Tomato Yellow Leaf Curl China Virus Impairs Photosynthesis in the Infected Nicotiana benthamiana with βC1 as an Aggravating Factor

  • Tomato yellow leaf curl China virus is a species of the widespread geminiviruses. The infection of Nicotiana benthamiana by Tomato yellow leaf curl China virus (TYLCCNV) causes a reduction in photosynthetic activity, which is part of the viral symptoms. ${\beta}C1$ is a viral factor encoded by the betasatellite DNA ($DNA{\beta}$) accompanying TYLCCNV. It is a major viral pathogenicity factor of TYLCCNV. To elucidate the effect of ${\beta}C1$ on plants' photosynthesis, we measured the relative chlorophyll (Chl) content and Chl fluorescence in TY-LCCNV-infected and ${\beta}C1$ transgenic N. benthamiana plants. The results showed that Chl content is reduced in TYLCCNV A-infected, TYLCCNV A plus $DNA{\beta}$ (TYLCCNV A + ${\beta}$)-infected and ${\beta}C1$ transgenic plants. Further, changes in Chl fluorescence parameters, such as electron transport rate, $F_v/F_m$, NPQ, and qP, revealed that photosynthetic efficiency is compromised in the aforementioned N. benthamiana plants. The presense of ${\beta}C1$ aggravated the decrease of Chl content and photosynthetic efficiency during viral infection. Additionally, the real-time quantitative PCR analysis of oxygen evolving complex genes in photosystem II, such as PsbO, PsbP, PsbQ, and PsbR, showed a significant reduction of the relative expression of these genes at the late stage of TYLCCNV A + ${\beta}$ infection and at the vegetative stage of ${\beta}C1$ transgenic N. benthamiana plants. In summary, this study revealed the pathogenicity of TYLCCNV in photosynthesis and disclosed the effect of ${\beta}C1$ in exacerbating the damage in photosynthesis efficiency by TYLCCNV infection.

피부 궤양이 발생한 금붕어(Carassius auratus)에서 분리한 Aeromonas veronii의 특성 및 병원성 분석 (A study on the characteristics and pathogenicity of Aeromonas veronii isolated from infected goldfish (Carassius auratus))

  • Aeromonas spp. 감염은 관상어 산업뿐만 아니라 양식업에서도 상당한 경제적 손실을 초래하는 것으로 보고되고 있다. 한편 2022년 12월부터 2023년 1월 사이에 국내에서 사육중인 금붕어(Crassius auratus)에서 Aeromonas병증을 보이는 어체에 대해서 발병 원인균을 규명하기 위해서 조사 하였다. 병증을 보이는 금붕어의 피부와 내부 장기에서 그람 음성균이 분리되었다. Biochemical test와 16S rRNA gene으로는 Aeromonas 속까지 동정할 수 있었으나 종레벨까지는 구분하기 어려웠다. 이에 가장 상동성이 높았던 A.veronii를 대상으로 species-specific PCR을 수행했고 명확히 종 수준으로 동정할 수 있었다. 아울러서 장독소 유전자, 감염 실험 및 항생제 내성 분석을 수행하였다. 금붕어에 대한 균주의 생체 내 병원성 시험 결과, 실험 주사 후 일주일 이내에 감염된 숙주에서 100%의 치사율을 나타냈다. 장독소 유전자인 act 독력 유전자가 검출되었다. 균주의 항균제 감수성 분석 결과 대부분의 항균제에 감수성을 보였으나, Ampicillin, Imipenem, Meropenem 및 Clindamycin 약제에는 내성을 보였다.

Identification of virulence-associated genes of Erwinia amylovora by transposon mutagenesis

  • Erwinia amylovora , which causes fire blight disease on apple and pear trees, is one of the most important phytopathogens because of its devastating impact. Currently, the only way to effectively control fire blight disease is through the use of antibiotics such as streptomycin, kasugamycin, or oxytetracycline. However, problems with the occurrence of resistant strains due to the overuse of antibiotics are constantly being raised. It is therefore necessary to develop novel disease control methods through an advanced understanding of the pathogenesis mechanism of E. amylovora . To better understand the pathogenesis of E. amylovora , we investigated unknown virulence factors by random mutagenesis and screening. Random mutants were generated by Tn5 transposon insertion, and the pathogenicity of the mutants was assessed by inoculation of the mutants on apple fruitlets. A total of 17 avirulent mutants were found through screening of 960 random mutants. Among them, 14 mutants were already reported as non-pathogenic strains, while three mutants, TS3128_M2899 (ΔSUFU ), TS3128_M2939 (ΔwcaG ), and TS3128_M3747 (ΔrecB ), were not reported. Further study of the association between E. amylovora pathogenicity and these 3 novel genes may provide new insight into the development of control methods for fire blight disease.

Virulence Reduction and Differing Regulation of Virulence Genes in rpf Mutants of Xanthomonas oryzae pv. oryzae

  • To define the functions of the rpf genes in Xanthomonas oryzae pv. oryzae (Xoo), which regulates pathogenicity factors in Xanthomonas campestris pv. campestris (Xcc), marker-exchange mutants of each rpf gene were generated. When the mutants were inoculated on a susceptible cultivar, the lesion lengths caused by the rpfB, rpfC, rpfF, and rpfG mutants were significantly smaller than those caused by the wild type, whereas those caused by the rpfA, rpfD, and rpfI mutants were not. Several virulence determinants, including extracellular polysaccharide (EPS) production, xylanase production, and motility, were significantly decreased in the four mutants. However, the cellulase activity in the mutants was unchanged. Complementation of the rpfB and rpfC mutations restored the virulence and the expression of the virulence determinants. Expression analysis of 14 virulence genes revealed that the expression of genes related to EPS production (gumG and gumM), LPS (xanA, xanB, wxoD, and wxoC), phytase (phyA), xylanase (xynB), lipase (lipA), and motility (pitA) were reduced significantly in the mutants rpfB, rpfC, rpfF, and rpfG. In contrast, the expression of genes related to cellulase (eglxob, clsA), cellobiosidase (cbsA), and iron metabolism (fur) was unchanged. The results of this study clearly show that rpfB, rpfC, rpfF, and rpfG are important for the virulence of Xoo KACC10859, and that virulence genes are regulated differently by the Rpfs.

The Roles of Two hfq Genes in the Virulence and Stress Resistance of Burkholderia glumae

  • The Hfq protein is a global small RNA chaperone that interacts with regulatory bacterial small RNAs (sRNA) and plays a role in the post-transcriptional regulation of gene expression. The roles of Hfq in the virulence and pathogenicity of several infectious bacteria have been reported. This study was conducted to elucidate the functions of two hfq genes in Burkholderia glumae, a causal agent of rice grain rot. Therefore, mutant strains of the rice-pathogenic B. glumae BGR1, targeting each of the two hfq genes, as well as the double defective mutant were constructed and tested for several phenotypic characteristics. Bacterial swarming motility, toxoflavin production, virulence in rice, siderophore production, sensitivity to $H_2O_2$, and lipase production assays were conducted to compare the mutant strains with the wild-type B. glumae BGR1 and complementation strains. The hfq1 gene showed more influence on bacterial motility and toxoflavin production than the hfq2 gene. Both genes were involved in the full virulence of B. glumae in rice plants. Other biochemical characteristics such as siderophore production and sensitivity to $H_2O_2$ induced oxidative stress were also found to be regulated by the hfq1 gene. However, lipase activity was shown to be unassociated with both tested genes. To the best of our knowledge, this is the first study to elucidate the functions of two hfq genes in B. glumae. Identification of virulence-related factors in B. glumae will facilitate the development of efficient control measures.

IVET-based Identification of Virulence Factors in Vibrio vulnificus MO6-24/O

  • Vibrio vulnificus is an opportunistic pathogen that causes septicemia in humans. To identify the genes associated with its pathogenicity, in vivo expression technology (IVET) was used to select genes specifically expressed in a host, yet not significantly in vitro. Random lacZ-fusions in the genome of V vulnificus strain MO6-24/O were constructed using an IVET vector, pSG3, which is a suicide vector containing promoterless-aph and -lacZ as reporter genes. A total of ${\sim}18,000$ resulting library clones were then intraperitoneally injected into BALB/c mice using a colony forming unit (CFU) of $1.6{\times}10^6$. Two hours after infection, kanamycin was administered at $200{mu}g$ per gram of mouse weight. After two selection cycles, 11 genes were eventually isolated, which were expressed only in the host. Among these genes, VV20781 and VV21007 exhibiting a homology to a hemagglutinin gene and tolC, respectively, were selected based on having the highest frequency. When compared to wild-type cells, mutants with lesions in these genes showed no difference in the rate of growth rate, yet a significant decrease in cytotoxicity and the capability to form a biofilm.

  • Phytophthora capsici is a pathogen on several economically important crops including pepper. In pepper growing areas in Korea, Phytophthora blight caused by p. capsici has been considered as the most serious problem in pepper production. The Oomycete attacks the roots, stems, leaves and fruits of the plant. To understand the molecular mechanisms involved in the disease development, the genes expressed doting pepper p. capsici interaction were explored by analyzing expressed sequence tags (ESTs). A complementary DNA (cDNA) library was constructed from total RNA extracted from pepper leaves challenged with p. capsici for 3 days resulting in early stage of symptom development. The comprehensive analysis on the single pass sequencing of over 4000 randomly selected cDNA clones with contig assembly, unique gene extraction, sequence comparison, and functional categorizing will be presented with an emphasis on the genes involved in plant defense and pathogenicity during disease development of the pepper Phytophthora blight.

Functional pathogenomics of Burkhozderia glumae (oral)

  • The aim of this study was to characterize the interactions of rice and Burkholderia glumae, a causal agent of bacterial grain rot of rice, at molecular levels using whole genomic sequences and to identify genes important for pathogenicity and symptom development. To do these, we sequenced whole genome of the bacterium and constructed cosmid clone profiles. We generated pools of mutants using various transposons and determined mutation sites by sequencing rescued plasmids. We focused on studying toxoflavin biosynthetic genes, quorum sensing regulation, and Hrp type III protein secretion systems. We found that two possible operons consisting of five genes are involved in toxoflavin biosynthesis and their expression is regulated by quorum sensing and LysR-type regulator, ToxR. We have isolated the nn PAI of B. glumae and characterized by mutational analyses. The hrp cluster resembled most the putative Type III secretion systems of B. pseudomallei, which is the causative agent of melioidosis, a serious disease of man and animals. The Hrp PAI core region showed high similarity to that of Ralstonia solanacearum and Xanthomonas campestris, however some aspects were dissimilar.

Fluctuation of Temperature Induces Pathogenicity of Streptococcus iniae and Changes of Immunology Related Genes of Korean Rockfish, Sebastes schlegeli

  • This study was designed to examine the immune response in Korean rockfish during water temperature fluctuation and to elucidate the factors contributing to streptococcal pathogenesis in cultured Korean rockfish, S. schlegeli. We investigated cumulative mortality against Streptococcus iniae (FP5228 strain) infection in the exposed Korean rockfish (39.7±5.8 g) to environmentally relevant temperature (Control, 23℃; High temperature, 28℃ and 23℃ and 28℃ with 12 hours interval exchange, 23↔28℃) for 48 hours. Also, the expression of the mRNA related to the immune response genes (heat shock protein 70, interleukin1β, lysozyme g-type and thioredoxin-like 1) were measured in spleen and head kidney by real-time PCR analysis in the exposed fish to thermal stress. In this study, the combined stress with bacterial challenge in fishes exposed to thermal stress lowered the survival rate than that of control (23℃). The cumulative mortality in the group of control, 28℃ and 23↔28℃ was 24%, 24% and 40% (P<0.05), respectively. Also, thermal stress modulated the mRNA level of immune related genes; heat shock protein 70, interleukin-1β, lysozyme g-type and thioredoxin-like 1 in Korean rockfish. The present study indicates that a high and sudden water temperature change affect immune responses and reduce the disease resistance in Korean rockfish.

Alternaria brassicifolii sp. nov. Isolated from Brassica rapa subsp. pekinensis in Korea

  • A new species belonging to the genus Alternaria was isolated from the necrotic leaf spots of Brassica rapa subsp. pekinensis in Yuseong district, Daejeon, Korea. It is an occasional isolate, not an etiological agent, which is morphologically similar to A. broccoli-italicae, but differs in conidial size and conidiophore shape. Phylogenetic analysis using the sequence datasets of the internal transcribed spacer (ITS) region of the rDNA, glyceraldehyde-3-phosphate dehydrogenase (gpd), and plasma membrane ATPase genes showed that it is distantly related to A. broccoli-italicae and closely related to Alternaria species in the section Pseudoalternaria, which belonged to a clade basal to the section Infectoriae. Morphologically, the species is unique because it produces solitary conidia or conidial chains (two units), unlike the four members in the section Pseudoalternaria that produce conidia as short branched chains. It exhibits weak pathogenicity in the host plant. This report includes the description and illustration of A. brassicifolii as a new species.
