Structural evaluation of degradation products of Loteprednol using LC-MS/MS: Development of an HPLC method for analyzing process-related impurities of Loteprednol

  • The current investigation entails the characterization of five degradation products (DPs) formed under different stress conditions of loteprednol using liquid chromatography-tandem mass spectrometry (LC-MS/MS). In addition, this study developed a stable high-performance liquid chromatography (HPLC) method for evaluating loteprednol along with impurities. The method conditions were meticulously fine-tuned which involved the exploration of the appropriate solvent, pH, flow of the mobile phase, columns, and wavelength. The method conditions were carefully chosen to successfully resolve the impurities of loteprednol and were employed in subsequent validation procedures. The stability profile of loteprednol was exposed to stress degradation experiments conducted under five conditions, and DPs were structurally characterized by employing LC-MS/MS. The chromatographic resolution of loteprednol and its impurities along with DPs was effectively achieved using a Phenomenex Luna 250 mm C18 column using 0.1 % phosphoric acid, methanol, and acetonitrile in 45:25:30 (v/v) pumped isocratically at 0.8 mL/min with 243 nm wavelength. The method produces an accurate fit calibration curve in 50-300 ㎍/mL for loteprednol and LOQ (0.05 ㎍/mL) - 0.30 ㎍/mL for its impurities with acceptable precision, accuracy, and recovery. The stress-induced degradation study revealed the degradation of loteprednol under basic, acidic, and photolytic conditions, resulting in the formation of seven distinct DPs. The efficacy of this method was validated through LC-MS/MS, which allowed for the verification of the chemical structures of the newly generated DPs of loteprednol. This method was appropriate for assessing the impurities of loteprednol and can also be appropriate for structural and quantitative assessment of its degradation products.

  • A new ionomeric polymer-metal composite (IPMC) was prepared using a cast membrane of acrylic copolymer, which was synthesized by radical copolymerization of fluoroalkyl acrylate and acrylic acid (AA). To examine its performance as a new electroactive polymer, the current and displacement responses to a step voltage applied across the IPMC were measured. The largest responses were observed when the AA content in the copolymer was $10.6\;wt\%$.

  • Heat release characteristics of live fire load are an important parameter for performance oriented fire safety design of a building. While investigations have been carried out on the fire load and its burning behavior in office, residential and commercial buildings and so on, little effort has been paid for the rational treatment of fire load in bookstore. In this report, burning behavior of typical combustible objects in bookstore are studied by measuring heat release rates of bookshelf with book.

  • In the design of a complementary electrochromic windows based on $WO_3/Li^+$ conducting electrolyte/$V_2O_5$ system, a characterization of electrochromic properties of $WO_3/V_2O_5$ complementary devices as a function of thickness combinations is necessary in order to predict such as the safe operating voltage, the optical modulation range and the optical switching response.   • Lithium dual complex grease was prepared through the second continuous saponification reaction of a complex gellant system whose essential components comprised of a selected hydroxy fatty acid, lithium hydroxide monohydrate and boric acid to have a fiber structure of chemical, thermal and mechanical stability at high temperatures. An optimum amount of complex gellant was found to be 14% (NLGI #2), and an addition of castor wax of 1.5% provided an excellent performance properties, especially. The oil separation, oxidation stability, water wash-out property, shear stability, extreme pressure and wear property of thus prepared were tested by the ASTM and KS methods, and a characteristic result was obtained.

  • The performance of membranes is governed their pore struture. Pore structures of porous materials can be determined by a number of techniques. However, The novel technique, capillary folw porometry has a number of advantages. In this technique, the sample is brought in contact with a liquid that fills the pores in the membrane spontaneously. Gas under pressure is used to force the liquid from the pores and increase gas flow. Gas flow rate measured as a function of gas pressure in wet and dry samples yield data on the largest pore size, the mean flow pore size, flow distribution and permeability. Pore characteristics of a number of membranes were measured using this technique. This technique did not require the use of any toxic material and the pressure employed was low. Capillary flow porometry is a suitable technique for measurement of the pore structure of many membranes.

  • To investigate schemes of secret key generation from Ultra-wideband (UWB) channel, we study a statistical characterization of UWB outdoor channel for a campus playground scenario based on extensive measurements. Moreover, an efficient secret key generation mechanism exploiting multipath relative delay is developed, and verification of this algorithm is conducted in UWB Line-of-sight (LOS) outdoor channels. For the first time, we compare key-mismatch probability of UWB indoor and outdoor environments. Simulation results demonstrate that the number of multipath proportionally affects key generation rate and key-mismatch probability. In comparison to the conventional method using received signal strength (RSS) as a common random source, our mechanism achieves better performance in terms of common secret bit generation. Simultaneously, security analysis indicates that the proposed scheme can still guarantee security even in the sparse outdoor physical environment free of many reflectors.

  • For a GNSS receiver's robustness against RFI and the high accuracy of navigation solution in GNSS, interference source detection and mitigation are needed. In this paper, an adaptive lattice IIR notch filter is employed to track single-tone continuous wave and swept continuous wave interference signals, and an interference detection method is proposed. Furthermore, this paper presents interference source characterization algorithm using multiple ground stations' interference detection results. The measurement of the signal powers from each ground station is used to build weighting factors to estimate the type of the interference. The performance of interference detection algorithm is simulated for scenarios of GPS signal in the presence of single-tone continuous wave interference and swept continuous wave interference.

  • In this paper, a rerouting-controlled ISL (Inter-Satellite link) handover protocol for LEO satellite networks (RCIHP) is proposed. Through topological dynamics and periodic characterization of LEO satellite constellation, the protocol firstly derives the ISL related information such as the moments of ISL handovers and the intervals during which ISLs are closed and cannot be used to forward packet. The information, combined with satellite link load status, is then been utilized during packet forwarding process. The protocol makes a forwarding decision on a per packet basis and only routes packets to living and non-congested satellite links. Thus RCIHP avoids periodic rerouting that occurs in traditional routing protocols and makes it totally unnecessary. Simulation studies show that RCIHP has a good performance in terms of packet dropped possibility and end-to-end delay.
