Characterization of the Causal Fungus of Citrus Melanose, Diaphorthe citri Isolated from Blighted Twigs of Citrus in Jeju (감귤나무 가지에서 분리한 검은점무늬병균 Diaporthe citri의 균학적 특성)

  • Mycological characteristics of Phomopsis citri isolates obtained from blighted twigs of citrus cultivation areas in Jeju island were all identified as Phomopsis citri known as the anamorph stage of Diaporthe citri. Pycnidia the isolates sized from 112.2 to 614.4${\mu}m$ (av. 328.8${\mu}m$) and shaped conical to lenticulata in black. Two types of phialospores, ${\alpha}$-and ${\beta}$-spores were observed from pycnidia. The ${\alpha}$-spores were hyaline, unicellula, fusiform to ellipsoidal and sized 4.7~8.7 1.9 ${\times}$ 3.5${\mu}m$ (av. 6.7 ${\times}$ 2.3 ${\mu}m$). The ${\beta}$-spores were hyaline, unicellula, filiform, curved and often strongly hooked and sized 13.2~27.1 ${\times}$ 0.8~1.6 ${\mu}m$ (av. 22.1 ${\times}$ 1.0 ${\mu}m$). All isolates grew well and produced abundant pycnidia and spore horns on PDA. In addition, all isolates showed strong pathogenicity to citrus fruit inducing melanose symptoms when artificially inoculated with the pycnidial spores.

Phytophthora Diseases of Apple in Korea: II. Occurrence of an Unusual Fruit Rot Caused by P. cactorum and P. cambivora (사과의 역병: II. Phytophthora cactorum과 P. cambivora에 의한 사과 과실역병의 발생)

  • An unusual young fruit rot of apple caused by two species of Phytophthora was epidemic from late May to early July of 1996 in Andong, Uisung and Chungwon areas of Korea. The disease spread to over 30 apple orchards in the areas and percent of the infected tree and fruit was ca. 10~90% and 1`15%, respectively. Water soaking lesions or spots on leaves and shoot blight were also developed by the pathogen. Among 39 isolates collected, 25 were identified as P. cactorum and the others were as zp. cambivora on the basis of their distinctive morphological characters. While the former fungus was homothallic, all isolates of the latter were A1 mating types. Koch's postulate was fulfilled. Both fungi showed strong pathogenicity not only to young fruits, leaves and shoots of apple but also to those of pear and peach. Several vegetables tested did not show symptoms even by wound inoculation. An Occurrence of young fruit rot of apple caused by Phytophthora has not been reported in Korea, especially, P. cambivora has not been recorded previously as the causal agent of the disease in the world.

Effect of Nitrogen Sources on Mycelial Growth and Sclerotial Formation of Rhizoctonia solani Causing Rice Sheath Blight (벼 잎집무늬마름병균의 균사생장 및 균핵형성에 미치는 질소원의 효과)

  • Effect of nitrogen sources on mycelial growth and sclerotial formation of Rhizoctonia solani causing rice sheath blight was studied by using sixty isolates from diseased samples of different localities in Chungbuk area. Based on the cultural characters, pathogenicity and sclerotial formation, three isolates of the fungus were selected. The virulence of the three isolates varied based on the percent of seeds germination, seedling blights and length and lesion number of lesions. As nitrogen sources, arginine, alanine, urea, ammonium sulfate and sodium nitrate were more effective for mycelial growth and sclerotial formation, than were cystine, methionine, lysine, histidine, tryptophan and thyrosine. The mycelium of R. so/ani grew well in a lower concentration of urea, sodium nitrate, arginine, ammonium sulfate and lysine, but in a higher concentration of proline, histidine, and alanine. In all nitrogen sources tested, high concentration of nitrogen nutrients greatly stimulated sclerotial formation of the fungus. Arginine, urea, and ammonium sulfate increased virulence of the fungus. All the isolates grown on the basic medium plus proline produced large sclerotia. Mycelial growth of the fungus was not well in the tryptophan-added medium, showing brown discoloration of medium.

Studies on the effects of culture filtrates of sesame-wilt organism(Fusarium oxysprum f. vasinfectum) on the germination of sesame seeds and the growth of sesame seedlings (참깨$\cdot$시들음병균(Fusarium oxysporum)의 배양여액이 참깨의 종자발아 및 유묘기의 생육에 미치는 영향)

  • 1) The purpose of the present study is to investigate the effects of culture filtrates of Fusarium oxysporum f. vasinfectum which is known to produce fusaric acid (wilt toxin) on the germination of sesame seeds and the growth of sesame seedlings. 2) Culture filtrates of Fusarium oxysporum 1. vasinfectum used in this study strongly or weakly inhibited the germination and bring about necrosis accompanying black discoloration of sesame seeds. 3) Varietal difference of sesame in the germination response on the culture filtrates of Fnsarium oxysporum f. vasinfectum is not shown in this study. 4) This study reveals that differential five strains of Fnsarium oxysoprum f. vasinfectum used in this study differ greatly in the toxicity of culture filtrates inhibiting the germination of sesame seeds. 5) In the seedling bed added with culture filtrates of Fnsarium oxysporum f. vasinfectum, the growth of shoot as well as root system of sesame seedlings are notably inhibited and necrotic black discoloration appear on both shoot and root system. But in the seedling beds added with weaker concentration of culture filtrates $/(10\%)$ the growth of shoot is slightly promoted. 6) In culture of sesame seedlings with Knop's solution containing 1 to 3 per cent culture filtrates, the growth of shoot as well as root system are slightly retarded" and till the time of development of the third leaves the whole stem and leaf petiole tissue are weakened so that they become thread like accompanying brown discoloration, interveinal light brown area appear in the second leaves, and the third leaves curl from both sides towards the middle with necrotic brown discoloration, especially symptoms of injury on the third leaves are nearly similar that of the leaves of wilted sesame in the field. 7) A pararell relationship is not found between toxicity of culture filtrates and pathogenicity of five differential strains of Fnsarium oxysporum f. vasinfectum used in this study.

  • Among 980 fungal strains isolated from the lesions of Trifolium repens L. and various weeds, three Penicillium sp. F40362, F40496, F40497 were selected by the first selection test and a pathogenicity test. The spores of these strains germinated readily 90 to 100% and readily infected the respective plant. The wheat bran-corn starch formulation of F40362 strain showed 100% mycoherbicidal activity against clover plant at $4{\times}10^8$ spores/pot. The same formulations of F40496 and F40497 strains showed 100% mycoherbicidal activity against clover plant at $12{\times}10^8{\sim}4{\times}10^8$ spores/pot. The same formulations of tee strains showed over 30% mycoherbicidal activity against Leguminosae plants. This method of pelletiation was potentially useful for the production of inoculum formulation as mycoherbicides and it was effective enough to treat $2{\sim}2.5\;g$ formulation($4.5{\times}10^7\;spores/mg$) to a $350\;cm^2$ pot. The three strains, F40362, F40496 and F40497 have selective mycoherbicidal activity between Trifolium repens L. and Zoysia japonica and nonselective mycoherbicidal activities against some other crop plants and weeds.

Genetic Organization of the hrp Genes Cluster in Erwinia pyrifoliae and Characterization of HR Active Domains in HrpNEp Protein by Mutational Analysis

  • The disease-specific (dsp) region and the hypersensitive response and pathogenicity (hrp) genes, including the hrpW, $hrpN_{Ep}$, and hrpC operons have previously been sequenced in Erwinia pyrifoliae WT3 [Shrestha et al. (2005a)]. In this study, the remaining hrp genes, including the hrpC, hrpA, hrpS, hrpXY, hrpL and hrpJ operons, were determined. The hrp genes cluster (ca. 38 kb) was comprised of eight transcriptional units and contained nine hrc (hrp conserved) genes. The genetic organization of the hrp/hrc genes and their orientation for the transcriptions were also similar to and collinear with those of E. amylovora, showing ${\geq}80%$ homologies. However, ORFU1 and ORFU2 of unknown functions, present between the hrpA and hrpS operons of E. amylovora, were absent in E. pyrifoliae. To determine the HR active domains, several proteins were prepared from truncated fragments of the N-terminal and the C-terminal regions of $HrpN_{Ep}$ protein of E. pyrifoliae. The proteins prepared from the N-terminal region elicited HR, but not from those of the C-terminal region indicating that HR active domains are located in only N-terminal region of the $HrpN_{Ep}$ protein. Two synthetic oligopeptides produced HR on tobacco confirming presence of two HR active domains in the $HrpN_{Ep}$. The HR positive N-terminal fragment ($HN{\Delta}C187$) was further narrowed down by deleting C-terminal amino acids and internal amino acids to investigate whether amino acid insertion region have role in faster and stronger HR activity in $HrpN_{Ep}$ than $HrpN_{Ea}$. The $HrpN_{Ep}$ mutant proteins $HN{\Delta}C187$ (D1AIR), $HN{\Delta}C187$ (D2AIR) and $HN{\Delta}C187$ (DM41) retained similar HR activation to that of wild-type $HrpN_{Ep}$. However, the $HrpN_{Ep}$ mutant protein $HN{\Delta}C187$ (D3AIR) lacking third amino acid insertion region (102 to 113 aa) reduced HR when compared to that of wild-type $HrpN_{Ep}$. Reduction in HR elicitation could not be observed when single amino acids at different positions were substituted at third amino acids insertion region. But, substitution of amino acids at L103R, L106K and L110R showed reduction in HR activity on tobacco suggesting their importance in activation of HR faster in the $HrpN_{Ep}$ although it requires further detailed analysis.

Relationship between production of exoenzymes and serotypes of Pseudomonas aeruginosa isolated from clinical specimens and hospital environments (녹농균(綠膿菌)의 균체외효소산생능(菌體外酵素産生能)과 그의 혈청형(血淸型)과의 관계(關係))

  • Exoenzymes, protease(P) and elastase(E) produced by Pseudomonas aeruginosa are reported to have close relationship with pathogenicity of Pseudomonas aeruginosa. Productibility of exoenzymes P and E were studied and compared in environmental isolates from hospital environments and clinical isolates from various clinical specimens, also, the relationship between their enzyme production and serotype were reviewed. 1. Clinical isolates were typed into nine serotypes A, B, C, D, E, F, G, H and I. Serotype E had the highest incidence of 24%, followed by B with 16.8%, G, 15.1% and C, 9.3%. 2. Environmental isolates were, typed as serotype B, C, E, F, G, H, I, K and M. Serotype I had the highest incidence of 26.6%, followed by C, F and M each incidence of 14.3%. 3. In the typing of the above two groups, serotypes A and D were found only in the clinical isolates and serotypes K, and M were found only in the environmental isolates. Serotypes J and L were found in neither clinical isolates nor environmental isolates. 4. In the distribution of serotypes from various clinical specimens, serotype G among isolates from pus showed incidence of 20.4%, and serotypes E and B were 19.5% separately. Serotype E had incidence of 22.6% and 20.0% in urine and sputa respectively, showing a high rate compared to the other serotypes. 5. The incidence of strains producing both exoenzymes P and E was 77.8% in the preserved strains of clinical isolates and 76.2% in the environmental isolates. There were no significant difference between the two groups. 6. Serotypes A and H, which are preserved strains from clinical isolates showed productibility of both exoenzymes P and E, the other serotypes showed productibility of various combination of exoenzymes. Among the environmental isoaltes, production of both exoenzymes P and E were seen in serotypes E, F, G, H, I and K and no serotype produced only P or E. 7. In ability to produce exoenzymes of isolates from sources of various clinical specimens, strains producing both exoenzymes P and E were found most frequently in pus with incidence rate of 82.0%, followed by 80.0% in sputum and urine. 8. Almost all the fresh strains of clinical isolates were producers of both exoenzymes P and E.

Identification and Ecological Characteristics of Bacterial Blossom Blight Pathogen of Kiwifruit (참다래 꽃썩음병균의 동정 및 발생생태)

  • Bacterial blossom blight is one of the most important diseases of kiwifruit (Actinidia deliciosa). The disease occurs during flowering in the late May and disease outbreaks associated with rainfall during the flowering season have resulted in a severe reduction in kiwifruit production. The causal organism isolated from diseased blossoms of kiwifruits was identified as Pseudomonas syringae pv, syringae based on the physiological and biochemical characteristics and pathogenicity test. Dead fruit stalks, dead pruned twigs, fallen leaves and soils mainly provided R syringae pv. syringae with overwintering places in the kiwifruit orchards, and the inocula also overwintered on buds, trunks, branches, and twigs on the kiwifruit trees. Among the overwintering places, the incula were detected in the highest frequencies from dead fruit stalks. The population density of P. syringae pv. syringae was speculated to be over $1{\times}10^4$cfu/ml for the bacterial infection, and the optimum temperature for the bacterial growth ranged 20 to $25^{\circ}C$. The highest population density of P. syringae pv. syringae on the overwintering places was detected in May and June when the daily average temperature coincided with the optimum temperature for bacterial growth of P. syringae pv. syringae.

Pink Rot of Palms Caused by Gliocladium vermoseni (Gliocladium vermoeseni에 의한 야자 분홍썩음병(가칭))

  • Pink rot of Palm was occurred at Yeuju area in 2001 and 2003. Infected plants showed rotting at the leaf-stock bases and killing of the terminal bud. The first symptoms are dark brown necrotic areas on the stem. Bases of infected frond may be covered with pink spores and the spots produced oozing gum pockets. Oozing lesions occur on the stems, and leaves turn brown and droop. The causal agent were isolated from salmon-pink spores sporulating on the leaf sheaths and necrotic stem tissues. Pathogen were isolated from freshly infected tissues were identified as Gliocladium vermoseni based on mycological characteristics. Fungus were grown plenty on PDA culture. Temperature for mycelial grown was tested at 5 to $40^{\circ}C$ and optimal temperature was $25^{\circ}C$ and was not nearly grew at temperature above $35^{\circ}C$. Artificial pathogenicity were tested on 9 species of Palm family in the wound inoculation and symptoms showed similar to those observed in the field. This is the first report on pink rot of palm in Korea.

Comparative Assay Fungus Population and Resistant Genes about Magnaporthe grisea between Korea and China (한국·중국 벼 도열병균 생리형 분석 및 품종 저항성 특성 비교)

  • One hundred isolates of Magnaporthe grisea from Korea and China were characterized for pathogenicity using eight Korean differential varieties(KDV), six Chinese differential varieties(CDV) and six near isogenic lines(NILs) developed in China. The restriction length polymorphism of M. grisea isolates from each country also was analyzed using MGR586 as a probe. One hundred Korean isolates classified into 17 races on KDV were grouped into 29 pathotypes on Chinese near isogenic lines(NILs). Virulence of 46% of Korean isolates against all the six Chinese NILs indicated that the current six Chinese NILs alone was not enough to be used as differential varieties in Korea. Especially, susceptibility of the BL1 carrying resistance gene Pi-b to 70% of tested Korean isolates suggested that BL1(Pi-b) may not be a useful resistance source to Korean blast. Based on the virulence assays of M. grisea populations from each country were divided into two groups. About 50% of Chinese isolates showed similarity to the 30% of the Korean isolates. Especially, the isolates from northern part of China, where Japonica rice cultivars were grown, showed high similarity to the Korean isolates, while isolates from southern part of China, where Indica rice were mainly grown, showed low similarity to Korean isolates. The genome RFLPs of Korean isolates were quite different from those of southern part of China using MGR586 as a probe. These data indicated that the physiological and genetical characteristics of M. grisea population might be determined by strong interaction with cultivated rice.
