Assessment of Resistance Induction in Mungbean against Alternaria alternata through RNA Interference

  • A comprehensive survey of mungbean-growing areas was conducted to observe leaf spot disease caused by Alternaria alternata. Alternaria leaf spot symptoms were observed on the leaves. Diversity of 50 genotypes of mungbean was assessed against A. alternata and data on pathological traits was subjected to cluster analysis. The results showed that genotypes of mungbean were grouped into four clusters based on resistance parameters under the influence of disease. The principal component biplot demonstrated that all the disease-related parameters (% disease incidence, % disease intensity, lesion area, and % of infection) were strongly correlated with each other. Alt a 1 gene that is precisely found in Alternaria species and is responsible for virulence and pathogenicity. Alt a 1 gene was amplified using gene specific primers. The isolated pathogen produced similar symptoms when inoculated on mungbean and tobacco. The sequence analysis of the internal transcribed spacer (ITS) region, a 600 bp fragment amplified using specific primers, ITS1 and ITS2 showed 100% identity with A. alternata. Potato virus X (PVX) -based silencing vector expressing Alt a 1 gene was constructed to control this pathogen through RNA interference in tobacco. Out of 50 inoculated plants, 9 showed delayed onset of disease. Furthermore, to confirm our findings at molecular level semi-quantitative reverse transcriptase polymerase chain reaction was used. Both phenotypic and molecular investigation indicated that RNAi induced through the VIGS vector was efficacious in resisting the pathogen in the model host, Tobacco (Nicotiana tabacum). To the best of our knowledge, this study has been reported for the first time.

코로나19 감염증 환자의 혈전 사례를 바탕으로 본 태양병 축혈증의 병리기전에 관한 문헌고찰 (Pathological Mechanism of Taeyang Blood Retention Pattern Based on Cases of Thrombosis in Patients with COVID-19 Infection : A Literature Review)

  • COVID-19 infection heightens the risk of thromboembolism. To see the similarities between the COVID-19 infection and Taeyang blood retention pattern, we conducted a PubMed search using specific terms related to blood circulation issues in the context of COVID-19, summarizing findings from 13 cases and 4 observational studies involving actual patients. Patients with COVID-19 are at risk of blood coagulation due to factors such as viral-induced cytokine storms, vascular endothelial dysfunction, reduced mobility in bedridden or isolated individuals, and resulting constipation. Additionally, cytokine storms and severe inflammation can lead to delirium in COVID-19 patients. The Taeyang blood retention pattern manifests as symptoms arising from delirium and an increased blood coagulation tendency in patients with a robust immune response. According to the Sanghan theory, certain herbal treatments can alleviate symptoms in patients with a tight lower abdomen who do not experience urinary issues. Studies show that components like Persicae Semen and Rhei Redix et Rhizoma in these prescriptions enhance blood circulation and reduce hypercoagulability. Additionally, these treatments aim to promote blood flow by relieving abdominal pressure through facilitating bowel movements. The excessive inflammation and heightened blood coagulation tendency in COVID-19 resemble the Taeyang blood retention pattern, although they are caused by different pathogens. Reinterpreting classical oriental medicine's principles in a modern context may enhance our understanding of traditional East Asian Medicine and foster future developments.

Preclinical Evidence and Underlying Mechanisms of Polygonum multiflorum and Its Chemical Constituents Against Cognitive Impairments and Alzheimer's Disease

  • Objectives: Cognitive impairments, ranging from mild to severe, adversely affect daily functioning, quality of life, and work capacity. Despite significant efforts in the past decade, more than 200 promising drug candidates have failed in clinical trials. Herbal remedies are gaining interest as potential treatments for dementia due to their long history and safety, making them valuable for drug development. This review aimed to examine the mechanisms behind the effect of Polygonum multiflorum on cognitive function. Methods: This study focused primarily on the effects of Polygonum multiflorum and its chemical constituents on cognitive behavioral outcomes including the Morris water maze, the passive avoidance test, and the Y maze, as well as pathogenic targets of cognitive impairment and Alzheimer's disease (AD) like amyloid deposition, amyloid precursor protein, tau hyperphosphorylation, and cognitive decline. Additionally, a thorough evaluation of the mechanisms behind Polygonum multiflorum's impact on cognitive function was conducted. We reviewed the most recent data from preclinical research done on experimental models, particularly looking at Polygonum multiflorum's effects on cognitive decline and AD. Results: According to recent research, Poligonum multiflorum and its bioactive components, stilbene, and emodin, influence cognitive behavioral results and regulate the pathological target of cognitive impairment and AD. Their mechanisms of action include reducing oxidative and mitochondrial damage, regulating neuroinflammation, halting apoptosis, and promoting increased neurogenesis and synaptogenesis. Conclusion: This review serves as a comprehensive compilation of current experiments on AD and other cognitive impairment models related to the therapeutic effects of Polygonum multiflorum. We believe that these findings can serve as a basis for future clinical trials and have potential applications in the treatment of human neurological disorders.

육계에서 발생한 흡수불량증후군 증례 (A case report of runting-stunting syndrome in a broiler farm)

  • A broiler farm in Jeonbuk province experienced high mortality due to decreased feed intake and poor growth, and pathological evaluation of 11- and 17-day-old broilers was performed, which led to the diagnosis of runting-stunting syndrome. From the start of the rearing period to shipment, 18.4% of the chickens in the three barns experiencing continuous culling and mortality were affected, compared to 7.7% in the other five barns. Gross findings on the 11-day-old broiler chicken revealed proventricular dilatation and hemorrhage, intestinal hemorrhage, urate deposition in the pericardium and renal tubule, nephropathy, and mild hepatic capsulitis. Similar proventricular dilatation and hemorrhage were observed in the 17-day-old broiler chickens. In addition, hepatitis and pericarditis were observed with the progression of secondary bacterial infection, and pathogenic Escherichia coli was isolated from these lesions. As a result of PCR, Newcastle disease virus, fowl adenovirus, chicken anemia virus and Marek's disease virus were not detected in the all tissue samples. In contrast, infectious bronchitis virus was detected in the proventriculus, kidney and cecal tonsil. chicken astrovirus was detected in the intestine, cecal tonsil and bursa of Fabricius, and chicken parvovirus was detected in proventriculus, intestine, cecal tonsil and bursa of Fabricius. By sharing the diagnostic process of a case of malabsorption syndrome through this case report, we hope that it can be widely utilized in the diagnostic process of livestock disease pathognomonic institutions.

좌심방 부속기 동맥류: 증례 보고 (Left Atrial Appendage Aneurysm: A Case Report)

  • 좌심방 부속기 동맥류는 심장 빗살근의 선천성 이형성이나 승모판 또는 심근의 후천적 병변에 의해 생길 수 있는 매우 드문 심장 질환이다. 주로 흉부 CT나 심초음파에서 좌심방이 비정상적 팽창되어 우연히 발견되며, 환자의 대부분은 무증상이지만 생명을 위협하는 합병증과 관련이 있어 외과적 치료를 필요로 한다. 그러므로 정확한 진단과 함께 이에 동반된 합병증을 찾아내는 것은 중요하다. 본 증례 보고에서는 수술적 치료로 확진 된 53세 여성의 좌심방 부속기 동맥류를 소개하고 병태생리학을 바탕으로 진단과 함께 놓치지 말아야 할 합병증에 대한 CT 및 MRI 검사의 소견을 소개하고자 한다.

미세낭종성 혈관종성 혼합 수막종의 영상 및 병리 소견: 증례 보고 (Imaging Features and Pathological Correlation in Mixed Microcystic and Angiomatous Meningioma: A Case Report)

  • 수막종은 가장 흔한 두개내 종양이지만, 그 아형인 미세낭종성 및 혈관종성 수막종은 매우 드물고 특이한 영상 소견으로 인해 영상의학적 진단이 어려울 수 있다. 이에 저자들은 미세낭종성 혈관종성 혼합 수막종으로 진단된 81세 남자 환자 증례를 보고하고자 한다. 자기공명 T2 강조영상에서 병변은 왼쪽 전두부에 약 1.5 cm 크기의 고신호강도의 축외 종양으로 보이고 있었고, 종양 내에 다수의 아주 작은 낭종들이 관찰되었으며, 종양 주위로 포획된 큰 낭을 형성하고 있었다. 수술적 절제를 통한 병리 검사상 미세낭종성 혈관종성 혼합 수막종으로 최종 진단되었다.

MR Imaging-Histopathologic Correlation of Radiofrequency Thermal Ablation Lesion in a Rabbit Liver Model: Observation during Acute and Chronic Stages

  • Objective: To determine the ability of MR imaging to detect the pathological changes occurring in radiofrequency (RF) thermal lesions and to assess its accuracy in revealing the extent of tissue necrosis. Materials and Methods: Using an RF electrode, thermal lesions were created in the livers of 18 rabbits. The procedure involved three phases. In the acute phase, six animals were killed the day after performing thermal ablation with RF energy, and two on day 3. In the subacute and chronic phases, eight rabbits underwent percutaneous hepatic RF ablation. After performing MR imaging, two animals were sacrificed at 1, 2, 4, and 7 weeks after the procedure, and MRpathologic correlation was performed. Results: In the acute phase, the thermal ablation lesions appeared at gross examination as well-circumscribed, necrotic areas, representing early change in the coagulative necrosis seen at microscopic examination. They were hypointense on T2-weighted images, and hyperintense on T1-weighted images. Gadolinium-enhanced MR imaging showed that a thin hyperemic rim surrounded the central coagulative necrosis. In the subacute phase, ablated lesions also showed extensive coagulative necrosis and marked inflammation at microscopic examination. Beyond two weeks, the lesions showed gradual resorption of the necrotic area, with a peripheral fibrovascular rim. The size of lesions measured by MR imaging correlated well with the findings at gross pathologic examination. Conclusion: MR imaging effectively demonstrates the histopathological tissue change occurring after thermal ablation, and accurately determines the extent of the target area.

Combination of Magnetic Resonance Spectroscopy and 11C-Methionine Positron Emission Tomography for the Accurate Diagnosis of Non-Enhancing Supratentorial Glioma

  • Objective: To evaluate whether the combination of magnetic resonance spectroscopy (MRS) and 11C-methionine positron emission tomography (11C-MET PET) could increase accurate diagnostic sensitivity for non-enhancing supratentorial gliomas. Materials and Methods: Between February 2012 and December 2017, 109 patients with non-enhanced supratentorial lesions on contrast-enhanced MRI were enrolled. Each patient underwent MRS and 11C-MET PET before treatment. A lesion was considered to be a glioma when either the MRS or 11C-MET PET results reached the diagnostic threshold. The radiological diagnosis was compared with the pathological diagnosis or medical diagnostic criteria. Results: The sensitivity and specificity were 60.0% and 50.0% for MRS and 75.8% and 50.0% for 11C-MET PET, respectively. Upon combining the two modalities, the sensitivity and specificity of the imaging-based diagnosis prior to surgery reached 89.5% and 42.9%, respectively. Statistically significant differences in the sensitivities were observed between the combined and individual approaches (MRS alone, 89.5% vs. 60.0%, p < 0.001; 11C-MET PET alone, 89.5% vs. 75.8%, p = 0.001). However, no significant differences in specificity were observed between the combined and individual modalities. Conclusion: The combination of MRS and 11C-MET PET findings significantly increases accurate diagnostic sensitivity for non-enhancing supratentorial gliomas without significantly lowering the specificity. This finding suggests the potential of the combined MRS and 11C-MET PET approach in clinical applications.

Anti-Inflammatory Activities of (+)-Afzelechin against Lipopolysaccharide-Induced Inflammation

  • In this study, we investigated the potential protective effects of (+)-afzelechin (AZC), a natural compound that is derived from Bergenia ligulata, on lipopolysaccharide (LPS)-induced inflammatory responses. AZC is known to have antioxidant, anticancer, antimicrobial, and cardiovascular protective properties. However, knowledge regarding the therapeutic potential of AZC against LPS-induced inflammatory responses is limited. Thus, we investigated the protective attributes of AZC against inflammatory damage caused by LPS exposure. We examined the effects of AZC on heme oxygenase (HO)-1, cyclooxygenase (COX)-2, and inducible nitric oxide synthase (iNOS) in LPS-activated human umbilical vein endothelial cells (HUVECs). In addition, the effects of AZC on the expression of iNOS, tumor necrosis factor (TNF)-α, and interleukin (IL)-1β were analyzed in the lung tissues of LPS-injected mice. Data revealed that AZC promoted the production of HO-1, inhibited the interaction between luciferase and nuclear factor (NF)-κB, and reduced the levels of COX-2/PGE2 and iNOS/NO, thereby leading to a decrease in the signal transducer and activator of transcription (STAT)-1 phosphorylation. Moreover, AZC facilitated the nuclear translocation of Nrf2, increased the binding activity between Nrf2 and the antioxidant response elements (AREs), and lowered the expression of IL-1β in the LPS-treated HUVECs. In the animal model, AZC significantly reduced the expression of iNOS in the lung tissue structure and the TNF-α level in the bronchoalveolar lavage fluid. These findings demonstrate that AZC possesses anti-inflammatory properties that regulate iNOS through the inhibition of both NF-κB expression and p-STAT-1. Consequently, AZC has potential as a future candidate for the development of new clinical substances for the treatment of pathological inflammation.

A comparison study of pathological features and drug efficacy between Drosophila models of C9orf72 ALS/FTD

  • Amyotrophic lateral sclerosis is a devastating neurodegenerative disease with a complex genetic basis, presenting both in familial and sporadic forms. The hexanucleotide (G4C2) repeat expansion in the C9orf72 gene, which triggers distinct pathogenic mechanisms, has been identified as a major contributor to familial and sporadic Amyotrophic lateral sclerosis cases. Animal models have proven pivotal in understanding these mechanisms; however, discrepancies between models due to variable transgene sequence, expression levels, and toxicity profiles complicate the translation of findings. Herein, we provide a systematic comparison of 7 publicly available Drosophila transgenes modeling the G4C2 expansion under uniform conditions, evaluating variations in their toxicity profiles. Further, we tested 3 previously characterized disease-modifying drugs in selected lines to uncover discrepancies among the tested strains. Our study not only deepens our understanding of the C9orf72 G4C2 mutations but also presents a framework for comparing constructs with minute structural differences. This work may be used to inform experimental designs to better model disease mechanisms and help guide the development of targeted interventions for neurodegenerative diseases, thus bridging the gap between model-based research and therapeutic application.
