Purification and Characterization of an Exo-polygalacturonase from Botrytis cinerea

  • Botrytis cinerea T91-1 has been shown to produce at least four different polygalacturonases into a liquid medium containing citrus pectin, a carbon sousrce. One of the enzymes, which had an apparent molecular weight of 66 kDa estimated by denatured polyacrylamide gel electrophoresis, was purified to electrophoretic homogeneity by a series of procedures including a cetone precipitation, ion exchange, heparin affinity, and reverse phase column chromatographies. The molecular weight of native enzyme was determined to be 64 kDa by gel permeation chromatography indicating the enzyme to be a single polypeptide chain. By viscometric analysis, the enzyme was revealed as exo-polygalacturonase. The enzyme activity was inhibited by divalent cations such as $Ca^{2+}$, $Mg^{2+}$, and Cu$^{2+}$. Km and Vmax for polygalacturonic acid hydrolysis were 0.33 mg/ml and 28.6 nM/min, respectively. The optimum temperature for enzymatic activity was 5$0^{\circ}C$. And the enzyme showed optimal pH values between 4.0 and 5.0. The enzyme was stable upto 12 hours in the range of pH 3 to 8 and at temperature below 3$0^{\circ}C$.

Byssochlamys fulca가 생성하는 펙틴질 분해효소에 관한 연구 II (Studies on the Pectolytic Enzymes from Byssochlamys fulva II)

  • Polygalacturonase of Byssochlamys fulva was purified and characterized. Specific activity increased from 2.21 units/mg protein to 10.47 units/mg protein through $(NH_4)_2SO_4$ treatment, SephadexG-100 gel filtration, and DEAE-Sephadex ion exchange chromatography. Divalent cations, such as $Ca^{++}\;and\;Cu^{++}$, increased polygalacturonase activity greatly. Added as $10^{-3}M$ concentration, $Ca^{++}$ ion enhanced enzyme activity 9.8folds. Optimum temperature was $50^{\circ}C$ and optimum pH was 5.0. Activation energy of reaction was 8.69 Kcal/mole. Michaelis-Menten $constant(K_M)\;and\;V_{max}$ of reaction were $6.27{\times}10^{-3}mole/l\;and\;2.85{$\mu}moles/min$. Polygalacturonase of Byssochlamys fulva preferred polygalacturonic acid to pectin as substrate and was presumed as endo-type on the basis of the relationship between viscosity reduction and substrate degradation. Molecular weight of polygalacturonase was estimated as 55,000.

  • This study was conducted to examine morphological characteristics and the content of cell wall polysaccharides of Brassica campestris var. pekinensis(baechu). First of all, the variety of scientific name and naming of parts of baechu in the literatures of kimchi showed, which will unify marks. So, we propose not so much mid-rib and leaf blade of baechu leaf as white part and green part, respectively. On the other hand, the forms of vessel elements of white part in baechu consist in ring, sclariform and reticulate thickening. The proximate compositions and contents of cell wall polysaccharides of baechu has significant differences between its cultivars. The cell wall pectin from baechu exhibited four peals with molecular weights of 2,000,000, about 100,000 and less than 10,000 by gel filteration chromatography and hemicellulose did two peaks with molecular weights of 2,000,000 and 10,000.

  • Polygalacturonase (PG) produced by Botrytis cinerea in the culture broth containing citrus pectin as a carbon source was partially purified and characterized. PG was produced on a range of carbon sources such as starch, glycerol, cellobiose, and Na+-PAG with total activities of 34.8, 32.0, 29.2, 27.8 units, respectively. The specific activity was highest with 2316.7 units on Na+-PGA. Proteins of culture filtrate were concentrated with polyethylene glycol and acetone and applied to a hydroxyapatite column. Among three active fractions collected from the column, the reaction containing the highest PG activity was resolved by a Q-sepharose column. The active fraction from the Q-sepharose column was further purified by HPLC Mono Q column. The partially purified enzyme was analyzed by 10% sodium dodecyl sulfate-polyacrylamide gel electrophoresis. Among a few protein bands revealed, the amount of the protein of which molecular weight estimated to be 43 kDa coincided with the PG activity. The partially purified PG had optimal temperatures between 35~55$^{\circ}C$ and pH between 4.5~5.5.

  • The protopectinase from the culture extract of a Verticillium sp. was purified about 1000 fold by ammonium sulfate fractionation, DEAE-Sephadex treatment and Sephadex G-75 column chromatography. The purified enzyme was homogeneous on electrophoresis and its molecular weight was estimated to be 38000 by Andrew's gel filtration, method. The enzyme was almost stable under the temperature of 4$0^{\circ}C$ and within the pH range of 3-5. Its optimum pH and temperature were 4 and 4$0^{\circ}C$, respectively. The activity was markedly inhibited by galacturonic acid. The purified enzyme was able to macerate various kinds of plant tissues, such as radish, cucumber, onion, carrot, and potato. It also reduced the viscosity of pectin solution more rapidly than that of pectic acid solution and showed no lyase or CMCase activity.

카테킨이 함유된 창상피복제용 하이드로젤의 개발 (Development of Hydrogel Containing Catechin for Wound Dressing)

  • Catechin(CTEC)은 천연항산화제로 알려져 있고 항염증에 효과적이며 피부 창상치료제로 응용되고 있다. 이 연구는 ${\beta}$-cyclodextrin(${\beta}$-CD)으로 나노입자화한 CTEC을 poly(vinyl alcohol)(PVA)/pectin(PT) 조성의 하이드로젤에 첨가하였다. 피부상처의 재상피화를 위해 CTEC과 ${\beta}$-CD을 분자복합(molecular complex) 방법에 의해 나노입자 제조를 수행하였다. 하이드로젤에 첨가된 CTEC 나노입자는 $250{\pm}17.5$ nm이며, 입자 내 CTEC의 담지 효율은 처음첨가량의 74%가 담지되었음을 확인하였다. CTEC는 하이드로젤 필름으로부터 pH7.4와 pH5.5 버퍼에서 72시간 동안 각각 $86.51{\pm}3.14%$$35.95{\pm}2.14%$의 방출을 보였다. 창상치료 실험에서 CTEC 나노입자가 적재된 PVA/PT 조성의 하이드로젤은 CTEC이 함유된 도포형 젤보다 빠른 치유력을 관찰할 수 있었다.

  • 양파 무름병 병원균인 Burkholder gladioli pv. allicola CH1을 양파에 접종했을 때 $20^{\circ}C$ 이상에서는 전형적인 양파 무름병 증상을 보이면서 심하게 부패하였지만, $10^{\circ}C$ 이하에서는 일부 조직이 무름증상을 보였지만 겉으로는 건전하게 보였다. B. gladioli pv. allicola CH1은 oxolinic acid WP, streptomycin + copper hydroxide WP와 Streptomycin WP에 강한 감수성을 보였다. 분비효소 중 펙틴질 분해효소의 활성은 검출할 수 있었지만, 섬유소 분해효소인 carboxymethylcellulase 활성은 검출되지 않았다. 그러나 펙틴질 분해활성 중에서 pectin(pectate) lyase의 활성은 검출되지 않고 polygalacturonase 활성만 검출할 수 있었다. PGA 액체배지에서 체외 효소는 18시간까지 급속한 효소반응을 보였으며, 24시간 후에 최고치를 가진 후 계속적인 활성을 가졌으며 효소의 생성은 균의 생육과 거의 일치하는 것으로 나타났다. PGA-SDS-PAGE에 의한 섬유소분해효소 직접활성 염색법을 이용하여 3종류의 polygalacturonase isozyme으로 추정되는 45 kDa, 35 kDa, 29 kDa 크기의 활성밴드를 관찰할 수 있었다.

Effective Use of Orange Juice Residue for Removing Heavy and Radioactive Metals from Environments

  • Large amounts of orange juice are produced in Japan every yea.. Accompanied by the production of orange juice, large amount of juice residues are also generated in nearly the same amounts with juice. Although, at present, some of these residues are marketed as a feed for cattle after drying and mixing with lime, the marketing price is lower than its production cost and the difference is paid by the consumers as a part of the price of orange juice. In the present work, we developed new innovative use of orange juice residue, a biomass waste, as adsorption gel for removing toxic heavy metals such as lead. arsenic, selenium and so on as well as radioactive elements such as uranium and thorium from environments. The major components of orange juice residue are cellulose. hemicellulose and pectin, which are converted into pectic. acid, an acidic polysaccharide, by means of saponification with concentrated sodium hydroxide solution. In the previous work, we found that crosslinked pectic acid gel strongly an selectively adsorbs lead over other metals such as zinc an copper. On the other hand. it is well known that polysaccharides such as cellulose can be easily phosphorylated and that phosphorylated polysaccharides have high affinity to uranium and thorium as well as some trivalent metals such as ferric iron and aluminum. Taking account of the noticeable characteristics of these polysaccharides, 2 types of adsorption gels were prepared from orange juice residue: one is the gel which was prepared by saponificating the residue followed by crosslinking with epichlorohydrin and another is that prepared by crosslinking the residue followed by phosphorylation. The former gel exhibited excellent adsorptive separation behavior for lead away from zinc owing to high content of pectic acid while the latter gel exhibited that for uranium and thorium. Both types of adsorption gels exhibited high affinity to ferric iron, which enables selective and strong adsorption for some toxic oxo-anions of arsenic (V and III), . selenium and so on via iron loaded on these gels. These results demonstrate that biomass wastes such as orange juice residue can be effectively utilized fer the purpose of removing toxic heavy or radioactive metals existing in trace or small amounts in environments.

Effects of Heat, Salt and Hydrocolloid Treatments on Flying Fish Cypselurus agoo Roe Analogs Prepared Using Calcium Alginate Hydrogels

  • The present study was conducted to elucidate effects of heat, salt and hydrocolloid treatments on flying fish Cypselurus agoo roe analogs prepared using calcium alginate gel. The changes in size, sphericity and rupture strength of the analogs as affected by treatments of heat, sodium chloride and hydrocolloids were investigated. The size (mm), sphericity (%), and rupture strength (kPa) of the analogs were $2.2{\pm}0.1$, $98.2{\pm}0.2$, and $74.7{\pm}1.7$, respectively. When the analogs were heated at $95^{\circ}C$ in water, the size was slightly decreased. The rupture strength by curing with 2% sodium chloride was slightly increased. Sphericity didn't show significant differences by sodium chloride and heat treatment. The rupture strength of the analogs was slightly decreased by heat treatment, whereas remarkably decreased by curing with sodium chloride. In order to prevent a remarkable decrease in rupture strength of the analogs by curing with sodium chloride, the analogs were treated with hydrocolloids such as xanthan gum, gum guar, glucomannan, pectin and gelatin. The hydrocolloids treated analogs showed an increment in size and no significant changes in sphericity. On the other hand, the rupture strength of the hydrocolloids treated analogs exhibited remarkable increase than that of untreated ones.

Enhancing the Viability Rate of Probiotic by Co-Encapsulating with Prebiotic in Alginate Microcapsules Supplemented to Cupcake Production

  • The objective of the study was to assess the survival of microencapsulated Lactobacillus plantarum ATCC8014 produced using the emulsion technique in alginate gel combined with pectin and maltodextrin components. The microcapsules were then added to cupcake dough that was further baked at 200℃ for 12 min. The viability of L. plantarum was assessed during baking and the 10 days of storage at 4℃ as well as in simulated gastrointestinal conditions. In addition, yeast-mold and water activity were investigated. After baking, the samples with microencapsulated L. plantarum contained more than 5 log CFU/g, which was higher compared to the bacterial concentration of the control samples. The concentration of L. plantarum was more than 6 logs CFU/g after the end of the storage; therefore, the probiotic functioned as a biopreservative in the cake. The prebiotic component strengthened the microcapsules network and helped protect the viability of L. plantarum in simulated gastric fluid (SGF) and simulated intestinal fluid (SIF) media. The results show that the addition of L. plantarum microencapsules did not affect the sensory scores of the cupcake while ensuring the viability of the probiotic during baking and storing.
