콩발효 종균후보 Enterococcus faecalis strain DM01의 유전체 염기서열 (Complete genome sequence of Enterococcus faecalis strain DM01, a potential starter culture candidate for soybean fermentation)

  • 메주로부터 분리한 Enterococcus faecalis DM01 균주는 8종의 항생제(ampicillin, chloramphenicol, ciprofloxacin, erythromycin, gentamicin, penicillin G, tetracycline, vancomycin) 저항성, 바이오필름 및 용혈현상을 나타내지 않았다. GC 함량 37.68%, 2,785,968 bp 크기의 단일 chromosome을 보유하고 있는 DM01 균주의 유전체 정보는 콩발효식품 제조용 종균 적용에 요구되는 항생물질 저항성, 바이오필름 및 용혈현상 관련의 안전성을 뒷받침하는 것으로 확인되었다.

  • Cancer, despite all the efforts, still causes one in five deaths worldwide. Surgery, chemotherapy and radiotherapy provide inadequate protection and instead affect normal cells along with cancer cells. The search for cancer cures from natural products (plants and animals) has been practice for over a decade and the use of purified chemical to treat cancer still continues. Several studies have been undertaken during last three decades to find the anti-cancerous property of various plant extract and toxins secreted by animals and micro-organism. These lead to the discovery of several promising molecule having anticancer activity, some of which are in clinical trial and may emerged to be a potential future drug in cancer therapy. In this study we have used penicillin to evaluate its anti-cancer activity. It shown significant effects at cellular and molecular levels against growth of HeLa and K562 cell lines.

성인성 치주염환자에서 분리한 Porphyromonas gingivalis와 Prevotella intennedia의 항생제 내성에 관한 연구 (Antibiotic susceptibility of Porphyromonas gingivalis and Prevotella intennedia from the Patients with Adult Periodontitis)

  • The purpose of this study was to an in estimate the antibiotic susceptibility of P. gingivalis and P. interrnedia isolate from the subgingival plaque to adult periodontitis. Six P. gingivalis and five P. intermedia bacterial strains were tested for their susceptibility to 10 antimicrobial agents under disc diffusion method and broth dilution method. Ten patients with deep pocket(6mm) were selected for this study. They had not taken antibiotics for 6 months and no history of dental treatment for 6 months before this study. The result were as follow : 1. For antibiotic disc diffusion method, six P. gingivalis and five P. interrnedia were tested with 10 antimicrobial agents which comprised penicillin, gentamycin, clindamycin, lincomycin, ampicillin, erythromycin, tetracycline, amikacin, chloramphenicol, vancomycin. The sensitive antibiotics were tetracycline, penicillin, ampicillin, vancomycin, chloramphenicol and resisitent antibiotics were lincomycin. The other antimicrobial agents were less active. 2. From the study of determination on the minimal inhibitory concentration(MIC) by broth dilution method, the MIC of tetracycline to P. gingivalis and P. intermedia were $0.5-1.0{\mu}g/ml$, $0.5{\mu}g/ml$, that of ampicillin were $1-8{\mu}g/ml$, that of clindamycin were $1-32{\mu]g/ml$, $8-16{\mu}g/ml$, that of lincomycin were $16-32{\mu}g/ml$, $2-32{\mu}g/ml$. These data suggest that tetracycline and ampicillin may be valuable drug in the elemination of P. gingivalis and P. interrnedia from the patients with adult periodontitis.

Identification and Antibiotic Susceptibility of the Bacteria from Non-odontogenic Infectious Lesions

  • The purpose of this study was to isolate and identify bacteria from the 4 patients with non-odontogenic infectious lesions (mucormycosis, chronic inflammation from wound infection, and two actinomycosis) and determine their antimicrobial susceptibility against eight antibiotics. Bacterial culture was performed under three culture conditions (anaerobic, $CO_2$, and aerobic incubator). The bacterial strains were identified by 16S rRNA gene (16S rDNA) sequence comparison analysis method. For investigating the antimicrobial susceptibility of the bacteria against eight antibiotics, penicillin G, amoxicillin, tetracycline, cefuroxime, erythromycin, clindamycin, vancomycin, and Augmentin$^{(R)}$ (amoxicillin + clavulanic acid), minimum inhibitory concentration (MIC) measurement was performed using broth microdilution assay. Nosocomial pathogens such as Enterococcus faecalis, Klebsiella pneumoniae, Bacillus subtilis, and Neisseria flavescens were isolated from mucormycosis. Veillonella parvula, Enterobacter hormaechei, and Acinetobacter calcoaceticus were isolated from chronic inflammatory lesion. Actinomyces massiliensis was isolated from actinomycosis in parotid gland. Capnocytophaga ochracea was isolated from actinomycosis in buccal region in anaerobic condition. There was no susceptible antibiotic to all bacteria in mucormycosis. Tetracycline was susceptible to all bacteria in chronic inflammation. C. ochracea was resistant to vancomycin and penicillin G; and other antibiotics showed susceptibility to all bacteria in actinomycosis. The results indicated that the combined treatment of two or more antibiotics is better than single antibiotic treatment in mucormycosis, and penicillin is the first recommended antibiotic to treat actinomycosis.

최근 충북지역에서 분리된 Gram 양성 세균종과 항생물질의 감수성 양상 (Gram-Positive Bacterial Species and Antimicrobial Susceptibility Patterns Isolated from Chungbuk Area)

  • 임상검체에서 분리되는 병원균을 주기적으로 분석하고 이에 대한 항균제 감수성의 변화 양상을 파악하는 것은 임상에서의 적절한 항균제의 선택이나 내성균의 감독과 조절을 위해 필수 적이라 할 수 있다. 저자들은 충북대학병원에 1996년 1년간 내원 환자의 미생물 배양 검사에서 분리, 동정된 1,689균주중 Gram양성인 542균주의 분리 빈도 및 주요 균종에 대하여 항생제 감수성 양상을 조사하였다. Gram 양성 균종은 S. aureus, S. pneumoniae, S. epidermidis, S. hemolyticus, coagulase negative Staphylococcus (CNS), Enterococcus faecalis 순으로 분리되었다. 한편, 항생제 감수성 시험 결과 S. aureus의 항생제 감수성은 penicillin과 gentamicin에 대한 저항성이 현저히 높았으며, teicoplanin 과 vancomycin (MIC$_{50}$ $\mu\textrm{g}$/ml)에 감수성이 높은 것으로 나타났다. CNS의 경우도 S. aureus와 대동소이 한 결과를 보였으나, S. epidermidis의 경우에는 ciprofloxacin과 clindamycin에 감수성 이 높은 것으로 나타났다. Enterococcus spp.는 vancomycin, penicillin, tetracycline 등에 저항성이 매우 높았으며, 검사한 모든 항생제에 대하여 대부분의 감수성이 zig-zag pattern을 보였다.

국내 새우젓에서 분리한 Lactobacillus brevis HLJ59의 Angiotensin Converting Enzyme 저해활성 및 생리적 특성 (Physiological Characteristics and Angiotensin Converting Enzyme Inhibitory Activity of Lactobacillus brevis HLJ59 Isolated from Salted Shrimp)

  • 본 연구는 우리나라 전통 발효식품인 장류, 김치류 및 젓갈류로부터 angiotensin converting enzyme 저해활성이 우수한 젖산균을 분리하고자 하였다. 젖산균을 분리하기 위한 선택배지로서 bromocresol purple (BCP) 한천배지를 사용하여 1차적으로 젖산균을 분리하였으며, 그 중 angiotensin converting enzyme 저해활성이 우수한 균주를 최종 선발하였다. 분리된 젖산균을 16S rRNA 유전자 염기서열 분석으로 동정한 결과 Lactobacillus brevis ATCC $14869^T$와 99.7%의 유사도를 나타냄에 따라 L. brevis HLJ59로 명명하였다. L. brevis HLJ59는 내산성의 경우 pH가 2.0, 3.0으로 보정된 DeMan Rogosa Sharpe 액체배지에서 접종 후 각각 90분, 30분이 경과하였을 때 배양초기와 비교 시 약 99% 감소하는 결과를 보였으나, 담즙산(Bile extract)의 경우 1% 첨가 시에도 생육에 저해를 받지 않는 것으로 조사되어 L. brevis HLJ59 균주는 담즙산에 대한 내성이 우수한 균주임을 확인하였다. 항생제 내성의 경우 20종의 항생제를 paper disc법으로 조사한 결과 본 균주는 cephalosporin계의 cefoxatin (30 ${\mu}g$), ceftnaxone (30 ${\mu}g$), penicillin계의 penicillin (10 units), quinolones계 cprofloxacin (5 ${\mu}g$), nalidixic acid (30 ${\mu}g$), lincosamid계의 lincomycin (2 ${\mu}g$) 및 기타 chloramphenicol (30 ${\mu}g$)에 대해서는 내성을 가지고 있음을 확인하였다.

Characterization and Food Application of a Potentiometric Biosensor Measuring $\beta$-Lactam Antibiotics

  • $\beta$-Lactam antibiotics such as penicillin G, amoxicillin, and ampicillin were determined by a potentiometric biosensor system which exploited penicillinase immobilized on Immobilon cellulose nitrate membrane and a flat-bottomed pH electrode-as the biological component and transducer. The optimum reaction buffer for maximum sensitivity was found as 2 mM of sodium phosphate buffer (pH 7.2). The detection limit of the biosensor could be extended to 1 $\mu{M}$ of the analytes by increasing the enzyme loading for immobilization to 100 units/$m\ell$. The model samples spiked with each of the standard penicillins were measured for their biosensor responses and HPLC peak area, resulting in the relative responses of 82.1-103.5% and 79.5-106.1% for the biosensor method along with HPLC analysis, respectively. This result showed a good precision of the current biosensor method for screening the penicillin compounds.

방선균의 일주가 생성하는 균체외 $\beta$-Lactamase의 특성 (Characters of Extracellular $\beta$-Lactamase Obtained from a Strain of Streptomyces sp.)

  • A strain of Streptomyces sp. isolated from soil was found to produce extra-cellular $\beta$-lactamase associated partially to the cell growth. The $\beta$-lactamase was purified from the culture supernatant through anmonium sulfate fractionation, ion-exchange chromatographies and gel filtration. The final purification fold and recovery yield were 57 and 6.2%, respectively. Molecular weight of the $\beta$-lactamase was estimated to be about 67, 000 by SDS-polyacrylamide gel electrophoresis. The optimal reaction condition was at pH 7-8 and at 35-$45^{\circ}C$. The $K_m$ and $V_{max}$ values of the enzyme for penicillin G were estimated to he 3 mM and $3\times 10^3$ $\mu\textrm{M}$/min/mg protein, respectively. The purified $\beta$-lactamase was classified to the class A enzyme hydrolyzing only penicillin.

임상 검체에서 분리된 Bacillus cereus의 성상, 장독소 생성 및 항균제 감수성 (Bacillus cereus Clinical Isolates : Characteristics, Enterotoxin Production and Antimicrobial Susceptibility)

  • Biochemical characteristics, enterotoxin production and antimicrobial susceptibility were determined for 30 strains of Bacillus cereus isolated from stool specimens of diarrhea patients at an university hospital in Chulabuk-do province. Positive rate for VP reaction and citrate utilization were lower, (33 % and 40 % respectively) while the rates of acid production from mannitol, arabinose, and xylose were higher (17 %, 13 % and 3 % respectively) than those obtained by other investigators. The enterotoxin gene was detected in 18 of 30 isolates (60 %) by PCR, and the toxin was detected from all of the toxin gene-positive isolates by RPLA test. The agar dilution test showed that all isolates were resistant to penicillin G and 73 % were to cephalothin, but all were susceptible to ciprofloxacin, clindamycin, erythromycin, fusidic acid, gentamicin, rifampin, teracycline and vancomycin. We conclude that B. cereus isolates producing acid from mannitol, arabinose and xylose exist, that PCR can be used to detect enterotoxin genes rapidly and accurately, and that this organism is susceptible to various antimicrobial agents though not penicillin G and cephalothin.

치면세균막에서 분리한 뮤탄스 연쇄상구균 및 Streptococcus anginosus의 수종 항생제에 대한 감수성 조사 (ANTIBIOTIC SUSCEPTIBILITY IN MUTANS STREPTOCOCCI AND STREPTOCOCCUS ANGINOSUS ISOLATED FROM DENTAL PLAQUE)

  • The aim of this study was to investigate the susceptibility of mutans streptococci (S. mutans and S. sobrinus) and Streptococcus anginosus, for seven antibiotics, penicillin G, amoxicillin, ciprofloxacin, cefuroxime, erythromycin, bacitracin, and vancomycin. The minimum inhibitory concentration (MIC) of seven antibiotics against 3 species (type strains) of mutans streptococci and S. anginosus, 10 strains (wild type) of S. mutans, 7 strains (wild type) of S. sobrinus, and 11 strains (wild type) of S. anginosus, were measured by broth dilution method. All of the type strains of mutans streptococci and S. anginosus had the same susceptibility for penicillin G, amoxicillin, cefuroxime and bacitracin. Type strain of S. anginosus was sensitive in ciprofloxacin, but those of mutans streptococci were not. All of the clinical isolates of mutans streptococci and S. anginosus had the same susceptibility for the seven antibiotics. Our data reveal that mutans streptococci and S. anginosus have similar antibiotic-resistant character. In addition. these results may offer the basic data to verify the antibiotic-resistant mechanism of mutans streptococci and S. anginosus.
