  • Thecodiplosis japonensis is an important insect pest in pine trees and hard to be controlled by chemical pesticides due to its characteristic life cycle. Experiments were conducted to determine the pathogenicity and growth temperature of several entomopathogenic fungi as potential microbial insectcides for T. japonensis. Beauveria bassiana SFB-SC and Paecilomyces fumosoroseus SFP-198 were highly pathogenic against T. japonensis. B. bassiana SFB-SC had also pathogenicity against a beneficial insect Bombyx mori. Growth rate and pathogenicity of P. funmosoroseus SFP-198 under 15$^{\circ}C$ was higher than those of B. bassians SFB-SC. These results indicated that P. fumosoroseus SFP-198 is a potential microbial control candidate for control of T. japonensis.

Storage of Phytopathogenic Fungal Cultures in Sterile Distilled Water (식물병원 진균 균주의 살균증류수 저장법)

  • About 450 phytopathogenic fungal cultures were stored in sterile distilled water ar room temperature by the sterile water storage method, which has been known as a simple, convenient, and long-term storage method of microorganisms. After 12 months, viability and pathogenicity of the stored isolates were tested. Among 205 tested, 175 isolates (84.5%) survived. Of these, Rhizoctonia solani, Botrytis cinerea, Pyricularia oryzae, Phytophthora infestans, and Sclerotinia sclerotiorum showed relatively lower survival rate; 92%, 74.1%, 62.5%, 45.8%, and 30%, respectively. Twenty seven isolates belonging to seven important phytopathogenic fungi were tested for pathogenicity, and all isolates tested maintained pathogenicity until at least 12 months after storage.

  • The serotypes and pathogenicity of 47 isolates of Erysipelothrix rhusiopathiae from pigs with acute septicemic swine erysipelas were investigated. The serotypes were determiend by agar gel precipitation test using autoclave extracted antigens. All of the 47 isolates(42 strains from Youngnam and fi fotrains from Kyunggi province) were serotype lao Five field isolates of Erhusiopathiae serovar la were tested for pathogenicity in mice. All of 5 selected isolates were pathogenic for mice and $LD_{50}$ was $3.7{\times}10^3$ viable cells. Mice immunized subcutaneously with live vaccine did not die after challenge with virulent isolates of E rhusiopathiae.

  • Mitogen-activated protein kinase (MAPK) cascades are conserved signaling modules in the eukaryotic cells. They are involved in many major cell processes in fungi such as stress responses, vegetative growth, pathogenicity, secondary metabolism and cell wall integrity. In this review, we summarized the advances of research on the MAPK signaling pathways in Alternaria species. As major phytopathogenic fungi, Alternaria species reduce crop production. In contrast to the five MAPK pathways known in yeast, only three MAPK pathways as Fus3/Kss1-type, Hog1-type, and Slt2-type have been characterized in Alternaria. The Fus3/Kss1-type MAPK pathway participates in regulation of vegetative growth, conidiation, production of some cell-wall-degrading enzymes and pathogenicity. The Hog1-type pathway is involved in osmotic and oxidative stress, fungicides susceptibility and pathogenicity. The Slt2-type MAP kinases play an important role on maintaining cell wall integrity, pathogenicity and conidiation. Although recent advances on the MAPK pathways in Alternaria spp. reveal many important features on the pathogenicity, there are many unsolved problems regarding to the unknown MAP kinase cascade components and network among other major signal transduction. Considering the economic loss induced by Alternaria spp., more researches on the MAPK pathways will need to control the Alternaria diseases.

Molecular characterization of Malaysian fowl adenovirus (FAdV) serotype 8b species E and pathogenicity of the virus in specific-pathogen-free chicken

  • Background: Inclusion body hepatitis (IBH) is an economically important viral disease primarily affecting broiler and breeder chickens. All 12 serotypes of fowl adenovirus (FAdV) can cause IBH. Objectives: To characterize FAdV isolates based on phylogenetic analysis, and to study the pathogenicity of FAdV-8b in specific-pathogen-free (SPF) chickens following virus inoculation via oral and intramuscular (IM) routes. Methods: Suspected organ samples were subjected to virus isolation and polymerase chain reaction (PCR) for FAdV detection. Hexon gene sequencing and phylogenetic analysis were performed on FAdV-positive samples for serotype identification. One FAdV-8b isolate, UPM/FAdV/420/2017, was selected for fiber gene characterization and pathogenicity study and was inoculated in SPF chickens via oral and IM routes. Results: The hexon gene phylogenetic analysis revealed that all isolates belonged to FAdV-8b. The fiber gene-based phylogenetic analysis of isolate UPM/FAdV/420/2017 supported the grouping of that isolate into FAdV species E. Pathogenicity study revealed that, chickens infected with UPM/FAdV/420/2017 via the IM route had higher clinical score values, higher percent mortality, higher degree of the liver lesions, higher antibody response (p < 0.05), and higher virus shedding amounts (p < 0.05) than those infected via the oral route. The highest virus copy numbers were detected in liver and gizzard. Conclusions: FAdV-8b is the dominant FAdV serotype in Malaysia, and pathogenicity study of the FAdV-8b isolate UPM/FAdV/420/2017 indicated its ability to induce IBH in young SPF chickens when infected via oral or IM routes.

  • In studies on Erysipelothrix rhusiopathiae infection of pigs, a series of investigations were conducted for isolation, biochemical properties, pathogenicity, serotyping and susceptibility to antimicrobial agents. The results could be summarized as follows: 1. A total of 14 isolates of E rhusiopathiae were made from an acute form of swine erysipelas and the incidence was high during the hot summer season. 2. The biochemical properties of the 14 isolates were identical to the reference strain of E rhusiopathiae. 3. Pathogenicity of the isolated strain(89368) were ascertained that $LD_{50}$ for mice was $1.8{\times}10 $ cfu and also typical urticarious lesions and acute septicemia for pigs were induced by intradermal and intravenous inoculation with isolates, respectively. 4. All isolates were serotyped by the agarose gel slide double-diffusion system and proved to be serotype 1. 5. In susceptibility test to antimicrobial agents, 14 isolates of E rhusiopathiae were highly sensitive to ampicillin, baytril, cephalothin, chloramphenicol, penicillin and tetracycline.

Cloning, Sequencing, and Characterization of Enterotoxin Pathogenicity Islet from Bacteroides fragilis 419

  • We have earlier reported on the cloning and identification of bft-k from an enterotoxigenic strain of Bacteroides fragilis 419, which was isolated from the blood of a Korean patient who suffered from systemic infections [4,5]. The bft-k gene encodes a 397-amino-acids metalloprotease enterotoxin, and the protein has been identified as a new isoform of B. fragilis enterotoxins (BFTs), which are cytopathic to intestinal epithelial cells to induce fluid secretion and tissue damage in ligated intestinal loops [4, 6, 18, 20]. This report describes the cloning and sequencing of the enterotoxin pahogenicity islet of B. fragilis 419 which contains the bft-k gene. the cloned enterotoxin pathogenicity islet was found to have 6,045 bp in length and to contain 120bp direct repeats near its end. In the pathogenicity islet, in addition to the BFR-K, two putative open reading frames (ORFs) were identified; (1) the t-3 gene encoding a 396-amino-acids protein of a putative metalloprotease; (2) the third gene encoding an ORF of a 59-amino-acids protein, whose function has not yet beenn characterized. The expression of the t-3 gene in B. fragilis 419 was verified by western blot analysis.

  • The strains of Colletotrichum gloeosporioides, C. coccodes, C. acutatum isolated from black raspberry were pathogenic to apple and strawberry after dropping inoculation, but showed weak pathogenicity in hot-pepper and tomato. The anthracnose pathogens of C. gloeosporioides, C. orbiculare, C. acutatum isolated from apple, hot-pepper and pumpkin showed pathogenicity in black raspberry. Moreover, the anthracnose pathogens isolated from apple caused disease symptoms in non-wounded inoculation.

Cultural and Pathogenic Characteristics between Colletotrichum gloeosporioides and Glomerella cingulata Isolated from Strawberry in Korea (국내 딸기 탄저병균 Colletotrichum gloeosporioides와 Glomerella cingulata의 배양적, 병원학적 특성)

  • Colletotrichum gloeosporioides and Glomerella cingulata are the most important pathogens causing anthracnose which may reduce the stand rate and yield on wide kinds of plants including strawberry. Average occurrence rate of anthracnose is 36.9% on major strawberry cropping areas in Korea. We newly found that C. gloeosporioides which does not forming a sexual stage, infects strawberry and differs in some characteristics concerning virulence, cultural and morphological properties to G. cingulata which has a sexual stage. C. gloeosporioides was mainly isolated from the crown with 35.2% rate, while G. cingulata was largely isolated from petiole, runner with 40.9% rate in infected strawberry plants. These two pathogens showed significant differences in cultural characteristics such as perfect stage formation, temperature response as well as benomyl resistance. It was demonstrated that C. gloeosporioides has significantly stronger pathogenicity than G. cingulata in pathogenicity test carried on strawberry plants to various strawberry cultivars. Akihime, Akaneko and Nyoho forcing cultured strawberry cultivars, considered to be susceptible, while semiforcing cultured cultivars, such as Suhong and Holowase, were shown resistant to both pathogens. In non-wound inoculation, C. gloeosporioides was shown pathogenicity on the apple fruit, but G. cingulata could not infect it.
