The role of peroxidases in the pathogenesis of atherosclerosis

  • Reactive oxygen species (ROS), which include superoxide anions and peroxides, induce oxidative stress, contributing to the initiation and progression of cardiovascular diseases involving atherosclerosis. The endogenous and exogenous factors hypercholesterolemia, hyperglycemia, hypertension, and shear stress induce various enzyme systems such as nicotinamide adenine dinucleotide (phosphate) oxidase, xanthine oxidase, and lipoxygenase in vascular and immune cells, which generate ROS. Besides inducing oxidative stress, ROS mediate signaling pathways involved in monocyte adhesion and infiltration, platelet activation, and smooth muscle cell migration. A number of antioxidant enzymes (e.g., superoxide dismutases, catalase, glutathione peroxidases, and peroxiredoxins) regulate ROS in vascular and immune cells. Atherosclerosis results from a local imbalance between ROS production and these antioxidant enzymes. In this review, we will discuss 1) oxidative stress and atherosclerosis, 2) ROS-dependent atherogenic signaling in endothelial cells, macrophages, and vascular smooth muscle cells, 3) roles of peroxidases in atherosclerosis, and 4) antioxidant drugs and therapeutic perspectives.

충격하중에 의한 용접구조물의 강도 증가에 관한 연구 (A Study on the strength improvement in weldment by the impact loading)

  • It is well known that during the oxygen cutting process residual thermal stresses are produced in weldment. The local non-uniform heating and subsequent cooling which takes place during any welding process causes complex thermal strains and stresses to finally lead to residual stresses exceed to the yield stress. High tensile stresses combined with applied structural load in the region near the welded joint can given rise to distortion brittle fracture change of the fatigue strength and stress corrosion cracking. The appropriate treatment of the welded component which reduces the peak of he welding residual stresses is believed to lower risk of the fracture during the service of the structure. In this study the impact loading in oxygen cutting frame was applied to reduce the residual stress. After applying the impact loading redistribution of resid-ual stress was measured by cutting method and the effect of fatigue was tested.

A Co-expression Network of Drought Stress-related Genes in Chinese Cabbage

  • Plants have evolved to adapt to abiotic stresses, such as salt, cold, and drought stress. In this study, we conducted an in-depth analysis of drought resistance mechanisms by constructing a gene co-expression network in Chinese cabbage (Brassica rapa ssp. pekinensis L.). This drought stress co-expression network has 1,560 nodes, 4,731 edges, and 79 connected components. Based on genes that showed significant co-expression in the network, drought tolerance was associated with the induction of reactive oxygen species removal by raffinose family oligosaccharides and inositol metabolism. This network could be a useful tool for predicting the functions of genes involved in drought stress resistance in Chinese cabbage.

Oxidative stress and endometriosis

  • Endometriosis is an estrogen-dependent chronic inflammatory condition that affects women in their reproductive period and is associated with pelvic pain and infertility. Oxidative stress (OS) occurs when reactive oxygen stress (ROS) and anti-oxidants are in imbalance. OS is a potential factor involved in the pathophysiology of endometriosis. Iron-induced ROS may trigger a chain of events resulting in the development and progression of endometriosis. Endogenous ROS are correlated with increased cellular proliferation and ERK1/2 activation in human endometriotic cells. An oxidative environment leads to stimulation of the ERK and PI3K/AKT/mTOR signaling pathways that facilitate endometriotic lesion progression through adhesion, angiogenesis, and proliferation. OS is also known to be involved in epigenetic mechanisms in endometriosis. We summarize the recent knowledge in our understanding of the role of oxidative stress in the pathogenesis of endometriosis.

Ingestion of Polystyrene Microplastics Acutely Induces Oxidative Stress in the Marine Medaka Oryzias javanicus

  • Larvae from the marine medaka fish Oryzias javanicus were exposed with polystyrene microplastics (MPs) for 24 h. Exposure to waterborne fluorescent MPs showed clear ingestion and egestion in feces. Under constant MPs, the concentration of dissolved oxygen significantly decreased in 24 h compared to the control. Significant intracellular reactive oxygen species and malondialdehyde contents were detected in larvae, indicating oxidative stress and lipid peroxidation. Significant elevations in mRNA expressions of heat shock protein 70 and antioxidant defense system genes (glutathione reductase, glutathione peroxidase, catalase, and superoxide dismutase) were measured with increases in enzymatic activity of oxidative stress-related proteins. Taken together, the alterations to the molecular and biochemical components suggested that waterborne MPs had an oxidative stress effect on marine medaka larvae.

Effect of Limited Oxygen Supply on Coenzyme $Q_{10}$ Production and Its Relation to Limited Electron Transfer and Oxidative Stress in Rhizobium radiobacter T6102

  • Coenzyme $Q_{10}$ ($CoQ_{10}$) production from Rhizobium radiobacter T6102 was monitored under various oxygen supply conditions by controlling the agitation speeds, aeration rates, and dissolved oxygen levels. As the results, the $CoQ_{10}$ production was enhanced by limited oxygen supply. To investigate whether the $CoQ_{10}$ production is associated with its physiological functions of electron carrier and antioxidant, the effects of sodium azide and hydrogen peroxide on the $CoQ_{10}$ production were studied, showing that the $CoQ_{10}$ contents were slightly enhanced with increasing sodium azide (up to 0.4 mM) and hydrogen peroxide (up to $10\;{\mu}M$) concentrations. These results suggest the plausible mechanism where the limited electron transfer stimulating the environments of limited oxygen supply and oxidative stress could accumulate the $CoQ_{10}$, providing the relationship between the $CoQ_{10}$ physiological functions and its regulation system.

Anthocyanin Extracts from Black Soybean (Glycine max L.) Protect Human Glial Cells Against Oxygen-Glucose Deprivation by Promoting Autophagy

  • Anthocyanins have received growing attention as dietary antioxidants for the prevention of oxidative damage. Astrocytes, which are specialized glial cells, exert numerous essential, complex functions in both healthy and diseased central nervous system (CNS) through a process known as reactive astrogilosis. Therefore, the maintenance of glial cell viability may be important because of its role as a key modulator of neuropathological events. The aim of this study was to investigate the effect of anthocyanin on the survival of glial cells exposed to oxidative stress. Our results demonstrated that anthocyanin extracts from black soybean increased survival of U87 glioma cells in a dose dependent manner upon oxygen-glucose deprivation (OGD), accompanied by decrease levels of reactive oxygen species (ROS). While treatment cells with anthocyanin extracts or OGD stress individually activated autophagy induction, the effect was signifi cantly augmented by pretreatment cells with anthocyanin extracts prior to OGD. The contribution of autophagy induction to the protective effects of anthocyanin was verifi ed by the observation that silencing the Atg5 expression, an essential regulator of autophagy induction, reversed the cytoprotective effect of anthocyanin extracts against OGD stress. Treatment of U87 cells with rapamycin, an autophagy inducer, increased cell survival upon OGD stress comparable to anthocyanin, indicating that autophagy functions as a survival mechanism against oxidative stress-induced cytotoxicity in glial cells. Our results, therefore, provide a rationale for the use of anthocyanin as a preventive agent for brain dysfunction caused by oxidative damage, such as a stroke.

응력부식균열을 고려한 고리 1호기 원자로냉각재계통의 배관 파손확률 평가 (Evaluation of Piping Failure Probability of Reactor Coolant System in Kori Unit 1 Considering Stress Corrosion Cracking)

  • The piping failure probability of the reactor coolant system in Kori unit 1 was evaluated considering stress corrosion cracking. The P-PIE program (Probabilistic Piping Integrity Evaluation Program) developed in this study was used in the analysis. The effect of some variables such as oxygen concentration during start up and steady state operation, and operating temperature, which are related with stress corrosion cracking, on the piping failure probabilities was investigated. The effects of leak detection capability, the size of big leak, piping loops, and reactor types on the piping failure probability were also investigated. The results show that (1) LOCA (loss of coolant accident) probability of Kori unit 1 is extremely low, (2) leak probability is sensitive to oxygen concentration during steady state operation and operating temperature, while not sensitive to the oxygen concentration during start up, and (3) the piping thickness and operating temperature play important roles in the leak probabilities of the cold leg in 4 reactor types having same inner diameter.

The Relationship between Oxidative Stress and Oxidative Markers

  • Metabolic syndrome such as, hypertension, diabetes, obesity, dyslipidemia etc are well known lifestyle diseases which threaten a health state. The purpose of this study is to evaluate antioxidant status and reactive oxygen species (ROS) in various stressful conditions. A group of persons (n=28) were enrolled in this study and stress status was evaluated using questionnaire of psychological well-being index (PWI-SF) and hematologic analysis of free oxygen radical defense (FORD), free oxygen radical test (FORT), lipid profiles and serum cortisol level. Analysis was done according to stress index, BMI status and overtime working. Blood samples from diabetics were used for control. Results are as in followings: Higher PWI-SF group showed higher FORT. Obese persons with BMI > $25kg/m^2$ showed statistically lower FORD (p<0.05) and higher FORT level (p<0.01). When compare with diabetics, an overweight group had similar levels in FORD and FORT while a normal body weight group showed quite different with diabetics in FORD (p<0.001). Higher ROS group has higher triglyceride, CRP and insulin levels but lower in FORD and HDL-cholesterol levels. Overtime working at night showed no meaningful result against our expectations. These results suggested that obesity showed most unfavorable correlations in an antioxidant status in various situations.

돌돔(Oplegnathus fasciatus)의 생존, 산소소비 및 생리학적 반응에 미치는 저수온의 영향 (Survival, Oxygen Consumption and Stress Response of Parrotfish Oplegnathus fasciatus Exposed to Different Lower Temperature)

  • The sudden drop of water temperature in winter is very threatening factor that affects the productivity of farmed fish and the management in aquafarm. In this study, we investigated the effect of low temperature on the survival, oxygen consumption and stress responses of parrotfish Oplegnathus fasciatus due to acute drop of water temperature. The survival rate of parrotfish Oplegnathus fasciatus was 5% at 6℃, 95% at 8℃ and 100% at 10℃ on the 4th day of exposure in each experimental temperature. Low-lethal temperature for 4days of parrotfish Oplegnathus fasciatus (4 day-LT50) was 6.99℃ (confidence limit, 6.55-7.42℃). Oxygen consumption rate was significantly decreased with decreasing water temperature. Temperature coefficient (Q10) was found to be 4.0 between 10℃ and 8℃ and 0.39 between 8℃ and 6℃. As a result of investigating the stress response according to the drop in water temperature, the concentration of SOD (Superoxide dismutase), cortisol, glucose, total Ig, AST (Aspartate) and ALT (Alanine aminotransferase) increased with decreasing of water temperature. This study would be useful for the management of temperature about cultured fish.
