Phenazine 1-carboxylic acid resistance in phenazine 1-carboxylic acid producing Bacillus sp. B-6

  • Phenazine 1-carboxylic acid (PCA) is an antifungal antibiotic isolated from a culture filtrate of Bacillus sp. B-6 producing an acyl CoA synthetase inhibitor. This antibiotic is reported as an inhibitor of an acyl CoA synthetase from Pseudomonas sp.. Bacillus sp. B-6 was resistant to PCA up to 350 ${\mu}g/ml$. We investigated the mechanism of the resistance of Bacillus sp. B-6 to PCA. The rate of growth in a medium containing up to 100 ${\mu}g/ml$ was as rapid as the PCA-free medium. At a PCA concentration of 300 ${\mu}g/ml$, the growth rate was more than half that of the control. In this work, we purified acyl CoA synthetase from Bacillus sp. B-6 and found that this acyl CoA synthetase was much less sensitive to PCA than the acyl CoA synthetase from other source. These findings suggested that the insensitivity of Bacillus sp. B-6 acyl CoA synthetase plays an important role in the PCA resistance of this bacterium.

A Study on Face Recognition Method based on Binary Pattern Image under Varying Lighting Condition (조명 변화 환경에서 이진패턴 영상을 이용한 얼굴인식 방법에 관한 연구)

  • In this paper, we propose a illumination-robust face recognition system using MCS-LBP and 2D-PCA algorithm. A binary pattern transform which has been used in the field of the face recognition and facial expression, has a characteristic of robust to illumination. Thus, this paper propose MCS-LBP which is more robust to illumination than previous LBP, and face recognition system fusing 2D-PCA algorithm. The performance evaluation of proposed system was performed by using various binary pattern images and well-known face recognition features such as PCA, LDA, 2D-PCA and ULBP histogram of gabor images. In the process of performance evaluation, we used a YaleB face database, an extended YaleB face database, and a CMU-PIE face database that are constructed under varying lighting condition, and the proposed system which consists of MCS-LBP image and 2D-PCA feature show the best recognition accuracy.

UVB-Shielding Effects of para-Coumaric Acid (파라-쿠마린산의 자외선B 차단 효과)

  • Recent studies have uncovered attractive properties of para-coumaric acid (PCA) as a potential skin hywhitening agent. The purpose of the current study was to examine its UVB-shielding effects. Effects of PCA on the viability of HaCaT cells exposed to UVB were assessed in vitro in comparison with other aromatic amino acid metabolites that have similar UV absorption spectra. For in vivo test, PCA cream (1.5 %) and cream base were topically applied to the dorsal skin of SKH-1 hairless mice and the inflammatory responses due to UVB exposure were monitored by changes in skin color (erythema) and thickness (edema). The cream application-UVB exposure regimen was repeated every other day for a total of 12 sessions. When HaCaT cells were irradiated with UVB, there was a dose-dependent decline in cell viability. The cell viability decline due to UVB exposure (10 mJ $cm^{-2}$) was significantly prevented by 100 ${\mu}M$ PCA, cinnamic acid, urocanic acid, or indole acrylic acid by 39, 27, 39, or 31 %, respectively. Topical application of PCA cream onto the dorsal skin of hairless mice (10 ${\mu}g\;cm^{-2}$) attenuated the changes of color parameters, $L^*$, $a^*$, $b^*$ values, and thickness of the UVB (150 mJ $cm^{-2}$)-exposed skin by 59, 50, 58, and 53 %, respectively. The current study, together with the previous studies that demonstrated the antimelanogenic effects of PCA, suggested that PCA may prevent not only dyspigmentation but also inflammatory reactions in the UVB-exposed skin.

Cloning of p-Hydroxybenzoate Degradation Genes and the Overexpression of Protocatechuate 4,5-Dioxygenase from Pseudomonas sp. K82

  • Pseudomonas sp. K82 cultured in p-hydroxybenzoate induces protocatechuate 4,5-dioxygenase (PCD 4,5) for p-hydroxybenzoate degradation. In this study, a 6.0-kbp EcoR1 fragment containing p-hydroxybenzoate degradation genes was cloned from the genome of Pseudomonas sp. K82. Sequence analysis identified four genes, namely, pcaD, pcaA, pcaB, and pcaC genes known to be involved in p-hydroxybenzoate degradation. Two putative 4-hydroxyphenylpyruvate dioxygenases and one putative oxidoreductase were closely located by the p-hydroxybenzoate degradation genes. The gene arrangement and sequences of these p-hydroxybenzoate degradation genes were similar to those of Comamonas testosteroni and Pseudomonas ochraceae. PcaAB (PCD4,5) was overexpressed in the expression vector pGEX-4T-3, purified using a GST column, and confirmed to have protocatechuate 4,5-dioxygenase activity. The N-terminal amino acid sequences of overexpressed PCD4,5 were identical with those of purified PCD4,5 from Pseudomonas sp. K82.

Genetic Polymorphism of Interleukin-1A (IL-1A), IL-1B, and IL-1 Receptor Antagonist (IL-1RN) and Prostate Cancer Risk

  • Purpose: We aimed to investigate the associations between polymorphisms of interleukin-1A (IL-1A), IL-1B, and IL-1 receptor antagonist (IL-1RN) and prostate cancer (PCa) risk. Materials and Methods: A comprehensive search for articles of MEDLINE and EMBASE databases and bibliographies of retrieved articles published up to August 3, 2014 was performed. Methodological quality assessment of the trials was based on a standard quality scoring system. The meta-analysis was performed using STATA 12.0. Results: We included 9 studies (1 study for IL-1A, 5 studies for IL-1B, and 3 studies for IL-1RN), and significant association was found between polymorphisms of IL-1B-511 (rs16944) as well as IL-1B-31 (rs1143627) and PCa risk. IL-1B-511 (rs16944) polymorphism was significantly associated with PCa risk in homozygote and recessive models, as well as allele contrast (TT vs CC: OR, 0.74; 95%CI, 0.58-0.94; P=0.012; TT vs TC+CC; OR, 0.79; 95%CI, 0.63-0.98; P=0.033; T vs C: OR, 0.86; 95%CI, 0.77-0.96; P=0.008). The association between IL-1B-31 (rs1143627) polymorphism and PCa risk was weakly significant under a heterozygote model (OR, 1.35; 95%CI, 1.00-1.80; P=0.047). Conclusions: Sequence variants in IL-1B-511 (rs16944) and IL-1B-31 (rs1143627) are significantly associated with PCa risk, which provides additional novel evidence that proinflammatory cytokines and inflammation play an important role in the etiology of PCa.

An Off-line Signature Verification Using PCA and LDA (PCA와 LDA를 이용한 오프라인 서면 검증)

  • Among the biometrics, signature shows more larger variation than the other biometrics such as fingerprint and iris. In order to overcome this problem, we propose a robust offline signature verification method based on PCA and LDA. Signature is projected to vertical and horizontal axes by new grid partition method. And then feature extraction and decision is performed by PCA and LDA. Experimental results show that the proposed offline signature verification has lower False Reject Rate(FRR) and False Acceptance Rate(FAR) which are 1.45% and 2.1%, respectively.

Effects of Protocatechuic Acid Derived from Rubus coreanus on the Lipid Metabolism in High Cholesterol Diet-induced Mice (복분자 유래 성분 protocatechuic acid 투여가 고콜레스테롤 식이로 유도된 생쥐의 지질대사에 미치는 영향)

  • Rubus coreanus has been used as a traditional medicine in Asia because of its various pharmacological properties. This study examined the effects of protocatechuic acid (PCA), one of phenolic compounds derived from R. coreanus on the lipid metabolism in high cholesterol diet-induced mice. A total of 30 male C57BL/6 mice were divided into 5 groups with 6 mice in each group as follows: (1) Control mice received normal diet (ND). (2) Mice received high-cholesterol diet (HCD) plus water, 10% sucrose solution and treated daily oral phosphate-buffered-saline (PBS) of equal volumes through gavage. (3) Mice received HCD and treated daily with 25 mg/kg b.w./day of PCA (4) with 50 mg/kg b.w./day or (5) with 10 mg/kg b.w./day of simvastatin via oral gavage for 12 weeks. Body weights were measured weekly for a period of experiment. After treatment, liver, thymus, spleen and kidney were harvested and weighed, and the lipid metabolite profiles (total cholesterol, triglyceride (TG), HDL-cholesterol (HDL-c), LDL-cholesterol (LDL-c) and liver-damaging markers (GOT and GPT) in serum were examined. PCA significantly reduced the total cholesterol, TG, LDL-c level and increased the HDL-c level. PCA administration also significantly reduced the levels of GOT and GPT. These results indicate that the PCA could be used as a functional material for lowering lipid and an adjuvant for the treatment of hyperlipemia.

A Study to Improve the Classification Accuracy of Mosaic Image over Korean Peninsula: Using PCA and RGB Indices (한반도 모자이크 영상의 분류 정확도 향상 기법 연구: PCA 기법과 RGB 지수를 활용하여)

  • Korea Aerospace Research Institute produces mosaic images of the Korean Peninsula every year to promote the use of satellite images and provides them to users in the public sector. However, since the pan-sharpening and color balancing methodologies are applied during the mosaic image processing, the original spectral information is distorted. In addition, there is a limit to analyze using mosaic images as mosaic images provide only Red, Green and Blue bands excluding Near Infrared (NIR) band. Therefore, in order to compensate for these limitations, this study applied the Principal Component Analysis (PCA) technique and indices extracted from R, G, B bands together for image classification and compared the classification results. As a result of the analysis, the accuracy of the mosaic image classification result was about 67.51%, while the accuracy of the image classification result using both PCA and RGB indices was about 75.86%, confirming that the accuracy of the image classification result can be improved. As a result of comparing the PCA and the RGB indices, the accuracy of the image classification result was about 64.10% and 74.05% respectively. Through this, it was confirmed that the classification accuracy using the RGB indices was higher among the two techniques, and implications were derived that it was important to use high quality reference or supplementary data. In the future, additional indices and techniques are needed to improve the classification and analysis results of mosaic images, and related research is expected to increase the utilization of images that provide only R, G, B or limited spectral information.

Face Recognition using Modified Local Directional Pattern Image (Modified Local Directional Pattern 영상을 이용한 얼굴인식)

  • Generally, binary pattern transforms have been used in the field of the face recognition and facial expression, since they are robust to illumination. Thus, this paper proposes an illumination-robust face recognition system combining an MLDP, which improves the texture component of the LDP, and a 2D-PCA algorithm. Unlike that binary pattern transforms such as LBP and LDP were used to extract histogram features, the proposed method directly uses the MLDP image for feature extraction by 2D-PCA. The performance evaluation of proposed method was carried out using various algorithms such as PCA, 2D-PCA and Gabor wavelets-based LBP on Yale B and CMU-PIE databases which were constructed under varying lighting condition. From the experimental results, we confirmed that the proposed method showed the best recognition accuracy.

Preprocessing and Facial Feature Robust to Illumination Variations (조명변화에 강인한 전처리 및 얼굴특징)

  • In this paper, we propose the face recognition method combining the ECSP preprocessing technique which is modified version of previous CS-LBP and the illumination-robust D2D-PCA feature. The performance evaluation of proposed method was carried out using various binary pattern operators and feature extraction algorithms such as well-known PCA and 2D-PCA on the Yale B database. As a results, the proposed method showed the best recognition accuracy compared to different approaches, and we confirmed that the proposed approach is robust to illumination variation.
