Antioxidant and anti-inflammatory effects of red garlic compositions (홍마늘 조성물의 항산화 및 항염증 효과)

  • Garlic (Allium sativum L.) and traditional herb has several functional properties and strong biological activities, making it useful as a functional food material. We investigated the antioxidant and anti-inflammatory activity of mixed compounds from red garlic and supplementary materials, including ginger (Zingiber officinale Roscoe), doraji (Platycodon grandiflorum), quince (Chaenomeles sinensis), citrus peel (Citri Pericarpium), and mint (Mentha arvensis). The extracts were prepared with water (W) and ethanol (E) at $70^{\circ}C$ (W-70, E-70) and $95^{\circ}C$ (W-95, E-95) for 3 h. The total content of phenolic compounds was the highest in E-70 (608.60 mg/100 g). Alliin, one of the active ingredients in red garlic, was contained at 1.18-1.29 mg/g and 0.81-0.85 mg/g in water and ethanol extract, respectively. Another active ingredient of red garlic, S-allyl-cysteine (SAC) had higher content in the water extract than in the ethanol extracts. DPPH radical scavenging activity was higher in E-70 (15.96-73.65%) at $313-5,000{\mu}g/mL$. ABTS radical scavenging activity was also higher in E-70 (5.71-77.19%) than in the others. The ROS production rate showed the same tendency as the NO production, with more efficacy in E-95. The expression level of iNOS and $IL-1{\beta}$ was decreased in the E-95 significantly at the concentration of $1,000{\mu}g/mL$ compared to the lipopolysaccharide (LPS) treated group. Based on the above results, the antioxidative and anti-inflammatory activities of the extracts of red garlic and supplementary materials were expressed by different useful substances. The contents of these useful substances were different according to the extraction solvent and temperature.

Effects of Onion Peel Extracts on Blood Circulation in Male Smokers (양파껍질추출물의 섭취가 남성 흡연자의 혈행개선에 미치는 효과)

  • A controlled and double blinded trial was performed to evaluate the effects of OPE supplementation on blood circulation in healthy male smokers. The study population consisted of 92 healthy male smokers aged $44.4{\pm}7.8$ years. The supplement group (n=49) received OPE containing 100 mg quercetin/day for 10 weeks, while the control group (n=43) received placebo powders. The prothrombin time and activated partial thromboplastin time increased significantly after OPE consumption, indicating the attenuated blood clot formation by OPE. In conclusion, these results indicate that the daily consumption of OPE (containing 100 mg quercetin) for 10 weeks exerts significant health benefits through the modulation of physiological functions including the blood circulation in male smokers.

Quinone Reductase Inductive Activity and Growth Inhibitory Effect against Hepatoma Cell of Oriental Melon Extract (참외 추출물의 Quinone Reductase 유도활성 및 간암세포 증식 억제효과)

  • This study was performed to elucidate anticancer activities of various parts, such as peel, flesh, placenta, seed, stalk and stem leaf of oriental melon. Chemopreventive and anticancer effects of oriental melon extract were evaluated by detoxifying enzyme, quinone reductase (QR) inductive activity, cytotoxicity and growth inhibitory effect against hepatoma cell. Stalk and stem leaf extracts of oriental melon showed the increment of QR inductive activity with dose-dependent manner and induced quinone reductase 3.9, 1.5-fold at $200{\mu}g/mL$ respectively compared to control. The growth inhibitory effect of oriental melon extract against mouse hepatoma cell (Hepa1c1c7) was investigated by crystal violet (CV) assay. Stalk and stem leaf of oriental melon showed potent growth inhibitory effect. Based on these result, the growth inhibitory effects of stalk, stem leaf at various concentration were examined in detail by MTT assay using human hepatoma cancer cell (HepG2). All of two parts showed growth inhibitory effects and expecially stalk exhibited inhibitory effect of 60.3% at maximum concentration. The above results suggest that stalk of oriental melon has a possibility as a source of natural cancer chemopreventive materials.

Screening of Effective Factor to Inhibition of NDMA Formation in Yuza (Citrus junos) (유자 중 NDMA 생성억제에 영향을 미치는 인자의 검색)

  • This study was conducted to investigate of inhibition activity of yuza (Citrus junos) extracts and juice on nitrite scavenging and N-nitrosodimethylamine (NDMA) formation in model system. Yuza juice was separated into organic acid, ascorbate and phenolic portion using sep-pak $C_{18}$ cartridge, respectively. Nitrite scavenging activity and inhibition of NDMA formation from yuza extract has shown higher inhibition with pH of reaction condition lower and amount of added sample more. And peel extract of yuza was higher than those of flesh extract. Nitrite scavenging activity was excellent juice portion rather that 3 portions. Of the 3 portions, it activity was the highest in the organic acid portion, and decreased in the following order; phenolic acid, ascorbate portion. Therefore, organic acid of yuza was suggested to expect effective ihhibitor of NDMA formation.

Isolation and Identification of 3 Low-molecular Compounds from Pear (Pyrus pyrifolia Nakai cv. Chuhwangbae) Fruit Peel (추황배(Pyrus pyrifolia Nakai cv. Chuhwangbae) 과피로부터 3종의 저분자 화합물의 단리·동정)

  • Three low-molecular compounds were isolated from methanol extracts of pear (Pyrus pyrifolia N. cv. Chuhwangbae) fruit peels using solvent fractionation, various types of column chromatogrphy (Diaion HP-20, Sephadex LH-20, and silica gel), and high performance liquid chromatography with an assay guided by 1,1-diphenyl-2-picrylhydrazyl radical-scavenging activity. The isolated compounds were identified as 2-carboxyl-4(1H)-quinolinone (kynurenic acid, 1) from butanol fraction, cis-p-coumaric acid (2) from ethyl acetate-acidic fraction, and vanillin (3) from the ethyl acetate-phenolic fraction, respectively. These isolated compounds were confirmed on the basis of the spectroscopic data of electrospray ionization mass spectrometry and nuclear magnetic resonance. This is the first time that compounds 1-3 were isolated and identified in pear.

Development of Sausage Using National Resource By-Product (천연 부산물을 이용한 소시지 개발)

  • These studies were carried out to develop sausage using extracts from waste resources. The changes of pH and moisture contents were tended to be a similar to control, respectively ranged in pH 6.0~6.4 and 62~60% in moisture content. However, the water holding capacity of sausage added with chitosan and sesamol gradually were tended to be a little high, compared to control. The treatments using natural resource by-product revealed a low TBARS value, compared to control. This means that sausage added with natural by-product seem to be extend its shelf-life during storage. At the same time, the TBARS values were very similar to sausage added with 0.5% potassium sorbate. The residual nitrite contents of sausage added with natural resource by-product were tended to be a little lower than that of control. As storage time goes by, the nitrite contents of sausage product gradually were a little more decreased during storage at 3$0^{\circ}C$ than those of storage at 1$0^{\circ}C$. The lightness of sausage color gradually were a little more decreased during storage at 3$0^{\circ}C$ than those of storage at 1$0^{\circ}C$. The redness of sausage added with chitosan were a little highter, compared to control Sensory test suggested that the changes of sausage color, flavor, texture and taste were tended to be decreased gradually. In conclusion, sausage added with natural resource by-product was effective on the basis of the reduction in residual nitrite content and lipid oxidation.

Effect of Removed Peel from Sweet Persimmon on Nutritional Ingredients and Antioxidant Activities (단감의 박피가 영양성분 및 항산화 활성에 미치는 영향)

  • This study was for the industrial application of functional food ingredients from whole fruits of sweet persimmon. Whole fruit and pulp of sweet persimmons were divided, and then lyophilized and powdered. Contents of crude fiber, vitamin C, and mineral were significantly higher in whole fruit than pulp of sweet persimmon. The amino acid content of whole fruit was 1.4 times higher than those of sweet persimmon pulp. In the biological activities of water and ethanol extracts from whole fruit and pulp of sweet persimmon, ethanol extract was higher than water extract, and whole fruit was higher than its pulp. The result which compared the biological activities of the water and ethanol extract from lyophilized sweet persimmon showed that total phenolic content was significantly higher in whole fruit of sweet persimmon, but flavonoid contents were not significantly different. Especially ABTS, NO radical scavenging activity, reducing power and tyrosinase inhibition activity were significantly higher in whole fruit extract than pulp extract of sweet persimmon. The relatively high content of fiber and vitamin C, and biological activity of whole fruit than pulp of sweet persimmon may be make it preferable as functional food materials for secondary processed goods.

Effect of Garlic Enzymatic Hydrolysates and Natural Color Resource Composites on Lipid Metabolism in Rat Fed a High Fat Diet (마늘 효소분해물과 천연색소 소재의 복합물이 고지방 식이성 흰쥐의 지질대사에 미치는 영향)

  • The study was performed to investigate the effects of enzyme treated garlic (EG) and its natural resources composites on lipid levels in serum and liver of rats fed a high fat diet. Four different types of EG-composite extracts prepared: EG and EG + grape peel (EGG), EG + Persimmon (EGP) and EG + Catechu (EGC) by mixed 9.5:0.5, 9:1 and 8:2 (w/w) ratios, respectively. DPPH and ABTS radical scavenging activity in vitro, show the highest in EG + Catechu (EGC) composite by mixed 8:2 (w/w). EG and EG-composites extracts (8:2, w/w) were administered orally to SD-male rats at a concentration of 2.5 g/kg/day for 5 weeks. Total lipid and cholesterol contents in serum were significantly lower in EGC group than control group, and triglyceride content was the lowest in EGP group by 54.29 mg/dL. HDL-cholesterol contents were significantly higher in EGP and EGC groups. LDL-cholesterol content was lower in EG group than EG-composite groups, and VLDL cholesterol content was the lowest in EGP group. GOT, GPT and ALP activity was significantly lower in EGP group. Total lipid, cholesterol and triglyceride contents in liver were significantly lower in EGP and EGC group than control group. Antioxidant activity in serum was the highest in EGC groups by 50.86%, in liver was the highest in EGP groups. TBARS content in serum and liver was the lowest in EGP group. In these results, we suggest that EGP composites could have hypolipidemic and anti-obesity effects in rats fed a high fat diet.

Correlation between phytochemical contents in peel and flesh of cold-stored Fuji apple (저온 저장 후지 사과의 과피와 과육 중 파이토케미컬 함량의 상관관계)

  • This study determined the relationships between the total anthocyanin content in apple skin and the total polyphenols, flavonoids, proanthocyanidins, and soluble solids contents in the flesh of cold-stored Fuji apples. Total anthocyanin content in apple skin ranged from 0.130±0.005 mg CE/g fw to 0.262±0.028 mg CE/g fw, and the overall average was 0.200±0.008 mg CE/g fw. The total polyphenolic compounds in the flesh was ranged from a 4.283±0.141 mM GAE/g fw to 8.207±0.234 mM GAE/g fw, and the average content was 6.275±0.177 mM GAE/g fw. The total flavonoid content ranged from 4.510±0.080 mM QE/g fw to 2.467±0.458 mM QE/g fw, and the average total flavonoid content was about 3.586 mM QE/g fw. The total proanthocyanidin content was relatively high, ranging from 3.475±0.577 mM EE/g fw to 6.816±0.277 mM EE/g fw, and the soluble solid in the flesh was about 12 °Brix to 14 °Brix. The DPPH radical scavenging activity of extracts from apple flesh ranged from 66.36% to 94.99%, and the ascorbate equivalent concentration was 0.482 mM. The ABTS radical scavenging activity was 99.12% to 99.9%, indicating a higher inhibitory activity than the DPPH inhibitory activity, and the ascorbate equivalent concentration was 0.486 mM. The correlation between the total anthocyanin and total polyphenolic compounds was y = 15.192x + 3.2169 (R2=0.2748), but the concentration of total polyphenolic compounds increased when the total anthocyanin content was increased. The correlation equation of total anthocyanin with total flavonoids was y = 15.18x + 0.5555 (R2=0.6226), with total proanthocyanin was y = 14.918x + 2.3422 (R2=0.3372), and with soluble solid was y = 10.558x + 11.126 (R2=0.1925), indicating that the correlation of total anthocyanin with total flavonoid was higher than that with soluble solid.
