Inactivation Kinetics of Listeria innocua ATCC 33090 at Various Temperature Heating-up and Pressure Building-up Rates

  • The effects of temperature heating-up rate and pressure building-up phase on the inactivation of Listeria innocua ATCC 33090 were evaluated in buffered peptone water. The number of L. innocua was reduced by 5.57 and 6.52 log CFU/mL during the nonisothermal treatment (the come-up time followed by isothermal process) and the isothermal treatment, respectively, at $60^{\circ}C$. When compared to the isothermal treatment (0.76$33.2^{\circ}C/min$ of temperature heating-rate. The effect of the combined high pressure and thermal processing on the inactivation of L. innocua increased with increasing pressure and temperature. At all temperature levels from 40 to $60^{\circ}C$ under 700 MPa, L. innocua was not detected by enrichment culture (>7 log reduction).

Pharmacokinetic Characteristics of Levosulpiride in Relation to the Genetic Polymorphism of MDR1: From Knockout Mouse to Human

  • The purposes of this study were to clarify the involvement of P-glycoprotein (P-gp) in the efflux of levosulpiride in knockout mice that lack the mdr1a1b gene and to evaluate the relationship between the genetic polymorphisms in MDR1 gene (exon 21) and levosulpiride disposition in healthy Korean subjects. After oral administration ($10\;{\mu}g/g$) of levosulpiride to mdr1a/1b(-/-) and wild-type mice, plasma and brain samples were obtained at 45 min. We also investigated the genotype for MDR1 (exon 21) gene in humans using a polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method. A single oral dose of 25 mg levosulpiride was administered to 58 healthy subjects, who were based on the MDR1 genotype for the G2677T SNP. Blood samples were taken up to 36 hr after dosing. The concentrations of levosulpiride in mouse plasma and brain were statistically significant difference between the two animal groups (P<0.05). In addition, the average brain-to-plasma concentration ratio (Kp) of levosulpiride was 3.4-fold (P<0.01) higher in the mdr1a/1b(-/-) mice compared with the wild-type mice. We also found that the values of $AUC_{0-{\infty}$, partial AUC ($AUC_{0-4h}$) and $C_{max}$ were significantly different between homozygous 2677TT subjects and the subjects with at least one wild-type allele (GG and GT subjects, P=0.012 for $AUC_{0-{\infty}$; P=0.008 for $AUC_{0-4h}$; P=0.038 for $C_{max}$). The results confirm that levosulpiride is a P-gp substrate in vivo, and clearly demonstrate the effect of SNP 2677G>T in exon 21 of the MDR1 gene on levosulpiride disposition.

Comparison of Body Composition and Serum Oxidized LDL Concentration between Middle-aged Women Exercising Aerobics and Sedentary (에어로빅댄스 수행과 비 운동 중년 여성의 신체 구성 및 혈청 Oxidized LDL 농도의 비교)

  • It is known that appropriate exercise changes body composition and improves coronary artery disease. This study was undertaken to evaluate the relationships between aerobic exercise, body composition, and the blood lipid levels in the middle-aged women(33 to 54 years old). The 57 subjects were divided into two groups based on their exercise: the aerobic exercise group(A group: 44) which performed at about $60\%$ of $VO_2max$ during over 6 months and the sedentary one(S group: 13). The percentages of body fat waist/hip ratio(WHR), body mass, and lean body mass of two groups were measured and compared in serum HDLC level and oxidized LDL level, the two most effective factors of coronary artery disease. The subjects in A group showed the lower percentages of body fat and WHR, when compared with the ones in S group. The body mass and lean body mass of A group subjects were higher than those of S group subjects. The serum HDL-C level$(58.6\pm12.7mg/mL)$ was significantly higher for A than S group subjects.(p<0.05) The sem ox-LDL level $(6.64\pm4.11Eu/mL)$ for A group subjects was lower than S group ones. The fat mass showed significantly positive correlations with atherogenic index(AI)(r=0.301, p<0.05), and with blood glucose levels(r=0.334, p<0.05). Also the WHR whowed significantly positive correlations with LDL-C levels(r=0.277, p<0.05), and with AI(r=0.466, p<0.01). In summary, the subjects in A group have the lower percentages of body fat and WHR, when compared with the ones in S group. Also, A group subject showed a tendency that exercise enhances serum HDL-C levels and decreases oxidized LDL levels. And aerobic exercise showed positive results which change body composition and improve blood lipid levels. There were significantly positive correlations among the percentages of body fat At and blood glucose level. These results suggest that moderately intensive exercise is a significant factor in reducing coronary artery disease.

Effects of Dry- and Wet-ageing on Flavor Compounds and Eating Quality of Low Fat Hanwoo Beef Muscles

  • The present study aimed at investigating the effects of dry- and wet-aging methods on flavor compounds and sensory properties of low fat Hanwoo beef muscles. All the beef samples were obtained from 2-grade carcasses of Hanwoo cows. The beef samples used in the dry- and wet-aging methods were prepared in the forms of quarter beef (bone-in) and cuts (boneless), respectively. The dry-aging was carried out at $2^{\circ}C-4^{\circ}C$ and humidity of 65%-85%, while the wet-aging was done at $1^{\circ}C$ for 0, 20, 40, and 60 d. At each aging time, three muscles: longissmus thoracis (LT), glutaeusmedus (GM) and semimembranosus (SM) were taken from the corresponding quarters and cuts, and used for the flavor compounds and sensory analyses. Results showed that both aging methods significantly increased the concentrations of flavor compounds and total amount of all classes of the flavor compounds as the aging time increased (p<0.05). In the dry-aging method, the GM and SM muscles presented significantly higher total amounts of pyrazines and sulfur-containing compounds compared to the LT muscle (p<0.05). Both the aging methods improved the eating quality attributes, indicating by increased scores of sensorial attributes with increased aging time for all the muscles studied (p<0.05). However, compared to the wet-aging, the dry-aging method resulted in significantly higher scores of tenderness and flavor for the GM and SM muscles after 40 to 60 d. Thus, postmortem aging, especially the dry-aging method could be used to improve eating quality attributes (tenderness and flavor) of low fat beef muscles such as GM and SM.

Physico-chemical Properties of Omija Extracts Made Prepared by Various Immersion Conditions (수침 조건을 달리하여 제조한 오미자 추출액의 이화학적 특성)

  • This study examined the physico-chemical properties of omija extracts that were manufactured using various water temperatures($0^{\circ}C$, $23^{\circ}C$, $70^{\circ}C$) and immersion times(12 hrs, 18 hrs, 24 hrs). The results were as follows: The pH levels of the extracts ranged from 2.86 to 2.95: however, there were no significant differences between the samples according to the various water temperatures and immersion times. The total organic acid contents of the samples ranged from 1.87 to 2.57%, and included citric acid, malic acid, and succinic acid at levels of $1.11{\sim}1.54%$, 0.49{\sim}0.66%$, and $0.25{\sim}0.38%$, respectively. As the immersion time and water temp. is erature increased the level of total organic acid significantly increased(p<0.01). The sugar contents of samples ranged from 3.01 to 3.90% brix, and did not show significant differences according to the immersion times at the different water temperatures(0, 23, $70^{\circ}C$). The Omija extract that was immersed in water for 24 hrs. at $70^{\circ}C$ had a significantly higher(p<0.05) sugar content than the other samples. The total free sugar contents of samples ranged from 0.714 to 0.833%, and included glucose, fructose, and sucrose at levels of $0.37{\sim}0.42%$, 0.34{\sim}0.41%$, and $0.003{\sim}0.004%$, respectively. The Omija extract that was immersed in water for 18 hrs. at $70^{\circ}C$ water had a significantly higher(p<0.05) total free sugar content than the other samples. The lightness, redness, and yellowness values of the samples were in ranges of $49.09{\sim}58.25$, 62.37{\sim}67.34$, and $19.76{\sim}24.57$, respectively; therefore, red was the predominant color of the extracts. Overall, as the immersion time and water temperatures increased is, lightness significantly decreased and redness significantly increased(p<0.001).

Study on $CuInTe_2$ Single Crystals Growth and Characteristics(I) ($CuInTe_2$ 단결정 성장과 특성연구(I))

  • CuInTe2 synthesised in a horizontal electric furnace was found to be polycrystalline. Single crystals of CuInTe2 were grown with the vertical Bridgman technique. The structure, Hall effect of the crystals were measured in the temperature range 30 to 293K. Both the polycrystals and single crystals of CuInTe2 were tetragonal in structure. The lattice constants of the polycrytals were measured as a=6.168Å and c=12.499Å, with c/a=2.026, these of the single crystals were measured as a=6.186Å and c=12.453Å, with c/a=2.013. The growth plane of the oriented single crystals was confirmed to be a (112) plane from the back-reflection Laue patterns. The Hall effect of the CuInTe2 single crystals was measured with the method of van der Pauw The Hall data of the samples measured at room temperature showed a carrier concentration of 2.14×1023holes/m3, a conductivity of 739.58Ω-1m-1, and a mobility of 2.16×10 -2m 2/V·s for the sample perpendicular to the c-axis. Values of 1.51×1023holes/m3, 717.55Ω-1m-1, and 2.97×10-2 m2/V·s were obtained for the sample parallel to the c-axis. The Hall coefficients for the samples both perpendicular and parallel to the c-axis in the temperature range 30K to 293K were always positive values. Thus the CuInTe2 single crystal was determined to be a p-type semiconductor.

Effect of Antioxidant Fortification on Preservability of Buffalo Semen

  • During the process of freezing, spermatozoa suffer cold shock which increases their susceptibility to lipid peroxidation which plays an important role in ageing of spermatozoa, shortening their life span and affecting the preservation of semen. An experiment was therefore conducted to study the effect of addition of natural antioxidants into semen diluents on the preservability of buffalo semen. Split semen samples were extended in milk egg yolk diluents fortified with vitamin E (MYE), vitamin C (MYC) and control group (MYO); Tris-egg yolk diluents fortified with vitamin E (TYE), vitamin C (TYC) and control group (TYO) and evaluated for their preservabilities at 4-7$^{\circ}C$ and $37^{\circ}C$. Overall least squares mean of percent motility observed after 0, 24, 48, 72 and 96 h of preservation at 4-7$^{\circ}C$ were 66.70, 54.00, 36.80, 21.90 and 12.50, respectively while the estimates for semen extended in MYE, MYC, MYO, TYE, TYC and TYO were 44.80, 42.70, 38.70, 36.00, 35.20 and 33.00 percent, respectively. The results showed that motility was significantly (p<0.01) affected by extender (extender-antioxidant combination) and preservation interval. Overall least squares mean percent motility observed after 0, 4, 8, 12 and 24 h of preservation at $37^{\circ}C$ were 68.50, 58.90, 45.00, 38.10 and 18.10 percent, respectively, while the estimates for semen extended in MYE, MYC, MYO, TYE, TYC and TYO were 48.20, 49.30, 46.80, 45.30, 42.30 and 42.50 percent, respectively. Extender and storage interval were found to be significantly (p<0.01) affecting spermatozoa motility on room temperature preservation. The results indicated that the incorporation of antioxidants, especially vitamin E, had beneficial effect on preservability of buffalo semen.

Purification and Comparison of Properties of the C-Terminus Truncated Agarase of Pseudomonas sp. W7

  • Three plasmids derived from the ${\beta}-agarase$ gene (PjaA) of Pseudomonas sp. W7 were expressed in Escherichia coli AD494(DE3) pLysS with lactose as an inducer. These products corresponded to the complete (PjaA) and the two C-terminal truncated (PjaAI and PjaAII) forms of ${\beta}-agarase$. The PjaAI and the PjaAII were originated from exonuclease L treatment from PjaA by deleting 127 and 182 amino acid residues-encoded nucleic acids at 3' region, respectively. The molecular weights of the purified proteins were 71 kDa, 58 kDa, and 50 kDa on SDS-PAGE, respectively. The $K_m$ value of PjaAI was lower than that of the PjaA, and the catalytic efficiency ($k_{cat}/K_m$) of PjaAI was increased to 5 times. The enzyme of PjaAI retained more than 90% activity at $50^{\circ}C$. In contrast to the PjaAI, the remaining activity of the PjaA was only 20% at the same temperature.

Cloning and Expression of $\beta$-Glucuronidase from Lactobacillus brevis in E. coli and Application in Bioconversion of Baicalin and Wogonoside

  • The $\beta$-glucuronidase (GUS) gene from Lactobacillus brevis RO1 was cloned and expressed in Escherichia coli GMS407. The GUS gene was composed of 1,812 bp, encoding a 603-amino-acid protein belonging to glycosyl hydrolase family 2 with three conserved domains. The amino acid similarity was higher than 70% with the $\beta$-glucuronidases of various microorganisms, yet less than 58% with the $\beta$-glucuronidase of L. gasseri ADH. Overexpression and purification of the GUS was performed in $\beta$-glucuronidase-deficient E. coli GMS407. The purified GUS protein was 71 kDa and showed 1,284 U/mg of specific activity at optimum conditions of pH 5.0 and $37^{\circ}C$. At $37^{\circ}C$, the GUS remained stable for 80 min at pH values ranging from 5.0 to 8.0. The purified enzyme exhibited a half-life of 1 h at $60^{\circ}C$ and more than 2 h at $50^{\circ}C$. When the purified GUS was applied to transform baicalin and wogonoside into their corresponding aglycones, $150\;{\mu}M$ of baicalin and $125\;{\mu}M$ of wogonoside were completely transformed into baicalein and wogonin, respectively, within 3 h.

A Study on the Development of a System for Measuring Dielectric Hysteresis (유전 히스테리시스 특성 측정장치의 연구 개발)

  • A computer-controlled system for measuring dielectric hysteresis has been developed. This system consists of the wave generation par\, the high voltage amplifier part, the measurement part, the data acquisition part and the related controll circuits and is interfaced to P.C(personal computer). The applied voltage and its frequency are controlled by P.C . Since the measured datas can be saved in the RAM of P.C, the analysis and the graphics of the datas are very convenient. As a accuracy test, the capcitances of the commercial mica and styroll capacitors were measured by applying high voltage and the results were good agreement with the rating values. In the test of PZT ceramic sample, the typical D-E hysteresis loops were obtained by applying a single frequency voltage to the sample, and $\varepsilon-E$ and D-E hysteresis loops could be measured at the same time by applying a double frequency voltage to the sample.sample.
