Repetitive Element-PCR (rep-PCR)을 이용한 Vibrio parahaemolyticus 의 분자유전학적 아형 분류 (Molecular Typing of Vibrio parahaemolyticus by Repetitive Element-PCR (rep-PCR))

  • The enterobacterial repetitive intergenic consensus (ERIC)-PCR is a recently described DNA fingerprinting technique based on amplification of repetitive element distributed in bacteria. We applied of ERIC-PCR to clinical isolates of Vibrio parahaemolyticus and other bacteria associated diarrhea. Twenty isolates of V. parahaemolyticus were used for intragenic genotyping, which were isolated from 2001 to 2002 in Chungbuk National University hospital. For interspecies genotyping, V. vulnificus, V. alginolyticus, V. parahaemolyticus, Escherichia coli, Salmonella and Shigella spp. were used. The genotyping were analyzed by ERIC-PCR. The genotyping of V. parahaemolyticus were grouped two major pattern (A, B) and were subdivided into 10 subtypes (A1, A2, B1-B8) by ERIC-PCR. These method distinctly differentiated bacterial species associated diarrhea. Those results show that ERIC-PCR can be reliable and efficient method for genotyping of V. parahaemolyticus and bacteria associated diarrhea.

인천연안 Vibrio parahaemolyticus의 분포 및 유산과 과산화수소 처리에 의한 항균효과 (Antimicrobial Effect of Lactic acid and Hydrogen Peroxide and Distribution of Vibrio parahaemolyticus from the Incheon Adjacent Sea)

  • This study was carried out to investigate the distribution of Vibrio parahaemolyticus in the Incheon adjacent sea, and antimicrobial effect on growth of Vibrio parahaemolyticus in lactic acid and hydrogen peroxide and combination of lactic acid and hydrogen peroxide. The detected strains were compared geographical, months and sample types. The distribution of Vibrio parahaemolyticus was high at Ganghwa county with 66.1%(336 samples), on 7-9 months with 72.4%(386 samples) and from tireland with 75.0%(90 samples), respectively. The minimun inhibitory concentration (MIC) of lactic acid in Vibrio parahaemolyticus were 1250 ppm at pH 6.5 and 7.0, 625 ppm at pH 6.0. respectively. The minimun inhibitory concentration (MIC) of hydrogen peroxide in Vibrio parahaemolyticus were 25 ppm at pH 6.5 and 7.0, 12.5 ppm at pH 6.0, respectively. MICs of combined treatment of lactic acid and hydrogen peroxide in Vibrio parahaemolyticus were 625 ppm of lactic acid with 12.5 ppm of hydrogen peroxide. The correlations between MICs of lactic acid and hydrogen peroxide in Vibrio parahaemolyticus were obtained through the coefficient of determination($R^2$). $R^2$ value were 1.0000. The antimicrobial effect of lactic acid and hydrogen peroxide in Vibrio parahaemolyticus could be confirmed from the result of this experiment.

  • Acute hepatopancreatic necrosis disease (AHPND), previously known as early mortality syndrome (EMS), is an emerging disease in shrimp caused by Vibrio parahaemolyticus. Some V. parahaemolyticus strains are associated with foodborne diseases in humans. To date, studies on the relationship between AHPND and pathogenic V. parahaemolyticus are very limited. In this study, we monitored the thermostable direct hemolysin-related hemolysin (trh) gene and AHPND-related genes, such as Photorhabdus insect-related (pir) genes, in 892 strains of V. parahaemolyticus isolated and identified in 24 areas of the West Coast of Korea from May to October 2019. The trh gene was detected in 9.6% of the isolates from short neck clam samples. However, the pirA and pirB genes related to AHPND were not found in any of the isolates despite using both duplex and nested PCR assays, suggesting that AHPND-related genes were nonexistent in the V. parahaemolyticus strains isolated. This study contributes to the current understanding of the relationship between AHPND and V. parahaemolyticus in Korea, as well as provides data on spatial and seasonal distributions of V. parahaemolyticus.

Food-borne outbreaks, distributions, virulence, and antibiotic resistance profiles of Vibrio parahaemolyticus in Korea from 2003 to 2016: a review

  • Background: Vibrio parahaemolyticus is one of the most common causes of seafood-borne illnesses in Korea, either directly or indirectly, by consuming infected seafood. Many studies have demonstrated the antibiotic susceptibility profile of V. parahaemolyticus. This strain has developed multiple antibiotic resistance, which has raised serious public health and economic concerns. This article reviews the food-borne outbreaks, distributions, virulence, and antibiotic resistance profiles of V. parahaemolyticus in Korea during 2003-2016. Main body: V. parahaemolyticus infections appeared to be seasonally dependent, because 69.7% of patient infections occurred in both August and September during 2003-2016. In addition, the occurrence of V. parahaemolyticus in marine environments varies seasonally but is particularly high in July, August, and September. V. parahaemolyticus isolated from aquaculture sources on the Korean coast varied in association with virulence genes, some did not possess either the tdh (thermostable direct hemolysin) or trh (tdh-related hemolysin) genes, and a few were positive for only the trh gene or both genes. The high percentage of ampicillin resistance against V. parahaemolyticus in the aquatic environment suggests that ampicillin cannot be used to effectively treat infections caused by this organism. Short conclusion: This study shows that the observed high percentage of multiple antibiotic resistance to V. parahaemolyticus is due to conventionally used antibiotics. Therefore, monitoring the antimicrobial resistance patterns at a national level and other solutions are needed to control aquaculture infections, ensure seafood safety, and avoid threats to public health caused by massive misuse of antibiotics.

Quantitative microbial risk assessment of Vibrio parahaemolyticus foodborne illness of sea squirt (Halocynthia roretzi) in South Korea

  • The annual consumption of fishery products, particularly sea squirt (Halocynthia roretzi), per person has steadily increased in South Korea. However, the quantitative risk of Vibrio parahaemolyticus following intake of sea squirt has not been analyzed. This study focuses on quantitative predictions of the probability of consuming sea squirt and getting of V. parahaemolyticus foodborne illness. The prevalence of V. parahaemolyticus in sea squirt was evaluated, and the time spent by sea squirt in transportation vehicles, market displays, and home refrigerators, in addition to the temperature of each of these, were recorded. The data were fitted to the @RISK program to obtain a probability distribution. Predictive models were developed to determine the fate of V. parahaemolyticus under distribution conditions. A simulation model was prepared based on experimental data, and a dose-response model for V. parahaemolyticus was prepared using data from literature to estimate infection risk. V. parahaemolyticus contamination was detected in 6 of 35 (17.1%) sea squirt samples. The daily consumption quantity of sea squirt was 62.14 g per person, and the consumption frequency was 0.28%. The average probability of V. parahaemolyticus foodborne illness following sea squirt consumption per person per day was 4.03 × 10-9. The objective of this study was to evaluate the risk of foodborne illness caused by Vibrio parahaemolyticus following sea squirt consumption in South Korea.

Preparation of Hetero-Chitooligosaccharides and Their Antimicrobial Activity on Vibrio parahaemolyticus

  • This study was performed to investigate the antimicrobial effects of hetero-chitosans and their oligosaccharides on the halophilic bacterium, Vibrio parahaemolyticus. Nine classes of hetero-chitosan oligosaccharides were prepared based on their molecular weights, using an ultrafiltration membrane reactor system with chitosanase and celluase, from partially different deacetylated chitosans, 90%, 75%, and 50% deacetylated chitosan, respectively. Thirty-two strains of V. parahaemolyticus were isolated from various marine organisms such as shellfish, shrimps, octopus, and seabirds. Seventy-five percent deacetylated chitosan showed the highest antimicrobial acitivity. The minimal inhibitory concentration (MIC) was 0.5 mg/ml on 14 strains of V. parahaemolyticus, and MIC of the rest strains (18 strains) was 1.0 mg/ml. In addition, MIC of most hetero-chitosan oligosaccharides was 8.0 mg/ml. The results revealed that the antimicrobial effects of hetero-chitosans and their oligosaccharides against V. parahaemolyticus depend on the degree of deacetylation, their molecular weights, and strains tested.

  • The author studied on the distribution of V. parahaemolyticus among sea water, sea mud, and various marine products in Jeju, Keoje, Namhae, Yockji, Busan, and Masan on the southern seas of Korea from winter in 1981 to summer in 1982. The author studies for the isolated strains to bacteriological identification Kanagawa phenomenon(hemolytic activity) on Modified Wagatuma blood agar plates and serotypes with anti V. parahaemolyticus. The results obtained were as follows:. 1. V. parahaemolyticus was isolated in 713 strains(28.3%) among 2519 total specimens of sea water, sea mud, and various marine products. 2. The isolation rates of V. parahaemolyticus in summer season were higher than in spring and winter season. Above results were 304 strains(32.6%) among 932 specimens in summer, 160 strains (28.1%) among 570 specimens in spring, 149 strains(14.6%) among 1017 specimens in winter, respectively. 3. The hemolysis on Modified Wagatuma agar added human erythrocytes was 66.0% of positive Kanagawa phenomenon, and was 34.0% of negative Kanagawa phenomenon, respectively. 4. The distributions of serotypes of V. parahaemolyticus isolated were from KI to K VIII of K pooling antisera. The results were 6 strains(6.6%) on K 1 of K I, 14 strains(l5.5%) on K 17 of K II, 26 strains(28.8%) on K 28 of K IV. 10 strains(11.1%) on K 32 of KV, 4 strains(4.4%) on K 39 of KV, 8 strains(8.8%) on K 42 of K VI, 2 strains(2.2%) on K 48 of K VII, 1 strain(1.1%) on K 50 of K II and 7 strains(7.7%) on K 55 of K VII, respectively. 5. V. parahaemolyticus was more frequently isolated from sea mud than sea water and various marine prdoucts in winter season. 6. There was no great difference as far as the distribution of V. parahaemolyticus concerned in Jeju, Keoje, Namhae, Yockji, Busan and Masan of the southern seas of Korea.

Pathogenic Vibrio spp. Isolated from the Gwangan Beach of Busan in 2003

  • A total of 52 pathogenic Vibrio strains was isolated from the Gwangan Beach during summer in 2003. The isolated vibrios were composed of 6 different species: V. parahaemolyticus, V. cholerae non O1, V. fluvialis, V. vulnificus, V. alginolyticus, and V. mimicus. V. parahaemolyticus was most predominant as $46\%$ (24/52), V. cholerae non O1 was the second with $23\%$ (12/52), and V. fluvialis was the third with $17\%$ (9/52). Among the isolated strains, 22 strains showed hemolytic, proteolytic or ureolytic activity. Eight strains showed both hemolysin and protease activities, and either 6 strains showed only hemolysin activities and 7 strains only protease activities. Only one strain of V. parahaemolyticus isolates showed urease activity. The urease-positive V. parahaemolyticus strain (V. parahaemolyticus S25) showed the same biochemical characteristics as the reference strain, V. parahaemolyticus KCTC 2471 (urease­negative) except for urease production. To compare the degree of virulence of Vibrio strains having different pathogenic factors, hemolysin, protease, or urease-positive strains were injected into groups of 10 each of ICR mice (7- to l0-week-old male). The lethal rate of urease-positive V. parahaemolyticus S25 was significantly high, being $70\%$. Protease-positive strains showed $40-60\%$ of lethal rate. Hemolysin-positive strains showed no mortality, similar to non-pathogenic V. parahaemolyticus KCTC 2471 and V. parahaemolyticus FM12.

MPN 및 H-NS 유전자를 표적으로 하는 PCR assay를 병용한 장염비브리오(Vibrio parahaemolyticus)의 정량 (Quantification of Vibrio parahaemolyticus Using a Most Probable Number-Polymerase Chain Reaction Assay Targeting the H-NS gene)

  • We applied a combination of most probable number-polymerase chain reaction (MPN-PCR) methods using a PCR procedure targeting the H-NS (VP1133) gene to detect Vibrio parahaemolyticus presence and density in seawater as well as within short-necked clam Ruditapes philippinarum tissues collected from Gomso Bay, Korea. In 30 seawater samples, V. parahaemolyticus levels ranged from less than 1.8 to $1.1{\times}10^3MPN/100mL$, and samples from August showed higher than those from other months. Furthermore, the levels of V. parahaemolyticus in six short-necked clam samples ranged from $7.8{\times}10^2$ to $2.1{\times}10^3MPN/100g$, approximately 2.5 times higher than in seawater samples from the corresponding month. Our results provide data on V. parahaemolyticus contamination in seawater and short-necked clam tissues, and help to improve quantitative methods of assessing V. parahaemolytcius levels.

비브리오 속의 단백질 분해능에 관한 연구 (Study on the Proteolytic Activities of Pathogenic Vibrio sp.)

  • V. parahaemolyticus 종류들이 gelatin분해 확인 실험에서 큰 clear zone을 형성했고, V. alginolyticus 분리균주도 V. parahaemolyticus와 비슷한 clear zone을 형성했다. V. fluvialis KCTC 2473은 clear zone을 형성하지 못했다. Hemolytic activity측정 실험에서는 V. parahaemolyticus 분리균주(2)가 가장 활성이 크게 나타났고, V. parahaemolyticus ATCC 17802, V. parahaemolyticus 분리균주(1)과 V. mimicus ATCC 33653도 높은 활성을 나타냈으며, 나머지 다른 비브리오균은 활성이 나타나지 않았다. Azocasein분해 활성 실험에서는 V. parahaemolyticus ATCC 17802, 분리균주(1), 분리균주(2)는 200EU/m1전후로 활성이 높게 나타났고, 나머지는 110EU/ml전후로 활성이 상대적으로 낮게 나타났다.
