8-60hIPP5m-Induced G2/M Cell Cycle Arrest Involves Activation of ATM/p53/p21cip1/waf1 Pathways and Delayed Cyclin B1 Nuclear Translocation

  • Protein phosphatase 1 (PP1) is a major serine/threonine phosphatase that controls gene expression and cell cycle progression. The active mutant IPP5 ($8-60hIPP5^m$), the latest member of the inhibitory molecules for PP1, has been shown to inhibit the growth of human cervix carcinoma cells (HeLa). In order to elucidate the underlying mechanisms, the present study assessed overexpression of $8-60hIPP5^m$ in HeLa cells. Flow cytometric and biochemical analyses showed that overexpression of $8-60hIPP5^m$ induced G2/M-phase arrest, which was accompanied by the upregulation of cyclin B1 and phosphorylation of G2/M-phase proteins ATM, p53, $p21^{cip1/waf1}$ and Cdc2, suggesting that $8-60hIPP5^m$ induces G2/M arrest through activation of the ATM/p53/$p21^{cip1/waf1}$/Cdc2/cyclin B1 pathways. We further showed that overexpression of $8-60hIPP5^m$ led to delayed nuclear translocation of cyclin B1. $8-60hIPP5^m$ also could translocate to the nucleus in G2/M phase and interact with $pp1{\alpha}$ and Cdc2 as demonstrated by co-precipitation assay. Taken together, our data demonstrate a novel role for $8-60hIPP5^m$ in regulation of cell cycle in HeLa cells, possibly contributing to the development of new therapeutic strategies for cervix carcinoma.

Correlation between Internal and External Egg Quality Indicators in the Early Phase of Hy-Line Brown Laying Hens

  • 계란의 내부 및 외부의 품질을 나타내는 요소들 사이의 상관 관계를 분석하여, 할란 검사를 통하지 않고도 계란 품질을 예측하는데 적합한 지표가 있는지 알아보기 위한 실험을 수행하였다. 케이지 계사에서 사육 중인 27주령의 하이라인 브라운 산란계 동일 계군이 같은 날에 산란한 180개의 계란을 수집하였다. 분석 항목으로는 호우유닛, 농후난백높이, 파각 강도, 난각 두께, 난각 색(CIE L*, CIE a*, CIE b*, 반사율), 계란의 중량, 계란의 장경, 계란의 폭, 난형 지수 및 난황색 등 총 13가지의 지표를 조사하였다. 피어슨 상관 계수(r)를 이용하여, 각 품질 지표 사이의 선형 관계성을 평가하였다. 호우유닛과 농후난백높이, 난각색 CIE L*값과 반사율, 그리고 계란 중량과 계란의 폭 사이에서 매우 강한 정의 상관 관계가 분석되었다(r=0.975, r=0.973, r=0.814, P<0.001). 계란의 중량과 계란의 장경, 난각색 CIE L*값과 난각색 CIE a*값, 난각색 CIE a*값와 반사율, 그리고 난형 지수와 계란의 장경 사이에는 높은 상관 관계가 나타났다(r=0.703, r=-0.726, r=-0.681, r=-0.681, P<0.001). 난각의 파각 강도와 난각의 두께, 난각색 CIE a*값과 난각색 CIE b*, 난형 지수와 계란의 폭 사이에는 중간 정도의 상관 관계가 나타났다(r = 0.417, r = 0.590, r = 0.405, P<0.001). 난각색 CIE a*값은 호우유닛, 농후난백높이와 약한 정의 상관 관계가 있었으며(r=0.217, r=0.202, P<0.01), 난각색 CIE L*값과 파각 강도 사이에는 약한 부의 상관 관계가 나타났다(r=-0.236, P<0.01). 난각색 CIE b*값은 난각 두께와 약한 부의 상관 관계를 보였고, 난각색 반사율은 파각 강도 및 난각 두께와 약한 부의 상관 관계에 있는 것으로 나타났다(r=-0.225, r=-0.249, r=-0.222, P<0.01). 그 외 나머지 품질 지표들 간의 상관성은 통계적으로 유의하지 않거나 관계가 없는 것으로 나타났다. 갈색란의 난각색은 반사율만으로 표현하는 것 보다, CIE L*값, CIE a*값과 CIE b*값을 함께 적용했을 때 갈색의 정도를 더 구체적으로 표현한다. 결론적으로 본 연구를 바탕으로 갈색란의 난각색 CIE L*a*b*값은 계란의 품질 지표인 호우유닛, 농후난백높이, 난각의 파각 강도와 두께를 간접적으로 예측하는데 사용할 수 있을 것으로 판단되었다.

  • 제7기 국민건강영양조사 자료(2016~2018)를 이용해서 고감도 C반응성 단백(hsCRP)과 요산의 비만에 대한 위험도와 상호 작용이 있는지 알아보고자 하였다. 비만은 체질량지수(BMI)가 25 kg/m2 이상으로, 중증 비만은 BMI가 30 kg/m2 이상으로, 병적 비만은 BMI가 35 kg/m2 이상으로 정의하였다. 복합표본 다중 로지스틱 회귀분석에서 주요한 위험인자를 추가하여 보정하였음에도 불구하고 고농도의 hsCRP와 고농도의 요산이 동시에 존재하는 그룹은 기준 그룹보다 비만의 오즈비(odds ratio, OR)가 모든 단계에서 높았다(비만, OR 1.89, P<0.001 vs. 심한 비만, OR 5.04, P<0.001 vs. 병적인 비만, OR 8.20, P<0.001). 비만의 단계가 높아질수록 비만에 대한 hsCRP와 요산의 오즈비가 1.89에서 8.20으로 높아졌다, 이는 BMI가 증가된 참가자일수록 hsCRP와 요산의 관련성이 크다는 것을 암시한다. 더욱이, 복합표본 일반선형분석에서 주요한 교란 요인을 보정한 후 비만에 대한 hsCRP와 요산 사이의 상호 작용이 분석되었다(P for interaction=0.009).

Econazole Induces p53-Dependent Apoptosis and Decreases Metastasis Ability in Gastric Cancer Cells

  • Econazole, a potent broad-spectrum antifungal agent and a Ca2+ channel antagonist, induces cytotoxicity in leukemia cells and is used for the treatment of skin infections. However, little is known about its cytotoxic effects on solid tumor cells. Here, we investigated the molecular mechanism underlying econazole-induced toxicity in vitro and evaluated its regulatory effect on the metastasis of gastric cancer cells. Using the gastric cancer cell lines AGS and SNU1 expressing wild-type p53 we demonstrated that econazole could significantly reduce cell viability and colony-forming (tumorigenesis) ability. Econazole induced G0/G1 phase arrest, promoted apoptosis, and effectively blocked proliferation- and survival-related signal transduction pathways in gastric cancer cells. In addition, econazole inhibited the secretion of matrix metalloproteinase- 2 (MMP-2) and MMP-9, which degrade the extracellular matrix and basement membrane. Econazole also effectively inhibited the metastasis of gastric cancer cells, as confirmed from cell invasion and wound healing assays. The protein level of p53 was significantly elevated after econazole treatment of AGS and SNU1 cells. However, apoptosis was blocked in econazole-treated cells exposed to a p53-specific small-interfering RNA to eliminate p53 expression. These results provide evidence that econazole could be repurposed to induce gastric cancer cell death and inhibit cancer invasion.

플라워 주얼리의 디자인 트렌드와 표현기법 분석에 관한 연구 (A Study on the Analysis of the Trends and Expression Techniques of Flower Jewelry)

  • 본 연구는 2011~2019년의 9년간 화훼장식 전문 월간지에 나타난 플라워 주얼리 작품을 추출하고, 추출된 플라워 주얼리 작품을 대상으로 표현유형, 표현기법, 사용된 식물재료와 비식물재료의 종류와 수 등을 작품별, 연도별로 분석하여 국내 플라워 주얼리의 트렌드를 알아보고자 실시하였다. 표현유형별로 살펴보면 머리장식의 경우 20.83%, 목걸이의 경우 57.29%, 귀걸이의 경우 5.21%, 리스트의 경우 10.42%, 기타 6.25%로 총 96작품으로 분석되었다(χ2=94.833, p<.001). 표현기법의 사용빈도를 분석한 결과 각 작품당 머리장식, 목걸이, 리스트는 5~6가지의 표현기법, 귀걸이는 2~4가지의 표현기법을 사용하여 제작되었다. 사용된 표현기법은 소재 결합기법 34.43%, 꽃과 잎의 이용기법 30.17%, 시각적 기법 16.63%, 집단화 기법 14.12%, 테크닉 강조기법 4.26%, 기타 0.39%로 나타났다(χ2=455.222, p<.001). 가장 많이 사용된 기법은 프레이밍 기법 16.63%, 노팅기법 16.44% 순으로 나타났다. 플라워 주얼리에 사용된 식물 재료는 절화의 경우 호접란 22.61%, 천일홍 13.48%, 글로리오사 9.57%, 에피덴드럼7.39%, 왁스플라워 6.96%, 골든볼 4.78% 등으로 나타났다(χ2=718.104, p<.001). 절지의 경우 말채나무 70.00%로 가장 많이 사용되었고, 다래덩굴, 노박덩굴, 능수버들이 각각 10.00% 사용되었다(χ2=10.800, p=.013). 절엽의 경우 엽란 24.65%, 아스파라거스 스마일락스 24.62%, 콩란 11.54%, 러브체인 6.15% 등으로 나타났다(χ2=269.385, p<.001). 열매의 경우 청미래덩굴의 열매 44.44%, 하이페리쿰 33.33%, 미국자리공 11.11% 등으로 나타났다(χ2=11.444, p=.022). 플라워 주얼리 제작에 사용된 비식물재료는 2mm 알루미늄와이어 47.34%, 카파와이어 33.73%, 1mm 알루미늄와이어 10.06% 등으로 나타났다(χ2=186.704, p< .001). 와이어 이외에 진주 53.57%, 리본 12.50%, 스팽글과 깃털 4.14% 등의 순으로 나타났다.

Ca+-(CO)n과 Ca+-(CO2)n (n=1,2)의 구조와 결합에너지에 대한 이론 연구 (Theoretical Study of the Structures and Binding Energies of Ca+-(CO)n and Ca+-(CO2)n (n=1,2))

  • $Ca^+-(CO)_n$,과 $Ca^+-(CO_2)_n$ (n=1,2) complex에 대한 구조와 결합 에너지를 MP2/6-311++G(2d,p) 방 법과 B3LYP/6-311++G(2d,p) 방법에 의해 계산하였고 vibrational frequencies도 계산하였다. $Ca^+-(CO)_n$의 경우 C-bonded complex와 O-bonded complex가 다 가능함을 보였고, $Ca^+-(CO)_2$에서는 선형과 $C_{2v}$ 형태가 나타남을 볼 수 있었으며 더 안정한 형태는 $C_{2v}$ 구조로 밝혀졌다. $Ca^+-(CO_2)_2$에서도 선형과 $C_{2v}$ 형태를 볼 수 있는데 이 경우는 선형이 근소한 에너지 차이로 더 안정한 것으로 나타났다.

Effect of Vitamin D on YKL-40: Rat Hypercholesterolemia Model

  • Background and Objectives: YKL-40 is considered to be associated with cardiovascular disease (CVD). In this study, the effect of serum 25(OH) vitamin D [25(OH)VitD] differences between groups on YKL-40 was evaluated on a hypercholesterolemia rat model. Methods: Thirty-two male rats (wistar albino) were equally divided into 4 groups. The first group was the control group; the second group was high-cholesterol (H-CH) adequate vitamin D (VitD) group (H-AdeVD). The third group was the H-CH deficient VitD group (H-DefVD), and the last group was designed with the H-CH supplement VitD (H-SupVD). The feeding process consisted of 2 stages. At the first stage (5 months), the H-DefVD group was fed on VitD deficient chow, while the other groups (control, H-AdeVD, H-SupVD) were fed on standard chow. At the second stage (3 months), the H-AdeVD and the H-SupVD groups were fed on the H-CH chow, whereas the H-DefVD group was fed on the H-CH-VitD deficient chow. Moreover, the H-SupVD group was given 100 IU/kg/day VitD along with the H-CH chow. Results: Compared with the control group, interleukin-6 (IL-6), soluble intercellular adhesion molecule-1 (sICAM-1), soluble vascular cell adhesion molecule-1 (sVCAM-1), and YKL-40 values in the H-DefVD groups increased significantly (p<0.001, p<0.001, p=0.009, p=0.005; sequentially). Conclusion: It can be concluded that VitD can suppress the YKL-40, thus, it will prevent CVD development in rat. Therefore, further clinical studies related with human will reveal the effect of VitD and YKL-40 on CVD development.

낙동강하구 을숙도 갯벌에서 새섬매자기의 생육 특성 (Growth Characteristics of Bolboschoenus planiculmis on the Eulsuk Tidal Flat of the Nakdong River Estuary, Korea)

  • Bolboschoenus planiculmis is an important prey species for winter migratory birds in the Nakdong River Estuary region, but its population has declined in the last decade due to a physio-chemical shift. To identify the growth and development characteristics of B. planiculmis, we carried out an experimental survey on the aboveground and belowground sections of B. planiculmis specimens in the Eulsuk tidal flat from April to September 2019. We divided the belowground area into two groups: a vegetation group in which B. planiculmis exists and a non-vegetation group in which B. planiculmis does not exist and measured the rate of tubers. We observed a 45% appearance frequency in the vegetation group and only a 5% frequency in the non-vegetation group, indicating that there were no supplies for plant growth. The mean density, length, and biomass of B. planiculmis aboveground were 166.90±149.62 n m-2, 44.39±5.45 cm, and 67.63±54.46 g DW m-2, respectively, which was approximately half of those obtained from the study in the Eulsuk tidal flat in 2002. Tuber number was the highest in the 15~25 cm deep layer(41.50%). The mean number of total tubers belowground was related to different sediment conditions(FS: flat surface, DS: disturbed surface) 251.75±16.86 n pot-1 for FS and 171.00±25.18 n pot-1 for DS, which was significantly different(total tubers: p<0.005). The mean biomass of B. planiculmis above-ground was 21.11±1.60 g DW pot-1 for FS and 15.41±1.38 g DW pot-1 for DS, which was significantly different(p<0.05). This research provides primary materials for the restoration of the Nakdong River Estuary, wintering sites for migratory birds, and protection of B. planiculmis.

  • In this paper, we establish the existence and uniqueness of the solution for a class of reflected backward stochastic differential equations with time delayed generator (RBSDEs with time delayed generator, in short) in the case when the terminal value and the obstacle process are $L^p$-integrable with p ${\in}$]1, 2[ for a sufficiently small Lipschitz constant of the generator and the time horizon T.

Studies on the Apoptosis-Inducing Effect of Ulmi Pumilae Cortex on Human Leukemia HL-60 Cells

  • The antiproliferative effect of the water extract of the branch and root bark of Ulmi Pumilae Cortex(WEUPC) was investigated on the p53-negative human leukemia cell line (HL-60). A dose- and time-dependent inhibition of cell growth was observed; this effect appears to be due to induction of apoptosis. Involvement of oxidative stress is indicated by a dose-dependent increase in intracellular reactive oxygen species levels. In addition. anti-apoptic effect was observed in the cells simultaneously treated with WEUPC and the anti-oxidant N-acetylcysteine. WEUPC did not affect the anti-apoptotic Bcl-2 and the pro-apoptotic Bax, whereas p21/sup WAF1/CIPl/ was enhanced in a dose- and time-dependent fashion; this effect was partially inhibited by N-acetylcysteine. The increase in p21/sup WAF1/CIPl/ was accompanied by a parallel accumulation of cells in the G1 phase of the cycle. These results suggest that the p53-independent induction of p21/sup WAF1/CIP/ and the induction of apoptosis may mediate the anti proliferative effect of WEUPC at least in this study; on the basis of this observation, WEUPC could be proposed as an useful adjunct to the treatment of p53-deficient tumors, which are often refractory to standard chemotherapy.
