작약추출물의 식품변패미생물에 대한 항균특성 (Antimicrobial Characteristics of Paeonia lactiflora Pall. Extract Tested against Food-putrefactive Microorganisms)

  • 한국산 작약(Paeonia lactiflora Pall.) 뿌리를 사용하여 작약추출물(PLE)을 조제 후, 변패미생물에 대한 작약추출물의 항균력을 측정하였다. Paper disk 확산법에 의한 농도별 항균력 검사에서는 작약추출물이 농도에 비례하는 항균력을 나타내었다. 생육저해곡선의 측정에서는 50 ${\mu}g$/mL이상에서 미생물의 생육이 완전히 억제되는 것을 확인하였다. 열($100^{\circ}C$에서 45분처리) 및 pH(3~11)의 넓은 범위에도 항균력을 보여, 열 및 pH에 대한 작약추출물에 포함된 항균력의 우수한 안정성을 확인할 수 있었다. 전자현미경(SAM, TEM)에 의한 미생물생태변화의 관찰에서도 작약추출물의 항균물질이 세포벽 또는 세포막의 삼투기능을 상실시켜 미생물의 생리가 중단되고 억제되는 것을 확인하였다. 그리고 ${\beta}$-galactosidase의 효소활성측정결과 작약추출물은 미생물의 세포막 기능파괴를 심하게 초래하는 것으로 예상할 수 있었다.

The root extract of Paeonia lactiflora Pall inhibits the oxidative damage via its anti-oxidant activity

  • Objectives : Reactive oxygen species (ROS) have been associated with pathogenic processes including carcinogenesis through direct effect on DNA directly and by acting as a tumor promoter. Therefore, it has been regarded that ROS may be a major target for cancer prevention. The root of Paeonia lactiflora pall (PL), a traditional Chinese herb, has been a component of effective prescriptions for treatment of liver disease. Also, there are some reports about the antioxidant activities of the extracts from PL. However, little has been known about the effects of PL against oxidative damage. This work aimed to elucidate the anti-oxidant effects of Paeonia lactiflora pall (PL) in the non-cellular system and cellular system. Methods : Antioxidant activities of PL were evaluated by hydroxyl radical scavenging assay and $Fe^{2+}$ chelating assay. Anti-oxidative effect of PL was evaluated by ${\varphi}X$-174 RF I plasmid DNA cleavage assay in non-cellular system. In addition, DNA migration assay, expression level of phospho-H2AX, MTT assay and lipid peroxidation assay were performed for evaluate the anti-oxidative effect of PL in cellular system. Results : PL had a dose-dependent hydroxyl radical scavenging and $Fe^{2+}$ chelating capacity. In addition, PL inhibited oxidative DNA and cell damage induced by hydroxyl radical in non-cellular system and cellular system. Conclusion : Taken together, P. lactiflora pall may be possible for the application to a potential drug for treating the oxidative diseases such as cancer.

작약근(芍藥根)의 열풍건조시(熱風乾燥時) 갈변(褐變)의 효과적(效果的) 억제(抑制) (Browning Inhibition of Paeonia lactiflora Root during Hot Air Dehydration)

  • 작약을 건조하는 동안 발생되는 갈변을 효과적으로 억제하기 위하여 본 연구를 수행한 결과는 다음과 같다. 1. 수분함량은 무처리와 연탄훈증 처리에서 시간 건조 후 각각 16.0, 16.2%로 높았고 산성 용액, 아황산염 용액, 및 blanching처리에서는 각각 13.5, 12.9, 14.8%로 낮았으며, 동결건조를 수행 한 시료에서는 18.8%로 가장 높았다. 2. 무처리 시료의 L, a, b, 및 ${\triangle}E$값이 61.60, 1.89, 10.20, 39.78로 갈변정도가 가장 심하였고, 연탄훈증과 아황산염 용액을 처리한 시료는 양호 하였으며 , 동결 건조를 수행한 시료는 전 건조기간 동안 갈변이 일어나지 않았다. 3. 작약의 paroniflorin은 연탄훈증과 아황산염 용액 처리에서 각각 2.41, 2.51%, 동결건조 처리에서 2.70%로 높게 나타났다. 4. $SO_{\2}$함량은 sulfites처리인 연탄훈증과 아황산염 용액 처리에서 0.63%로 차이가 없었는데 비하여 무처리와 동결건조 처리 보다는 각각 0.1%, 0. 17%가 높았다. 5. 작약 건조시 건조활동의 생력화 및 경제성, 건조시간, 상품성 등을 고려할 때는 건조온도 $60{\circ}C$에서 sulfite salts용액에 침지한 후 건조하는 것이 바람직 하지만 안전성의 측면에서 볼 때 작약 건조시 아황산 처리에 대한 안전사용기준 설정에 대한 충분한 연구가 수행되어야 할 것으로 사료된다.

Antioxidant Activity and Its Mechanism of Paeonia lactiflora Pall Extract

  • Paeonia lactiflora Pall (PL) has been used as a traditional herbal medicine in China, Korea, and Japan for more 1,200 years. PL has reported to have antioxidant activity and protective effect of cells from oxidative stress, although the mechanism has not been verified. FOXO3a is a transcription factor that binds to its target gene's consensus FOXO binding site. FOXO3a protein modulates the various biological functions including cell cycle control, apoptosis, DNA repair, and ROS detoxification. Therefore, FOXO3a activity is associated with cancer, aging, diabetes, infertility, neurodegeneration, and immune system dysfunction. Here we found that FOXO3a was activated by PL extract. Transcriptional target genes such as MnSOD, p27, and GADD45 were activated by PL extract. Protein levels of MnSOD and catalase were increased, consequently, ROS level was reduced in HEF cells by PL extract. These findings suggest that PL extract has an antioxidant activity through FOXO activation and thereby activation of FOXO target genes, MnSOD and catalase.

작약(Paeonia lactiflora Pall.)에서 paeoniflorin 추출방법 및 HPLC 분석조건 (Extraction Methods and HPLC Analysis Conditions of Paeoniflorin in Peony, Paeonia lactiflora Pall.)

  • 경북 영천군 고경면 작약 재배농가의 정식 4년차인 의성품종(Paeonia lactiflora Pall.)을 1993년 6월 17일 수확하고, 굵기가 27mm 이상인 뿌리를 선별하여 paeoniflorin 추출방법 및 HPLC 분석조건에 대한 조사결과를 요약하면 다음과 같다. 1. 초음파추출 및 환류추출법에 의한 작약근 추출물의 HPLC chromatogram은 상호간의 차이가 없었으며, 두 추출법 모두에서 paeoniflorin retention time이 10.2분대에 peak가 존재하였다. 2. 환류추출법에 의한 paeoniflorin 추출은 60min., 1회로 충분하였다. 3. 초음파추출에서 추출시간이 1hr.∼2hr. 까지의 paeoniflorin함량은 동일시간의 환류추출보다 각각 0.28%, 0.32% 낮게 평가되었고 3hr.∼4hr.의 추출에서는 두 방법간에 차이가 없었다. 4. 초음파 추출법은 분석작업이 용이함으로 다수계통의 상대적 비교시에는 환류추출법보다 효과적이다. 5. 작약추출물 중 paeoniflorin, albiflorin을 HPLC를 이용하여 분석할 때 높은 감도를 보이는 가장 효과적인 UV Detector 파장은 각각 240nm, 254nm이고, 동시정량시에는 254nm가 비교적 안정적이었다.

Anti-Inflammatory Effects of Paeoniflorin Derivatives

  • We previously showed that the root extract of Paeonia lactiflora might have anti-inflammatory effects. Paeoniflorin (PF) has been identified as one of the main bioactive components of Paeonia lactiflora, however its role has been well characterized. In this study, we tested whether PF and its derivatives, which is removed the hydroxy group from PF, might have anti-inflammatory effects. In the Nitric Oxide assay, PF and Paeoniflorin's derivative (PFD) showed 55% and 56% more anti-inflammatory effect, compared to LPS control, respectively at 250ug/ml. To further confirm, we examined the effect of PF on tyrosine phosphorylation of Erk MAP Kinase. It is well established that tyrosine phosphorylation of Erk MAP Kinase is related to NF-kB mediated inflammation pathway. We therefore examined whther PF and PFD might regulate Erk activity. PF and PFD showed 35% and 22% less tyrosine phosphorylation compared to Paeonia lactiflora Red Charm extract control, respectively at 500ug/ml. Taken together, these results suggest that PF and PFD may play a role in anti-inflammatory effects in the root extract of Paeonia lactiflora. This study will provide the basis to develop a platform for the inflammation-mediated diseases therapeutics in the near future.

작약(Paeonia lactiflora Pall.) 동아의 경정배양을 통한 기내증식 (In vivo propagation of Paeonia lactiflora Pall. Through Shoot-Tip Culture of Winter Buds)

  • 작약동아의 정아 및 액아의 기내배양에 의한 유묘의 증식에 필요한 배양조건을 구명코자 실험한 결과는 다음과 같다. 정아배양의 경우, 생장조절재의 조성에 무관하게 두지방종에서 100% 신초가 신장하였으나 생장은 의성지방종 은 NAA 0.1 mg/L 단용배지, 영천지방종은 NAA 0.01 mg/L 단용배지에서 가장 양호하였다. 액아배양의 경우, NAA 0.01 mg/L와 zeatin 5.0 mg/L 혼용배지에서 의성지방종은 100%, 영천지방종은 50%의 가장 높은 신초신장율을 보였고 신초의 생장도 양호한 경향이었다. 정아 및 액아배양으로부터 유도된 신초는 vermiculite를 지지물로 한 NAA 0.1 mg/L 첨가배지에서 30.0%의 발근율을 보였으나 뿌리의 생장은 양호하였다.

작약 촉성재배 시기가 절화와 뿌리품질 및 수량에 미치는 영향 (Influence of Forcing Cultivation Time on Cut Flower, Root Quality, and Yield in Peony (Paeonia lactiflora Pall. cv. Taebaek))

  • Three year-old peony (Paeonia. lactiflora Pall. cv. Taebaek) was cultivated in green-house at Jan. 15, Feb. 15, or Mar. 15, respectively. The mean of temperature during the forcing cultivation was higher (air; $1.0{\sim}11.1^{\circ}C$, soil; $1.1{\sim}7.4^{\circ}C$) than that of open-field condition. From sprouting to flowering in peony cultivated at Jan. 15 was about 54 days, which is shorted the cut flower periods (ca. 26 days) compared with the open-field cultivation. However, earlier forcing cultivars were very susceptible to pathogens such as powdery mildew or gray mold. The yield in green house was also lower than in the open-field cultivation. The content of bioactive compounds such as paeoniflorin and albiflorin in green-house cultivars was similar that of open-field cultivars. These results showed the forcing cultivation time of peony at Feb. 15 in green-house was most desirable for commercialization.

First Report of Botrytis Mold Caused by Botrytis cinerea on Peonies (Paeonia lactiflora Pall.)

  • In 2019, symptoms of Botrytis mold on the peony (Paeonia lactiflora Pall.) 'Sarah Bernhardt' were observed during a survey of the commercial greenhouses of Gangjin County, South Korea. The initial symptoms, small brown spots, were observed mainly at the leaf margins. The lesions extended to the interior of leaves forming irregular spots in which abundant conidia developed. Fungal colonies were obtained from surface-sterilized tissue excised from growing edges of the lesions that were transferred to potato dextrose agar. Melanized irregular sclerotia were formed in these colonies after 40 days at 8℃. Molecular phylogeny based on sequences of genes for glyceraldehyde-3-phosphate dehydrogenase, heat-shock protein 60, and RNA polymerase subunit II were highest for the PBC-2 isolate to the type strains of Botrytis cinerea, rather than other Botrytis species associated with peony diseases. Following Koch's postulates, healthy Sarah Bernhardt plants were inoculated with a foliar application of conidial suspensions of the isolate PBC-2. Following incubation under humidity with a 12 hr photoperiod for 7 days, symptoms developed on the leaf margins that were identical to those observed in the greenhouses. This study is the first report of Botrytis blight caused by B. cinerea on peonies grown in commercial greenhouses in South Korea.

2종(種) 작약류(芍藥類)의 내외부형태(內外部形態)와 패턴분석 연구(硏究) (A Study on Morphological and Pattern Analysis in 2 kinds of Paeoniae Radix)

  • The following is a taxonomic list of the specific features of external and internal shape and pattern analysis of Paeonia lactiflora PALL. and P. obovata MAX as the standard of herbal medicine. 1. External shape of original plant P. lactiflora has lancelate and elliptical leaves with no hair on both sides. It bears a flower bud at the shoot apex or leaf axil. On the other hand, P. obovata has obovate leaves with hair on the back side. It has a single flower bud at the shoot apex. 2. Physical characteristics of herbal states P. lactiflora has a bright and smooth external surface whose color is light red brown, light gray on its section and it is not easy to cut. On the other hand, P. obovata has a coarse external surface whose color is dark brown, pinkish on its section and it is easy to cut. 3. Physical characteristics of herbal state in currents Paeonia Radix Alba is brown on the external surface, and whitish on its section. On the other hand, Paeonia Radix Rubra is dark brown on the external surface, and yellowish-white on its section. 4. Internal characteristics Internal shape is correspond to that of Paeonia Radix described in literatures. Only P. lactoflora has a concentrated vascular bundle toward centeral cylinder. On the other hand, P. obovata has a scattered vascular bundle at the cortex. 5. Physicochemical pattern by HPLC Both P. lactoflora and P. obovata showed the same pattern. Paeoniforin, a main component, appears upon 13.68 in retention time (Rt) in both. In addition, both contain a large amount of paeoniflorin. Such results will, in the future, be used as basic source for the additional research, such as biological reactions and genetic discrimination.
