Effects of chitosan on growth of tomato plant, and suppression of Fusaruim wilt caused by Fusarium oxysporum f. sp. lycopersici and late blight casued by Phytophthora infestans, were examined. Both late blight and fusarium wilt were suppressed by spray and irrigation of chitosan, respectively. Inhibition of mycelial growth was not greatly affected by molecular size of chitosan but, concentration dependent effects was observed. Ninty percent of P. infestans and 80% of F. oxysporum f. sp. lycopersici of mycelial growth was inhibited by 1,000 ppm of chitosan (MW 30,000~50,000) when amended in plate media. Induction of defense-related gene expression in plant by chitosan treatments were observed when chitosan treated tobacco and tomato RNA samples were hybridized with several defense-related genes as probes. The results revealed that $\beta$-1,3-glucanase and chitinase genes were strongly induced, while pathogenesis-related protein-1, 3-hydroxy-3-methylglutaryl coenzyme A reductase, anionic peroxidase, phenylalanine ammonia lyase genes were weakly induced by chitosan treatment. These results suggest that chitosan have dual effects on these host-pathogen interactions. Possible roles of chitosan in suppression of tomato diseases by inhibition of mycelial growth and activation of plant defense responses are discussed.

Recently, Immune therapies of gene which use cytokines and HLA-B7 are carrying out. 2. In oriental medicine, development of disease is closely related to up and down of healthy qi, so healthy qi operates as a immune factor and resistance factor. 3. On the base of theory "Increasing healthy qi reduces mass(養正則積自除)", strengthening body resistance is emphasized in cancer therapy. Also strengthening body resistance activates cellular immune response and promote killing tumor facility of T-cell. 4. In clinical view, using immune therapy after operation, radiation, and chemotheraphy is more effective than immune therapy itself, so it is expected that east-west cooperation will be effective in cancer therapy. 5. The study of oncological immunity is progressed on emphasizing T-cell and it is related to oriental medical theory "strengthening healthy qi to eliminate pathogen(扶定祛邪)" and advanced study is expected in future.

Certain beneficial microorganisms isolated from rhizosphere soil promote plant growth and induce resistance to a wide variety of plant pathogens. We obtained 49 fungal isolates from the rhizosphere soil of paprika plants, and selected 18 of these isolates that did not inhibit tomato seed germination for further investigation. Based on a seed germination assay, we selected four isolates for further plant tests. Treatment of seeds with isolate JF27 promoted plant growth in pot tests, and suppressed bacterial speck disease caused by Pseudomonas syringae pathovar (pv.) tomato DC3000. Furthermore, expression of the pathogenesis-related 1 (PR1) gene was higher in the leaves of tomato plants grown from seeds treated with JF27; expression remained at a consistently higher level than in the control plants for 12 h after pathogen infection. The phylogenetic analysis of a partial internal transcribed spacer sequence and the b-tubulin gene identified isolate JF27 as Aspergillus terreus. Taken together, these results suggest that A. terreus JF27 has potential as a growth promoter and could be used to control bacterial speck disease by inducing resistance in tomato plants.
The filamentous fungus Fusarium graminearum is an important cereal pathogen. Although quantitative realtime PCR (qRT-PCR) is commonly used to analyze the expression of important fungal genes, no detailed validation of reference genes for the normalization of qRT-PCR data has been performed in this fungus. Here, we evaluated 15 candidate genes as references, including those previously described as housekeeping genes and those selected from the whole transcriptome sequencing data. By a combination of three statistical algorithms (BestKeeper, geNorm, and NormFinder), the variation in the expression of these genes was assessed under different culture conditions that favored mycelial growth, sexual development, and trichothecene mycotoxin production. When favoring mycelial growth, GzFLO and GzUBH expression were most stable in complete medium. Both EF1A and GzRPS16 expression were relatively stable under all conditions on carrot agar, including mycelial growth and the subsequent perithecial induction stage. These two genes were also most stable during trichothecene production. For the combined data set, GzUBH and EF1A were selected as the most stable. Thus, these genes are suitable reference genes for accurate normalization of qRT-PCR data for gene expression analyses of F. graminearum and other related fungi.
Akabane disease is caused by an arthropod-borne viral pathogen and leads to congenital abnormalities of the central nervous system in infected ruminants. One isolate, KV0505, showed cytopathic effect in Vero cells. The KV0505 isolate was obtained from plasma, which was collected from a cattle raised on Jeju Island in May 2005. Jeju Island is located near the southern part of the Korean peninsula. The isolate was confirmed as Akabane virus (AKAV) by immunofluorescence assay using AKAV specific monoclonal antibodies and reverse transcription polymerase chain reaction (RTPCR). Suckling mice inoculated with the isolate showed signs of paralysis and died within 10 days postinoculation. Comparisons of the KV0505 N gene sequence with 39 other known AKAV strains revealed nucleotide homologies ranging from 83.6% (MP496 strain) to 99.7% (M171 strain). When compared with the K-9 strain, which was isolated from a cow in Korea in 1994, the nucleotide sequence homology with the N gene was 99.7%. Thus, genes of the KV0505 isolate were closely related to those of the M171 strain, which were clustered into the Ic group of AKAV.
Human hepatitis B virus (HBV) is a pathogen related to the development of liver diseases including chronic hepatitis, cirrhosis, and hepatocellular carcinoma (HCC). However, the efficient methods to suppress HBV replication have not been developed yet. Therefore, we have used RNA interference (RNAi) as a potential tool for the suppression of HBV replication. Here, we designed a 21 nt small intefering dsRNA (siRNA) against hepatitis B virus X (HBx) RNA with 3' overhanging ends derived from T7 promoter. It has been reported that HBV X protein plays an important role in HBV gene expression and viral replication. The suppression of HBx gene expression by the 21 nt siRNA was investigated by Northern blot analysis and chloramphenicol acetyl transferase (CAT) assay. The level of HBx mRNA was decreased by siRNA in a dose-dependent manner. We also found that the 21 nt siRNA inhibited the HBV replication in hepatocellular carcinoma cell.
The objective of this research was introduction of chit42 to tuber mustard plants through Agrobacteriummediated transformation against white mold caused by Sclerotinia sclerotiorum. The binary plasmid pGisPEC1 was used in this study. Polymerase chain reaction analysis detected the transgene in 27 transformants with a transformation efficiency of 6.9%. Southern blot test was used to assess the copy number of transgene in tuber mustard plants. One, two, two, and two chit42-related bands were observed in the transformed lines TMB4, TMB7, TMB12, and TMB18, respectively. Enzymatic tests showed a significant increase in the activity of endochitinase in protein isolated from leaf tissues of chit42 transgenic 75-day tuber mustard lines. The pathogenicity of three pathogen isolates was tested on the leaves of transformed plans. The results of current study showed that expression of the gene chit42 in tuber mustard plants markedly reduced infection radius on the leaves 7 days after inoculation with the fungus.
Acanthamoeba is an opportunistic pathogen known to cause granulomatous amoebic encephalitis and amebic keratitis. Acanthamoeba exhibits life cycle consisting of trophozoite and cyst, and the cyst is highly resistant to variable antibiotics and therapeutic agents. To understand the encystation mechanism of Acanthamoeba, the expression profiles of trophozoite and cyst were compared by gene ontology (GO) analysis. Ribosomal proteins and cytoskeletal proteins were highly expressed in trophozoite. In cyst, various protease, and signal transduction - and protein turnover - related proteins were highly expressed. These results correlated with eukaryotic orthologous groups (KOG) assignment and microarray analysis of Acanthamoeba trophozoite and cyst ESTs. The information of differential expression profiles of trophozoite and cyst would provide important clues for research on encystation mechanism of cyst forming protozoa including Acanthamoeba.
The diseases caused by Xylella fastidiosa are associated with aggregation of the bacteria m xylem vessels, formation of a gummy matrix and subsequent blockage of water uptake. In the closely related pathogen, Xanthomonas campestris, xanthan gum is known to be an important virulence factor, probably contributing to bacterial adhesion, aggregation and plugging of xylem. Xanthan gum, produced by X. campestris, is an extra-cellular polysaccharide consisting of a cellulose backbone ($\bate$-1,4-linked D-glucose) with trisaccharide side chains composed of mannose, glucuronic acid and mannose attached to alternate glucose residues in the backbone. We had constructed a mutant of X. campestris lacking gumI gene that is responsible for adding the terminal mannose for producing modified xanthan gum which is similar to xanthan gum fromX. fastidiosa. The modified xanthan gum degrading endgphytic bacterium Acineto-bacter johnsonii GX123 isolated from the oleander infected with leaf scorch disease.
Here, we show that an endophytic bacterial strain, Enterobacter asburiae B1 exhibits the ability to elicit ISR in cucumber, tobacco and Arabidopsis thaliana. This indicates that strain B1 has a widespread ability to elicit ISR on various host plants. In this study, E. asburiae strain B1 did not show antifungal activity against tested major fungal pathogens, Colletotrichum orbiculare, Botrytis cinerea, Phytophthora capsici, Rhizoctonia solani, and Fusarium oxysporum. Moreover, the siderophore production by E. asburiae strain B1 was observed under in vitro condition. In greenhouse experiments, the root treatment of strain B1 significantly reduced disease severity of cucumber anthracnose caused by fungal pathogen C. orbiculare compared to nontreated control plants. By root treatment of strain B1 more than 50% disease control against anthracnose on cucumber was observed in all greenhouse experiments. Simultaneously, under the greenhouse condition, the soil drench of strain B1 and a chemical inducer benzothiadiazole (BTH) to tobacco plants induced GUS activity which is linked with activation of PR promoter gene. Furthermore, in Arabidopsis thaliana plants the soil drench of strain B1 induced the defense gene expression of PR1 and PDF1.2 related to salicylic acid and jasmonic acid/ethylene signaling pathways, respectively. In this study, for the main focus on root colonization by strain B1 associated with defense responses, bacterial cells of strain B1 was tagged with the gfp gene encoding the green fluorescent protein in order to determine the colonization pattern of strain B1 in cucumber. The gfp-tagged B1 cells were found on root surface and internal colonization in root, stem, and leaf. In addition to this, the scanning electron microscopy observation showed that E. asburiae strain B1 was able to colonized cucumber root surface.
