The 1, 1- diphenyl 2-picrylhyorazyl (DPPH) is a well-known radical and a trap (scavenger) for other radicals. Hyaluronidase (HAase) is an enzyme that depolymerizes the polysaccharide hyaluronic acid (HA) in the extracellular matrix of connective tissue. Lipoxygenase (LOX) enzyme was reported to convert the arachidonic, linoleic and other polyunsaturated fatty acid into biologically active metabolites involved in the inflammatory and immune responses. The purpose of the present study is to evaluate plant extracts as sources of natural antioxidants and to examine whether Achyranthes japonica having significant DPPH, HAase and LOX inhibitory activity. The inhibitory effect of HAase by A. japonica was assayed using a Morgan microplate assay. The antioxidant activity of the A. japonica extracts was measured on the basis of the scavenging activity of the stable 1, 1- diphenyl 2-picrylhyorazyl (DPPH) free radical. DPPH scavenging activity of matured roots of A. japonica was evaluated at 4.0 mg/ml was 87.8% and that of young roots was 86.2% at same concentration. The roots of A. japonica showed maximum inhibition of HAase activity (IC50 = 27.7 μg/ml). The highest LOX inhibition was recorded in the root extract among three vegetative parts. Inhibition of HAase activity of roots may contribute towards the development of herbal medicines. Although percent inhibition of lipoxygenase by Achyranthes japonica for all young and matured groups for leaves, stems, and roots at different concentrations, there were not show a statistically significant difference (p<0.05).

We determined a portion of mitochondrial 16S rRNA gene sequences (416 bp) to investigate the genetic structure of the octopus (Octopus minor Ssaki) population in Korea and China. Samples were obtained from Korea (Yeosu, Namhae, Jindo, Muan, Geomundo and Seosan) and China (Sandong) during the period of August 2006 to September 2007. Sequence analyses of 28 individual specimens collected from 7 localities revealed 11 haplotypes, ranging in a sequence divergence of 0.2% - 1.2%. Phylogenetic analyses using PHYLIP and networks subdivided the octopus into two clades (termed clade A and B) and the nucleotide divergence between them was 0.4%. This haplotype subdivision was in accordance with geographic separation: one at Yeosu, Namhae, Muan and Jindo, and the other at Seosan, Geomundo and Sandong. On the basis of hierarchial genetic analysis, genetic distance between localities in Korea and China were also found, but a significant population differentiation was not shown in this study (p>0.05). Consequently, most of the octopus populations in Korea had considerable distribution due to the mitochondrial gene flow that resulted in a formation of a genetically homogenous structure, whereas some of the Korean and Chinese populations had different genetic structures. Gene flow among populations may be restricted due to impassable geographic barriers that promote genetic differentiation.
Recently, two formulas of sulgidduk added to pine needle juice (PNJ) with various physiological activities were developed for metabolic syndrome patients in our lab. According to previous studies, cooking may alter antioxidant properties by initiating destruction, release or transformation of antioxidant compounds contained in food. Therefore, the aim of this study was to compare the antioxidant activities and antigenotixic effects of sulgidduk with/without PNJ and to note changes in these activities after cooking. The ingredients of sulgidduk was added on the basis of 100% rice flour as follows: conventional sulgidduk (S): 1.5% salt, 30.0% sugar; PNJ added to sulgidduk A (PS-A): 1.4% salt, 30.0% sugar, and 1.0% PNJ; PNJ added to sulgidduk B (PS-B): 1.5% salt, 21.4% sugar, and 1.4% PNJ. Ethanol and water extracts of sulgidduk were analyzed for the total phenolic content (TPC), DPPH radical scavenging activity (DPPH RSA), total radical-trapping antioxidant potential (TRAP), oxygen radical absorbance capacity (ORAC), and antigenotoxic effect by comet assay. The ethanol extracts PS-A and PS-B showed higher TPC and antioxidant activities (DPPH RSA, TRAP, and ORAC) than did the S ethanol extract before cooking. The more PNJ was added, the higher TPC and anitoxidant activities were observed in sulgidduk (PS-A$200{\mu}M$ of $H_2O_2$. Taken together, this study suggests that sulgidduk added to 1.44% of pine needle juice may be a good option antioxidant and antigenotoxic source.
Highly Pathogenic Avian Influenza (HPAI) is a very acute systemic disease in poultry, particularly in chickens and turkeys caused by HPAI viruses. An outbreak of HPAI caused by subtype H5N1, was first reported in a broiler breeder farm on December 10, 2003 in Korea, although there had been twenty one outbreaks of the disease reported in the world before. Since mid-December 2003, eight Asian countries have confirmed outbreaks of HPAI due to the same subtype. The outbreak has also resulted in at least twenty three fatal human cases in Vietnam and Thailand as of May 17, 2004 according to the WHO. Regarding the first outbreak of recent Asian HPAI, it has been suspected that some Asian countries with the exception of Korea and Japan veiled the fact of HPAI outbreaks since the last half of 2003, even though it was first reported in Korea. There have been total nineteen outbreaks of HPAI among chicken and duck farms in 10 provinces in Korea since Dec. 2003 and approximately 5,280,000 birds were slaughtered from 392 farms for eradication of the disease and preemptive culling. The origin of the H5Nl HPAI virus introduced into the country are unknown and still under epidemiological investigation. Current status of outbreaks and characteristics of HPAI will be reviewed and discussed on the basis of genetic, virological, clinicopathological, and ecological aspect, as well as future measures for surveillance and prevention of the disease in Korea.
In 60 dairy cows with inactive ovaries, follicular cyst, luteal cyst, persistent corpus luteum and silent heat as diagnosed by rectal palpation, and those that had not resumed ovarian cycles until 60 days postpartum, progesterone concentrations for differential diagnosis of reproductive disorders were measured and were compared in matched plasma, skim milk and milk fat samples at 10 days interval. The incidence rate of reproductive disorders were as follows; inactive ovaries 20(33.3%), silent heat 11(18.3%), follicular cyst 7(11.7%), luteal cyst 7(11.7%), persistent corpus luteum 7(11.7%), pyometra 4(6.7%), vaginitis 2(3.3%), cystic corpus luteum 1(1.7%), and endometritis 1(1.7%), respectively. Cows having a progesterone concentration in plasma and skim milk < 1.0 ng/ml, and in milk fat < 80.0 ng/ml were considered to have inactive ovaries or follicular cyst. Those with concentrations in plasma and skim milk ${\geq}1.0ng/ml$, and in milk fat ${\geq}80.0ng/ml$ were regarded as the cases of luteal cyst or persistent corpus luteum. Progesterone concentrations in above cows did not differ significantly between the time of initial determination and the 10 days after initial determination. But progesterone concentrations in cows with silent heat did differ significantly between the time of initial determination and the 10 days after initial determination(P<0.05). The accuracy of rectal palpation for making a differential diagnosis of ovarian dysfunction, as defined on basis of progesterone concentrations, were as follows; follicular cyst 55.6%, luteal cyst 50.0%, inactive ovaries 90.5% and persistent corpus luteum 60.0%, respectively. It may be concluded that progesterone determinations at 10 days interval is practical as an aid to diagnosing ovarian dysfunction, particularly follicular cyst and luteal cyst.
Six hundred fourteen consecutive cases of bioprosthetic cardiac valve replacement performed during the period from March 1976 through December 1982 were reviewed. A total of 748 tissue valves [534 Ionescu-Shiley valves, 144 Hancock valves, 46 Angell-Shiley, and 24 Carpentier-Edwards] were implanted in 610 patients. Of these, 477 had single valve replacements [403 mitral, 60 aortic, and 14 tricuspid] including three REDO MVR and one REDO AVR. The remaining 129 had double valve replacements [95 AVR and MVR and 34 MVR and TVR] and 8 had triple valve replacement.592 cases were evaluated. Overall early mortality rate [within 30 days of operation] was 7.1% [6.2% in single valve replacement, 10.2% in double valve replacement, and 16.7% in triple valve replacement]. Leading causes of mortality were low cardiac output or myocardial failure and ventricular arrhythmias. The follow-up period was from one month to 7 years with a cumulative follow-up of 906.6 patient-years [mean 1.53 years]. The late mortality was 1.6%, 3.9%, 0%, 2.6%, 6.6% and 2.0% per patient-year for MVR, AVR, TVR or triple valve replacement, AVR+MVR, MVR+TVR and total, respectively. Actuarial analysis of late results including early mortalities indicates an expected survival rate of 87.6+1.8% at 3 years and 85.92.4% at 7 years for all cases. We also analyzed actuarial survival rate between groups of each valve replacement [AVR, TVR, Double valve, and Triple valve] and the tissue valve groups in MVR. We experienced 7 cases [0.77% per patient-year] of confirmed endocarditis, two of which were fatal. Valve failure-free rates calculated according to the confirmed cases were 97.5% at 4 years, 87.5% at 7 years, and 88.3% at 6 years for Ionescu-Shiley, Hancock and Angell-Shiley valves, respectively. The occurrence rate of thromboembolism was 2.0% per patient-year in total cases, although almost all the patients were given anticoagulant therapy for one year. The occurring rate in MVR was 1.5% and 2.7% per patient-year for Ionescu-Shiley and Hancock valve groups, respectively. The difference in actuarial rate free from thromboemboli between Ionescu-Shiley and Hancock groups was statistically significant [P value less than 0.001]. Thromboembolic events beyond the period of anticoagulation therapy mainly occurred in patients with atrial fibrillation. The actuarial thromboemboli free survival was 95.71.4% at 3 years and 80.17.3% at 7 years. The incidence of hemorrhagic complications was 1.2% per patient-year [fatality 0.55% per patient-year] for anticoagulated patients. Although our clinical data favorably compares with results from other reports, our results suggest that anticoagulant therapy be given on a short-term basis or not at all to hemodynamically stable patients. Long-term therapy with antiplatelet drugs is probably inevitable with patients who have thromboembolic risk factors [such as atrial fibrillation].
The purpose of this study was to investigate the dietary intake, obesity index and psychosocial factors influencing obesity among 200 high school girls in Seoul. The Social Cognitive Theory provided the Conceptual basis for this study. A cross-sectional survey was conducted to examine factors related to obesity, including self-efficacy for controlling overeating, social support for eating behavior, perception of body image and weight control, nutrition knowledge, and attitudes toward obesity. The data were analyzed using t-test and multiple logistic regression. The results of this study are as follows : 1) The mean age of the girls was 16.4 years, and the rate of overweight and obesity(measured by obesity index) was 27.0$\%$. 2) The mean energy intake of subjects was 1832.3$\pm$384.0kcal. The energy derived from carbohydrates, proteins and fats was 62.7$\%$, 13.8$\%$, and 23.5$\%$, respectively. There was no significant difference between the obese and the comparison group in energy intake. 3) The result of multivariate analysis indicated that obesity had a significant relation to the perception of ideal body image, social support for eating behavior, and self-efficacy for controlling overeating(p〈0.01). As subjects preferred thinner body images(OR=0.39) and received less social support(OR=0.93), the odds of being classified as obese increased. The odds of being obese were also associated with self-efficacy, however, the relation was not strong(OR=1.04). 4) Specific social support was related to obesity among adolescent girls. As subjects received more support from family member, the odds of being obese decreased. The emotional support as well as family member's positive nutrition behavior plays a significant role. In addition, instrumental support from friends was associated with obesity. With repect to self-efficacy, the odds of being obese were increased as subjects felt less confident in controlling overeating when tempting food was placed in front of them or after an argument. In contrast, the obese group felt more confident in controlling overeating for the rest of the specific situations examined. These findings suggest that educational interventions for weight control should incorporate strategies to help participants realize their degree of obesity, to reduce the discrepancy between current and ideal body image, to elicit and maintain social support from friends and family, and to increase the self-efficacy for changing eating behaviors. The human adipose and milk samples analyzed in this study were collected at hospitals in Seoul, Masan and Jinju. The mean values of International Dioxin Toxic Equivalents (I-TEQ) of PCDDs and PCDFs in human adipose samples from the three regions were calculated as 9 pg I-TEQ/g(0.2 ~ 30 pg TEQ/g, lipid weight basis) and 8 pg I-TEQ/g (0.8 ~ 25 pg TEQ/g), respectively. The residue levels of PCDDs-TEQ and PCDFs-TEQ in human milk collected from Masan were 13 pg I-TEQ/g (lipid wt.) and 4.8 pg I-TEQ/g, respectively. On the whole, the contamination levels of these compounds in the Korean population were lower than those in the other countries. Based on the analytical data and assuming a daily intake consumption of 800 mL milk with 3% fat, the average daily intake of PCDDs/DFs via human milk for a baby weighing 5 Kg could be calculated. The daily intake of PCDDs/DFs via breast-feeding was estimated to be 39 pg/kg body weight/day for 2,3,7,8-TeCDD and 86 pg/kg/day for TEQ. These levels are far above all virtually safe dose(VSD) or tolerable daily intake(TDI) values proposed by health authorities in various countries, ranging from 0.001(US EPA) to 4 pg/kag/day (WHO).
Objectives : This study was performed to understand smoking condition and to investigate the relations among knowledge regarding smoking and oral hygiene, awareness of anti-smoking policy, and self-esteem, to provide fundamental basis, developing programs for smoking prevention and anti-smoking. Methods : From September 7, 2010 to September 16, 2010, questionaire survey was conducted for 566 female university students studying dental hygienics at a local district in Jeollanam-do. Collected data was analyzed by Chi-square test, t-test, one way ANOVA, Scheffe multiple range test, Perason's correlation test, and stepwise multiple regression test. Results : 1. Smoking condition of study subjects on general characteristics showed 17.1%, where 14.0% of previous smoking history, and 68.9% of currently nonsmoker. 2. Analysis on the knowledge related to smoking and oral hygiene, awareness of anti-smoking policy, and self esteem on the general characteristics of study subjects have shown better awareness of anti-smoking policy in non-alcoholics. Significant difference was found in knowledge about smoking and awareness of anti-smoking policy according to current resident status, but no difference found in self-esteem. Higher academic grade was related to higher awareness of anti-smoking policy and better ics. aalcell. Better health, but no diffeoral hygiene were related to better awareness of anti-smoking policy, and bewhich were statistically significant. Favorable interpersonal self-estehip was related to better awareness of anti-smoking policy. 3. Analysis on knowledge regarding smoking related health and oral hygiene, awareness of anti-smoking policy, and self-esteem showed higher level of anti-smoking knowledge in non-smoking group(60.44) compared to currently smoking group or group with previous history of smoking, and better awareness of anti-smoking policy in non-smoking group(53.54) than other groups, which was statistically significant. 4. Correlation analysis among smoking related knowledge, awareness of anti-smoking policy, and self-esteem has found relatively high correlation between smoking related knowledge and awareness of anti-smoking policy, which was statistically significant(r=0.481, p<0.001). 5. Among factors influencing awareness of anti-smoking policy, higher level of smoking related knowledge and satisfaction to specialty was related to higher awareness of anti-smoking policy which was lower in current smoker than past smoker, and higher academic grade was related to higher awareness of anti-smoking policy. Conclusions : Following results emphasize the need to establish a novel curriculum by which effective programs for smoking prevention and education of anti-smoking specialist could be provided as well as promoting coaching anti-smoking activity.
Green algae are important primary producers in aquatic ecosystem, and they are sensitive test species in bioassay. Green algae are broadly used to assess the adverse effects of various chemicals by measuring the inhibition of metabolism, reproduction and survival. In this study, we extensively gathered domestic and foreign toxicity test methods conducted using green algae, which are distributed in Korean water environment. Selected eight domestic green algae were Chlamydomonas reinhardtii, Desmodesmus subspicatus (=Scenedesmus subspicatus), Scenedesmus abundans, Scenedesmus acutus, Scenedesmus quadricauda, Podohedriella falcata (=Ankistrodesmus falcatus), Pseudokirchneriella subcapitata (=Selenastrum capricornutum), and Chlorella vulgaris. Forty four test methods were collected from the standard test ones, government reports, SCI papers and Korean research papers. P. subcapitata and D. subspicatus are the most common test species recommended by the standard test methods. Initial cell density and dilution water were the main differences among the test methods we collected. We proposed the suitable ecotoxicity test methods based on domestic green algae in Korea. This study could be a fundamental basis to establish the ecotoxicity test methods by green algae distributed in Korea.
