A New Multiplex-PCR for Urinary Tract Pathogen Detection Using Primer Design Based on an Evolutionary Computation Method

  • This work describes a new strategy for optimal design of Multiplex-PCR primer sequences. The process is based on the Particle Swarm Optimization-Simplex algorithm (Mult-PSOS). Diverging from previous solutions centered on heuristic tools, the Mult-PSOS is selfconfigured because it does not require the definition of the algorithm's initial search parameters. The successful performance of this method was validated in vitro using Multiplex-PCR assays. For this validation, seven gene sequences of the most prevalent bacteria implicated in urinary tract infections were taken as DNA targets. The in vitro tests confirmed the good performance of the Mult-PSOS, with respect to infectious disease diagnosis, in the rapid and efficient selection of the optimal oligonucleotide sequences for Multiplex-PCRs. The predicted sequences allowed the adequate amplification of all amplicons in a single step (with the correct amount of DNA template and primers), reducing significantly the need for trial and error experiments. In addition, owing to its independence from the initial selection of the heuristic constants, the Mult-PSOS can be employed by non-expert users in computational techniques or in primer design problems.

  • This proposes an online loss minimization algorithm for series PI(SPI) based interior permanent magnet synchronous motor(IPMSM) drive to yield high efficiency and high dynamic performance over wide speed range. The loss minimization algorithm is developed based on the motor model. In order to minimize the controllable electrical losses of the motor and thereby maximize the operating efficiency, the d-axis armature current is controlled optimally according to the operating speed and load conditions. For vector control purpose, a SPI is used as a speed controller which enables the utilization of the reluctance torque to achieve high dynamic performance as well as to operate the motor over a wide speed range. Also, this paper proposes current control of model reference adaptive fuzzy controller(MFC), and estimation of speed using artificial neural network(ANN) controller. The proposed efficiency optimization control, SPI, MFC, ANN in this paper is applied to IPMSM drive system, the validity of this paper is proved by analyzing response characteristics in variety operating conditions.

  • A study on the flow-structure characteristics of marine 3/2WAY pneumatic valve is essential for optimizing the performance of ship engines. It is important that the valve has desirable safety factor and reduced weight from safety and economic point of view. In this paper, flow-structure characteristics of pneumatic valve is obtained by being optimized based on the proper design criteria. The air with the pressure of 30 bar is the working fluid which is made to fill in the tack in short time. This time is defined as the filling time. On optimum design by considering the flow-structure characteristics, the approach is based on (1) the mathematical formulation of design decisions using the compromise decision-making method, and (2) the approximation technique of response surfaces. The methodology is demonstrated as the multi-objective optimization tool to improve the performance of marine 3/2WAY pneumatic valve.

  • The study is conducted on the subject of optimization of components sizing for series hybrid electric propulsion systems. The components sizing of series type hybrid system is very important because each component of series type is larger than the corresponding component of the parallel type or series-parallel type. If the components sizing is greater or less than what is required to this system, the performance of the system is getting worse. The methodology for the sizing of a driving motor is introduced based on the foundation of determined system configuration and performance target. And the sizing of an engine/generator and a battery is achieved based on simulation results using Dynamic Programming. It is possible to find the optimal sizing of these components by comparing fuel efficiency of hybrid electric propulsion system for 8 driving cycles.

  • Minimizing construction cost and reducing seismic damage are two conflicting objectives in the design of any new structure. In the present work, we try to develop a framework in order to solve the optimum performance-based design problem considering the construction cost and the seismic damage of steel moment-frame structures. The Park-Ang damage index is selected as the seismic damage measure because it is one of the most realistic measures of structural damage. The non-dominated sorting genetic algorithm (NSGA-II) is employed as the optimization algorithm to search the Pareto optimal solutions. To improve the time efficiency of the proposed framework, three simplifying strategies are adopted: first, simplified nonlinear modeling investigating minimum level of structural modeling sophistication; second, fitness approximation decreasing the number of fitness function evaluations; third, wavelet decomposition of earthquake record decreasing the number of acceleration points involved in time-history loading. The constraints of the optimization problem are considered in accordance with Federal Emergency Management Agency's (FEMA) recommended seismic design specifications. The results from numerical application of the proposed framework demonstrate the efficiency of the framework in solving the present multi-objective optimization problem.

A Cloud-Edge Collaborative Computing Task Scheduling and Resource Allocation Algorithm for Energy Internet Environment

  • To solve the problems of heavy computing load and system transmission pressure in energy internet (EI), we establish a three-tier cloud-edge integrated EI network based on a cloud-edge collaborative computing to achieve the tradeoff between energy consumption and the system delay. A joint optimization problem for resource allocation and task offloading in the threetier cloud-edge integrated EI network is formulated to minimize the total system cost under the constraints of the task scheduling binary variables of each sensor node, the maximum uplink transmit power of each sensor node, the limited computation capability of the sensor node and the maximum computation resource of each edge server, which is a Mixed Integer Non-linear Programming (MINLP) problem. To solve the problem, we propose a joint task offloading and resource allocation algorithm (JTOARA), which is decomposed into three subproblems including the uplink transmission power allocation sub-problem, the computation resource allocation sub-problem, and the offloading scheme selection subproblem. Then, the power allocation of each sensor node is achieved by bisection search algorithm, which has a fast convergence. While the computation resource allocation is derived by line optimization method and convex optimization theory. Finally, to achieve the optimal task offloading, we propose a cloud-edge collaborative computation offloading schemes based on game theory and prove the existence of Nash Equilibrium. The simulation results demonstrate that our proposed algorithm can improve output performance as comparing with the conventional algorithms, and its performance is close to the that of the enumerative algorithm.

  • This paper presents a development of an educational simulator of particle swarm optimization (PSO) and application for solving the test functions and economic dispatch (ED) problems with nonsmooth cost functions. A particle swarm optimization is one of the most powerful methods for solving global optimization problems. It is a population-based search algorithm and searches in parallel using a group of particles similar to other AI-based heuristic optimization techniques. In developed simulator, lecturers and students can select the functions for simulation and set the parameters that have an influence on PSO performance. To improve searching capability for ED problems, a crossover operation is proposed to the position update of each individual (CR-PSO). To verify the feasibility of CR-PSO method, numerical studies have been performed for two different sample systems. The proposed CR-PSO method outperforms other algorithms in solving ED problems.

  • In the ground-structure-based topology optimization, beam elements are regarded to be rigidly connected to each other, and joints are assumed to have infinite stiffness. Thus the optimized topology of a structure is obtained according to the assumption of no joint effect, and the resulting structure should be manufactured in one piece if the joint effect is to be excluded as much as possible. The underlying problems are that 1) the performance of the structure might be seriously decreased if the members of the structure are connected through welding or bolting, not manufactured in one piece, and 2) the topology of the structure will be changed if the joint effect is taken into account. In the paper, the assemblage issue is considered on topology optimization, and a new formulation based on the joint stiffness-varied ground beam structure is developed. Joints of a beam structure are modeled by elastic spring elements whose stiffnesses are controlled by design variables during the optimization.

  • This paper describes an implementation of a graph-based simultaneous localization and mapping(SLAM) method called the General Graph Optimization. The General Graph Optimization formulates the SLAM problem using nodes and edges. The nodes represent the location and attitude of a robot in time sequence, and the edge between the nodes depict the constraint between the nodes. The constraints are imposed by sensor measurements. The General Graph Optimization solves the problem by optimizing the performance index determined by the constraints. The implementation is verified using the measurement data sets which are open for test of various SLAM methods.

  • This research presents a multi-material topology optimization for functionally graded material (FGM) and nonFGM with elastic buckling criteria. The elastic buckling based multi-material topology optimization of functionally graded steels (FGSs) uses a Jacobi scheme and a Method of Moving Asymptotes (MMA) as an expansion to revise the design variables shown first. Moreover, mathematical expressions for modified interpolation materials in the buckling framework are also described in detail. A Solid Isotropic Material with Penalization (SIMP) as well as a modified penalizing material model is utilized. Based on this investigation on the buckling constraint with homogenization material properties, this method for determining optimal shape is presented under buckling constraint parameters with non-homogenization material properties. For optimal problems, minimizing structural compliance like as an objective function is related to a given material volume and a buckling load factor. In this study, conflicts between structural stiffness and stability which cause an unfavorable effect on the performance of existing optimization procedures are reduced. A few structural design features illustrate the effectiveness and adjustability of an approach and provide some ideas for further expansions.
