Effect of Transport Stocking Density on the Physicochemical Characteristics of Muscle from Olive Flounder Paralichthys olivaceus (수송밀도가 넙치(Paralichthys olivaceus) 근육의 물리화학적 변화에 미치는 영향)

  • The effects of transport stocking density (500, 600 and 700 kg of olive flounder Paralichthys olivaceus/per water tank) on the physicochemical characteristics of muscle were investigated. Transport of the animals took, on average, 10 days from Busan, Korea, to Long Beach, CA, USA. There was no significant difference in the breaking strength of muscle among the three groups. There was also no significant difference in the ATP, lactate, or glycogen content of muscle from the 500 and 600 kg/tank stocking density groups at the beginning and after transport. However, the ATP content decreased sharply while the lactate content increased in muscle from fish transported at a density of 700 kg/tank at the beginning of transport, and the ATP and lactate contents in this group were stable after transport.

  • This study was performed to estimate biomass and provide management guidance through population ecological characteristics, including growth parameters, instantaneous coefficients of natural and fishing mortalities, and age at first capture of the starry flounder, Platichthys stellatus and olive flounder, Paralichthys olivaceus of Korea. For describing growth of this species, a von Bertalanffy growth model was adopted. The von Bertalanffy growth parameters estimated from a non-linear regression for starry flounder were $L_{{\infty}}=48.25cm$, K=0.16/yr, and $t_0=-1.48$, respectively and those for olive flounder were $L_{{\infty}}=86.46cm$, K=0.26/yr, and $t_0=-0.29$, respectively. Biomass of Platichthys stellatus was estimated by direct biomass estimation method was 2.6 M/T, that was estimated by indirect method was 13.4 M/Tt. Those of Paralichthys olivaceus were estimated as 10.1 M/T, 19.3 M/T, respectively. An yield per recruit analysis showed that the current yield per recruit on Platichthys stellatus was about 48.2 g with F=0.646/yr and the age at first capture ($t_c$) 1.35yr, that on Paralichthys olivaceus was about 167.6 g with F=1.121/yr and the age at first capture ($t_c$) 1yr.

Changes of hematological parameters and plasma constituents in the olive flounder Paralichthys olivaceus exposed to TBT (TBT 노출에 따른 넙치, Paralichthys olivaceus의 혈액학적 성상 및 혈장성분의 변화)

  • Juvenile Paralichthys olivaceus (mean length $17.3{\pm}2.2cm$, mean weight $46.5{\pm}6.3g$) were exposed for 10 days to tributyltin (TBT) at various concentrations (0, 1, 2, 4 and $8{\mu}g/L$). Hematological parameters, such as red blood cell (RBC) count, hematocrit (Ht), and hemoglobin (Hb) concentration, were considerably decreased by high TBT concentrations. In plasma constituents, inorganic components such calcium and magnesium were also significantly altered. Plasma calcium was notably decreased, whereas plasma magnesium was increased. The enzyme components, glutamic oxalate transaminase (GOT) and glutamic pyruvate transaminase (GPT), were significantly decreased by TBT exposure. The present findings suggest that TBT exposure can cause significant alterations in hematological parameters and plasma constituents of flatfish Paralichthys olivaceus.

Effects of Nonylphenol on Gonadal Development and Growth of Olive Flounder, Paralichthys olivaceus (Nonylphenol이 넙치, Paralichthys olivaceus의 생식소 발달과 성장에 미치는 영향)

  • Effects of nonylphenol (NP) on gonadal development and growth of olive flounder, Paralichthys olivaceus were investigated. NP treatment was carried out to fry fish (during 55 to 64 days after hatching) using oral adminstration at nominal NP concentrations of 50, 100 and 150 tig/g BW Gonad before NP treatment was sexually undifferentiated as observed with mostly gonia cells. At 159 days after hatching, ovarian lamella of ovary were filled with oogonia and perinucleolus oocytes. On the other hand, testicular lobules of the testis were occupied by spermatogonia, spermatocytes and spermatids. Histological observation of ovary and testis development was any different between the control and NP treated groups. However, growth was significantly increased in NP treated groups than control groups (P< 0.05). These results considerate that NP has any effect for sex differentiation and gonadal development, but act for early growth in olive flounder.

Performance Evaluation of Object Detection Deep Learning Model for Paralichthys olivaceus Disease Symptoms Classification (넙치 질병 증상 분류를 위한 객체 탐지 딥러닝 모델 성능 평가)

  • Paralichthys olivaceus accounts for a large proportion, accounting for more than half of Korea's aquaculture industry. However, about 25-30% of the total breeding volume throughout the year occurs due to diseases, which has a very bad impact on the economic feasibility of fish farms. For the economic growth of Paralichthys olivaceus farms, it is necessary to quickly and accurately diagnose disease symptoms by automating the diagnosis of Paralichthys olivaceus diseases. In this study, we create training data using innovative data collection methods, refining data algorithms, and techniques for partitioning dataset, and compare the Paralichthys olivaceus disease symptom detection performance of four object detection deep learning models(such as YOLOv8, Swin, Vitdet, MvitV2). The experimental findings indicate that the YOLOv8 model demonstrates superiority in terms of average detection rate (mAP) and Estimated Time of Arrival (ETA). If the performance of the AI model proposed in this study is verified, Paralichthys olivaceus farms can diagnose disease symptoms in real time, and it is expected that the productivity of the farm will be greatly improved by rapid preventive measures according to the diagnosis results.

Effect of the Burrowing Substratum on the Growth and Ambicoloration of Juvenile Flounder Paralichthys olivaceus Cultured at High Density (넙치(Paralichthys olivaceus) 치어의 성장 및 양면착색 현상에 있어 잠입기질의 효과)

  • To evaluate the influence of burrowing substrate on the rearing performance and ambicoloration of cultured flounder, Paralichthys olivaceus, we compared the daily food intake (DFI), feed efficiency (FE), survival, growth, proportion of pigmented skin on the blind side, and proportion of ambicolored fish. We reared juvenile flounders [total length (TL) $4.46{\pm}0.06cm$, body weight (BW) $0.77{\pm}0.03g$] in dark-green fiberglass-reinforced plastic (FRP) aquariums without (control) or with gravel substrate at a density of 200 fishes/ton for 120 days. While there was no difference in survival rate or growth, the DFI was lower and FE higher in the group raised with substrate than in the control. The proportions of pigmented area on the blind side and ambicolored fish were significantly higher in the control tank. Therefore, the supplement of substrate on the aquarium bottom positively affects the feeding efficiency, and inhibits abnormal pigmentation on the blind side in flounder farming at high density.

Monitoring Kudoa septempunctata in Cultured Olive Flounder Paralichthys olivaceus in Different Regions of Korea in 2013 (우리나라 양식 넙치(Paralichthys olivaceus) 육성어와 종묘의 지역별 Kudoa septempunctata 감염 현황)

  • A nationwide survey was conducted to study the rate of Kudoa septempunctata parasitization in cultured olive flounder, Paralichthys olivaceus, in Korea in 2013. Of 1107 olive flounder sampled randomly from 89 culture farms in five different regions in Korea, K. septempunctata was detected only in Jeju, where 10 of 318 fish (3.14%) were PCR positive, with genomic concentrations of $4.67{\times}10^5$ to $1.48{\times}10^{11}$ rDNA copies/g by real-time PCR. Of the ten Kudoapositive fish, K. septempunctata spores with 5-7 polar capsules were detected only in four fish. No samples of K. septempunctata were detected in olive flounder from the other regions surveyed. Furthermore, K. septempunctata was not detected in 326 samples of olive flounder seeds sampled from 39 hatcheries in seven different regions in Korea. Therefore, the parasite infection is restricted to Jeju and K. septempunctata infection is not spread from hatcheries.

The Physiological Changes in the Olive Flounder Paralichthys olivaceus Caused by a Diet Containing CaO (CaO (oxide calcium)가 첨가된 사료 공급이 넙치(Paralichthys olivaceus)에 미치는 생리적 변화에 대한 연구)

  • This study investigated the physiological effects of a diet containing calcium oxide (CaO) on the olive flounder, Paralichthys olivaceus. The results indicate that the amount of calcium in the blood was higher in the calcium group compared with the controls. The aspartate amino transferase (AST) and alanine amino transferase (ALT) levels were lower in the calcium group, while lysozyme activity was higher in the calcium group. Histologically, fish in the calcium group had more hepatocytes in the liver and more intact scales. In addition, an attack experiment using Vibrio anguillarum (KCTC-2711) and Edwardsiella tarda (KCTC-3657) showed that the survival was higher in the calcium group compared with the controls. These results suggest that feeding fish CaO when they require it, such as during periods of gonadal maturation and rapid growth, will increase their immunity and resistance to pathogens.

  • The effects of diluents composition on cold storage for olive flounder (Paralichthys olivaceus) sperm were examined in terms of the swimming speed of sperm, the percentage of mobile sperm, and fertilization rates. The following results indicated that cold storage methods with fresh conditions could be employed in olive flounder milt preservation. The preserved sperm of olive flounder that was diluted I: 10 with artificial seminal plasma II (ASP II) and Stein's solution (SS) at $0^{\circ}C$ remained motile for 30 days. The most effective condition for cold storage was ASP II and SS at 0 $0^{\circ}C$ for the sperm, although there is no significant difference statistically. No difference in the fertilization rate was found between fresh sperm and the preserved ones with ASP I, II and SS at 10 days post-storage.

Changes in Respiratory Metabolism and Blood Chemistry of Olive Flounder Paralichthys olivaceus Exposed to Hypoxia (저산소에 노출된 넙치(Paralichthys olivaceus)의 호흡대사와 혈액의 화학적 변화)

  • This experiment investigated changes in metabolic rate (MO2), critical oxygen saturation (Scrit), and blood chemistry of olive flounder Paralichthys olivaceus exposed to progressive hypoxia and returned to normoxic water at 20°C. The normoxic standard metabolic rate (SMR) and routine metabolic rate (RMR) were 69.5-83.9 and 70.2-156.4 mg O2 kg-1h-1, respectively based on fish weight. Scrit was 31.0% dissolved oxygen (DO) at 20°C. After returning the fish to 70% DO following exposure to hypoxia (20% DO), MO2 increased two-fold compared to the normoxic SMR and then decreased into the range of the RMR with time. Blood PO2 and plasma lactate decreased significantly after exposure to hypoxia (20% DO) and then increased as ambient oxygen saturation decreased. Cortisol levels increased as ambient oxygen saturation decreased, but the levels decreased rapidly in the range of the normoxic control when the fish were returned to ambient water with 70% DO. Plasma glucose levels increased when the fish were returned to normoxic water after exposure to a progressively more hypoxic condition.
