A cross-examination between KT&G Central Research Institute and Guangzhou Institute for Drug Control was carried out in order to select optimum conditions for extraction, separation and determination of ginsenosides in red ginseng and to propose a better method for the quantitative analysis of ginsenosides. The optimum extraction conditions of ginsenosides from red ginseng were as follows: the extraction solvent, $70\%$ methanol; the extraction temperature, $100^{\circ}C;$ the extraction time, 1 hour for once; and the repetition of extraction, twice. The optimum separation conditions of ginsenosides on the SepPak $C_{18}$ cartridge were as follows: the loaded amount, 0.4 g of methanol extract; the washing solvents, distilled water of 25 ml at first and then $30\%$ methanol of 25 ml; the elution solvent, $90\%$ methanol of 5 ml. The optimum HPLC conditions for the determination of ginsenosides were as follows: column, Lichrosorb $NH_2(25{\times}0.4cm,$ 5${\mu}m$, Merck Co.); mobile phase, a mixture of acetonitrile/water/isopropanol (80/5/15) and acetonitrile/water/isopropanol (80/20/15) with gradient system; and the detector, ELSD. On the basis of the optimum conditions a method for the quantitative analysis of ginsenosides were proposed and another cross-examination was carried out for the validation of the selected analytical method conditions. The coefficient of variances (CVs) on the contents of ginsenoside-$Rg_{1}$, -Re and $-Rb_1$ were lower than $3\%$ and the recovery rates of ginsenosides were $89.4\~95.7\%,$ which suggests that the above extraction and separation conditions may be reproducible and reasonable. For the selected HPLC/ELSD conditions, the CVs on the detector responses of ginsenoside-Rg, -Re and $-Rb_1$) were also lower than $3\%$, the regression coefficients for the calibration curves of ginsenosides were higher than 0.99 and two adjacent ginsenoside peaks were well separated, which suggests that the above HPLC/ELSD conditions may be good enough for the determination of ginsenosides.
Objectives Diagnosis System of Oriental Medicine (DSOM) was made as a computerized assistant program for oriental medicine doctors to be able to diagnose with statistical basis. Then DSOM uses questionnaires filled out by respondents with explanatory guide. But if the respondents misunderstand the meaning of the passages, the results were quite the opposite. Methods This study was designed to investigate the diagnostic correspondent rates between DSOM and TKM practitioners. First, let the respondents answer to DSOM. After that, three doctors diagnosed the respondents and marked 'p' when they diagnose that the respondent had the pathogenic factors, marked 'n' when they diagnose that the respondent had the pathogenic factors but not severs, and did not marked when they diagnose that the respondent didn't have the pathogenic factors. Finally, this study was investigated the correspondent rates of diagnosis between DSOM and doctors. Results In the pathogenic factor of three including insufficiency of Yin (陰虛), the correspondent rates were 90%. In the pathogenic factor of nine including deficiency of qi (氣虛), the correspondent rates were 80%. In the pathogenic factor of four including blood stasis (血瘀), the correspondent rates were 70%. In HH and HL, they showed the correspondent rates of 61.77%. The correspondent rate of heat (熱) was highest (96.88%). The correspondent rate of insufficiency of Yang (陽虛) was lowest (0%). In LH and LL, they showed the correspondent rates of 88.31%. The correspondent rate of blood stasis (血瘀) was lowest (71.76%). They all showed the correspondent rates of over 70%. Conclusions In DSOM and Doctors' diagnose, they showed the correspondent rates of 83.60%.
Langmuir adsorption maximum was used as a basis for the estimation of phosphate requirement as related to the application method to a newly reclaimed hilly land. The application rates of phosphate fertilizer were 4 levels corresponding to 5, 10, 20, and 30 percent of the Langmuir adsorption maximum which was determined by the laboratory experiment. The phosphate fertilizer was applied in two different methods of broadcast and band application. Yield of soybean on a newly reclaimed hilly land was greatly affected by the amount of phosphate fertilizer and application method. With the same amount of phosphate fertilizer, the soybean yield in the broadcast was always lower than that in the band application even though the yield in the broadcast increased with the application rate up to the highest level of this experiment. However, the yield in the band application showed steep increase up to 20 percent level and then the yield decreased at the highest level. The yield at the rate of 5 percent with the band application was equal to the yield at the rate of 20 percent with broadcast, and the yield at the rate of 10 percent with band application was higher by 17 percent than that in the highest broadcast rate. When manure was applied at the rate of 1000 kg/10a to the plot of the 10 percent band application, the yield was 14.6 percent higher than the yield of the plot of the 10 percent band application without the manure and was equivalent to the yield of the plot of the 20 percent band application.
High salt content in food waste limit use as the potential source of compost. Since sodium chloride content should be lower than one percent to meet requirement for agricultural use of compost, we attempted use of three kinds of co-composting material for food wastes compost, which were fermented, dried, and fresh types of amendments which included pig manure, sawdust and puffed rice hull. Food wastes were composted well and stabilized after around the 40th days of composting. Little difference were found between composts amended sawdust and puffed rice hull. The result indicated that puffed rice hull could be used as a good alternative of sawdust. Because sawdust or puffed rice hull were mixed as amendments to control water contents and to dilute high NaCl content of compost material, the C/N ratios of the final products were significantly high over 40. However, NaCl contents of final products were dropped to less than percent on the fresh weight basis by mixing food wastes and the above amendments with the optimum rates.
Objective: Inbreeding depression of reproduction is a major concern in the conservation of native chicken genetic resources. Here, based on the successful development of strongly inbred (Sinb) and weakly inbred (Winb) Langshan chickens, we aimed to evaluate inbreeding effects on reproductive traits and identify candidate genes involved in inbreeding depression of reproduction in Langshan chickens. Methods: A two-sample t-test was performed to estimate the differences in phenotypic values of reproductive traits between Sinb and Winb chicken groups. Three healthy chickens with reproductive trait values around the group mean values were selected from each of the groups. Differences in ovarian and hypothalamus transcriptomes between the two groups of chickens were analyzed by RNA sequencing (RNA-Seq). Results: The Sinb chicken group showed an obvious inbreeding depression in reproduction, especially for traits of age at the first egg and egg number at 300 days (p<0.01). Furthermore, 68 and 618 differentially expressed genes (DEGs) were obtained in the hypothalamus and ovary between the two chicken groups, respectively. In the hypothalamus, DEGs were mainly enriched in the pathways related to vitamin metabolism, signal transduction and development of the reproductive system, such as the riboflavin metabolism, Wnt signaling pathway, extracellular matrix-receptor interaction and focal adhesion pathways, including stimulated by retinoic acid 6, serpin family F member 1, secreted frizzled related protein 2, Wnt family member 6, and frizzled class receptor 4 genes. In the ovary, DEGs were significantly enriched in pathways associated with basic metabolism, including amino acid metabolism, oxidative phosphorylation, and glycosaminoglycan degradation. A series of key DEGs involved in folate biosynthesis (gamma-glutamyl hydrolase, guanosine triphosphate cyclohydrolase 1), oocyte meiosis and ovarian function (cytoplasmic polyadenylation element binding protein 1, structural maintenance of chromosomes 1B, and speedy/RINGO cell cycle regulator family member A), spermatogenesis and male fertility (prostaglandin D2 synthase 21 kDa), Mov10 RISC complex RNA helicase like 1, and deuterosome assembly protein 1) were identified, and these may play important roles in inbreeding depression in reproduction. Conclusion: The results improve our understanding of the regulatory mechanisms underlying inbreeding depression in chicken reproduction and provide a theoretical basis for the conservation of species resources.
In order to enhance overall quality of the Backsulki, a sweet potato flour (SPF) which is rich in sugar and dietary fiber was added into the rice flour at 0, 5, 15, and 25% (w/w, db) ratios. Quality changes of the products were evaluated by storing at 5, 15, and 25$^{\circ}C$ for 4 days. Water binding capacity, swelling power, and solubility of the products increased as the SPF increased. Hunter colorimetric a- and b-value increased while L value of the products decreased as the SPF increased. Retarded retrogradation of the flour mixtures containing the SPF was shown from the DSC thermogram as indicated by the decreased $\Delta$H values. Increases in $T_{i}$ and $T_{p}$ values in relation with the starch gelatinization were shown from the DSC thermogram with a temperature margin of 15-20$^{\circ}C$ as the SPF increased. Increases in softness along with decreases in springiness and chewiness of the products were shown as the SPF increased. It was not so much the storage temperature as the added SPF affected the final texture of the products. Retarded growth of the total microbes of the products was noted by the added SPF during storage at 25$^{\circ}C$ for 3 days. Regarding the above and additional sensory preference scores of the products, a 15%(w/w) level of the SPF was suggested as optimum for the best Backsulki with improved qualities.
The purpose of this study was to investigate the Q - method, which is one of the qualitative analysis methods to approach deep and intrinsic meaning about the perception of college students about food in traditional market. Recently, local governments have been developing diverse tourism products aimed at young people in order to revitalize traditional markets. In this study, we classify the perception of food in traditional market by university students, And to suggest strategic implications by using it as basic data for establishing marketing strategy so that young people can visit in the futures. In order to analyze the perception of college students' subjective perception of traditional market food, Factor analysis was used to conduct an exploratory study. To do this, a Q-sort, Program, and Q factor analysis. The results were classified into three types. The first type (N = 21): Memories seeking type, the second type (N = 6): Local culture resource seeking type, the third type(N = 5). Each of these subjective perceptions can be used as a basis for future research. Through the establishment of marketing strategies for each of the three types of classifications, the direction of traditional markets is presented, and a variety of food items that are valuable as local tourism resources are accommodated by accepting university students' to contribute to the revitalization of traditional markets.
The purpose of this study was to find out the general type of consumer perception by defining and structuring consumers' self - awareness through the Q methodological approach to the types of youth perceived by HMR. This study focuses on the recognition of HMR as a major customer of HMR, and aims to provide information that can be considered in the HMR market by identifying consumption characteristics of youth consumers in the HMR market. To do this, the respondent classifies the statement card and analyzes the Q-sort. Q-sort, which is obtained by selecting and constructing the Q-population, Q factor analysis. (N = 10): price seeking type], the second type is [(N = 10)] which pursues the purchase by trust, (N = 2): seeking convenience], and the fourth type [(N = 3): seeking the appearance of the packaging)] Each type has its own characteristics. Each subjective opinion detected through this analysis is utilized as a basis for the follow-up study of HMR and will be provided as a marketing data of the HMR market in a future research direction.
BACKGROUND: Silver grass (Miscanthus sinensis) No. 1 was developed for production of bio-ethanol, and for the purpose the silver grass growing sector was established in Geumgang basin, Iksan, Jeonbuk, in 2011. However, the other application potentials except for using as the bio-energy resources should be considered because of the drop in international oil prices. Therefore, there is the necessity of a scientific basis to use the silver grass instead of rice straw as the organic matter source that is used for improvement of soil quality in the plastic film house. METHODS AND RESULTS: The silver grass was applied at 5, 10, 15 and 20 Mg/ha and tilled before the watermelon was planted in the plastic film-house. The control plot was treated with 10 Mg/ha with rice straw, and watermelons have been cultivated for 3 years(2017~2019). Soil aggregation, soil chemistry, and the growth characteristics were investigated, when the watermelon was harvested every year. Soil aggregation levels at the 2nd and 3rd year of watermelon harvest were similar from the plot applied with the silver grass at 5 Mg/ha and the control plot, and increased in the silver grass treated plots with more than 10 Mg/ha. However, there was no statistically significant difference between the plots. The nitrogen mineralization of silver grass in the control plot tended to be similar to the 5 Mg/ha plot, but the silver grass treated plots with over 10 Mg/ha showed low nitrogen mineralization. Soil EC on harvest stage was proportional to the applied mass of the silver grass, but pH was in inverse with the applied mass. Soil organic matter content, available phosphate, and exchangeable cations increased with the continued use of silver grass. Watermelon weight found to be the best on more than 15 Mg/ha of silver grass, and the sugar content was highest when 10 Mg/ha was treated. CONCLUSION: The use of the silver grass at 10 Mg/ha annually as the organic source was effective in replacing rice straw while growing fruits and vegetables on the plastic film house.

Bacterial leaf blight(BLB), caused by X. oryzae pv. oryzae(Xoo), is one of the most destructive diseases of rice due to its high epidemic potential. Understanding BLB resistance at a genetic level is important to further improve the rice breeding that provides one of the best approaches to control BLB disease. In the present investigation, a collection of 192 accessions was used in the genome-wide association study (GWAS) for BLB resistance loci against four Korean races of Xoo that were represented by the prevailing BLB isolates under Xoo differential system. A total of 192 accessions of rice germplasm were selected on the basis of the bioassay using four isolated races of Xoo such as K1 and K2. The selected accessions was used to prepare 384-plex genotyping by sequencing (GBS) libraries and Illumina HiSeq 2000 pairedend read was used for GBS sequencing. GWAS was conducted using TASSEL 5.0. The TASSEL program uses a mixed linear model (MLM). The results of the bioassay using a selected set of 192 accessions showed that a large number of accessions (93.75%) were resistant to K1 race and K2 resistant germplasm proportion remained between 66.67. The genotypic data produced SNP matrix for a total of 293,379 SNPs. After imputation the missing data was removed, which exhibited 34,724 SNPs for association analysis. GWAS results showed strong signals of association at a threshold of [-log10(P-value)] more than 5 (K1 and K2) for nine of the 39 SNPs, which are plausible candidate loci of resistance genes. These SNP loci were positioned on rice chromosome 2, 9, and 11 for K1 and K2 races. The significant loci detected have also been illustrated and make the CPAS markers for NBS-LRR type disease resistance protein, SNARE domain containing protein, Histone deacetylase 19, NADP-dependent oxidoreductase, and other expressed and unknown proteins. Our results provide a better understanding of the distribution of genetic variation of BLB resistance to Korean pathogen races and breeding of resistant rice.
