the environmental-friendly materials against major diseases of pear and to elucidate factors related to spore disperse of pear scab and pear rust. Spore disperse was deeply related ambient temperature in the early stage of disease occurrence and the maximum spore disperse was affected greatly by the air humidity, Percent of infected leaves of pear scab in plot treated with Trichoderma harzianum-YC459 was 29.3% and 43.3% in plot treated with Sulfur-80%. Two formulations above-mentioned were less effective to control pear scab than the chemical pesticide, hexaconazole. Control effect in plots single- and mixed-treated with Trichoderma harzianum-YC459, Sulfur-80% and hexaconazole against pear scab were compared with that in plot treated with chemicals by conventional application schedule in pear orchard. The incidence of pear scab was lower in plot treated with the environmental-friendly materials than that of untreated plot. However there was no significant difference among the treatments at the 5% level. Application of Sulfur-80% reduced significantly the incidence of pear scab (7.8%) compared to that (10.3%) in plot treated by conventional application schedule in organically cultivated pear orchard. Alternative application of Trichoderma harzianum-YC459 and Sulfur-80% (4.3%) suppressed the incidence of pear rust than that (7.1%) in plot treated by conventional application schedule in organically cultivated pear orchard.
