Correlation of Gene Expression between Adiponectin and Glucose Transporter 4 in Mouse Adipose Tissue (생쥐 지방조직에서의 아디포넥틴과 포도당수송체-4 유전자 발현의 상관관계)

  • Adiponectin has been known to improve insulin sensitivity and elicit glucose uptake via increased glucose transporter 4 (GLUT4) translocation. In the current study, mRNA expression levels of adiponectin and GLUT4 were measured in subcutaneous adipose tissue from C57BL/6 mice fed normal (ND) or high-fat diet (HFD) until 16, 26, 36, 47, or 77 weeks of age starting from 6 weeks of age. Expression levels were also measured in mice with calorie restriction (CR) and in thiazolidinedione (TZD) treated mice. Using quantitative real-time PCR, we demonstrated that GLUT4 expression in adipose tissue significantly decreased in HFD mice groups and increased in CR (p<0.05) and TZD (p=0.007) groups while there was no difference in adiponectin mRNA expression levels between experimental and control groups. General linear regression models were used to assess the association of gene expression levels between adiponectin and GLUT4 and to determine whether adiponectin affects GLUT4 transcription. mRNA expression levels of adiponectin and GLUT4 are significantly associated each other in mice fed a ND (p<0.0001) or HFD (p<0.0001), in groups separated into each age and diet, and CR group (p=0.002), but not in TZD group (p=0.73). These results demonstrated that gene expression of adiponectin and GLUT4 is strongly associated, suggesting that there is a common regulatory mechanism for adiponectin and GLUT4 gene expression and/or adiponectin has a direct role in GLUT4 gene expression in adipose tissue.

Risk Factors Affecting the Graft and Patient Survival in Kidney Transplant Patients (신이식환자에서 이식과 환자의 생존에 영향을 끼치는 위험요인 분석)

  • Purpose: To determine the short (1 year of transplant) and long-term (1-5 years of transplantation) risk factors affecting the graft and patient survival in kidney transplantation recipients. Methods: Records of 149 patients who received kidney transplantation in 1996 from Asan Medical Center were followed for 5 years retrospectively. Results: All patients initiated triple immunosuppressive therapy with cyclosporine, prednisone and azathioprine. One, two, three, four, five year patient and graft survival rates were 98.7%, 98.0%, 98.0%, 97.3%, 97.3%, and 96.6%, 95.2%, 94.6%, 92.5%, 91.8%, respectively. There were 30 cases of acute rejection (AR) and 6 cases of chronic rejection (CR) within $2.1{\pm}3.2$ months and $42.1{\pm}13.2$ months of transplantation, respectively. The risk factors for AR were donor's age older than 30 years (p=0.02) and cardiovascular disease (p=0.05). The risk factors for CR were AR (p=0.0169) and episode of complications (p=0.0330). Increasing period of dialysis (p=0.0473), episodes of AR (p<0.0001) and complication (p=0.0317) were significant factors for graft loss. Seven grafts were lost from noncompliance during 1-5 year period. The most com- mon cause of the graft loss for both periods was the graft rejection. The graft survival rate was significantly lower in patients with than without rejection episodes (77.4% vs. 90.0%, p=0.002). Conclusions: Survival rate of the graft with rejection was significantly lower. The risk factors affecting AR were donor's age older than 30years and CVD. AR and episode of complications within 1year were the risk factors for CR and graft loss.

Optimization of coagulant dosage using response surface methodology with central composite design (반응표면분석법-중심합성계획을 이용한 최적 응집제 주입량 산정 연구)

  • The determining the appropriate dosage of coagulant is very important, because dosage of coagulant in the coagulation process for wastewater affects removing the amount of pollutants, cost, and producing sludge amount. Accordingly, in this study, in order to determine the optimal PAC dosage in the coagulation process, CCD (Central composite design) was used to proceed experimental design, and the quadratic regression models were constructed between independent variables (pH, influent turbidity, PAC dosage) and each response variable (Total coliform, E.coli, PSD (Particle size distribution) (< $10{\mu}m$), TP, $PO_4$-P, and $COD_{cr}$) by the RSM (Response surface methodology). Also, Considering the various response variables, the optimum PAC dosage and range were derived. As a result, in order to maximize the removal rate of total coliform and E.coli, the values of independent variables are the pH 6-7, the influent turbidity 100-200 NTU, and the PAC dosage 0.07-0.09 ml/L. For maximizing the removal rate of TP, $PO_4$-P, $COD_{cr}$, and PSD(< $10{\mu}m$), it is required for the pH 9, the influent turbidity 200-250 NTU, and the PAC dosage 0.05-0.065 ml/L. In the case of multiple independent variables, when the desirable removal rate for total coliform, E.coli, TP, and $PO_4$-P is 90-100 % and that for $COD_{cr}$ and PSD(< $10{\mu}m$) is 50-100 %, the required PAC dosage is 0.05-0.07 ml/L in the pH 9 and influent turbidity 200-250 NTU. Thus, if the influent turbidity is high, adjusting pH is more effective way in terms of cost since a small amount of PAC dosage is required.

A Study on Cochineal Dyeing by Various Mordants and pH Conditions I -Treatment on Cotton Fabric- (염욕의 pH와 매염제의 변화에 따른 코치닐의 염색성 연구 I -면직물을 중심으로-)

  • In this study, cotton fabric specimens were pre-mordanted using Sn, Al, Cu, Cr, and Fe, and subsequently dyed using cochineal, maintaining the pH of the dye bath constant using pH buffer solutions of 4, 5, 6, 7, and 8. In the cases of Al, Cr, and Sn mordanting, the dye-uptake was well accomplished in the acidic range of pH 4 and 5. However, the dye-uptake was not accomplished in the range over pH 6. In the cases of Cu and Fe mordanting, however, the pH value did not affect the dye-uptake and the dyeability in the alkaline range was not decreased remarkably. In the case of non-mordant, the dye-uptake was not accomplished at all, and the Cu and Fe mordanting gave rise to the best dyeability. In the cases of non-mordanting and Al, Cu mordanting, there was no change in the $\lambda_{max}$, and in the cases of Sn and Cr mordanting, the $\lambda_{max}$ moved toward the shorter wavelength. In the case of Fe mordanting only, the $\lambda_{max}$ moved gradually toward the longer wavelength, and due to the pH effect deep-dyeing effect was revealed.

Sewage Treatment using Membrane Bioreactor(MBR) and Reverse Osmosis(RO) Process (Membrane bioreactor(MBR)과 Reverse osmosis(RO) 공정을 이용한 하수처리)

  • The objective of this study was to investigate the effect of hydraulic retention time (HRT) on removal efficiencies of organic matter, nitrogen and phosphorus in MBR-RO process for treating synthetic sewage. In MBR process, turbidity was less than average 2 NTU and average removal efficiency showed more than 99% during the operation period(MBR 105 day). As a result of HRT variation, average removal efficiencies of $COD_{Cr}$ on HRT 6, 12, 18 and 24hour were about 72.4, 84, 88.6 and 92.5%, respectively. The $NH_4{^+}-N$ removal efficiency was about 60.2 85.5, 91.3 and 92.2%, respectively. T-N and T-P removal efficiencies increased from 53.7 and 56.8 to 82.5 and 86.4%, respectively as the HRT increased from 6 hour to 24 hour. In RO process, average removal efficiencies of color and $COD_{Cr}$ in RO permeate were about 99.9% and 96.8%, respectively. Also, removal efficiencies of T-N, $NH_4{^+}-N$, $NO_3{^-}-N$ and T-P were all above average 90%.

Metabolic Changes in Pericontusional Edematous Areas in Mild Head Injury Evaluated by Proton MRS (경도 두부외상 환자에서 뇌좌상주위 부종영역에서의 자기공명분광법을 이용한 대사변화)

  • Objectives : In order to evaluate the metabolic changes associated with pericontusional edematous area in mild head injury, proton magnetic resonance spectroscopy(1H-MRS) was performed in mild head injury patients (initial GCS score 13-15) with focal brain contusion. Methods : Seven head injury patients with initial GCS 13-15(3 males and 4 females : age range 15-65 years, mean age 33 years) have underwent 1H-MRS evaluations. The patients were examined within 7 days after injury(n=7) and 2 months after injury(n=5). The region of interest(ROI) was selected in the edematous area adjacent to traumatic brain contusion upon T2-weighted MR images and a corresponding region of the contralateral hemisphere (ROC, region of contralateral corresponding hemisphere) was examined as well. The metabolic ratios of NAA/Cr and lactate/Cr were compared between ROIs, ROCs and control values. Results : In initial NAA/Cr ratios, the values of ROIs were significantly lower than those of the controls(p=0.009), but there was no difference either between ROIs and ROCs(p=0.410) or between ROCs of patients and the control (p=0.199). In lactate/Cr ratios, the ROIs in all seven patients and the ROCs in two showed increased lactate signals. The lactate/Cr ratios of the ROIs were significantly elevated as compared to those of the ROCs(p=0.02) and the control(p=0.015). In two months follow-up, lactate signals were absent or significantly reduced(p=0.015). In no patients, clinical or radiological deterioration has been observed. Conclusion : Our 1H-MRS results demonstrate that there are significant ischemic changes in pericontusional edematous areas as indicated by elevated lactate signals in the patients with mild head injury. But there were no consistent neural loss or dysfunction in these area. There findings suggest that pericontusional edematous areas can be vulnerable to secondary brain insults even in the patients with mild head injury.

Effects of Alloying Elements on Corrosion Resistance of Low Alloyed Steels in a Seawater Ballast Tank Environment (Seawater ballast tank 환경에서 저합금강의 내식성에 미치는 합금원소의 영향)

  • Co-application of organic coating and cathodic protection has not provided enough durability to low-alloyed steels inseawater ballast tank (SBT) environments. An attempt has made to study the effect of alloy elements (Al, Cr, Cu, Mo, Ni, Si, W) on general and localized corrosion resistance of steels as basic research to develop new low-allowed steels resistive to corrosion in SBT environments. For this study, we measured the corrosion rate by the weigh loss method after periodic immersion in synthetic seawater at $60^{\circ}C$, evaluated the localized corrosion resistance by an immersion test in concentrated chloride solution with the critical pH depending on the alloy element (Fe, Cr, Al, Ni), determined the permeability of chloride ion across the rust layer by measuring the membrane potential, and finally, we analyzed the rust layer by EPMA mapping and compared the result with the E-pH diagram calculated in the study. The immersion test of up to 55 days in the synthetic seawater showed that chromium, aluminium, and nickel are beneficial but the other elements are detrimental to corrosion resistance. Among the beneficial elements, chromium and aluminium effectively decreased the corrosion rate of the steels during the initial immersion, while nickel effectively decreased the corrosion rate in a longer than 30-day immersion. The low corrosion rate of Cr- or Al-alloyed steel in the initial period was due to the formation of $Cr_2FeO_4$ or $Al_2FeO_4$, respectively -the predicted oxide in the E-pH diagram- which is known as a more protective oxide than $Fe_3O_4$. The increased corrosion rate of Cr-alloyed steels with alonger than 30-day exposure was due to low localized corrosion resistance, which is explained bythe effect of the alloying element on a critical pH. In the meantime, the low corrosion rate of Ni-alloyed steel with a longer than 30-day exposure wasdue to an Ni enriched layer containing $Fe_2NiO_4$, the predicted oxide in the E-pH diagram. Finally, the measurement of the membrane potential depending on the alloying element showed that a lower permeability of chloride ion does not always result in higher corrosion resistance in seawater.

Characteristics of CrOx Thin-films for High Precision Resistors (고정밀저항용 크롬산화박막의 특성)

  • This paper presents characteristics of CrOx thin-film, which were deposited on $Al_2$O$_3$ wafer by DC reactive magnetron sputtering in an argon-oxide atmosphere for high temperature applications. The present paper deals with a study of the technological characteristics of thin film resistors to provide a control in obtaining temperature coefficients of resistance of given value. The optimized condition of CrOx thin-film were thickness range of 2500 $\AA$ and annealing condition(350 $^{\circ}C$, 1 hr) in oxide partial pressure(3.5${\times}$10$^{-4}$ torr). Under optimum conditions, the CrOx thin-films is obtained a high resistivity, p=340 $\mu$Ωcm, a low temperature coefficient of resistance, TCR=-55 ppm/$^{\circ}C$. The CrOx thin films resistors which were fabricated in this paper had excellent characteristics as high precision resistors.

Cloning of hexavalent chromium reductase gene from E.coli ATCC 33456

  • E.coli ATCC 33456 has relatively higher activity of Cr(VI) reduction than other microorganism. The purpose of this research is cloning of Cr(V) reductase from E.coli ATCC 33456. Using colony and southern hybridization, we selected two condidates. Among candidates, pNCR9 is higher Cr(VI) reduction activity than E.coli ATCC 33456. Purified Cr(VI) reductase antibody was reacted at estimated 42Kda protein band of candidate's crude extract on 12% SDS-PAGE. This results showed cloned gene's product is very similar to purified Cr(VI) reductase from E.coli ATCC 33456.

Comparison of the bond strength of ceramics to Co-Cr alloys made by casting and selective laser melting

  • PURPOSE. Considering the importance of metal-ceramic bond, the present study aimed to compare the bond strength of ceramics to cobalt-chrome (Co-Cr) alloys made by casting and selective laser melting (SLM). MATERIALS AND METHODS. In this in-vitro experimental study, two sample groups were prepared, with one group comprising of 10 Co-Cr metal frameworks fabricated by SLM method and the other of 10 Co-Cr metal frameworks fabricated by lost wax cast method with the dimensions of $0.5{\times}3{\times}25\;mm$ (following ISO standard 9693). Porcelain with the thickness of 1.1 mm was applied on a $3{\times}8-mm$ central rectangular area of each sample. Afterwards, bond strengths of the samples were assessed with a Universal Testing Machine. Statistical analysis was performed with Kolmogorov-Smirnov test and T-test. RESULTS. Bond strength in the conventionally cast group equaled $74.94{\times}16.06\;MPa$, while in SLM group, it equaled $69.02{\times}5.77\;MPa$. The difference was not statistically significant ($P{\leq}.05$). CONCLUSION. The results indicated that the bond strengths between ceramic and Co-Cr alloys made by casting and SLM methods were not statistically different.
