고온성 세균의 $\beta$-Galactosidase에 관한 연구 ( I ) - 분리고온균의 생리적 특성 - (Studies on the $\beta$-Galactosidase from Thermphilic Bacterium - Physiological Characteristics of the Selected Thermophile -)

  • 고온균의 생리적 특성자 내열기구에 관한 기초 자료를 얻고자 온천토양에서 $\beta$-galactosidase를 생산하는 고온균을 분리하여 동정하고 몇가지 주요한 생리적 특성을 검토하였으며 그의 균체 지방산 조성을 gas chromatography로 분석하여 다음과 같은 결과를 얻었다. 1. $\beta$-galactosidase를 생산하는 공시균주는 Thermus sp. 으로 동정되었다. 2. 분리선정한 균주의 최적 생육온도는 $65^{\circ}C$이었고, 37$^{\circ}C$에서 생육하지 않는 절대고온균 이었으며 최적pH는 6.5내외이었고 pH에 민감하였다. 3. NaCl에 대한 내성은 NaCl 1% 이상에서는 생육하지 못했다. 4. 항생물질에 대한 내성은 penicillin G는 10$\mu\textrm{g}$/$m\ell$, chloramphenicol은 0.5$\mu\textrm{g}$/$m\ell$ 이었다. 5. Vitamin 요구성은 Ca-pantothenate와 pyridoxine-HCI를 절대적 생육인자로 요구하였고niacin을 자극적 생육인자로 요구하였다. 6. 공시균주의 균체 지방산 조성은 palmitic acid 60.20%, lauric acid 11.80%, myristic acid 7.56%. behenic acid 4.25%, Capric acid 1.77%, stearic acid 2.13%, arachidic acid 1.53% 이었다.

영남지방 돼지에서 분리한 Erysipelothrix rhusiopathiae의 세균학 (Bacteriological Study of Erysipelothrix rhusiopathiae Isolated from the Pigs in Youngnam Area)

  • The present study was conducted to investigate biochemical, serologic, and pathogenic characteristic of E. rhusiopathiae isolated from the cases of acute septicemic swine erysipelas in Youngnam provinces during the period from June 1988 to September 1990. The majority of biochemical and cultural properties of E. rhusiopathiae isolated from pigs affected with acute erysipelas were identical to those of the standard strain employed. All of the 45 isolates were serotype la. All isolates were highly susceptible to penicillin G, lincomycin, cephalothin, ampicillin, erythromycin (MIC : 0.025-0.78IU or ${\mu}g$ / ml ), and moderately susceptible to oleandomycin, oxytetracycline, chloramphenicol (MIC : 0.78-25${\mu}g$ / ml ). Kanamycin and sulfadimethoxine showed no activity against the isolates(MIC : >400${\mu}g$ / ml ). The MICs of dihydrostreptomycin presented two distribution peaks ; of 45 strains, 5(11.1%) were resistant to dihydrostreptomycin(MIC : 400${\mu}g$ / ml ). All of 5 selected isolates were pathogenic for mite and $LD_{50}$ was $3.7{\times}10^3$viable cells. Mice immunized subcutaneously with live vaccine did not die after challenge to virulent isolates of E. rhusiopathiae.

  • The present study was conducted to investigate biochemical properties and antimicrobial drug susceptibilities of 47 strains of E rhusiopathiae isolated from the cases of acute septicemic swine erysipelas in Youngnam and Kyunggi provinces during the period from June 1988 to December 1989. The isolants were identified as E rhusiopathiae on the basis of cellular and colonial morphology, and characteristic reactions in some biochemical tests. All the organisms produced hydrogen sulfide in triple sugar iron agar and showed the characteristic "pipe cleaner" type of growth in gelatin stab cultures. The majority of biochemical and cultural properties of E rhusiopathiae isolated from pigs affected with acute erysipelas were identical to those of the reference strains employed. All the isolates were highly susceptible to penicillin G, ampicillin, erythromycin (MIC:0.025~0.39IU or ${\mu}g/ml$), and moderately susceptible to oleandomycin, oxytetracycline, chloramphenicol(MIC:$0.78{\sim}25{\mu}g/ml$). Kanamycin and sulfadimethoxine showed no activity against the isolates(MIC:>$400{\mu}g/ml$). The MICs of dihydrostreptomycin presented two distribution peaks; of 47 strains, 5(10.6%) were resistant to dihydrostreptomycin (MIC:$400{\mu}g/ml$), while the majority of them were susceptible to the drug.

  • Eighteen strains of Clostpidium perfringens were isolated from the piglets with hemorrhagic enteritis. The characteristics of the outbreaks, clinical signs and lesions were examined. The biochemical properties, type of toxins and susceptibility to antimicrobial agents against the isolates were investigated. 1. The incidence of diarrhea was appeared in 97(22.4%) of 432 piglets examined. 2. The isolation rate of Cl perfingens from the 97 diarrheal faeces were 18.5%(18 strains) 3. The population of Cl perfingens in feces were ranged $10^{8-9}$cfu/g in 5(32.5%) and $10^{3-7}$cfu/g in 13(67.4%) of 18 samples. 4. The toxin type of the 18 isolates investigated by mouse inoculation test was all type C strains of Cl perfringens. 5. As a results of antimicrobial susceptibility test, 18 isolates were higly susceptible to cephalothin, tetracycline and penicillin.

  • Thirteen strains of Bacillus thuringiensis were isolated from soil in Korea and characterized. The all strains produced parasporal crystals and spores in their cells. Two strains had bipyramidal crystals, seven strains contianed round ones and four strains had unregular ones. Only minor biochemical characteristics of the thirteen isolates were different and distinctive, however general characteristics were similar to the known serotypes of B. thuringiensis. Two strains were resistant to ampicilin. Three strains were resistant to bacitracin, six strains were resistant to cephalothin, two strains were resistant to colistin, HL-68 strain was resistant to gentamycin, HL-67 strain was resistant to kanamycin and HL-71 was resistant to tetracycline. Two strains were resistant to penicillin G. Four strains were toxic to Bombyx mori larvae and eleven strains were toxic to Culex pipiens larvae.

Production and Characterization of Extracellular $\beta$-Lactamase from Streptomyces aureofaciens SMF14

  • A strain SMF14 producing an extracellular $\beta$-lactamase was isolated from soil and identified to be a strain of Streptomyces aureofaciens. $\beta$-Lactamase was purified from the cell free culture broth through batchwise hydroxyapatite adsorption, anion exchange chromatography on DEAE Sephadex A-50, gel filtration on Sephadex G-75, and adsorption chromatography on hydroxyapatite. The molecular mass was estimated to be about 43 kDa by SDS-PAGE. The $\beta$-lactamase had substrate specificity to penicillins and it was inhibited by clavulanic acid, being classified to the group 2a of penicillinase.. The optimal reaction pH and temperature were pH 6.0~7.5 and $50^{\circ}C$. The $K_m, and V_{max}$ values of $\beta$-lactamase for penicillin G were calculated to be 1.72 mM and 5.4$\times$$10^5 \mu M \cdot min^{-1}$, respectively.

시판 냉동식품에서 분리한 장구균의 항생제 내성 양상 (Antibiotic Resistance Patterns of Enterococcus spp. Isolated From Commercial Frozen Foods)

  • 2009년 4월부터 12월까지 서울시 보건환경연구원 미생물 관리팀에 의뢰된 냉동식품 100건을 대상으로 일반세균수, 대장균군수, 대장균과 장구균의 오염도를 조사였으며, 분리된 장구균을 대상으로 하여 항생제 감수성 검사를 실시하였다. 일반세균수는 평균 검출량이 $4.3{\times}10^4CFU/g$ 이고 대장균군수는 평균 검출량이 $4.3{\times}10^3CFU/g$ 이고 대장균은 100건 모두에서 불검출되어 0.0%의 검출률을 보였다. 100건의 검체 중 총 22건에서 장구균이 분리되었으며 이 가운데 12주는 E. faecium, 7주는 E. faecalis, 2주는 E. gallinarum, 1주는 E. hirae이었다. 분리균주의 항생제 내성률을 검사 한 결과, 내성을 나타내는 항생제들은 erythroycin, rifampin, teracycline ciprofloxacin, chlorampenicol, penicillin이며, vancomycin, ampicillin, gentamicin, strepomycin, linezolid에 대해서는 모두 감수성이었다. 또 22건의 분리 균주 중에서 2종 이상의 항생제에 다제 내성을 나타낸 균주가 15주나 되며 가장 많은 형태로는 6균주의 장구균에서 나타난 E-RA에 대한 것이었다.

  • A total of 204 strains of Staphylococcus hyicus subsp. hyicus isolated from piglets suffering from exudative epidermitis and healthy pigs were tested for antibiotic susceptibility. The growth of all isolates of Staph. hyicus subsp. hyicus tested was inhibited by cloxacillin(Cx) at a concentration of $0.8{\mu}g/ml$ and by cefazolin (Cf) at a concentration of $0.4{\mu}g/ml$, and by gentamicin and methicillin at a minimum inhibitory concentration of $3.2{\mu}g/ml$. Of total isolates obtained from the diseased and healthy pigs, 69.6% were resistant to penicillin G, 53.4% to tetracycline, 11.8% to kanamycin and 0.5% to chloramphenicol, with considerable variations among farms in the occurrence of resistant strains and resistance patterns to the drugs. By application of Cx or Cf which was active to the organism, piglets exposed to infection were successfully protected from the disease, but piglets suffering from already severe exudative epidermitis did not respond to treatment.

  • The $H^+$-selective membrane consists of tridodecylamine, 2-nitrophenyl octyl ether and a little amount of additive in carboxylated PVC matrix, where penicillinase(Pcase ) and glutaraldehyde may be covalently attached to the matrix. When the $H^+$-selective electrode was used as a detector of biosensor, calibration curve calculated from Nernst equation was not linear. So we investigated the characteristic responses of the $H^+$-selective electrode to the product H+ from hydrolysis of penicillin-G(Pc-G) and plotted calibration curve correlating potential to concentration of Pc-G linearly. The optimal concentration of buffer solution was theoretically calculated and was also experimented. We tried to explain the linear curve of potential to concentration of Pc-G by using Henderson-Hasselbach equation. This method is more effective in calibration curve plotting than any other previous methods. The results obtained may help in further developing pH electrodes with improved analytical preformance.

치과병원 진료실 내에서 메티실린 또는 반코마이신 저항성 Staphylococcus aureus의 검출 (DETECTION OF METHICILLIN OR VANCOMYCIN-RESISTANT STAPHYLOCOCCUS AUREUS FROM DENTAL HOSPITAL)

  • 본 연구는 조선대학교 치과병원의 진료환경 및 진료요원으로부터 기회감염성 병원체로 알려진 methicillin 또는 vancomycin 저항성 황색포도상 구균 (methicillin-or vancomycin-resistant Staphylococcus aureus. MRSA or VRSA)의 존재 여부를 조사하여, 이를 광주지역 개원치과와 비교분석을 통해 현재 조선대학교 치과병원의 MRSA와 VRSA의 오염정도를 파악하고자 하였다. 이를 위해 진료실 환경 및 진료요원으로부터 분리한 S. aureus 균주들의 8종 항생제에 대한 감수성 조사를 시행하고, 기존에 알려진 항생제 내성 유전자 존재 여부를 PCR법을 이용하여 확인하였다. 그 결과, 조선대학교 치과병원의 진료요원에서 채취한 샘플 중 1개 (2.3%), 개원 치과에서는 2명 (10%)의 진료요원의 샘플에서 S. aureus가 분리되었으며, 진료환경에서는 두 곳 모두에서 S. aureus가 검출되지 않았다. 조선대학교 치과병원과 개원치과에서 분리된 S. aureus는 amoxicillin, penicillin G, ciprofloxacin clindamycin, vancomycin에 내성을 보이며 oxacillin, cefuroxime에는 균주에 따라 감수성 또는 내성을 보였다. 조선대학교 치과병원에서 분리된 S. aureus는 erythromycin과 clindamycin에 내성 유전자인 ermA가 존재하였으며, 개원치과에서 분리된 3개의 S. aureus 중 2개에서 penicillin과 oxacillin에 내성 유전자 mecA가 존재하는 것으로 나타났다. Vancomycin 내성 유전자인 vanA, vanB는 어떠한 샘플에서도 검출되지 않았다. 이상의 결과를 종합할 때, 본 연구는 조선대학교 치과병원과 개원치과의 S. aurues분포 및 MRSA 또는 VRSA의 존재여부를 조사하여 MRSA와 VRSA의 확산예방을 위한 치과진료 환경의 개선과 적절한 항생제 사용에 대한 기초 자료를 제공할 것으로 사료된다.
