ANALYSIS OF FLUIDIC BEAD CUBE EMBEDDED PORTABLE CMOS SENSING SYSTEM FOR IMMUNO REACTION MONITORING (유체소자가 집적화된 면역검사용 휴대용 CMOS 바이오칩의 분석)

  • This paper describes the novel immunoassay sensing system for a portable clinical diagnosis system. It consists of a bead cage reactor and a CMOS integrated biosensor. It showed the simple and easy antibody coating method on beads by flow-through avidin biotin complex technology in a microfluidic device. It showed just 90 nL sample consumption and good result for the application of alpha feto protein. The bead cage reactor has the role of the antibody coating, antigen binding and enzyme linking for the electrochemical sensing method. The CMOS biosensor consists of ISFET (ion selective field effect transistor) biosensor and temperature sensor for detecting pH that is the byproduct of enzyme reaction. The sensitivity is 8 $kHz/^{\circ}C$ in a temperature sensor and 33 mV/pH in a pH sensor. After filling the 15 um polystyrene beads in bead cage, antibody flowed and reacted to beads. Subsequently, the biotinylated antigen flowed and bound to the antibody and GOD (glucose oxidase)-avidin conjugate flowed and reacted to the biotin of the biotinylated antigen. After this reaction process, glucose solution flowed and reacted to the GOD on beads. The hydrogen was generated by glucose-GOD reaction. And it was detected by the pH sensor.

Time-course Analysis of Biofilm Formation in Quorum Sensing-deficient Bacteria (Quorum sensing 결핍 세균에서 생물막 형성의 시간적 추이 분석)

  • Pseudomonas aeruginosa and Vibrio vulnificus are Gram-negative human pathogens, which exert their virulence through quorum sensing (QS) regulation. The infection of these pathogens have been known to be mediated by biofilm formation in many cases and this study carried out the time-course analysis of biofilm formation depending on the QS regulation in P. aeruginosa and V. vulnificus. In P. aeruginosa, our results demonstrated that QS-deficient mutant better attached to surface at initial stage of biofilm formation, but poorly proceeded to the maturation of the biofilm structure, while wild type less attached at initial stage but developed highly structured biofilm at late stage. Because of this, the quantitative comparison of biofilm formation between wild type and the QS mutant showed the reversion; the QS mutant formed more biofilm until 10 h after inoculation than wild type, but wild type formed much more biofilm after 10 h than QS mutant. V. vulnificus has been reported to form more biofilm with the mutation on QS system. When we performed the same time-course analysis of the V. vulnificus biofilm formation, the reversion was not detected even with prolonged culture for 108 h and the QS mutant always forms more biofilm than wild type. These results indicate that the QS regulation negatively affects the attachment at early stage but positively facilitates the biofilm maturation at late stage in P. aeruginosa, while the QS regulation has a negative effect on the biofilm formation throughout the biofilm development in V. vulnificus. Based on our results, we suggest that the developmental stage of biofilm and bacterial species should be considered when the QS system is targeted for biofilm control.

  • $Si_3N_4$ 박막을 동일한 공정 파라메터로 저압 화학기상증착법(LPCVD)으로 증착하고, IS, LOCOS- IS 및 ISFET의 세 가지 각각 다른 구조로 하여 용약 중 pH 농도 감지용 센서소자를 제작하였다. 이 세 가지 다른 센서소자에 대하여 pH 농도변화에 따른 감지도, 감지특성곡선의 선형성, 히스테리시스 등 주요 특성을 각각 조사한 후 비교 분석하였다. LOCOS-IS 구조의 pH 센서는 ISFET 구조의 pH 센서와 유사한 우수한 제반 pH 감지특성을 보였으나, 간단한 IS 구조의 pH 센서는 이들에 비해 상대적으로 열악한 pH 감지특성을 보였다. 동일공정으로 제작된 Si3N4 박막으로 제작되었음에도 불구하고 간단한 IS 구조의 pH 센서의 비교적 열악한 특성을 보이는 원인을 규명하기 위하여, pH 농도 변화에 따른 C-V특성 변화에 의한 pH 감지특성 조사시의 IS 및 LOCOS-IS 구조의 정전용량의 변화를 비교하고 고찰하였다.

Disposable Solid-State pH Sensor Using Nanoporous Platinum and Copolyelectrolytic Junction

  • A disposable solid-state pH sensor was realized by utilizing two nanoporous Pt (npPt) electrodes and a copolyelectrolytic junction. One nanoporous Pt electrode was to measure the pH as an indicating electrode (pH-IE) and the other assembled with copolyelectrolytic junction was to maintain constant open circuit potential ($E_{oc}$) as a solid-state reference electrode (SSRE). The copolyelectrolytic junction was composed of cationic and anionic polymers immobilized by photo-polymerization of N,N'-methylenebisacrylamide, making buffered electrolytic environment on the SSRE. It was expected to make. The nanoporous Pt surrounded by a constant pH excellently worked as a solid state reference electrode so as to stabilize the system within 30 s and retain the electrochemical environment regardless of unknown sample solutions. Combination between the SSRE and the pH-IE commonly based on nanoporous Pt yielded a complete solid-state pH sensor that requires no internal filling solution. The solid state pH sensing chip is simple and easy to fabricate so that it could be practically used for disposable purposes. Moreover, the solid-state pH sensor successfully functions in calibration-free mode in a variety of buffers and surfactant samples.

Poly(Dimethylaminoethyl Methacrylate)-Based pH-Responsive Hydrogels Regulate Doxorubicin Release at Acidic Condition

  • Stimuli-responsive biomaterials that alter their function through sensing local molecular cues may enable technological advances in the fields of drug delivery, gene delivery, actuators, biosensors, and tissue engineering. In this research, pH-responsive hydrogel which is comprised of dimethylaminoethyl methacylate (DMAEMA) and 2-hydroxyethyl methacrylate (HEMA) was synthesized for the effective delivery of doxorubicin (Dox) to breast cancer cells. Cancer and tumor tissues show a lower extracellular pH than normal tissues. DMAEMA/HEMA hydrogels showed significant sensitivity by small pH changes and each formulation of hydrogels was examined by scanning electron microscopy, mechanical test, equilibrium mass swelling, controlled Dox release, and cytotoxicity. High swelling ratios and Dox release were obtained at low pH buffer condition, low cross-linker concentration, and high content of DMAEMA. Dox release was accelerated to 67.3% at pH 5.5 for 6-h incubation at $37^{\circ}C$, while it was limited to 13.8% at pH7.4 at the same time and temperature. Cell toxicity results to breast cancer cells indicate that pH-responsive DMAEMA/HEMA hydrogels may be used as an efficient matrix for anti-cancer drug delivery with various transporting manners. Also, pH-responsive DMAEMA/HEMA hydrogels may be useful in therapeutic treatment which is required a triggered release at low pH range such as gene delivery, ischemia, and diabetic ketoacidosis.

Self-assembled Micelle-based Fluorescence Sensor for Extremely Acidic pH Range (강산성 용액의 pH를 측정할 수 있는 미셀기반의 형광센서 개발)

  • In this study, an effective fluorescence pH sensor based on conjugated polyelectrolyte micelles (CPMs) was devised for detecting extremely acidic conditions. An amphiphilic coumarin derivative (CC12-N), a building block, was prepared, into which an ionizable amino group, aryl amine, was incorporated as a potential hydrophilic moiety. This monomer displays self-assembled micelle formation in extremely acidic pH ranges, giving a hydrophobic π-extended conjugated system at the inner part and hydrophilic functionality at the periphery, resulting in efficient fluorescence intensity enhancement. This new micelle-based fluorescence provides an efficient sensing platform for detecting very low pH values in the presence of competing substances.

  • The measurement of pH is of significant importance in chemistry, life sciences, and environmental monitoring. Unlike conventional pH sensors that utilize glass electrodes, thin-film transistor (TFT)-based pH sensors offer distinct advantages, including enhanced response speed and additional circuit functions. In this study, we developed a pH sensor that incorporates biocompatible parylene-C as both the substrate and sensing layer, thereby enhancing flexibility, transparency, and biological compatibility. We conducted tests to measure the voltage-current characteristics of the pH solutions and assessed their performance in terms of drift and hysteresis. Using InGaZnO (IGZO) as the channel material, our pH sensor demonstrated an average sensitivity of approximately 82 mV/pH, albeit with certain drift limitations. The initial pH measurements exhibited good reversibility over time. IGZO- and parylene-C-based TFT pH sensors are well suited for various applications, including wearable health monitoring, owing to their flexibility and biocompatibility.

Enhanced pH Response of Solution-gated Graphene FET by Using Vertically Grown ZnO Nanorods on Graphene Channel

  • We observe enhanced pH response of solution-gated field-effect transistors (SG-FET) having 1D-2D hybrid channel of vertical grown ZnO nanorods grown on CVD graphene (Gr). In recent years, SG-FET based on Gr has received a lot of attention for biochemical sensing applications, because Gr has outstanding properties such as high sensitivity, low detection limit, label-free electrical detection, and so on. However, low-defect CVD Gr has hardly pH responsive due to lack of hydroxyl group on Gr surface. On the other hand, ZnO, consists of stable wurtzite structure, has attracted much interest due to its unique properties and wide range of applications in optoelectronics, biosensors, medical sciences, etc. Especially, ZnO were easily grown as vertical nanorods by hydrothermal method and ZnO nanostructures have higher sensitivity to environments than planar structures due to plentiful hydroxyl group on their surface. We prepared for ZnO nanorods vertically grown on CVD Gr (ZnO nanorods/Gr hybrid channel) and to fabricate SG-FET subsequently. We have analyzed hybrid channel FETs showing transfer characteristics similar to that of pristine Gr FETs and charge neutrality point (CNP) shifts along proton concentration in solution, which can determine pH level of solution. Hybrid channel SG-FET sensors led to increase in pH sensitivity up to 500%, compared to pristine Gr SG-FET sensors. We confirmed plentiful hydroxyl groups on ZnO nanorod surface interact with protons in solution, which causes shifts of CNP. The morphology and electrical characteristics of hybrid channel SG-FET were characterized by FE-SEM and semiconductor parameter analyzer, respectively. Sensitivity and sensing mechanism of ZnO nanorods/Gr hybrid channel FET will be discussed in detail.

The Studies for the Malate Tissue Biosensor Using Malate Dehydrogenase(Decarboxylating) in the Bundle Sheath Cell of the Corn Leaf (옥수수잎의 유관속초세포내에 들어 있는 Malate Dehydrogenase(Decarboxylating)을 이용한 Malate 측정용 조직바이오센서에 관한 연구)

  • A biosensor for the measurement of malate has been constructed by the sodium-alginate immobilized bundle sheath cell tissue of corn leaf containing malate dehydrogenase (decarboxylating) (EC 1. 1. 1. 40) on the CO2 gas-sensing electrode. The proposed tissue sensor had the linear in the range of malate concentration $5.5{\times}10^{-5}M∼2.5{\times}10^{-2}M$ with a slope of 53.5 mV/decade in 0.02M Tris-HCl buffer solution at optimum pH 8.0, and $25^{\circ}C$. A response time was 16∼18min. The present L-malate sensing tissue sensor is stable for more than one week. At pH 7.4, Km value was $0.6{\times}10^{-5}M$. The various kinds of salt did not effect the signal of malate tissue biosensor as the inhibitor. We can measure the malate by the CO2 electrode at the pH=8.0. Thus, the proposed tissue sensor will be useful for the measurement of malate.

Evaluation of DoP-CPD Classification Technique and Multi Looking Effects for RADARSAT-2 Images

  • This paper give further assessment on the original DoP-CPD classification scheme. This paper provides some additional comparative study on the DoP-CPD with H/A/alpha classifier in terms of multi look effects and classification performances. The statistics and multi looking effects of the DoP and CPD were analyzed with measured polarimetric SAR data. DoP-CPD is less sensitive to the number of averaging pixels than the entropy-alpha technique. A DoP-CPD diagram with appropriate boundaries between six different classes was then developed based on the data analysis. A polarimetric SAR image DoP-CPD classification technique is verified with C-band polarimetric RADARSAT-2 images.
