Performance of a Novel Bioreactor Equipped with Moving Membrane Tube-Aeration System (회전하는 산소전달장치가 부착된 동물세포 배양기의 조업 성능에 관한 고찰)

  • The optimal conditions for operating a moving-aeration bioreactor were determined as 30rpm and 150 (ml/min) of air flow rate, which can yield ca. 7.3 (l/h)of maximum mass transfer coefficient. It was also found that the agitation speed played much much important role than air input rate in oxgen transfer into the medium. $2.6{\times}10^6$ (cells/ml) and 0.6 (ml/l) of maximum cell denisty and IL-2 production were observed in batch cultivation of IL-2 producing BHK cell line. 0.53 (mM/l/h) of oxygen uptake rate was also estimated. The performance of a moving-aeration bioreactor (specific growth rate and oxygen uptake rate, etc.) was superior to other culture systems, such as cell-life and static membrane aeration bioreactors. Ii must be useful to apply this reactor to many culture processes by improving structural limitations in scaling-up the system.

  • The general objectives of this study are to develop a new biofilm reactor equipped with agitating mixer and to evaluated the treatment efficiency of the reactor. The experimental tests were conducted to estimate the oxygen transfer rate of agitating mixer system. Results are as follows. 1. The oxygen transfer coefficient, KLa, was $8.94hr{-1}$ and $7.50hr{-1}$ at 500rpm and 250rpm of agitating mixer speed, respectively. When the agitating mixer was used in the biofilm reactor, 22.5% and 18.8% of oxygen transfer rates were increased at 500rpm and 250rpm, respectively. 2. The removal rate of BOD and CODcr was decreased by 5.0% when the agitating mixer speed was varied from low (250rpm) to high level (500rpm). 3. The concentration of attached biomass had a difference of 5.0% to 7.3%, whereas that of suspended biomass had a difference of about 15.0%, depending upon variation of the agitating mixer speed.

  • In order to improve the bacterial cellulose(BC) production yield, centrifugal and inclined centrifugal impellers were developed. A 6 flat-blade turbine impeller was used as a control system. The flow pattern in the fermenter and volumetric oxygen transfer coefficient($k_La$) of these fermentation systems were studied. Fermentations were carried out for the production of BC by G. hansenii PJK in a 2-L jar fermenter equipped with new impellers. Liquid medium was circulated from the bottom, through the cylinder of the impeller and to the wall. The volumetric oxygen transfer coefficients, $k_La$, of inclined centrifugal and centrifugal impeller systems at 100 rpm were 23 and 15% of the conventional turbine impeller system, respectively. However, the conversion of microbial cells to cellulose non-producing mutant decreased and this results in the increase in BC production at low rotating speed of impellers.

Active Reaction Sites and Oxygen Reduction Kinetics on $La_1_{-x}Sr_xMnO_{3+\delta}$(x=0.1-0.4)/YSZ (Yttria-Stabilized Zirconia) Electrodes for Solid Oxide Fuel Cells

  • Active reaction sites and electrochemical O2 reduction kinetics on La_{1-x}Sr_xMnO_{3+{\delta}} (x=0.1-0.4)/YSZ (yttria-stabilized zirconia) electrodes are investigated in the temperature range of 700-900 ℃ at $Po_2=10^{-3}$-0.21 atm. Results of the steady-state polarization measurements, which are formulated into the Butler-Volmer formalism to extract transfer coefficient values, lead us to conclude that the two-electron charge transfer step to atomically adsorbed oxygen is rate-limiting. The same conclusion is drawn from the $Po_2$-dependent ac impedance measurements, where the exponent m in the relationship of $I_o$ (exchange current density) ∝ $P_{o_{2}}^m$ is analyzed. Chemical analysis is performed on the quenched Mn perovskites to estimate their oxygen stoichiometry factors (δ) at the operating temperature (700-900 ℃). Here, the observed δ turns out to become smaller as both the Sr-doping contents (x) and the measured temperature increase. A comparison between the 8 values and cathodic activity of Mn perovskites reveals that the cathodic transfer coefficients $({\alpha}_c)$ for oxygen reduction reaction are inversely proportional to δ whereas the anodic ones $({\alpha}_a)$ show the opposite trend, reflecting that the surface oxygen vacancies on Mn perovskites actively participate in the $O_2$ reduction reaction. Among the samples of x= 0.1-0.4, the manganite with x=0.4 exhibits the smallest 8 value (even negative), and consistently this electrode shows the highest ${\alpha}_c$ and the best cathodic activity for the oxygen reduction reaction.

Enhanced Production of Valuable Bioactive Metabolites in Submerged Cultures of Medicinal Mushroom Ganoderma lucidum by Manipulation of Oxygen Supply

  • Submerged cultures of Ganoderma lucidum, a valuable mushroom in traditional Chinese medicine, were used for production of bioactive Banoderic acids and Ganoderma polysaccharides. The significant effects of oxygen supply were demonstrated in both shake flasks and bioreactors. By changing the medium loading volume in a shake flask, a different value of initial volumetric oxygen transfer coefficient ($K_L$a) was obtained, and a higher $K_L$a value led to a higher biomass density and a higher productivity of both intracellular polysaccharide and ganoderic acid. In a stirred bioreactor, at an initial $K_L$a of 78.2 $h^{-1}$, a maximal cell concentration of 15.6 g/L by dry weight was obtained, as well as a maximal intracellular polysarcharide (IPS) production of 2.2 g/L and its maximal productivity of 220 mg/(L.d). An increase of initial $K_L$a led to a higher production and productivity of GA, and the GA production and productivity at an initial $K_L$a of 96.0 $h^{-1}$ was 1.8-fold those at an initial $K_L$a of 16.4 $h^{-1}$. The fundamental information obtained in this study may be useful for efficient large-scale production of these valuable bioactive products by the submerged cultures.

Residence Time Distributions of Liquid pbase Flow and Mass Transfers in the Trickle Bed Reactor (점적상 반응기에서 액상흐름의 체류시간 분포 및 물질전달)

  • The residence time distribution of liquid flow in a 4.0cm diameter column packed with porous $Al_2O_3$ spheres of 0.37cm diameter were measured with pulse injections of a tracer under cocurrent trickling flow conditions. The mean residence time of liquid flow and liquid hold-up calculated by the transient curve of tracer were unaffected by gas flow rates under experimental ranges of liquid flow rates from 2.4 to $4.5(kg/m^2\;sec)$ and gas flow rates from 0 to $0.13(kg/m^2\;sec)$. The axial dispersion coefficient of liquid stream and apparent diffusivity of tracer in a micropore of solid particle were estimated from the response curve of tracer. The calculated Peclet No. were increased in ranges of 68-to 82 with a increasing of liquid mass velocity, and the external effective contacting efficiency between liquid and solid which can be expressed. by $(D_i)_{app}/D_i$ varied in ranges of 0.54 to 0.68 depending on the liquid flow rates. The gas to liquid(water) volumetric mass transfer coefficient were determined from desorption experiments with oxygen at $25^{\circ}C$ and 1 atm. The measured mass transfer coefficients were increased with liquid flow rates and the effect of gas flow rates on the mass transfer coefficient was insignificant.

Effects of Stireed Tank Bioreactor Scale-up on Cell Growth and Alkaloids Production in Cell Cultures of Eschscholtzia californica (탱크 교반형 생물반응기의 scale-up이 Eschscholtzia californica 세포생장 및 알칼로이드 생성에 미치는 영향)

  • Studies were made to investigate effects of the scale-up of stirred tank bioreactors on cell growth and alkaloids production for suspension cultures of Eschscholtzia californica. In the 1.5 L STR, cell lysis was observed at 110 rpm or higher agitation speed. The agitation speed of 30 L STR was 43.7 rpm to maintain the same shear stress developed in 1.5 L STR of 100 rpm. As a result of scale-up from 1.5 L to 30 L STR, the specific growth rate was decreased from 0.12 to 0.07 day-1. The alkaloids productivity was also decreased from 0.24 to 0.14 mg/L-day. Changes of mixing performance and oxygen transfer were studied to explain the decrease of cell growth and alkaloids production. Decreased oxygen transfer rate coefficient(KLa) and increased mixing time by the scale-up was observed at various aeration rates.

Oxygen Transfer and Hydraulic Characteristics in Bubble Column Bioreactor Applied Fine Bubble Air Diffusing System (미세기포 산기장치를 적용한 타워형 생물반응기의 산소전달 및 수력학적 특성)

  • For improving performance of conical air diffuser generating fine bubble, both experimental and numerical simulation method were used. After adapting diffusers inner real scale bubble column, suitable for various diffuser submergence, the effect of diffuser submergence on oxygen transfer performance such as Oxygen Transfer Coefficient ($K_{L}a_{20}$) and Standard Oxygen Transfer Efficiency (SOTE) was investigated empirically. As flow patterns for various diffuser number and submergence were revealed throughout hydrodynamic simulation for 2-phase fluid flow of air-water, the cause of the change for oxygen transfer performance was cleared up. As results of experimental performance, $K_{L}a_{20}$ was increased slightly by 7% and SOTE was increased drastically by 39~72%, 5.6% per meter. As results of numerical analysis, air volume fraction, air and water velocity in bioreactor were increased with analogous flow tendency by increasing diffuser number. As diffuser submergence increased, air volume fraction, air and water velocity were decreased slightly. Because circulative co-flow is determinant factor for bubble diffusion and rising velocity, excessive circulation intensity can result to worsen oxygen transfer by shortening bubble retention time and amount.

Determination of Oxygen Transfer Coefficient in Fed-Batch Culture of Streptomyces avermitilis with Concentrated Medium Control (농축 배지 조절 유가식 배양에 의한 Streptomyces avermitilis의 산소전달계수 측정)

  • The large-scale production of antibiotics by filamentous mycelial organism requires and adequate supply of dissolved oxygen. In terms of productivity, it means that oxygen transfer is the rate-limiting step. Therefore, the oxygen transfer coefficients(K$\_$L/A) were determined in a broth involving a filamentous mycelial organism such as Streptomyces avermitilis for use in fermentations. To determine (K$\_$L/A) inn a stirred vessel, a great deal of effort is required to provide all the cells with a sufficient oxygen supply. To overcome the oxygen limitation in a batch culture, a fed-batch culture was applied to control the growth rate by an intermittent supply of nutrients. Thus, it was possible to maintain a suitable dissolved oxygen concentration at a low agitation rate. The optimal agitation speed was 350 rpm at low cell concentrations (below 7 g/L) by considering the efficiency of agitation and shear stress. The (K$\_$L/A) was found to decrease from 64.26 to 29.21h.$\^$-1/ when the biomass concentration was increased from 9.82 to 12.06 g/L. In addition, and increase in viscosity was also observed during the growth phase. By comparing the (K$\_$L/A) values for the various agitation and aeration rates, it was found that the effect of an increase in (K$\_$L/A) by aeration was reduced dramatically at high biomass concentrations. However, this effect was not observed when altering the agitation rate. This suggests that controlling the dissolved oxygen concentration by altering the agitation rate was more efficient than increase the aeration rate.

Enhancement of Hyaluronic Acid Production by Batch Culture of Streptococcus zooepidemicus via the addition of n-Dodecane as an Oxygen Vector

  • This study aimed to examine the influence of adding an oxygen vector, n-dodecane, on hyaluronic acid (HA) production by batch culture of Streptococcus zooepidemicus. Owing to the high viscosity of culture broth, microbial HA production during 8-16 h was limited by the oxygen transfer coefficient $K_La$, which could be enhanced by adding n-dodecane. With the addition of n-dodecane to the culture medium to a final concentration of 5% (v/v), the average value of $K_La$ during 8-16 h was increased to $36{\pm}2h^{-1}$, which was 3.6 times that of the control without n-dodecane addition. With the increased $K_La$ and dissolved oxygen (DO) by adding 5% (v/v) of n-dodecane, a 30% increase of HA production was observed compared with the control. Furthermore, the comparison of the oxygen mass transfer in the absence and presence of n-dodecane was conducted with two proposed mathematical models. The use of n-dodecane as an oxygen vector, as described in this paper, provides an efficient alternative for the optimization of other aerobic biopolymer productions, where $K_La$ is usually a limiting factor.
