식이 지방의 P/S비율이 흰쥐의 혈장 및 조직의 지질함량에 미치는 영향 (Effects of Dietary P/S Ratio on Lipid Content of Plasma and Tissues in Rats)

  • 식이 지방의 P/S비율을 0.05, 0.5, 1.0, 2.0, 3.0, 4.0으로 조절한 실험식이로 Wistar계 흰쥐를 사육하면서 급식 2주 및 4주에 혈장 총 콜레스테롤과 중성지방 함량, 간, 소장 및 동맥의 콜레스테롤과 중성지방 함량을 측정하여 식이지방의 P/S비율이 체내 지방대사에 미치는 영향을 검토한 바 다음과 같은 결과를 얻었다. 1. 식이효율은 2주 급식시에는 여섯 실험군간에 차이가 없었으나 4주 급식시에는 P/S 비율이 높을수록 높은 경향을 나타내었다. 2. 혈장의 총 콜레스테롤 함량은 P/S비율이 높아감에 따라 2주 급식시에는 증가하는 경향을 보였으나, 4주 급식시에는 유의하게 감소하였다. (p<0.05) 3. 혈장의 중성지방 함량은 4주 급식시 P/S비율이 높을수록 유의하게 감소하였다.(p<0.05) 4. 간 및 소장의 콜레스테롤 함량은 4주 급식시 P/S비율이 높아감에 따라 증가하였다. 그러나 동맥의 콜레스테롤 함량은 P/S 비율의 변화에 따른 영향을 보이지 않았다. 5. 간 및 소장의 중성지방 함량은 P/S비율이 높아감에 따라 현저하게 증가하였으나(p<0.05), 동맥의 중성지방 함량은 P/S비율의 변화에 따른 영향이 뚜렷하지 않았다.

n-6/n-3 비율과 P/S 비율을 변화시킨 식이지방이 나이가 다른 흰쥐의 체내 지방대사에 미치는 영향 (The Effects of n-6/n-3 and P/S Ratio of Dietary Lipid on Lipid Metabolism of Rats at Different Age)

  • The effects of age and dietary fatty acid composition on lipid metabolism were investigated in Sprague-Dawley strain male rats. These animals weighing 88.6$\pm$2.2g were fed 10% dietary fat(W/W, 20% of total energy) with 0.5, 1, 2 P/S ratio and in each P/S ratio there were three different levels of n-6/n-3 fatty acid ratio ; 2, 4, 8. The experimental period was 1 month, 6 months and 12 months. The results of this study were as follows. The body weight of rats increased rapidly for the first two months, then increased slowly until 7 to 8 months. After 10 months of dietary regimen their weight decreased. The weight of liver, kidney and epidydimal fat pad increased along with the body weight and then decreased in the 12 months. Plasma total lipid increased with age and it decreased significantly when P/S ratio of dietary fatty acid was high. In creased with age and it decreased significantly when P/S ratio of dietary fatty acid was high. In creasing n-3 fatty acid intake in each P/S ratio resulted in lower plasma total lipid although was not statistically significant. The amount of plasma total cholesterol increased at 6 months, but decreased at 12 months. In case of 1, 12 months, increasing P/S ratio significantly plasma total cholesterol and LDL-cholesterol were decreased and hepatic cholesterol was increased, VLDL-HDL-cholesterol did not changed. The n-6/n-3 ratio did not affect any of theses. The amount of plasma triglyceride and VLDL-triglyceride increased at 6 month then decreased. When the rats consumed higher amount of n-3 fatty acid in each P/S ratio, their plasma triglyceride and hepatic triglyceride increased at 1, 12months.

  • The aim of this study was to investigate the effects of P/S ratio of fatty acid and antioxidant (vitamin E, selenium) supplements on the serum lipid levels and hepatic antioxidant enzyme activity in rats. Female 16-week-old Sprague-Dawley rats were fed 6 different experimental diets for 4 weeks. While the peroxidizability index (PI) levels of fatty acids in the experimental diets were fixed at 81.22, the levels of P/S ratio of fatty acids were formulated at 0.38, 1.00, 4.81 (LP, MP, HP). These diets were supplemented with vitamin E (1,000 mg/kg diet) and selenium (2.5 mg/kg diet) (LP-S, MP-S, HP-S). This study showed that the serum concentrations of total-cholesterol and HDL-C increased with the increasing of the P/S ratio in the diet (p <0.05). Antioxidant supplementation significantly lowered the concentrations of triglyceride (TG) and VLDL-C of serum (p<0.05). Levels of thiobarbituric acid reactive substance (TBARS) in the liver tended to decrease with the increasing of the P/S ratio in the diet (p<0.001), but antioxidant enzyme activity in the liver was not significantly different. In addition, antioxidant supplementation significantly lowered TBARS level in the liver (p<0.05), but had no effect on antioxidant enzyme activity except for glutathione reductase (p<0.05). In conclusion, it is necessary to consider the properties of dietary fatty acids and antioxidants supplementation for the prevention of cardiovascular diseases.

  • The present studies were designed to compare the effects of both dietary fat levels and P / S ratio on lipid components in plasma and tissues. Changes in plasma HDL-cholesterol, cholesterol and TG, and also in tissue cholesterol and TG were determined in young rats fed diets providing total dietary fat as 10%, 25% or 45% of calories and P / S ratio as 0.2 or 4.0. Plasma cholesterol levels were getting higher as dietary fat levels increased at P / S 0.2. Plasma cholesterol was lower in rats fed dietary fat either 25% or 45 %, each with P / S 4.0. But at 10% no change in plasma cholesterol were observed by P / S 4.0 because of a possible insufficiency of the absolute amount of PUFA. HDL-cholesterol was rather less sensitive to the modification of dietary fat level, but was reduced in rats fed diets of P / S 4.0 at either 25% or 45% fat, even though HDL-cholesterol were increased in the group of 10% with P / S 0.2. Total cholesterol per g- liver were significantly increased as dietary fat levels increased. Liver cholesterol levels were higher in rats fed diets of P / S 4.0 at higher fat levels (25% or 45%) which possibly suggested that a reduction of plasma cholesterol by high PUFA diet was not at least from a decreased synthesis of cholesterol in liver. However, in muscle no significant differences were found by feeding high P / S ratio at each levels of fat. At 10% fat level, compared to 25% or 45%, cholesterol level was lower in g-liver but higher in g- muscle. Plasma TG was decreased as more dietary fat were supplied at P / S ratio, but no consistant response obtained at low P / S ratio. TG per g-liver were reduced by feeding P / S 4.0 diet at 10% or 45% fat level but no differences were found in muscle. P / S 4.0 diet was more efficient in lowering plasma cholesterol TG and HDL-cholesterolt levels only if fat level was more than 25% of the total calories And young rats were more resistant to dietary fat modification.

고불포화 지방식이가 흰쥐의 Plasma High Density Lipoprotein Cholesterol 량과 혈청 및 조직 내의 지방성분에 미치는 영향 (Effects of Dietary Polyunsaturated Fat on HDL - Cholesterol, Total Cholesterol and Triglyceride in Plasma and Tissues of Adult Rats)

  • The study was designed to observe the effects of dietary intake of three different levels of polyunsaturated to saturated fatty acid (P/S) ratio on plasma HDL - cholesterol and triglyceride in plasma and tissues of adult rats. Rats were assigned into the three diet groups, composed of protein 24%, carbohydrate 46%, and fat 30% of total caloric intake. However, the P/S ratio of fat was adjusted by using beef tallow, corn oil and perilla oil to give 0.2, 1.1 and 6.0. All groups were fed ad libitum for 4 week. Plasma cholesterol level was decreased but net significantly with increasing P/S ratio of dietary fat, and showed significant negative correlation coefficient. Plasma TG level was also decreased with increasing PUFA level of dietary fat. However, the effect of P/S ratio (6.0) was not greater than that of P/S ratio (1.1) on plasma TG concentration. This implied that high level of PUFA was not required to change plasma TG level as much in the case of HDL-cholesterol. Plasma HDL-cholesterol was significantly increased only in the group of P/S ratio 6.0 which suggested that high level of dietary PUFA was required to increase HDL level. There was a decreasing trend in the levels of total cholesterol and TG per g liver with increasing P/S ratio of dietary fat. There was no effect in the levels of TG per g skeletal muscle by P/S ratio but the level of cholesterol per unit skeletal muscle was rather increased with increased P/S ratio.

  • 동일한 파원에서 발생된 P, SH, SV 파를 3성분 지오폰으로 동시에 기록하는 방법을 연구하기 위해서, 94 m의 측선을 따라 2 m 간격으로 설치한 수신점에서 기록된 24채널 자료를 512 ms동안 기록하였다. 원시자료의 수직성분에는 P파가 초동으로 뚜렷하게 나타나며, 측선방향과 수직한 수평성분은 P파가 약화된 반면 S파의 신호/잡음비가 향상된 모습을 보인다. 토모그래피 역산을 통해 계산된 P파 및 S파의 속도 토모그램을 이용하여 동포아송비의 2차원적 분포를 산출한 결과, 동포아송비는 $0.2{\~}0.3$의 범위로 분포하며, 이는 한번의 타격으로 P파 및 S파의 속도분포와 더불어 및 동포아송비의 2차원적 분포까지 획득할 수 있다는데 큰 의의가 있다고 판단된다.

Effects of Combining Feed Grade Urea and a Slow-release Urea Product on Characteristics of Digestion, Microbial Protein Synthesis and Digestible Energy in Steers Fed Diets with Different Starch:ADF Ratios

  • As a result of the cost of grains, the replacement of grains by co-products (i.e. DDGS) in feedlot diets is a common practice. This change produces diets that contain a lower amount of starch and greater amount of fibre. Hypothetically, combining feed grade urea (U) with slow release urea (Optigen) in this type of diet should elicit a better synchrony between starch (high-rate of digestion) and fibre (low-rate of digestion) promoting a better microbial protein synthesis and ruminal digestion with increasing the digestible energy of the diet. Four cannulated Holstein steers ($213{\pm}4$ kg) were used in a $4{\times}4$ Latin square design to examine the combination of Optigen and U in a finishing diet containing different starch:acid detergent fibre ratios (S:F) on the characteristics of digestive function. Three S:F ratios (3.0, 4.5, and 6.0) were tested using a combination of U (0.80%) and Optigen (1.0%). Additionally, a treatment of 4.5 S:F ratio with urea (0.80% in ration) as the sole source of non-protein nitrogen was used to compare the effect of urea combination at same S:F ratio. The S:F ratio of the diet was manipulated by replacing the corn grain by dried distillers grain with solubles and roughage. Urea combination did not affect ruminal pH. The S:F ratio did not affect ruminal pH at 0 and 2 h post-feeding but, at 4 and 6 h, the ruminal pH decreased as the S:F ratio increased (linear, p<0.05). Ruminal digestion of OM, starch and feed N were not affected by urea combination or S:F ratio. The urea combination did not affect ADF ruminal digestion. ADF ruminal digestion decreased linearly (p = 0.02) as the S:F ratio increased. Compared to the urea treatment (p<0.05) and within the urea combination treatment (quadratic, p<0.01), the flow of microbial nitrogen (MN) to the small intestine and ruminal microbial efficiency were greater for the urea combination at a S:F ratio of 4.5. Irrespective of the S:F ratio, the urea combination improved (2.8%, p = 0.02) postruminal N digestion. As S:F ratio increased, OM digestion increased, but ADF total tract digestion decreased. The combination of urea at 4.5 S:F improved (2%, p = 0.04) the digestible energy (DE) more than expected. Combining urea and Optigen resulted in positive effects on the MN flow and DE of the diet, but apparently these advantages are observed only when there is a certain proportion of starch:ADF in the diet.

  • The effects of age and dietary fatty acid composition on prostagladin production was investigated in Sprague-Dawley strain male rats. Animals weighing 88.6$\pm$2.2g were fed 10% dietary fat(W/W, 20% of total energy). The P/S ratios of dietary lipid were three levels(0.5, 1, 2) and there were three different levels of n-6/n-3 fatty acid ratio(2, 4, 8) in each P/S ratio. The experimental period were 1 month and 12 months, respectively. The results of this study were as follows. As the age of rats increased, the plasma thromboxane B2 production increased, but aorta 6-keto prostaglandin F1$\alpha$ decreased. When a higher amount of n-3 fatty acid was fed in each P/S ratio, the relative percentage of linolenic acid and EPA in platelet increased.

Effects of the P/S Ratio of Dietary Lipids and Antioxidant Vitamin Supplements on the Level of Serum Lipids and Liver. Lipid Peroxidation in Rats Treated with DMBA

  • This study an analyzes the effects of the P/S ratio of dietary lipids and antioxidant vitamin supplements on serum lipids level and fatty acid profile, the degree of lipid peroxidation, and the antioxidant enzyme activities in the liver of rats treated with 7,12-dimethylbenz($\alpha$) anthracene(DMBA). P/S ratio of dietary lipids was made into 0.5, 1 and 2 by mixing palm oil, soybean oil, sesame oil and perilla oil at 10%(w/w) fat level and n-6/n-3 ratio was fixed to 4. Antioxidant vitamin of $\alpha$-tocopherol or $\beta$-carotene was supplemented in addition to vitamin mixture which was given at 1 % of the standard diet. female Sprague-Dawley strain rats, about 60 days old, were divided into three groups(LP : low P/S ratio(0.5), MP : medium P/S ratio (1.0), HP , high P/S ratio(2.0)) and each group was sub-divided into three groups(S ; standard, T ; tocopherol supplemented, C : carotene supplemented): Two weeks after feeding experimental diets, all groups were treated with a single dose of DMBA(2mg/100g BW) by gastric intubation and fed experimental diet for 9 week. The results were as follows ; 1) Serum total cholesterol(TC) level was not significantly influenced by diet but tended to be lower in HP groups compared to LP and MP groups. Triglyceride level was the highest in LP groups and the lowest in $\alpha$-tocopherol supplemented groups. 2) Thiobarbituric acid reactive substance(TBARS) level, representing lipid peroxidation in hepatic microsome, tended to be increased as the unsaturation of dietary lipids increases. $\alpha$-Tocopherol supplement significantly decreased TBARS level. 3) The activities of superoxide dismutase(SOD) and glutathione peroxidase(GSHPx) in hepatic cytosol showed the tendency to be high with increasing P/S ratio of dietary lipids. SOD activity was not significantly influenced by antioxidant vitamin, but GSHPx activity was significantly increased in $\alpha$-tocopherol supplemented groups. In summary, high polyunsaturated fat diet was effective on reducing the serum level of total cholesterol and triglyceride, while it increased unsaturation and peroxidizability of serum fatty acid. With increasing P/S ratio of dietary lipids, lipid peroxidation was increased in the liver and antioxidant enzyme system was induced to inhibit lipid peroxidation against oxidative damage. $\alpha$-Tocopherol supplement was effective in lowering lipid peoxidation, but $\beta$-carotene supplement did not exhibit antioxidant effect.   • The present studies were designed to observe the effects of both dietary fat levels and P/S ratio on lipid components in plasma and tissues of young rats when cholesterol was supplemented at 1%(w/w) to four dietary groups providing total fat as 10%( LF ) or 45% ( HF ) of calories and P/S ratio as 0.2 or 4.0, Low Fat-0.2 : Low Fat-4.0 : High Fat-0.2 : High Fat-4.0. Plasma total cholesterol was increased almost to the same level in all dietary groups after the cholesterol supplement but the value of HF -0.2 was slightly higher than that of LF -0.2. Plasma TG was also increased in all dietary groups when cholesterol was supplemented but was more significantly increased in LF group than in HF group. HDL-cholesterol was slightly increased by cholesterol supplement but there was no effect by the total dietary fat level of previous diet. However, plasma HDL-cholesterol, total cholesterol and TG levels were slightly reduced in high fat diet of P/S 4.0. Total cholesterol per g -liver was higher in HF group than in LF group before and after cholesterol supplement even though it was increased more in LF group with cholesterol addition. Liver cholesterol was also higher in rats fed diets of P/S 4.0 at high fat level before and after cholesterol supplement. In contrast, total cholesterol per g-muscle was reduced by cholesterol supplement in all groups and it was significantly higher in LF than in HF. There was no significant effect in liver TG by total fat level and P/S ratio when cholesterol was supplemented. Muscle TG was lowered in all groups by cholesterol supplement but no effect by total fat level and P/S ratio.
