The present study was performed to identify pathogenic factors of Chlamydia trachomatis, which invade the host cell membrane. We prepared monoclonal antibody against C. trachomatis and searched for pathogenic factors using this antibody, and subsequently identified the surface components of the elementary body of C. trachomatis, i.e., major outer membrane protein (MOMP), lipopolysaccharide (LPS), and two other surface exposure proteins. These proteins are believed to be important in the pathogenesis of host cell chlamydial infection. Additionally, to identify factors related to the host cell and C. trachomatis, we prepared C. trachomatis infected and non-infected McCoy cell extracts, and reacted these with anti-chlamydial LPS monoclonal antibody. We found that anti-chlamydial LPS monoclonal antibody reacted with a 116 kDa proteinaceous McCoy cell membrane component.
As neurofilament proteins are major cytoskeletal components of neuron, abnormality of neurofilament is proposed in brain with neurodegenerative disorders such as Parkinson's disease (PD). Since oxidative stress might play a critical role in altering normal brain proteins, we investigated the oxidative modification of neurofilament-L (NF-L) induced by the reaction of cytochrome c with H2O2. When NF-L was incubated with cytochrome c and H2O2, the protein aggregation was increased in cytochrome c and H2O2 concentrationsdependent manner. Radical scavengers, azide, formate and N-acetyl cysteine, prevented the aggregation of NFL induced by the cytochrome c/H2O2 system. The formations of carbonyl group and dityrosine were obtained in cytochrome c/H2O2-mediated NF-L aggregates. Iron specific chelator, desferoxamine, prevented the cytochrome c/H2O2 system-mediated NF-L aggregation. These results suggest that the cytochrome c/H2O2 system may be related to abnormal aggregation of NF-L which may be involved in the pathogenesis of PD and related disorders.
Together with neuronal loss, the existence of insoluble inclusions of alpha-synuclein (α-syn) in the brain is widely accepted as a hallmark of synucleinopathies including Parkinson's disease (PD), multiple system atrophy, and dementia with Lewy body. Because the α-syn aggregates are deeply involved in the pathogenesis, there have been many attempts to demonstrate the mechanism of the aggregation and its potential causative factors including post-translational modifications (PTMs). Although no concrete conclusions have been made based on the previous study results, growing evidence suggests that modifications such as phosphorylation and ubiquitination can alter α-syn characteristics to have certain effects on the aggregation process in PD; either facilitating or inhibiting fibrillization. In the present work, we reviewed studies showing the significant impacts of PTMs on α-syn aggregation. Furthermore, the PTMs modulating α-syn aggregation-induced cell death have been discussed.
Foot-and-Mouth Disease (FMD) is a highly contagious trans-boundary viral disease caused by FMD virus, which causes huge economic losses. FMDV infects cloven hoofed (two-toed) mammals such as cattle, sheep, goats, pigs and various wildlife species. To control the FMDV, it is necessary to understand the life cycle and the pathogenesis of FMDV in host. Especially, the protein-protein interaction between FMDV and host will help to understand the survival cycle of viruses in host cell and establish new therapeutic strategies. However, the computational approach for protein-protein interaction between FMDV and pig hosts have not been applied to studies of the onset mechanism of FMDV. In the present work, we have performed the prediction of the pig's proteins which interact with FMDV based on RNA-Seq data, protein sequence, and structure information. After identifying the virus-host interaction, we looked for meaningful pathways and anticipated changes in the host caused by infection with FMDV. A total of 78 proteins of pig were predicted as interacting with FMDV. The 156 interactions include 94 interactions predicted by sequence-based method and the 62 interactions predicted by structure-based method using domain information. The protein interaction network contained integrin as well as STYK1, VTCN1, IDO1, CDH3, SLA-DQB1, FER, and FGFR2 which were related to the up-regulation of inflammation and the down-regulation of cell adhesion and host defense systems such as macrophage and leukocytes. These results provide clues to the knowledge and mechanism of how FMDV affects the host cell.

Cyclic dipeptides (CDPs) are one of the simplest compounds produced by living organisms. Plant-growth promoting rhizobacteria (PGPRs) also produce CDPs that can induce disease resistance. Bacillus vallismortis strain BS07 producing various CDPs has been evaluated as a potential biocontrol agent against multiple plant pathogens in chili pepper. However, plant signal pathway triggered by CDPs has not been fully elucidated yet. Here we introduce four CDPs, cyclo(Gly-L-Pro) previously identified from Aspergillus sp., and cyclo(L-Ala-L-Ile), cyclo(L-Ala-L-Leu), and cyclo(L-Leu-L-Pro) identified from B. vallismortis BS07, which induce disease resistance in Arabidopsis against Pseudomonas syringae infection. The CDPs do not directly inhibit fungal and oomycete growth in vitro. These CDPs require PHYTOALEXIN DEFICIENT4, SALICYLIC ACID INDUCTION DEFICIENT2, and NONEXPRESSOR OF PATHOGENESIS-RELATED PROTEINS1 important for salicylic acid-dependent defense to induce resistance. On the other hand, regulators involved in jasmonate-dependent event, such as ETHYLENE RECEPTOR1, JASMONATE RESPONSE1, and JASMONATE INSENSITIVE1, are necessary to the CDP-induced resistance. Furthermore, treatment of these CDPs primes Arabidopsis plants to rapidly express PATHOGENESIS-RELATED PROTEIN4 at early infection phase. Taken together, we propose that these CDPs from PGPR strains accelerate activation of jasmonate-related signaling pathway during infection.
Clubroot disease is one of the most wide-spread and devastating diseases in the cultivation of Chinese cabbage. To develop a protein marker for resistance to clubroot disease in Chinese cabbage, a comparative proteome analysis was performed between a sensitive line, 94SK, and a resistant line, CR Shinki DH. Three proteins of two fold or higher accumulation that are specific to each line were found 3 days after innoculation of the Plasmodiphora brassicae. They are glutamine synthetase, malate dehydrogenase/oxidoreductase and fructose-bisphosphate aldolase in the 94SK and actin, phosphoglycerate kinase, and Cu/Zn superoxide dismutase in the CR Shinki line. From the comparison of the synthesized proteins in the 94SK and the CR Shinki, CR Shinki was found to produce more ATP-binding protein for the ABC transporter while 94SK showed a higher level of pathogenesis-related protein 1 production. All of these proteomic variations may lead to the development of molecular markers to accelerate the breeding process.
Parkinson's disease (PD) is a neurodegenerative disease caused by selective degeneration of dopaminergic neurons in the substantia nigra. Mutations in ${\alpha}$-synuclein have been causally linked to the pathogenesis of hereditary PD. In addition, it is a major component of Lewy body found in the brains of sporadic cases as well. In the present study, we examined whether overexpression of wild type or PD-related mutant ${\alpha}$-synuclein induces unfolded protein response (UPR) and triggers the known signaling pathway of the resulting endoplasmic reticulum (ER) stress in SH-SY5Y cells. Overexpression of wild type, A30P, and A53T ${\alpha}$-synuclein all induced XBP-1 mRNA splicing, one of the late stage UPR events. However, activation of ER membrane kinases and upregulation of ER or cytoplsmic chaperones were not detected when ${\alpha}$-synuclein was overexpressed. However, basal level of cytoplsmic calcium was elevated in ${\alpha}$-synuclein-expressing cells. Our observation suggests that overexpression of ${\alpha}$-synuclein induces UPR independent of the known ER membrane kinase-mediated signaling pathway and induces ER stress by disturbing calcium homeostasis.
구제역(Foot-and-Mouth Disease: FMD)이란 소, 돼지, 양, 염소 등의 cloven-hoofed 동물에서 나타나는 바이러스성 질병으로 입, 코, 유두, 발굽 등에 수포가 형성되는 것이 특징이다. 일곱 가지 혈청형(O, A, C, Asia1, SAT1, SAT2 and SAT3)으로 분류되는 구제역바이러스(Foot-and-Mouth Disease Virus: FMDV)는 single stranded positive RNA virus로 nonenveloped capsid virus이다. Viral genome은 8.2 Kb로 하나의 ORF인 polyprotein으로 되어있으며, 크게 capsid protein coding region인 P1, replication related protein coding region인 P2, RNA dependent RNA polymerase coding region인 P3로 구성된다. FMDV는 respiratory tract의 pharynx epithelial cell에 감염되며, lung epithelial cell에서 replication을 한다. 구제역바이러스는 감염율은 높지만 낮은 치사율을 가진다. 2002년 한국에서 구제역이 발병하여 많은 경제적 손실을 입었다. FMDV의 감염을 조절할 수 있는 조절방법이 없는 실정이며, 현재 많은 나라에서는 구제역바이러스의 감염을 막을 수 있는 효과적인 방법을 연구하고 있다. 본 보고서에서는 FMD에 대한 보다 효과적인 예방법인 DNA vaccine, edible vaccine, peptide vaccine에 대해 고찰하였다.
As obesity is known to be related to hyperlipidemia, diabetes and coronary heart disease, and other chronic diseases, many researches have focused on functional food materials showing anti-obesity activity. The adipokines secreted by adipose tissue, resistin and adiponectin are known to play an important role in the pathogenesis of chronic diseases directly. C-reactive protein and homocysteine are molecules regulated by adipose tissue indirectly also relate to the chronic diseases. This study was performed to study of the anti-obesity effects of Sasa borealis in diet-induced obese mice (C57/BL6J). The mice were divided into four group: NFD (Normal fat diet), HFD (High fat diet), BSE (High fat diet containing 5% of 70% ethanol extract of Sasa borealis leaves), BLW (High fat diet containing 5% of water extract of Sasa borealis leaves). The experimental diets were fed for 11 weeks. The final body weight of the mice in the groups of BSE and BLW groups were significantly lower than the HFD group. The effects of weight reduction were due to reduced body fat accumulation. The adiponectin levels are significantly decreased in HFD group compared than NFD group and increased taken by Sasa borealis containing diet. The resistin levels are not significantly different between experimental groups. The CRP and homocyteine levels are significantly higher in HFD group than NFD group and significantly decreased by Sasa borealis containing diet, especially BLW group. These results indicate that orally administered Sasa borealis not only has the effect of reducing the body weight and total fat weight, but preferable effect in adiponectin levels and related molecules as CRP and homocysteine. Therefore we expect the Sasa borealis may have an anti-obesity function and anti-metabolic syndrome effect in diet-induced obese mice.

We reported previously that overexpression of a salicylic acid (SA) methyltransferase1 gene from rice (OsBSMT1) or a SA glucosyltransferase1 gene from Arabidopsis thaliana (AtSAGT1) leads to increased susceptibility to Pseudomonas syringae due to reduced SA levels. To further examine their roles in the defense responses, we assayed the transcript levels of AtBSMT1 or AtSAGT1 in plants with altered levels of SA and/or other defense components. These data showed that AtSAGT1 expression is regulated partially by SA, or nonexpressor of pathogenesis related protein1, whereas AtBSMT1 expression was induced in SA-deficient mutant plants. In addition, we produced the transgenic Arabidopsis plants with RNAi-mediated inhibition of AtSAGT1 and isolated a null mutant of AtBSMT1, and then analyzed their phenotypes. A T-DNA insertion mutation in the AtBSMT1 resulted in reduced methyl salicylate (MeSA) levels upon P. syringae infection. However, accumulation of SA and glucosyl SA was similar in both the atbsmt1 and wild-type plants, indicating the presence of another SA methyltransferase or an alternative pathway for MeSA production. The AtSAGT1-RNAi line exhibited no altered phenotypes upon pathogen infection, compared to wild-type plants, suggesting that (an)other SA glucosyltransferase(s) in Arabidopsis plants may be important for the pathogenesis of P. syringae.
