Nano-emulsion Containing Parthenocissus tricuspidata Stem Extracts for Enhanced Skin Permeation and the Antibacterial Activity of the Extracts (피부 흡수 증진을 위한 담쟁이덩굴 줄기 추출물 함유 나노에멀젼 및 이의 항균활성 연구)

  • In a previous study, we investigated the antioxidative and cellular protective effects of Parthenocissus tricuspidata stem extracts. In this study, we prepared nano-emulsion containing P. tricuspidata stem extract to improve skin permeation. The particle size of the nano-emulsion using the microfluidizer was 302 nm. Its loading efficiency was over 86%. The size distribution of the nano-emulsion took a monodispersed form and the nano-emulsion was more stable than typical emulsion without using microfluidizer during a 2 week period. In vitro skin permeation study of nano-emulsion containing P. tricuspidata stem extracts was carried out using Franz diffusion cell. The 1,3-butylene glycol used as a control group had 32.59% skin permeation efficiency. The skin permeation efficiency of the nano-emulsion was 42.47%. Also, we observed the antibacterial activity of the ethyl acetate fraction on skin flora for prospective applications as a natural antimicrobial. The ethyl acetate fraction had antibacterial activities higher than methyl paraben on Staphylococcus aureus, and Bacillus subtilis. These results indicate that nano-emulsion containing P. tricuspidata stem extracts could possess valued applications in cosmetic formulations for improving skin permeation. Also, based on the antibacterial activities on skin flora, antioxidative and cellular protective effects shown in our previous study, we suggest that P. tricuspidata stem extracts could be used as functional cosmetic materials.

Antioxidant, Antidiabetic, and Anti-Inflammatory Effects of Extracts and Fractions from Parthenocissus tricuspidata Stems (담쟁이덩굴 추출물과 분획물의 항산화, 항당뇨 및 항염증 효과)

  • This study was carried out to analyze the effects on antioxidative, antidiabetic, and anti-inflammatory activities of Parthenocissus tricuspidata (PT) stem extracts. The total phenolic contents of hot water and ethanol extracts from PT stems were 61.5 mg TAE/g and 122.1 mg TAE/g, respectively. The antioxidative activities of hot water and ethanol extracts from PT stem were measured by using 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity and superoxide dismutase (SOD) assay. The DPPH radical scavenging activities of ethanol extract and butanol fraction were approximately 95% and 92% at 1 mg/ml, respectively, and the SOD activities of ethanol extract and butanol fraction were about 91% and 97% at 1 mg/ml, respectively. The DPPH radical scavenging and SOD activities of ethanol extract and butanol fraction from PT stem increased remarkably increased in a dose-dependent manner and were higher than in the hot water extracts. Compared to the acarbose, a known anti-diabetic drug, which was used as a positive control, the ${\alpha}$-glucosidase inhibitory capacity of PT stem showed a strong inhibitory rate in ethanol extract and in butanol and hexane fractions. We investigated the effect of hot water extract from PT stem on lipopolysaccharide (LPS)-induced nitric oxide (NO) production in RAW 264.7 cells. Hot water extract from PT stem inhibited LPS-induced NO production up to 40% at a treatment of 1 mg/ml. These results suggest that PT stem extracts have an effect on antioxidative, antidiabetic, and anti-inflammatory activities and thus have great potential as antidiabetic materials and a source for natural health products.

Anti-Obesity Effect of Parthenocissus tricuspidata (Siebold & Zucc.) Planch Stem Extracts (소나무 담쟁이덩굴(Parthenocissus tricuspidata (Siebold & Zucc.) Planch) 줄기 추출물의 항비만 효과)

  • In this study, we investigated in vitro anti-obesity activity of Parthenocissus tricuspidata (Siebold & Zucc.) Planch stem (PTS) in mouse adipocytes 3T3-L1 cells. PTS inhibited lipid accumulation and reduced the expression level of the proteins such as C/EBPα, PPARγ and FABP4 associated with adipogenesis in 3T3-L1 cells. In addition, PTS increased lipolysis-related protein expression such as p-AMPK, ATGL and p-HSL, and reduced perilipin-1 in 3T3-L1 cells. Taken together, PTS may have anti-obesity activity through inhibiting adipogenesis and inducing lipid lipolysis in adipocytes. Based on these results, PTS is expected to be used as a potential functional agent for anti-obesity.

Screening of the Antioxidant Defense Systems from Parthenocisuss tricuspidata PLANCH (담쟁이덩굴의 항산화 방어계의 탐색)

  • The non-enzymatic antioxidants and antioxidant enzyme from the extracts of Parthenocissus tricuspidata PLANCH. were examined in order to utilize natural product for cerchemopreventive agents. The antioxidant potential and enzyme activities on plant positions in the extracts of Parthenocisuss tricuspidata PLANCH. showed considerable differences. The antioxidant activity of the leaf extracts by Ethyl acetate fractions of Parthenocisuss tricuspidata PLANCH. was the highest among three positions ($7.57\mu\textrm{g}/m\ell$). The highest activities showed in S-5 (in leaf), S-4 (in stem) and S-3 (in root) fraction by Silicagel column chromatography and the antioxidant activity showed, in purified extract of each positions, $7.06\mu\textrm{g}/m\ell$ (in leaf), $6.99\mu\textrm{g}/m\ell$ (in stem) and $12.39\mu\textrm{g}/m\ell$ (in root) respectively. The activities of DPPH by LH-20 column chromatography revealed much higher than those by silica-gel column chromatography. These were identified as the phenolic compounds known as antioxidant compounds such as Benzoic acid(Gallic acid), 1-methyl-3-(2-phenylethen) benzene, phloroglucinol and 1,2-dihydroxy-4-(1-propyl)benzene by GC/MS. POD activities in the stem and root were higher than in the leaf. SOD activity was highest in the leaf, stem and root activity was comparatively low. Especially, SOD activity in leaf was over 2 times higher than root.

Cellular Protective and Antioxidative Acivities of Parthenocissus tricuspidata Stem Extracts (담쟁이덩굴 줄기 추출물의 세포 보호 작용과 항산화 활성)

  • In this study, the cellular protective effects on HaCaT cells and human erythrocytes and antioxidative effects of P. tricuspidata stem extracts were investigated. The ethyl acetate ($50{\mu}g/mL$) and aglycone fraction ($25{\mu}g/mL$) of P. tricuspidata stem extracts doesn't show any characteristics of cytotoxicity. When HaCaT cells were treated with 10 mM $H_2O_2$ and $30{\mu}M$ rose bengal, the ethyl acetate ($6.25{\sim}50{\mu}g/mL$) and aglycone ($6.25{\sim}25{\mu}g/mL$) fraction protected the cells against the oxidative damage in a concentration dependent manner. The P. tricuspidata stem extracts showed more prominent cellular protective effect than (+)-${\alpha}$-tocopherol, known as lipid antioxidant at $10{\mu}g/mL$. The ethylacetate fraction of P. tricuspidata stem extracts ($18.5{\mu}g/mL$) showed more free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity ($FSC5_{50}$). Reactive oxygen species (ROS) scavenging activity ($OSC_{50}$) of P. tricuspidata stem extracts on ROS generated in $Fe^{3+}$-EDTA/$H_2O_2$ system was investigated using the luminol-dependent chemiluminescence assay. The ethyl acetate ($1.72{\mu}g/mL$) and the aglycone fraction ($1.53{\mu}g/mL$) showed similar ROS scavenging activity of L-ascorbic acid ($1.50{\mu}g/mL$). These results indicate that extract/fractions of P. tricuspidata stem extracts can function as natural cytoprotective agents and antioxidants in biological systems, particularly skin exposed to UV radiation by protecting cellular membrane against ROS.

Antioxidant and Cellular Protective Effects of Parthenocissus tricuspidata Stem Extracts Fermented by Lactobacillus pentosus (Lactobacillus pentosus 발효에 의한 담쟁이덩굴 줄기 추출물의 항산화 및 세포보호 효과)

  • In this study, the antioxidant activities, cellular protective effects, and inhibitory effects on elastase of non-fermented and fermented extracts of Parthenocissus tricuspidata (P. tricuspidata) stem using Lactobacillus pentosus were investigated. The free radical scavenging activities ($FSC_{50}$) of non-fermented and fermented extracts were 42.3 and $34.5{\mu}g/mL$, respectively, in which the activity after fermentation was approximately 18.4% higher. Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) in $Fe^{3+}-EDTA/H_2O_2$ system of non-fermented and fermented extracts were 2.6 and $2.5{\mu}g/mL$, respectively. The activity after fermentation was approximately 4.2% higher. In the $^1O_2$-induced cellular damage of erythrocytes, the cellular protective effects (${\tau}_{50}$) of non-fermented and fermented extracts were 126.4 and 173.0 min at $50{\mu}g/mL$, respectively. The activity after fermentation was approximately 34.0% higher. The effect of fermented extract was 3.9 times higher than $(+)-{\alpha}$-tocopherol (${\tau}_{50}=43.4min$), known as a lipophilic antioxidant at $50{\mu}g/mL$. The inhibitory effect of elastase was investigated to predict the anti-wrinkle efficacy using Hs68 human fibroblasts cells. The elastase inhibitory activities ($IC_{50}$) of non-fermented and fermented extracts were 873.6 and $687.8{\mu}g/mL$, respectively, and the activity after fermentation was approximately 21.3% higher. These results indicated that fermented extract of P. tricuspidata stem has potentials as natural cosmetic ingredients with antioxidant and anti-wrinkle effect.
