실규모의 고도처리공정에서 NADH변화 유형과 이를 활용한 공정제어 (NADH Variation and Process Control with NADH Fluorometer in Full Scale Biological Nutrient Removal Process)

  • In this study, we investigated the possibility of auto control and the proper operating factors in the BNR(Biological Nutrient Removal) process using an NADH(Reduced Nicotinamide Adenine Dinucleotide) fluorometer, which characterized the emitted fluorescence when activated by flashes of UV light at 460 nm. In terms of finding adequate operating parameters, results indicted that nitrification efficiency decreased in the controlled DO while denitrification efficiency decreased in the controlled pH. The above results indicated that controlled operating condition after combination with NADH, DO and pH was resonable. Result obtained from the correlation between NADH and pH showed that variation trend of influent loading was similar to those of NADH and pH, and also the variation cycle was repeated on a daily basis. Consequently, this result showed the increase of BOD loading caused the nitrification efficiency to decrease because air-flow, required for nitrification, was reduced, and so the NADH value was increased. From these results, it is possible to use NADH flourimetry to assess the variation of organic load and nitrification efficiency in the case of small change in influent pH such as in sewage and also to handle and operate the load variation in the auto control system using the NADH fluorometer.

  • This study was peformed to understand permeability characteristic of clays with various pH values. A serious of physical properties and permeability tests, consolidation tests was performed on a clay and a marine clay. Results of the study are as follows. As pH values were decreased, coefficient of permeability was increased because of increase in effective void caused by decrease in thickness of diffuse double layer besides change in soil structures and effective grain size. As pH values were increased, coefficient of permeability of marine clay was increased. Variation of coefficient of permeability of marine clay was increased. Variation of coefficient of permeability of marine clay was largely related because of higher clay contents.

pH 변화와 인산 제한이 Closoidium acetobutylicum의 연속발효에 미치는 영향 (Effects of p variation and phosphate limitation on the continuous fermentation of Clostridium acetobutylicum)

  • Clostridium acetobutyloicum KCTC 1037의 용매 생성조건을 알기 위하여 pH변화와 인산제한의 조건 하에서 회분 배양과 연속 배양을 실시하였다. C. acetobut-ylicum KCTC 1037은 pH5.5나 6.5에서 보다 pH 4.5에서 잘 자라는 호산성이었으며, pH4.5의 진리되지 않은 부티르산이 약 20mM인 조건에서 용매 발효로 전환되었으나, pH5.5의 전리되지 않은 부티트산이 17mM인 조건에서는 용매 발효가 일어나지 않았다. 인산 제한 조건에서 배지에 인산이 KH$_2PO_4$$K_2HPO_4$0.1g/l이하인 경우 용매 생성량이 증가하였으나, 생육 속도에는 영향을 미치지 않았다. 인산제한 이단 연속배양에서 pH5.5인 stage1에서는 산생성기가 유지 되었고 한편 pH4.5인 stage2에서는 초기에는 용매가 생성 되었으나 150시간 이후에는 용매의 양이 감소되었으며 산생성기로 전환되는 등 불규칙한 발효 양상을 보였다. 또 pH5.0, 희석율 $0.05h^{-1}$인 stage2에서 전리되지 않은 유기산이 36mM인 조건에서도 용매 발효가 일어나지 않았다. 이러한 결과로서 C.acelobutylicumd의 용매 생성 조건은 산성 pH나 인산 제한 조건외에 균의 최대 에너지 생성을 유지하려는 대사와 관련 되었을 것으로 추측하였다.

ORP와 pH 측정에 의한 간헐폭기 고도처리 공정 제어의 평가 (State Evaluation of Nutrient Removal in an Intermittent Aeration Process by Monitoring ORP & pH)

  • In this study, to evaluate the applicability of ORP and pH as process control parameters for an intermittent aeration process, a 200L/d bench-scale plant was installed and operated for 90 days. It was fed with synthetic wastewater which contained $COD_{Cr}$ : 400mg/L, TN : 40mg/L and TP : 7mg/L. ORP & pH were measured on-line and compared with the variation of nutrient concentrations. As the results, both of the ORP and pH were able to monitor successfully nitrification and denitrification. Bending-points on the ORP curve and peak points on the pH curve corresponded to the termination of nitrification and denitrification. For P uptake and release, pH was the best indicator for performance evaluation. The aerobic pH apex was appeared when P uptake was accomplished and there was a relationship between the P release and pH variation. But the pH curve needed filtering because there were many noises on it. In this study, the shape of the ORP & pH curves were varied as the operating conditions such as aeration rates and organic loads were changed. It allowed the operating states of biological systems to be effectively evaluated. If it is properly managed to show the bending points and peak points clearly, the on-line monitoring of ORP & pH will be a reliable and effective technique for process control of intermittent aeration processes.

김치숙성중 숙성온도, pH 및 소금 농도가 니트로스아민의 생성에 미치는 영향 (Effect of Temperature, pH and Salt Concnetration on formation of N-nitrosamines during Kimchi Fermentation)

  • 김치숙성중 숙성온도, pH 및 소금농도가 NDMA의 생성량에 미치는 영향과 또한 in vitro에서 온도, pH 및 소금농도가 NDMA의 생성에 미치는 영향을 각각 연구하였다. 배추김치를 저온 냉장온도($4^{\circ}C$)와 실온 ($16{\pm}2^{\circ}C$)에서 6주간 숙성할 때 아질산염과 DMA의 함량변화는 실온에서는 숙성기간에 따라 일정하지 않았으나 저온 냉장 온도에서는 숙성기간 동안 큰 변화 없이 거의 일정하였고,NDMA의 최대 생성량도 $2.69\;\mu\textrm{g}/kg$으로 저온 냉장 때 보다 약 18배 이상 더 많이 생성되었다. 김치 숙성 중 아질산염과 DMA의 함량변화와 NDMA의 생성량과는 관계가 없었으나 pH의 변화는 NDMA 생성량과 같은 관계가 있어 김치 숙성 중 가장 낮은 pH 4.0 부근에서 NDMA의 생성량도 가장 높았다. in vitro 시험에서 NDMA의 생성량은 저온보다는 실온에서 그리고 pH가 낮을수록 더 많이 생성되었고 그 생성량은 온도보다는 pH의 영향이 더 컸다. 보통 소금농도(2.5%)의 김치와 낮은 소금농도(1.5%)의 김치를 각각 숙성할 때 NDMA 의 생성량 변화는 보통 소금농도에서는 숙성기간동안 큰 변화 없이 일정하였고 최대 생성량도 $5.86\;\mu\textrm{g}/kg$으로 낮았으나, 낮은 소금 농도에서는 3주째 최고 $90.0\;\mu\textrm{g}/kg$ 생성되었다. in vitro에서도 소금농도가 높을수록 NDMA의 생성량도 낮았다.

전기화학적 방법에 의한 HRP의 최적 pH 도출 (Electrochemical Determination of the Optimum pH of HRP)

  • A carbon paste electrode was constructed with peroxidase extracted from Horseradish and the variation of the response of the sensor with pH was investigated. Current profiles showed two highest sensitivities at two pH values respectively. In addition, two bands were observed in the electrophoretic expansion. A coincidence of the two experimental results added support to the possibility that the biosensor has two different isozymes. Assuming that current profiles are the sum of two gaussians, we deconvoluted them and determined the optimum pH of peroxidase isozymes.

  • In order to investigate quantitative behavior of galvanic corrosion in the weld joint between stainless steel and carbon steel, electrochemical polarization experiments are performed for various pH, water temperature with boric acid concentration 4000ppm. The corrosion rate of stainless steel was almost independent of the variation of pH. The significant corrosion rates of carbon steel and the weld joint of carbon-carbon steel were observed at pH 4. The corrosion rates in the weld joint of the carbon-stainless steel were observed depending on the variation of pH level.

천연염료에 관한 연구(7) -코치닐색소의 견섬유에 대한 염색성- (Studies on the Natural Dyes(7) -Dyeing properties of cochineal colors for silk fibers-)

  • In order to study the properties of cochineal color, variation of uv, visible spectra by pH, dyeing properties on the silk in several dyeing conditions and thermodynamic parameter were investigated. Cochineal colors had an unusual to pH, especially had instability in alkali condition. An increase in the dyeing temperature and in time resulted in an increase in the dye content of silk fibers. Concentration of cochineal color in the silk fiber was related to pH and the maximum exhaustion of cochineal colors showed at about pH 3. The value of apparent diffusion coefficients and standard affinities of dyeing increased with the increase of dyeing temperature. The standard heats of dyeing(ΔH°), variation of entropy(ΔS°) and activation energy(E/sub act/) were caculated to be -1.72kcal/mo1, -3.77cal/mo1ㆍdeg and 1.26kcal/mo1, respectively. Silk fabrics were dyed bright red by tin chloride, reddish purple by copper sulfate, and bluish gray by iron sulfate, respectively. Lightfastness of silk fabrics mordanted by metal ion was weak.

한국산 퉁가리속 어류의 유전 및 형태적 변이에 관한 연구 (Genetic and Morphological Variation of the Genus Liobagrus in Korea)

  • 한국산 퉁가리속 어류의 종간및 종내 집단간의 유전적 근연관계와 변이정도를 알아보기 위하여 남한의 6개 수계의 7개 지역에서 채집된 420개분 표본을 대상으로 전기영동에 의한 유전자분석과 형태비교를 실시하였다. 집단간 유전적 변이정도를 분석한 결과 L. andersoni 3개 집단에서는 P=21.3%, H=0.068이였고 L. mediadiposalis 의 경우는 P=16.3%, H=0.056으로 나타났다. 또 L.andersoni 및 L. mediadiposalis 의 집단간 유전적 평균근연치는 각기 S=0.592, S=0.675이었고 그 중 금강집단이 가장 근연치가 먼 것으로 나타났으며 종간 근연치는 S=0.530이었다. 외부형태형질에 대한 discriminant function analysis를 실시한 결과 L. andersoni 의 금강 집단이 뚜렷하게 구분되어 전기영동결과와 일치함을 보였다. 이상의 결과에서와 같이 종내 집단간에서도 현저한 유전적 차이를 나타내고 었어 같은 수 계안의 집단간에 대하여서도 더욱 자세한 유전적, 형태적 비교분석이 있어야 할 것으로 사료된다.

니트로페놀화합물의 호기성생물분해시 pH 영향에 관한 연구 (pH Effect on the Aerobic Biodegradation of Nitrophenolic Compound in SBR)

  • Dinitrophenol is preventing cells from making energy for growth and it has been suggested that pH may be important in mitigating effects of uncouplers. The effect of pH on toxicity of dinitrophenol at high concentration was investigated, over a pH range of 5.7 to 8.7. DNP inhibition was found to be strongly dependent on mixed liquor pH. The DNP degradation rate was highest in the pH range of 7.0 to 7.8; at pH 6.0 degradation of 0.41 mM dinitrophenol was significantly inhibited; at pH <5.7, dinitrophenol degradation was completely inhibited after approximately 25% of the dinitrophenol was degraded. However no significant effect of pH variation was seen on glucose uptake by the activated sludge mixed culture.
