Inhibitory Effects of Ginseng Extracts on Histamine-release from Rat's Mast Cell (인삼추출물의 랫트 비만세포 히스타민 유리 억제 효과)

  • We investigated inhibitory effects of ginseng extracts against compound 48/80-induced responses in rat peritoneal mast cells. Initially, we optimized extraction condition with various temperature and time for recovery of high saponin contents in extracts. Using a primary rat peritoneal mast cells, we examined whether ginseng extracts inhibit compound 48/80-induced histamine release form rat mast cells. High red ginseng-spercific saponin containing extracts were recovered at $85^{\circ}C$ for 48 hr, and had no cytotoxicity with relatively high dose of extracts on rat peritoneal mast cells(<0.5 mg/ml). For examine of ameliorate effects of mast cells responses by ginseng extract, we pre-treated the extracts or saline to mast cells and treated compound 48/80. In results, compound 48/80 treatment was increased histamine release (approximately 30%) from mast cells than normal group, whereas ginseng treatment was completely inhibited histamine release. These results suggested that ginseng extracts inhibits the compound 48/80-induced mast cell activation, and ginseng extracts is a candidate for effective therapeutic tools of allergic diseases.

Studies on Inhibitory Effect of Immediate-Type Allergic Reaction of Mahwang-Shin-Gung-San (麻黃辛芎散의 卽時型 알레르기 反應 抑制 效果에 關한 硏究)

  • Mahwang-Shin-Gung-San has been found inhibiting the mast cell-mediated anaphylactic reaction. This report describes an inhibitory effect of Mahwang-Shin-Gung-San (MSGS) on the immediate-type cutaneous allergic reactions. MSGS has concentration -dependently inhibited the ear swelling response induced by compound 48/80 in mouse by intradermal injection. The mast cells in mouse ear tissue undergone ear-swelling response by compound 48/80 were stained by alcian blue/nuclear fast red. MSGS significantly inhibited the compound 48/80-induced degranulation from mast cells in ear tissue. MSGS concentration-dependently inhibited the histamine release from the rat peritoneal mast cells (RPMC) by compound 48/80. We also studied the effect of MSGS on mast cell-dependent passive cutaneous anaphylaxis (PCA) activated by dinitrophenyl IgE antibody. MSGS showed potent inhibition of PCA by oral administration. These results indicate that MSGS inhibits immediate-type allergic reactions by inhibition of mast cell degranulation in vivo and in vitro.

  • The immediate-type allergic reaction (anaphylaxis) is involved in many allergic diseases such as asthma, allergic rhinitis, and sinusitis. The discovery of drugs for the treatment of immediate-type allergic diseases is a very important subject in human health. In this study, we investigated the effect of Rubus coreanus Miq.(Rosaceae) unripe fruits (RCF) on mast cell-mediated allergic reaction and inflammatory cytokine secretion. RCF inhibited compound 48/80-induced systemic reactions in mice. RCF attenuated immunoglobulin (Ig) E-mediated local allergic reactions. In addition, RCF dependently reduced histamine release from rat peritoneal mast cells local allergic reactions. In addition, RCF dependently reduced histamine release from rat peritoneal mast cells activated by compound 48/80 or IgE. Furthermore, RCF decreased the phorbol 12-myristate 13-acetate plus calcium ionophore A23187-stimulated tumor necrosis factor $(TNF)-{\alpha}$ and interleukin (IL)-6 secretion in human mast cells. Our findings provide evidence that RCF inhibits mast cell-derived immediate-type allergic reactions.

  • Objective : Mast cells are a potent source of mediators that regulate inflammatory response in allergies and asthma. The author studied the effect of Bojungikgitanggamibang(BITB) on mast cell-mediated anaphylactic reaction. Method : When BITB was given as pre-treatment at concentrations ranging from 0.01 to 1 mg/ml, the histamine release from rat peritoneal mast cells induced by compound 48/80 was reduced in a dose-dependent manner. Result : BITB dose-dependently inhibited compound 48/80-induced systemic anaphylactic shock. BITB also inhibited passive cutaneous anaphylaxis activated by anti-dinitrophenyl IgE. In addition, BITB inhibited phorbol 12-myristate 13-acetate and A23187-induced interleukin-6 secretion from human mast cell line HMC-1 cells. Conclusion : These results indicate that BITB may be actively anti-allergic.

Shini-San Inhibits Mast Cell-Dependent Immediate-Type Allergic Reactions

  • Shini-San has been used for treatment of allergic disease in Korea. However, its effect in experimental models remains unknown. The mast cell plays a pivotal role in initiating al1ergic response by secreting intracytoplasmic granular mediators such as histamine. The present report describes an inhibitory effect of Shini-San on mast cell-mediated immediate-type al1ergic reactions. Topical application of compound 48/80 can induce an ear swelling response in normal ($WBB6F_1-+/+$) mice but not in congenic mast cell-deficient $WBB6F_1-W/W^v$ mice. Shini-San inhibited concentration dependent mast cell-dependent ear swelling response induced by compound 48/80 in normal mice. Shini-San inhibited concentration-dependent passive cutaneous anaphylaxis induced by anti-dinitrophenyl (DNP) immunoglobulin E (IgE) in rats by topical application. Shini-San also inhibited in concentration-dependent fashion the histamine release from the rat peritoneal mast cells by compound 48/80 or anti-DNP IgE. Moreover, Shini-San had a significant inhibitory effect on compound 48/80-induced systemic anaphylactic reaction. These results indicate that Shini-San inhibits immediate-type allergic reactions by inhibition of mast cell degranulation in vivo and in vitro.

Mechanism of Inhibitory Action of Anaphylaxis by Aqueous Extract of Poncirus trifoliata (즉시형(卽時型) 알레르기 반응(反應)에 있어서 물추출액(抽出液)의 억제작용기전(抑制作用機轉))

  • The dried unripe fruit of Poncirus trifoliata L. is widely used to treat urticaria, itch and indigestion in folk medicine. And recently it was reported the component of the fruit was found to exhibit an inhibitory effect on histamine release from mast cells. So to evaluate the effect of aqueous extract of Poncirus trifoliata L.(PTFE) on compound 48/80-induced histamine release, the study was carried out in rat peritoneal mast cell. PTFE $(10^{-3}{\sim}1mg/ml)$ inhibits the histamine release induced by compound 48/80 $(5{\mu}g/ml)$ in rat peritoneal mast cells. To clarify the mechanism of these inhibition, we investigated the effect of PTFE on cAMP and intracellular calcium content. The increase in cAMP content, when PTFE was added, was transient. At concentration of 1mg/ml, the cAMP content of mast cells was significantly increased at a rate of 53 times of basal cells at 10sec. PTFE inhibits histamine release by augmenting the cAMP content in mast cells. Moreover, PTFE inhibits intracellular calcium release induced by compound 48/80. This result suggests that PTFE may be useful for the prevention and treatment of allergy-related disease.

  • Mast cells play an important role in the pathophysiologlcal changes observed in acute cutaneous and inflammatory diseases In order to see whether Kum-Hwang-San has an influence on mast cell- mediated immediate cutaneous reactions, the author has undertaken an animal study. Ears of mice were treated with a compound 48/80 solution topically at 30 min after the cutaneous application of Kum-Hwang-San. At each point, an ear swelling response was measured with a digimatic thickness micrometer. Ear swelling response induced by compound 48/80 was significantly suppressed dose-dependently by Kum-Hwang-San 30 min before topical application as compared with that in nonapplicated control mice, and the value returned to normal levels by 120 min. Compound 48/80- induced mast cell degranulation by Kum-Hwang-San was also remarkably decreased in accordance with the suppression in ear swelling response. Kum-Hwang-San dose-dependently inhibited histamine release from the rat peritoneal mast cells activated by compound 48/80. Another way to test acute cutaneous reaction is to induce passive cutaneous anaphylactic reaction. Kum-Hwang-San significantly inhibited passive cutaneous anaphylactic reaction activated by anti-dinitrophenyl IgE on both topical application and intradermal injection. Kum-Hwang-San also inhibited histamine release from the rat peritoneal mast cells induced by anti-DNP IgE. This study provides evidence that Kum-Hwang-San will be beneficial in the treatment of acute cutaneous diseases.

  • A 7-year-old female maltese dog with anorexia and vomiting for a week, and left axillary and perineal mass was admitted to Gyeongsang National University Veterinary Teaching Hospital. CBC revealed mild leukocytosis and severe anemia. Abdominal radiography revealed multiple gas opacities in upper abdomen. On ultrasonography, large amount of echogenic free abdominal fluid were detected. In addition, There are enhanced peritoneal strip sign (EPSS) indicating peumoperitoneum in the upper abdomen. Fine needle aspiration of the mass was performed and the result was mast cell tumor. Duodenal perforation caused by mast cell tumor was identified on laparotomy.

Anti-allergic Effects of Artemisia iwayomogi on Animal Models of Allergic Reactions

  • The effects of aqueous extract of Artemisia iwayomogi (Compositae) (AIAE) on the mast cell-dependent allergic and inflammatory reactions were investigated. AIAE (0.05 to 1 g/kg) dose-dependently inhibited systemic allergic reaction induced by compound 48/80 in mice. AIAE (0.1 and 1 g/kg) also significantly inhibited local allergic reaction activated by anti-DNP IgE. AIAE (0.001 to 1 mg/ml) dose-dependently inhibited the histamine release from rat peritoneal mast cells (RPMC) activated by compound 48/80. Moreover, AIAE inhibited the secretion of interleukin (IL)-6 in phorbol 12-myristate 13-acetate (PMA) plus calcium ionophore A23187-stimulated human mast cell line (HMC-1) cells. These results provide evidence that AIAE may be beneficial in the treatment of allergic diseases.

Suppression of Anaphylactic Reaction in Murine by Siegesbeckia pubescens

  • The aqueous extract of Siegesbeckia pubescens (SPAE) inhibited compound 48/80-induced systemic anaphylaxis 100% with the dose of 1.0, 0.5 mg/g body weight (BW) at 1 h before or 5 min, 10 min after intraperitoneal injection of compound 48/80. The passive cutaneous anaphylaxis (PCA) reaction also inhibited to 78.5% by oral administration of SPAE(1.0 mg/g BW). When SPAE pretreated on mice at concentrations ranging from 0.0001 to 1.0 mg/g BW, the serum histamine levels were reduced in a dose-dependent manner. Moreover, SPAE ($100-800{\mu}g/ml) dose-dependently inhibited the histamine release from peritoneal mast cells (RPMC) by compound 48/80 $(5{\mu}g/ml)$. Analysis by microscopic appearance observation revealed that SPAE $(500{\mu}g/ml) stabilized the RPMC membrane. Therefore, these findings indicate that SPAE inhibits anaphylactic reactions through stabilization of mast cell membrane.
