Gum류의 연고제제와 흡수촉진제가 Riboflavin의 경피흡수에 미치는 영향 (Transdermal Permeation of Riboflavin in Ointment Bases Using Gums & Enhancers)

  • We investigated characters of transdermal therapeutic system(TTS) and the skin permeability of that with applying drug delivery system(DDS). Natural gums were selected as material of TTS. The permeation of natural gums ointment containing drug in rat skin using diffusion cell model. Permeation properties of materials were investigated for water soluble drug such as riboflavin in vitro. We used glycerin, PEG 600 and oleic acid as enhancers. Since dermis has more hydration than the stratum corneum, skin permeation rate at steady state was highly influenced when glycerin was used in riboflavin. The permeation rate of content enhancer and drug was found to be faster than that of content riboflavin only. These results showed that skin permeation rate of drug across the composite was mainly dependent on the property of ointment base and drug. All the gum ointment tested showed good safety. Proper selection of the materials which resemble and enhance properties of the delivering drug was found to be important in controlling the skin permeation rate.

In vitro 경피흡수 실험시 Donor와 Receptor용액중의 글리세린과 PEG 400이 약물의 경피투과도에 미치는 영향 (Effects of Glycerin and PEG 400 in Donor and Receptor Solutions upon Skin Permeation of Drug)

  • Effects of glycerin and PEG 400 in donor and receptor solutions upon skin permeation of drug were investigated. Deoxycortisone was used as a model compound. In vitro skin permeation study with freshly excised hairless mouse skin was performed and the steady-state skin permeation rates of the drug were determined in different fractions of glycerin or PEG 400 in donor and receptor solutions. Glycerin in donor solution didn't show any effect on the skin permeation rate of deoxycortisone. However glycerin in receptor solution showed significant effect on the skin permeation rate of the drug. In glycerin, there's a critical concentration for balancing hydration and dehydration of skin. At low concentration, less than 20 %, glycerin showed the enhancement of the flux due to the hydration effect of skin. At high concentration, more than 30 %, glycerin retard the permeation rate which might be due to the dehydration effect on the dermis layer. Since dermis has more water content than the stratum corneum, the steady state skin permeation rates were more influenced when glycerin was in receptor solution than that of in donor solution. PEG 400 aqueous solutions doesn't affect the steady state permeation rate of deoxycortisone significantly.

질산술코나졸겔의 제조 및 약물방출에 관한 연구 (Study on Preparation and Drug Release of Sulconazole Nitrate Gels)

  • Sulconazole nitrate(SCN), an imidazole derivative which has been effective in the treatment of dermatophytosis, tinea versicolor and candidiasis, was formulated as a gel containing drug, poloxamer 407, ethanol and propylene glycol. The resulting SCN gels were evaluated with respect to their viscosity, drug release rate, skin permeation rate. The apparent viscosity of SCN gel increased in proportion to poloxamer 407, drug and propylene glycol concentration. In case ethanol was added, the apparent viscosity decreased. The drug release rate of SCN gel increased in proportion to temperature and ethanol concentration. But the drug release rate decreased as the concentration of poloxamer 407 increased. The increase of drug concentration induced nonlinear increase of drug release rate. When propylene glycol was added at the level of 10%, the drug release rate increased but from 15% it decreased. The skin permeation rate decreased in high concentration of poloxamer 407. The skin permeation rate of SCN gel containing 15% ethanol increased about twice than that of gel without ethanol. The increase of drug concentration induced nonlinear increase of skin permeation rate. When propylene glycol was added at the level of 10%, the skin permeation rate increased but from 15% it decreased.

항에이즈 약물의 경피흡수에 미치는 합성고분자 멤브레인의 영향 (The Effect of Synthetic Polymer Membranes on the Skin Permeation of Anti-AIDS Drugs)

  • The effect of synthetic polymer membranes on the permeation rate of dideoxynucleoside-type anti-HIV drugs through hairless rat skin was studied using ethylene/vinyl acetate copolymer (EVA) and ethylene/methyl acrylate copolymer (EMA) membranes fabricated by solvent casting method. In vitro skin permeation kinetics study of DDC (2',3'-dideoxythymidine), DDI (2',3'-dideoxyinosine) and AZT (3'-azido-3'-deoxythymidine) across the (membrane/skin) composite was conducted for 24 hours at $37^{\circ}C$ using the Valia-Chien skin permeation system. The results showed that skin permeation rate of each drug across the (skin/membrane) composite was mainly dependent on the property of the membrane. Proper selection of the polymeric membrane which resembles hydrophilicity/lipophilicity of the delivering drug was important in controlling the skin permeation rate.

  • Transdermal therapeutic system(TTS) is often used as the method of drug dosage into the epidermic skin. Natural polymer were selected as ointment material of TTS. We investigated the permeation of natural polymer ointment containing drug in rat skin using horizontal membrane cell model. Permeation properties of materials were investigated for water-soluble drug such as Nicotinic acid N-oxide in vitro. These results showed that skin permeation rate of drug across the composite was mainly dependent on the property of ointment base and drug. Proper selection of the polymeric materials which resemble and enhance properties of the delivering drug was found to be important in controlling the skin permeation rate. This result suggests a possible use of natural polymer ointment base as TTS of antihyperlipoproteinemic agent.

표면 개질된 나노복합막의 투과 특성 (Permeation Properties of Surface Modified Nanofiltration Membrane)

  • 본 연구에서는 복합막의 제조공법으로 가장 광범위하게 쓰이는 계면중합법을 이용하여 나노복합막을 제조하였다. Monomer의 농도 및 조성, curing 조건, 후처리 조건과 같은 복합막 제조조건과, 운전압력, 공급액의 농도와 같은 운전인자에 의한 막성능 변화를 조사하였고, 계면중합 과정중에 첨가제를 사용하여 막의 유효면적을 확대하여 투과유속을 증가시키고자 하였다. Monomer의 농도가 증가함에 따라 배제능은 일정하였지만 투과유속이 감소하였고, curing 온도가 증가함에 따라서는 오히려 안정적인 박막층의 형성이 저해되어서 배제능과 투과유속이 모두 감소하였고, curing 시간에 따른 막성능의 변화는 나타나지 않았다. Test solution의 농도가 올라감에 따라서 투과유속과 배제능 모두 감소하였으며, 운전압력이 증가함에 따라서는 배제능과 투과유속 모두 향상하는 경향을 보였다. 계면중합과정중에 사용하는 아민단량체(amine monomer) 중 방향족 다이아민(aromatic amine)인 MPD의 함량을 높임에 따라서 배제능은 증가하였지만 투과유속은 현저하게 떨어졌다. 첨가제의 농도에 따른 표면 거칠기 증가 경향은 MPD가 포함된 amine 조성일 때가 더 높았지만 투과유속은 piperazine만 단독으로 사용하였을 때보다 떨어졌다.

  • The molecular weights of surfactant SDS(M.W. 288) and SLS(M.W. 420) are smaller than the molecular weigh cut-off of cellulose acetate membrane used in this study. However, 20% of SDS and 67% of SLS were rejected by cellulose aectate membrane. The higher rejection rate of SLS than that of SDS is due to the longer hydrophobic chain and greater molecular weight. There was no resistance to permeation rate by membrane fouling. Most of permeation resistance was due to the concentration polarization. Permeation rate was declined with operating time and with increase in concentration of surfactant due to concentration polarization.

겔제제로부터 인도메타신의 피부투과 (Skin Permeation of Indomethacin from Gels)

  • In order to reduce the systemic side effects and the gastrointestinal irritation of indomethacin following its oral administration, the drug was formulated as a transdermal gel using poloxamer 407. In vitro diffusion cells fitted with excised rat skins were used to evaluate the effects of formulation variables on skin permeation of indomethacin from poloxamer gels. The formulation variables were the concentrations of indomethacin, poloxamer 407 and ethanol, and the gel pH. The increase of the drug amount in the gel from 0.5% to 2.0% induced a direct but nonlinear increase in the skin permeation rate of indomethacin. The increase of poloxamer concentration from 17.5% to 25% in the gel resulted in a decrease of skin permeation rate of indomethacin, which was due to a reduction in the amount of free drug molecules available for permeation through skin by entrapping more drug molecules within the micelles formed by poloxamer. The increase of ethanol concentration from 10% to 20% in the gel resulted in a linear increase of permeation rate of indomethacin through skin, possibly due to the penetration enhancing effect of ethanol. The skin permeation of indomethacin was substantially influenced by the gel pH, exhibiting a maximum at pH 4.

Utilization improvement of PDMS and fluoropolymers by mutual application

  • We investigated about the utilization improvement of the PDMS and fluoropolymers by mutual application in environmental analysis. We were conducted the direct fluorination with mild condition on the PDMS films and analyzed its surfaces before and after fluorination. The results of FTIR and SEM analysis on the PDMS films showed that the film surfaces were fluorinated without irreversible deformation by the fluorination. During the fluorination, the measured contact angles of water and several alcohols on the PDMS films decreased with time and that of most alcohols decreased to 0 after 30 minutes. The surface energy of fluorinated PDMS films has increased by 2 times. Also, we investigated the influence factors on the change of permeation rate of water through PDMS tubes with time. It was observed that the change of permeation rate of water through PDMS tube was affected by temperature, water phase and spatial distribution of water. From these results, we could verify the principal causes of the decrease of permeation rate of water through PDMS tube with time and proposed a new experimental setup for reducing the variation of permeation rate of water less than 2%.
