Clinical and bacteriological studies on infectious atrophic rhinitis of swine (돼지 전염성 위축성 비염의 임상학적 및 세균학적 연구)

  • Clinical and bacteriological observations on infectious atrophic rhinitis of swine were conducted in order to obtain some basic information for the clinical and immnnological control of the disease prevailing in the republic. Samples were collected from nasal cavities of 135 4~12 week old pigs from 12 herds and from turbinates of 199 slaughtering pigs from 14 swine herds to investigate the prevalence of Bordetella bronchiseptica and/or Pasteurella multocida in the nasal cavities of the pigs. On the examination of nasal swabs by cultural techniques and of gross lesion of snouts of slaughtering pigs, all the swine herds investigated were found to be affected by atrophic rhinitis and a total of 84 B bronchiseptica and 139 P multocida cultures were isolated from the nasal cavities of the pigs. Of the 199 slaughtering pigs, some 48% revealed gross pathological lesion typical of atrophic rhinitis and the prevalence of B bronchiseptica and P multocida were 27.6% and 46.7%, respectively. Biochemical properties, antimicrobial susceptibilities, serological characteristics and toxigenicity of the isolates of B bronchiseptica and P multocida were investigated.

Prevalence of bacterial respiratory diseases in cattle (소 세균성 호흡기 질병 분포도 조사)

  • The objective of this study was to determine the prevalence of pneumonic bacteria in lungs and nasal swabs of cattle with respiratory diseases. From 95 pneumonic lungs of slaughtered cattle, 41 (43.2%) positive lungs were yielded with 54 pneumonic bacteria, which corresponded to P. multocida (n = 34), A. pyogenes (n = 14) and P. haemolytica (n = 6). One-hundred sixty seven pneumonic bacteria were isolated from 195 nasal swabs in calves, 64.7% (108 isolates) belonged to P. multocida, 16.2% to A. pyogenes, 13.8% to P. haemolytica and 5.4% to H. somnus. Fifty percents (n = 6) of isolates from pneumonic lungs of calves were identified as P. multocida. All isolates of P. multocida belonged to type A according to hyaluronidase test. Antimicrobial susceptibility tests showed that most isolates of P. multocida and P. haemolytica were sensitive to amoxicillin/clavulanic acid, cephalothin, ciprofloxacin, enrofloxacin, fluorophenicol and norfloxacin. The emergence of antimicrobial resistant Pasteurella spp. observed in this study, however, might limit such application. According to histopathological examination, pneumonia by mycoplasma or/and bacteria accounted for 92.8% among 69 pneumonic lungs of slaughtered cattle.

Killing Rate Curve and Antivacterial Activity against Various Pathogenic Bacteria in the Presence of Enrofloxacin and Colistin (병원성미생물에 대한 Enrofloxacin과 Colistin의 배합비육에 따른 항균작용과 균의 사멸속도)

  • Enrofloxacin-colistin combination, widely used in Gram negative infections in veterinary sector, was investigated in terms of MIC and initial killing rate using E coli k 88ab, Salmonella typhimurium, Pasteurella multocida type A, Bordetella bronchiseptica and Staphylococcus aureus as test organisms. On the basis of MICs of enrofloxacin-colistin combination against the above bacteria, killing rates of the combination of enrofloxacin and colistin at the ratio of 5:0, 4:1, 3:2, 1:1, 2:3, 1:4 and 0:5, indicated high and rapid antibacterial acitivities against all but Staphylococcus aureus R-209, with the number of bacteria reducing to less than one percent within two hours. At the MIC of enrofloxacin or colistin, both antibacterials showed the highest killing rates during 2-4 hours against Gram negatives such as E coli K88ab,Pasteurella multocida type A and Bodetella bronchiseptica but allowed the regrowth of the same pathogens thereafter. On the while, the combination of two antibacterials at a fourth MIC resulted in high killing rate without bacterial regrowth during 24 hours, suggesting the synergistic antivacterial effects. The combination, however, did not show favourable activity against Gram negatime S typhimurium and Gram positive S aureus ergistic antibacterial activity against Gram negatime pathogens but also colistin showed LPS-neutraization, we could suggest the combination should provide clinically positive therapeutic armarium in Gram negative infections.

Epidemiological study on prevalence of Iung lesions of slaughtered pigs in winter (동절기 도축돈 폐병변 발생 양상에 관한 역학조사)

  • This study was carried out to investigate and analyze the gross lesions of lungs in slaughtered pigs. Pigs were collected from the areas of Kyounggj, Chungbuk, Chungnam and Kangwon provinces from November to December 1999. One hundred-eleven pigs(17.4%) had lung lesions with various degrees among 639 pigs tested. By the standard scoring system, mean score of the lung lesion was 25.6+13.2. Regional prevalence of lung lesions were 23.5%(8/34)) in Kangwon, 17.3%(61/352) in Kyunggi, 15.7%(32/204) in Chungnam and 20.4%(10/49) in Chungbuk. Mean number of pulmonary lesions per pig were 2.87. Most frequent region with pulmonary lesions was right cranial lobe(30.1%) and the decreasing prevalence rates were followed by 23.8% in right middle lobe, 21.05 in right accessory lobe, 15.0% in left cranial lobe, and 5.0% in left middle and accessory lobe(5.0%) and also there was the same prevalent tendency on pulmonary lesions in each lobes of 111 pigs with gross lesion. Isolation rate of bacteria from the affected lungs was 72.1% and main pathogen was Pasteurella multocida. Gross and histological examination of pulmonary lesions in some pigs suggested that there were no marked changes regarded as the correlation with specific diseases except fibropurulent bronchopneumonia which was suspective of some respiratory bacteria including Pasteurella multocida and peribronchiolar lymphoid hyperplasia in varying degrees which was strongly associated with mycoplasmal infection. Consequently, the results in the study were suggested that there was consistently exposed against many causative factors including bacteria in the considerable number of pig herds rearing in the middle area in Korea.

Immunological Characterization of Full and Truncated Recombinant Clones of ompH(D:4) Obtained from Pasteurella multocida (D:4) in Korea

  • We cloned a gene of ompH(D:4) from pigs infected with P. multocida D:4 in Korea [16]. The gene is composed of 1,026 nucleotides coding 342 amino acids (aa) with a signal peptide of 20 aa (GenBank accession number AY603962). In this study, we analyzed the ability of the ompH(D:4) to induce protective immunity against a wild-type challenge in mice. To determine appropriate epitope(s) of the gene, one full and three different types of truncated genes of the ompH(D:4) were constructed by PCR using pET32a or pRSET B as vectors. They were named ompH(D:4)-F (1,026 bp [1-1026] encoding 342 aa), ompH(D:4)-t1 (693 bp [55-747] encoding 231 aa), ompH(D:4)-t2 (561 bp [187-747] encoding 187 aa), and ompH(D:4)-t3 (540 bp [487-1026] encoding 180 aa), respectively. The genes were successfully expressed in Escherichia coli BL21(DE3). Their gene products, polypeptides, OmpH(D:4)-F, -t1, -t2, and -t3, were purified individually using nickel-nitrilotriacetic acid (Ni-NTA) affinity column chromatography. Their $M_rs$ were determined to be 54.6, 29, 24, and 23.2 kDa, respectively, using SDS-PAGE. Antisera against the four kinds of polypeptides were generated in mice for protective immunity analyses. Some $50{\mu}g$ of the four kinds of polypeptides were individually provided intraperitoneally with mice (n=20) as immunogens. The titer of post-immunized antiserum revealed that it grew remarkably compared with pre-antiserum. The lethal dose of the wild-type pathogen was determined at $10{\mu}l$ of live P. multocida D:4 through direct intraperitoneal (IP) injection, into post-immune mice (n=5, three times). Some thirty days later, the lethal dose ($10{\mu}l$) of live pathogen was challenged into the immunized mouse groups [OmpH(D:4)-F, -t1, -t2, and -t3; n=20 each, two times] as well as positive and negative control groups. As compared within samples, the OmpH(D:4)-F-immunized groups showed lower immune ability than the OmpH(D:4)-t1, -t2, and -t3. The results show that the truncated-OmpH(D:4)-t1, -t2, and -t3 can be used for an effective vaccine candidate against swine atrophic rhinitis caused by pathogenic P. multocida (D:4) isolated in Korea.

  • In domestic rabbits, pasteurellosis is a primary respiratory disease entity caused by Pasteurella multocida. Respiratory pasteurellsos in rabbits is highly contagious due to dissemination of the organism in the nasal discharge (Benirschke, et al., 1978). The disease reflects a varied and complex host-parasite relationship. Acute infection with high mortality occurs sporadically especially in stressed rabbits less than 10 weeks old, in the form either an enzootic pneumonia, infection of the upper respiratory tract or septicemia. In addition to respiratory form, infection with P. multocida results in a variety of disease processes in rabbits including rhinitis, otitis media, otitis interna, abscesses, metritis, orchitis and meningoencephalaitis (Manning et al., 1989). The deleterious effects of pasteurellosis are common and pose serious problems for rabbit breeders and researchers (Flatt, 1974). (omitted)

Exchange method of the nasal cavity spraying amikacin instead of kanamycin and gentamicin (가나마이신 및 겐타마이신 비강 분무액을 아미카신 비강 분무액으로 바꾸는 방법)

  • Atrophic rhinitis (AR) is the one of important respiratory diseases and causes severe economic losses in pig industry. Severe attempts have been made to reduce the economic losses by preventing the disease. One of the methods is the spraying of antibiotics into nasal cavity of piglets. Recently, the efficacy of the spraying with kanamycin and gentamicin was reduced in the Korean swine industry. Therefore, the preventive methods have been required to be changed based on the antimicrobial susceptibility profiles of causative agents of swine AR. Based on the current situations of this disease, Bordetella (B.) bronchiseptica and Pasteurella (P.) multocida 4D were isolated from pigs with clinical signs of AR. The isolation rates of B. bronchiseptica and P. multocida 4D were 58.5% and 32.9%, respectively. In the antimicrobial susceptibility test, the bacteria were resistant to kanamycin and gentamicin which have been used as the spraying agents, but they were susceptible to amikacin. A new spraying agent was developed using amikacin using ${\beta}$-glucan and yakbaltag as supplementary agents. Field efficacy of the agent was carried out with different schedule. The results from this study suggested that the newly developed spraying agents might be helpful to prevent AR in swine.

Efficacy of atropic rhinitis vaccine in pigs (돼지 위축성 비염백신의 효과에 관한 연구)

  • Atropic rhinitis (AR) is one of major respiratory diseases in pigs. AR causes a great economic losses and is considered to be a multifactorial disease in which herd management, heredity, and environment. Several vaccines against have been developed commercially and used in pig farms but the efficacy of each vaccine is still questionable. In this study, one of commercial AR vaccines, which contains inactivated Bordetella bronchiseptica, Pasteurella multocida type D and their toxoid was evaluated for vaccine efficacy by challenge test. Twenty piglets were divided into four groups as follows; group I was piglets from vaccinated sows (twice before parturition); group II was piglets from vaccinated sows (same as group I) and were vaccinated at 1 day old; group III and IV were piglets without any vaccination. Groups I, II, and III were challenged by intranasal instillation of $5.3{\times}10^7$ CFU of B bronchiseptica twice and $1{\times}10^9$ CFU of P multocida five times. Group IV was control group without any vaccination and any challenge. We compared serological results, recovery rate of P multocida by polymerase chain reaction, clinical signs and pathological findings between vaccinated groups and unvaccinated groups for efficacy of the vaccine, Serological responses against B bronchiseptica and toxigenic P multocida type D were not showed evident discrepancy between vaccinated groups and unvaccinated groups assuming that the antibody responses against the vaccine is very delayed. However, growth rate, clinical signs and snout lesion grading in vaccinated groups showed more favorable than those in unvaccinated group. Therefore, AR vaccination in this study is considered to be effective in the prevention of AR in pigs.

Annual Distribution of Heterotrophic Bacterial Community in the Marine Ranching Ground of Tongyeong Coastal Waters (통영 바다목장 해역의 종속영양세균 군집의 연차적 분포)

  • The cell numbers of heterotrophic bacteria inhabiting the surface and bottom sea water harvested from the 5 stations in the marine ranching ground of Tongyeong coastal waters in $2003{\sim}2007$ were examined, and species composition of the heterotrophic bacterial population and dominant species were analyzed as well. Sea water samples collected in summer season contained much higher number of heterotrophic bacteria than those harvested in winter, spring and autumn seasons due to the higher sea water temperature. However the cell number of heterotrophic bacteria did not show a significant dependence on the location of the sampling stations. The cell number of heterotrophic bacteria in the surface sea water harvested in October 2003 and in September 2004 was not discernibly different from that in the bottom sea water and sometimes the former was even fewer than the latter because of the typhoon and localized torrential downpour. The number of heterotrophic bacteria decreased every year. The main bacterial species were Pseudomonas fluorescens TY1, Pseudomonas stutzeri TY2, Acinetobacter lwoffii TY3, Sphingomonas paucimobilis TY4, Burkholderia mallei TY5, Pasteurella haemolytica TY6, Pasteurella multocida TY7, Comamonas acidovorans TY8, Actinobacillus ureae TY9 and Chryseobacterium indologenes TY10. P. fluorescens TY1 and A. lwoffii TY3 were found to be the dominant species.
